Total organic carbon (TOC) contents in agricultural soil are presently receiving increased attention, not only because of their relationship to soil fertility, but also due to the sequestration of organic carbon in soil to reduce carbon dioxide emissions. In this research, the spatial patterns of TOC and its relationship with pH at the European scale were studied using hot spot analysis based on the agricultural soil results of the Geochemical Mapping of Agricultural Soil (GEMAS) project. The hot and cold spot maps revealed the overall spatial patterns showing a negative correlation between TOC contents and pH values in European agricultural soil. High TOC contents accompanying low pH values in the north-eastern part of Europe (e.g., Fennoscandia), and low TOC with high pH values in the southern part (e.g., Spain, Italy, Balkan countries). A special feature of co-existence of comparatively low TOC contents and low pH values in north-central Europe was also identified on hot and cold spot analysis maps. It has been found that these patterns are strongly related to the high concentration of SiO 2 (quartz) in the coarse-textured glacial sediments in north-central Europe. The hot spot analysis was effective, therefore, in highlighting the spatial patterns of TOC in European agricultural soil and helpful to identify hidden patterns. A special co-existence of both low soil total organic carbon contents and low pH values was identified in agricultural soil in north-central Europe, which is highly correlated with the high concentration of SiO 2 in those soil. • The General Soil Quality Index (GISQ) is used for the first time to assess reclamation of a coal mine. • Maximum differences between the 1-y site and forests were evident, verifying the sensitivity of the indicator. • Rehabilitation operations of the Cerrejón mine allowed recovery of soil macrofauna. The evaluation and success of ecological reclamation can be assessed by measuring soil physical, chemical and biological variables, either in isolation or combined into composite indicators. In this study we tested the suitability of biological, chemical and physical quality indicators–and their combination in a General Indicator of Soil Quality (GISQ)—to monitor soil quality in restored areas of an open cast coal mine. These composite indicators were computed with principal component analyses (PCA) and co-inertia multivariate analyses (CoIA). Our biological indicator showed a significant recovery of soil invertebrate communities along the chronosequence. Taxonomic richness increased from 7 taxa the 1st year to 13–17 taxa in 6–20-y sites, far less than in forests (19–20). Soil pH, bulk density and proportion of physical and macro aggregates were highest in the 1-y site, while soil organic matter (SOM), total N and proportion of biogenic aggregates were highest in older sites. In general, the three sub-indicators and the GISQ yielded the lowest values in the 1-y site (from 0.1 to 0.3 on a scale of 0.1 to 1.0), intermediate in the 16- and 20-y sites, (0.4–0.7) and reached highest values in the two forests (0.4–1.0). Abstract: Restoration ecology that maximizes ecosystem services (ES) requires planning at large spatial scales, which are often the most meaningful for ecosystem functioning and ES supply. As economic resources to undertake ecological restoration at large scales are scarce, prioritizing sites to enhance multiple ES supply is critical. We present the Relative Aggregated Value of Ecosystem Services (RAVES) index, to prioritize sites for ecological restoration based on the assessment of multiple ES. We tested the spatial heterogeneity of ES to identify the relevant scale to managing ES and to apply the RAVES index using a local case study. We also used the RAVES index to compare three alternative restoration scenarios to enhance ES based on the availability of socio‐economic resources. The highest RAVES values were found in areas with natural vegetation and in gorges with riparian forests. The lowest values were found in crop fields, steep slopes and river stretches without riparian forest. The multiscale spatial analysis indicated that most ES showed significant heterogeneity at multiple spatial scales, especially at broad (20–30 km) and very broad (40–50 km) scales. For spatial scales smaller than 2 km, only biological control showed significant heterogeneity. The optimal socio‐economic conditions to enhance ES supply were met when both private and public land, together with economic funds, were available to implement ecological restoration. As most areas with low RAVES were in private lands, even with limited funds restoration of private lands would result in a large increase in RAVES. Synthesis and applications. The Relative Aggregated Value of Ecosystem Services (RAVES) index is a practical tool to hierarchically prioritize