402. Formulation of secretome derived from mesenchymal stem cells for inflammatory skin diseases

Abstract

Background: Growing evidence has demonstrated that the administration of mesenchymal stem cells (MSCs) in various inflammatory skin diseases caused by autoimmune or allergic reactions can be served as promising therapeutics owing to their immunomodulatory effect as well as tissue regenerative capacity. Area covered: In this review, we summarize the therapeutic application of MSCs and their secretome targeting inflammatory skin diseases and current secretome engineering strategies for the clinical translation of cell-free therapeutics. Expert opinion: Considering that direct application of MSCs often fails to reproduce stable therapeutic outcomes, partially due to harsh in vivo microenvironment leading to insufficient survival of injected cells, the concept of cell-free approaches utilizing MSC-derived paracrine factors including secretome and exosome has emerged to overcome present issues. Furthermore, various bioengineering techniques with biocompatible materials can be applied to modulate naïve secretome to improve their delivery efficiency and bioavailability in vivo.

Abstract

Curcumin, used as a spice and traditional medicine in India, exerts beneficial effects against several diseases, owing to its antioxidant, analgesic, and anti-inflammatory properties. Evidence indicates that curcumin might protect against heavy metal-induced organ toxicity by targeting biological pathways involved in anti-oxidation, anti-inflammation, and anti-tumorigenesis. Curcumin has received considerable attention owing to its therapeutic properties, and the mechanisms underlying some of its actions have been recently investigated. Cadmium (Cd) is a heavy metal found in the environment and used extensively in industries. Chronic Cd exposure induces damage to bones, liver, kidneys, lungs, testes, and the immune and cardiovascular systems. Because of its long half-life, exposure to even low Cd levels might be harmful. Cd-induced toxicity involves the overproduction of reactive oxygen species (ROS), resulting in oxidative stress and damage to essential biomolecules. Dietary antioxidants, such as chelating agents, display the potential to reduce Cd accumulation and metal-induced toxicity. Curcumin scavenges ROS and inhibits oxidative damage, thus resulting in many therapeutic properties. This review aims to address the effectiveness of curcumin against Cd-induced organ toxicity and presents evidence supporting the use of curcumin as a protective antioxidant. [ABSTRACT FROM AUTHOR]

411. Clinical and computed tomography factors associated with sepsis in women with clinically uncomplicated pyelonephritis.

Abstract

Objective: Sepsis is major determinants of prognosis in acute pyelonephritis (APN). This study aimed to assess factors associated with the development of sepsis among patients with clinically uncomplicated APN. Subjects and methods: We examined 463 patients presenting to our hospital without complications. We assessed clinical factors including demographic and laboratory features. Renal and extrarenal features on computed tomography (CT) were also analyzed. Risk factors of sepsis are assessed. Results: The study included 361 patients without (78.0%) and 102 patients with sepsis (22.0%). Crude and attributable mortality rates were 3.9% and 2.0% versus 1.4% and 0.6%, respectively, among patients with and without sepsis. Clinical risk factors for sepsis were age >65 years (odds ratio [OR] 1.79, P = 0.02), absence of flank pain (OR 1.59, P = 0.04), absence of costovertebral tenderness (OR 1.89, P = 0.03), diabetes mellitus (OR 2.25, P = 0.02), bacteremia (OR 2.8, P = 0.01), C-reactive protein level >100 mg/L (OR 1.42, P = 0.02), and lack of previous APN history (OR 1.76, P = 0.04). APN grade IV (OR 3.16, P = 0.01), high grade hydronephrosis (OR 1.50, P = 0.03), diffuse peritoneal thickening (OR 4.12, P = 0.01), and acute interstitial pulmonary edema (OR 3.73, P = 0.01) were the CT features predictive of septic shock. Conclusions: Although uncomplicated APN was largely non-fatal, several clinical and CT features could lead to sepsis. Our findings may be useful for predicting sepsis risk and deciding whether intravenous antibiotic treatment and intensive management should be initiated for uncomplicated APN. [ABSTRACT FROM AUTHOR]

Abstract

Research was conducted to examine the Kerr rotation angle spectrum and magnetic hysteresis loop of CrPt3 film prepared by depositing Cr/Pt multilayers using a rf magnetron sputterer followed by heat treatment. It was shown that CrPt3 films displayed the perpendicular magnetic anisotropy to the film surface, large coercivity of 3.7 kOe and maximum Kerr rotation angle of 0.65 degrees at about 1100 nm.

Interest in nanomedicine due to minimal systemic toxicity and a high degree of target-specific drug-delivery have pulled the attention of health scientists in this area of therapeutics.Area coveredThe review covers the incidence and epidemiology of HCC, proposed molecular drug targets, mechanistic approach and emergence of nanomedicines including nanoparticles, lipidic nanoparticles, vesicular-based nanocarrier, virus-like particles with momentous therapeutic aspects including biocompatibility, and toxicity of nanocarriers along with conclusions and future perspective, with an efficient approach to safely cross physiological barriers to reach the target site for treating liver cancer.Expert opinionRemarkable outcomes have recently been observed for the therapeutic efficacy of nanocarriers with respect to a specific drug target against the treatment of HCC by existing under trial drugs.
