416. Zero kinetic energy photoelectron spectroscopic studies of the ground electronic states of methyl bromide and furan cations

Abstract

High-resolution zero kinetic energy (ZEKE) photoelectron spectrum of methyl bromide has been obtained using vacuum ultraviolet laser single photon excitations. The dominant feature of the spectrum is the origin band of the transition −X ~ 2 E 3/2 −X 1 A 1 −X~2E3/2−X1A1. The excitation of the totally symmetric vibrational mode, ν 2 (CH 3 ν2(CH3 symmetruc deformation), is observed. The 3 1 1 311 hot band is also present. All the bands obtained show similar rotational contours. Simulation of the rotational contour of the origin band yields the accurate first ionization potential of methyl bromide (85022.4±1.0cm −1 85022.4±1.0cm−1) and also the rotational constants of the X ~ 2 E 3/2 X~2E3/2 ground electronic state of the cation. The non-resonant two-photon ZEKE photoelectron spectrum of furan is also presented.

Abstract

Premature termination codons (PTCs) are an important reason for the silence of high-molecular-weight glutenin subunits in Triticumspecies. Although the Glu-A1ygene is generally silent in common wheat, we here isolated an expressed Glu-A1ygene containing a PTC, named 1Ay8.3, from Triticum monococcumssp. monococcum(AmAm, 2n = 2x = 14). Despite the presence of a PTC (TAG) at base pair positions 1879–1881 in the C-terminal coding region, this did not obviously affect 1Ay8.3expression in seeds. This was demonstrated by the fact that when the PTC TAG of 1Ay8.3was mutated to the CAG codon, the mutant in Escherichia colibacterial cells expressed the same subunit as in the seeds. However, in E. coli, 1Ay8.3containing the PTC expressed a truncated protein with faster electrophoretic mobility than that in seeds, suggesting that PTC translation termination suppression probably occurs in vivo(seeds) but not in vitro(E. coli). This may represent one of only a few reports on the PTC termination suppression phenomenon in genes.

418. Development of subcellular fractionation techniques to determine the intracellular cobalamin transit in vitro and the impact of lysosomal dysfunction

Abstract

Intracellular cobalamin (Cbl) utilization is critically dependent on its efficient transit through the lysosome and subsequent delivery to cytosol and mitochondria. We propose that age-related pathological processes may inhibit lysosomal function and impair intracellular Cbl transport. Purpose: To investigate alterations of intracellular [57Co]Cbl trafficking in subcellular organelles when lysosome function is interrupted it is essential to develop an optimal subcellular fractionation method to isolate pure lysosomes, mitochondria and cytosol. Methods: Approximately 100 million human SH-SY5Y neurons or HT1080 fibroblasts were labelled with [57Co]Cbl, homogenised using a ball-bearing homogeniser and the lysates fractionated using an Optiprep gradient and reagent kits from either Pierce or Sigma. [57Co] in each fraction was measured using a gamma counter and subcellular fractions were probed by western blotting. Results: Both protocols separated subcellular organelles to a certain extent. The Pierce method seemed to be superior, separating pure lysosomes from mitochondrial fractions without cytosol contamination. SH-SY5Y [57Co]Cbl lysosome / mitochondria / cytosol distribution was 5.2 + 0.4 / 13.2 + 0.6 / 81.6 + 0.8, respectively, and this was changed (all p < 0.01) to 54.2 + 1.9 / 7.0 + 0.9 / 35.5 + 2.7, respectively (all mean + SE, n = 3), when cells were treated for 48 h with 25 μM chloroquine (to increase lysosomal pH). Similar results were obtained using HT1080 fibroblasts. Conclusion: Development of subcellular fractionation methods provides a useful tool for investigating intracellular Cbl trafficking. This method can be adapted to study the impact of age- or pathology-related lysosomal dysfunction on intracellular [57Co]Cbl transport.

419. Nonlinear wave propagation in a vegetated sandbar-lagoon system

Abstract

The sandbar-lagoon system presents a rich variety of values. At the same time, the degradation of ecosystems and coastal erosion caused by human activities and climate change are even more severe. A series of ecological conservation and restoration projects focused on vegetation rehabilitation have been implemented. However, there is insufficient scientific evidence to guide coastal engineering in the presence of vegetation. In this paper, ten tests were presented to experimentally investigate nonlinear wave propagation in a vegetated sandbar-lagoon system. The following conclusions can be obtained. In the absence of vegetation, the low-frequency wave energy proportion gradually increases with the rise of incident wave height. Double-peaked and even multi-peaked spectra are notable throughout the propagation in tests with larger incident wave heights. Wave height decay and wave energy dissipation due to vegetation effects increase as the incident wave height increases. The dependence of the wave energy dissipation induced by vegetation on the incident wave energy (the square of wave height) presents a linear relationship. Vegetation plays an important and complex role in both sea-swell wave and infragravity wave attenuation.

449. Nicotinamide phosphoribosyltransferase enhances beta cell expansion during pregnancy.

Abstract

OBJECTIVE: Many factors contribute to the development of gestational diabetes mellitus (GDM). Among these factors, nicotinamide phosphoribosyltransferase (NAMPT) plays a critical role. Previous studies have demonstrated the effects of NAMPT on insulin resistance during GDM, while the effects of NAMPT on beta cells remain unknown. Here we addressed this question. PATIENTS AND METHODS: Mouse islets were isolated and transduced with adeno-associated vectors carrying NAMPT. Glucose-stimulated insulin secretion, beta cell proliferation, and beta cell apoptosis were analyzed in NAMPT-overexpressing islets, compared to controls. RESULTS: Overexpression of NAMPT in primary mouse islets increased glucose-stimulated insulin secretion and increased beta cell expansion through augmentation of beta cell proliferation, without affecting beta cell apoptosis. CONCLUSIONS: NAMPT enhances expansion of beta cell mass during pregnancy. Inadequate NAMPT may contribute to the development of GDM partially through reduced beta cell expansion in the gestational period. [ABSTRACT FROM AUTHOR]