sites for ecological restoration across large spatial scales. The RAVES index integrates both ecological information and societal values by weighting ecosystem services (ES) via a multicriteria analysis and can be used in scenario analysis to identify optimal management scenarios. Soil micromorphology in thin section and near infrared reflectance spectroscopy (NIRS) are useful techniques for assessing the participation of soil macrofauna in the formation of aggregates and soil structure. The purpose of this study was to use micromorphological analysis and NIRS techniques to assess the role of soil fauna in the recovery of soil aggregates and in the modification of soil microstructure in a chronosequence of rehabilitated areas at Cerrejon coal mine (La Guajira, Colombia). 64 soil samples were taken from rehabilitated areas (from 1 to 20 years ago) and from natural dry tropical forests Soil macroaggregates were subdivided into three categories: biogenic (BA), physical (PA), and non-aggregated soil (NAS). 32 samples were used for NIRS analysis, while ten thin sections of resin-impregnated soil blocks were micromorphologically analyzed. Principal component analysis of NIRS spectra showed a clear separation between BA, PA, and NAS. Likewise, an increase in BA was observed in the intermediate and advanced stages of rehabilitation. Respect to the micromorphological features, there was a clear change from a matrix of silt-sized quartz, unaccomodated peds, and unstructured materials with non-existent biogenic activity at the 2-year site to the formation of consolidated aggregates, more homogenized soil and increased biological activity at the 20-year site. Soil biological activity, principally the footprint of macrofauna, was recognized using the two techniques as well as at both the micro- and macro-morphology scales. Riparian forests nestled in agricultural landscapes represent a small proportion in crop-intensive areas, while contributing remarkably to their biodiversity. This biodiversity supports several ecological processes crucially involved in the supply of ecosystem services (ES) complementary to that provided by agricultural lands and also relevant for designing biodiverse and multifunctional landscapes. Riparian forest is one of the most threatened ecosystems due to land-use intensification and associated water extraction, especially in Mediterranean semi-arid areas, and proper evaluation of the success of riparian restoration projects is usually lacking. Furthermore, there is little empirical evidence of the effects of riparian restoration on ES supply. In this study, we first investigated the effect of hydrological and soil features on survival and growth of saplings planted in degraded riparian areas in two Mediterranean watersheds. Then, we evaluated how riparian restoration affected the supply of ES, comparing nine regulating and supporting ES on these restored areas with other riparian areas spanning a gradient of conservation status, and with other natural and agricultural land-uses in the same watershed. We found that restoration success mainly depended on water table depth, soil salinity and soil nutrients (namely Mg+2 and Olsen P). Moreover, we detected an antagonistic interaction between the latter two, and a synergetic interaction between water table depth and soil salinity. Forest patches provided meaningful regulating and supporting ES in agricultural landscapes. In particular, riparian restoration zones increased the supply of regulating and supporting ES (water purification, habitat provision, microclimate regulation and soil C storage) in comparison with degraded natural land-uses and crops. Nevertheless, they were still far from the magnitude and range of ES provided by mature riparian forests. In recent years, honeybees ( Apis mellifera) have been strangely disappearing from their hives, and strong colonies have suddenly become weak and died. The precise aetiology underlying the disappearance of the bees remains a mystery. However, during the same period, Nosema ceranae, a microsporidium of the Asian bee Apis cerana, seems to have colonized A. mellifera, and it's now frequently detected all over the world in both healthy and weak honeybee colonies. For first time, we show that natural N. ceranae infection can cause the sudden collapse of bee colonies, establishing a direct correlation between N. ceranae infection and the death of honeybee colonies under field conditions. Signs of colony weakness were not evident until the queen could no longer replace the loss of the infected bees. The long asymptomatic incubation period can explain the absence of evident symptoms prior to colony collapse. Furthermore, our results demonstrate that healthy colonies near to an infected one can also become infected, and that N. ceranae infection can be controlled with a specific antibiotic, fumagillin. Moreover, the administration of 120 mg of fumagillin has proven to eliminate the infection, but it cannot avoid reinfection after 6 months. We provide Koch's postulates between N. ceranae infection and a syndrome with a long incubation period involving continuous death of adult bees, non-stop brood rearing by the bees and colony loss in winter or early spring despite the presence of sufficient remaining pollen and honey. Abstract: The chiral resolution of four antifungal compounds, three imidazoles (miconazole, econazole and sulconazole) and one triazole (itraconazole) using supercritical fluid chromatography on the amylose-based chiral stationary phase Chiralpak AD, is presented in this work. The influence of pressure, type and percentage of organic modifier and temperature on retention times and resolution was studied. The enantiomeric separation of the three imidazoles was achieved with resolutions higher than two and analysis times lower than 10min, obtaining the best results using methanol as modifier. However, the analysis time of the triazole was higher than 80min due to the existence of a high number of functional groups that were able to interact with the chiral stationary phase. In this case, the resolution of the four stereoisomers was achieved only partially with mixtures of ethanol and 2-propanol as modifier. The isoenantioselective temperatures were obtained from the study of the influence of the temperature, they were above the range of temperatures assayed, except for sulconazole using 2-propanol. Abstract: Andean hillsides dominate the landscape of a considerable proportion of Cauca Department in Colombia. The typical cropping cycle in the region includes monocrops or intercrops of maize (Zea mays L.), beans (Phaseolus vulgaris L.) and/or cassava (Manihot esculenta Crantz). Cassava is usually the last crop before local farmers leave plots to natural fallow until soil fertility is recovered and a new cropping phase can be initiated. Previous studies on land use in the Río Cabuyal watershed (6500ha) show that a considerable proportion of land (about 25–30%) remains under natural fallow every year. The focus of our studies is on systems of accelerated regeneration of soil fertility, or improved fallow systems, as an alternative to the natural regeneration by the native flora. Fallow improvement studies were conducted on plots following cassava cultivation. The potential for soil fertility recovery after 12 and 28 months was evaluated with two fast growing trees, Calliandra calothyrsus Meissn (CAL) and Indigofera constricta L.(IND), and one shrub, Tithonia diversifolia (Hemsl.) Gray (TTH), as slash/mulch fallow systems compared to the natural fallow (NAT). All planted slash/mulch fallow systems produced greater biomass than the natural fallow. Greatest dry biomass (16.4Mgha−1 year−1) was produced by TTH. Other planted fallows (CAL and IND) produced about 40% less biomass than TTH and the control (NAT) about 75% less. Nutrient levels in the biomass were especially high for TTH, followed by IND, CAL, and NAT. The impact of fallow management on soil chemical, physical and biological parameters related to residual soil fertility during the cropping phase was evaluated. Soil parameters most affected by slash/mulch fallow systems included soil total N, available N (ammonium and nitrate), exchangeable cations (K, Ca, Mg and Al), amount of P in light fraction, soil bulk density and air permeability, and soil macrofauna diversity. Results from field studies suggest that the Tithonia slash/mulch fallow system could be the best option to regenerate soil fertility of degraded volcanic-ash soils of the Andean hillsides. Plant-soil feedback mechanisms influence the abundance and rarity of plant species and can favour invasive processes, including those of native species. To explore these mechanisms, we analysed correlations between spatial distributions of plant biomass and soil properties in two neighbouring grasslands at different phases of expansion of the native Eurasian tall-grass Brachypodium rupestre (Host) Roem & Schult (B. rupestre cover: >75 and 25–50%). For this, we applied spatially explicit sampling, geostatistical analysis and structural equation models (SEM) to probe causal relationships among measured variables involved in nutrient accumulation. We hypothesized that if litter accumulates as a result of reduced grazing, 'fertility islands' (spots of high SOM and nutrient contents) will form under B. rupestre clumps because the increase in resource inputs from litter will trigger SOM build-up and promote microbial growth. Our results show that 'fertility islands' of P and amino acids occurred under the patchy clumps of B. rupestre in the less invaded grassland. In addition, the SEMs indicated that nutrient accumulation was partially due to mineralization of the SOM and modulated by the soil microbial biomass. However, there was no correlation between spatial patterns of B. rupestre biomass, SOM and microbial biomass. Moreover, the SEMs explained small amounts of variance in them (SOM r2 = 0.22 and microbial biomass r2 = 0.08), suggesting that factors other than B. rupestre biomass were responsible for the high fertility below the patches. Our spatially explicit approach demonstrated that litter inputs in dense temperate grassland communities can generate 'fertility islands' that may favour the stability and expansion of a tall-grass invader and suggest that herbivory may enhance or inhibit this phenomenon. Introduction: The use of medications by emergency medical services (EMS) is increasing. Conventional scores are time-consuming and therefore difficult to use in an emergency setting. For early decision-making, an easy-to-use score based on the medications administered by the EMS may have prognostic value. The primary objective of this study was to develop the prehospital drug-derived score (PDDS) for 2-day mortality., Methods: A prospective, multicenter, ambulance-based cohort study was conducted in adults with undifferentiated acute diseases treated by EMS and transferred to the emergency department. Demographic data, prehospital diagnosis data, prehospital medication and variables for the calculation of the National Early Warning Score 2 (NEWS2), Rapid Emergency Medicine Score (REMS), and Rapid Acute Physiology Score (RAPS) were collected. The PDDS was developed and validated, establishing three levels of risk of 2-day mortality. The predictive capability of each score was determined by the area under the curve of the receiver operating characteristic curve (AUROC) and compared using the Delong's test (p-value)., Results: A total of 6401 patients were included. The PDDS included age and the use of norepinephrine, analgesics, neuromuscular blocking agents, diuretics, antihypertensive agents, tranexamic acid, and bicarbonate. The AUROC of PDDS was .86 (95% CI: .816-.903) versus NEWS2 .866 (95% CI: .822-.911), p = .828; versus REMS .885 (95% CI: .845-.924), p = .311; versus RAPS .886 (95% CI: .846-.926), p = .335, respectively., Conclusion: The newly developed easy-to-use prehospital drug-derived PDDS score has an excellent predictive value of early mortality. Objective: This study aimed to assess the impact of combining transcranial direct current stimulation with end-effector robot-assisted treatment on upper limb function, spasticity, and hand dexterity in chronic stroke patients., Design: This was a prospective, double-blind randomized trial with 20 equally allocated stroke patients. The experimental group received dual transcranial direct current stimulation (anode over affected M1, cathode over contralateral M1) alongside robot-assisted treatment, while the control group received sham transcranial direct current stimulation with the same electrode placement + robot-assisted treatment. Each patient underwent 20 combined transcranial direct current stimulation and robot-assisted treatment sessions. The primary outcome measure was the Fugl-Meyer Upper Limb motor score, with secondary outcomes including AMADEO kinematic measures, Action Research Arm Test, and Functional Independence Measure. Assessments were conducted at baseline, after rehabilitation, and 3 mos later., Results: Combining bilateral transcranial direct current stimulation with robot-assisted treatment did not yield additional improvements in Fugl-Meyer Upper Limb motor score, Functional Independence Measure, or Action Research Arm Test scores among stroke patients. However, the real transcranial direct current stimulation group showed enhanced finger flexion in the affected hand based on AMADEO kinematic measures., Conclusions: The addition of transcranial direct current stimulation to robot-assisted treatment did not result in significant overall functional improvements in chronic stroke patients. Purpose: Pseudomyxoma peritonei (PMP) is a rare and poorly understood malignant condition characterized by the accumulation of intra-abdominal mucin produced from peritoneal metastases. Currently, cytoreductive surgery remains the mainstay of treatment but disease recurrence and death after relapse frequently occur in patients with PMP. New therapeutic strategies are therefore urgently needed for these patients., Experimental Design: A total of 120 PMP samples from 50 patients were processed to generate a collection of 50 patient-derived organoid (PDO) and xenograft (PDX) models. Whole exome sequencing, immunohistochemistry analyses, and in vitro and in vivo drug efficacy studies were performed., Results: In this study, we have generated a collection of PMP preclinical models and identified druggable targets, including BRAFV600E, KRASG12C, and KRASG12D, that could also be detected in intra-abdominal mucin biopsies of patients with PMP using droplet digital PCR. Preclinical models preserved the histopathological markers from the original patient sample. The BRAFV600E inhibitor encorafenib reduced cell viability of BRAFV600E PMP-PDO models. Proof-of-concept in vivo experiments showed that a systemic treatment with encorafenib significantly reduced tumor growth and prolonged survival in subcutaneous and orthotopic BRAFV600E-PMP-PDX mouse models., Conclusions: Our study demonstrates for the first time that systemic targeted therapies can effectively control PMP tumors. BRAF signaling pathway inhibition represents a new therapeutic opportunity for patients with BRAFV600E PMP who have a poor prognosis. Stroke is the third leading cause of disability in the world, and effective rehabilitation is needed to improve lost functionality post-stroke. In this regard, robot-assisted therapy (RAT) and transcranial direct current stimulation (tDCS) are promising rehabilitative approaches that have been shown to be effective in motor recovery. In the past decade, they have been combined to study whether their combination produces adjuvant and greater effects on stroke recovery. The aim of this study was to estimate the effectiveness of the combined use of RATs and tDCS in the motor recovery of the upper extremities after stroke. After reviewing 227 studies, we included nine randomised clinical trials (RCTs) in this study. We analysed the methodological quality of all nine RCTs in the meta-analysis. The analysed outcomes were deficit severity, hand dexterity, spasticity, and activity. The addition of tDCS to RAT produced a negligible additional benefit on the effects of upper limb function (SMD -0.09, 95% CI -0.31 to 0.12), hand dexterity (SMD 0.12, 95% CI -0.22 to 0.46), spasticity (SMD 0.04, 95% CI -0.24 to 0.32), and activity (SMD 0.66, 95% CI -1.82 to 3.14). There is no evidence of an additional effect when adding tDCS to RAT for upper limb recovery after stroke. Combining tDCS with RAT does not improve upper limb motor function, spasticity, and/or hand dexterity. Future research should focus on the use of RAT protocols in which the patient is given an active role, focusing on the intensity and dosage, and determining how certain variables influence the success of RAT.

This work reports on an alternative and advantageous procedure to attain VO 2 -based thermochromic coatings on silicon substrates. It involves the sputtering of vanadium thin films at glancing angles and their subsequent fast annealing in an air atmosphere. By adjusting thickness and porosity of films as well as the thermal treatment parameters, high VO 2 (M) yields were achieved for 100, 200, and 300 nm thick layers treated at 475 and 550 °C for reaction times below 120 s. Comprehensive structural and compositional characterization by Raman spectroscopy, X-ray diffraction, and scanning-transmission electron microscopies combined with analytical techniques such as electron energy-loss spectroscopy bring to the fore the successful synthesis of VO 2 (M) + V 2 O 3 /V 6 O 13 /V 2 O 5 mixtures. Likewise, a 200 nm thick coating consisting exclusively of VO 2 (M) is also achieved. Conversely, the functional characterization of these samples is addressed by variable temperature spectral reflectance and resistivity measurements. The best results are obtained for the VO 2 /Si sample with changes in reflectance of 30-65% in the near-infrared at temperatures between 25 and 110 °C. Similarly, it is also proven that the achieved mixtures of vanadium oxides can be advantageous for certain optical applications in specific infrared windows. Finally, the features of the different structural, optical, and electrical hysteresis loops associated with the metal-insulator transition of the VO 2 /Si sample are disclosed and compared. Incorporating an intentional strain compensating InSb interface (IF) layer in InAs/GaSb type-II superlattices (T2SLs) enhances device performance. But there is a lack of studies that correlate this approach's optical and structural quality, so the mechanisms by which this improvement is achieved remain unclear. One critical issue in increasing the performance of InAs/GaSb T2SLs arises from the lattice mismatch between InAs and GaSb, leading to interfacial strain in the structure. Not only that but also, since each side of the InAs/GaSb heterosystem does not have common atoms, there is a possibility of atomic intermixing at the IFs. To address such issues, an intentional InSb interfacial layer is commonly introduced at the InAs-on-GaSb and GaSb-on-InAs IFs to compensate for the strain and the chemical mismatches. In this report, we investigate InAs/GaSb T2SLs with (Sample A) and without (Sample B) InSb IF layers emitting in the mid-wavelength infrared (MWIR) through photoluminescence (PL) and band structure simulations. The PL studies indicate that the maximum PL intensity of Sample A is 1.6 times stronger than that of Sample B. This could be attributed to the effect of migration-enhanced epitaxy (MEE) growth mode. Band structure simulations understand the impact of atomic intermixing and segregation at T2SL IFs on the bandgap energy and PL intensity. It is observed that atomic intermixing at the IFs changes the bandgap energy and significantly affects the wave function overlap and the optical property of the samples. Transmission electron microscopy (TEM) measurements reveal that the T2SL IFs in Sample A are very rough compared to sharp IFs in Sample B, indicating a high possibility of atomic intermixing and segregation. Based on these results, it is believed that high-quality heterostructure could be achieved by controlling the IFs to enhance its structural and compositional homogeneities and the optical properties of the T2SLs.

Background and Objective: This paper presents an empirical study of a gamified mobile-based assessment approach that can be used to engage students and improve their educational performance., Method: A gamified audience response system called G-SIDRA was employed. Three gamification elements were used to motivate students in classroom activities: badges for achievements to increase engagement, points to indicate progression and performance in the subject and ranking for promoting competitiveness. A total of 90 medical students in a General and Descriptive Anatomy of the Locomotor System course were taught using G-SIDRA in the academic year 2019/2020. Smart bracelets were configured to collect heart rate measurements from 30 students with the aim of evaluating the impact of the gamification elements. The control group consisted of a sample of 110 students enrolled on the same course in the academic year 2016/2017 using non-gamified SIDRA., Results: Statistically significant differences were found between multiple choice questions (MCQ) scores obtained by using SIDRA and G-SIDRA in the four experiments (U = 1.621,50, p < 0,01 for Exp1; U = 1.950,00, p < 0,01 for Exp2; U = 955,00, p < 0,01 for Exp3; U = 2.335,00, p < 0,01 for Exp4). In the students' final exam grades, statistically significant differences between students that used G-SIDRA as opposed to SIDRA (T(157) = 3.992; p = 0.044) were obtained. Concerning gamification elements, statistically significantly differences were found in comparing the pulse increases after and before the badge event in the four experiments (U = 2.484,00, p = 0,038 for Exp1; U = 2.109,50, p = 0,046 for Exp2; U = 1.790,50, p = 0,025 for Exp3; U = 1.557,0, p = 0,048 for Exp4). However, there are not statistically significant differences between the pulse increases after and before the ranking event in the four experiments. In a 5-point Likert-type scale, the students expressed satisfaction with G-SIDRA (M = 4.552) and thought the system helped to better understand both theoretical and practical concepts (M = 4.092). Their global assessment of the G-SIDRA platform was 4.471., Conclusions: Of the three gamification elements used in the study, only badge has an effect on heart rate. Better student responses and academic performance were achieved when using G-SIDRA. Nevertheless, more research is required to evaluate the impact of the gamification elements on the motivation, engagement and performance of students. Concentration data of veterinary drugs in microalgae biomass collected from photobioreactors fed with piggery wastewaters are presented for the first time in this work. To this aim, a QuEChERS methodology and an ultrasound-assisted solid-liquid extraction have been assessed as sample preparation procedures with the purpose of determining 20 veterinary drugs, mainly antibiotics of different physico-chemical properties in addition to dexamethasone, fenbendazole and progesterone. Some critical operation parameters of the QuEChERS procedure were optimized by an experimental design but tetracycline, oxytetracycline, doxycycline, marbofloxacin and ciprofloxacin were not detected by the QuEChERS sample preparation. The use of a longer and thorough approach, a solid-liquid extraction with water/methanol in presence of primary secondary amine as a clean-up agent followed by solid-phase extraction on Oasis HLB cartridges, is recommended to monitor all intended analytes. The determination in extracts is carried out by ultra-high performance liquid chromatography-tandem mass spectrometry in selected reaction monitoring mode. Limits of detection about 0.2-42 ng per g of lyophilized microalgae sample, and repeatabilities about 6-46% (n = 5, RSDs) are reached. The solid-liquid extraction method was applied to microalgae biomass samples collected from a photobioreactor. Nine drugs were detected in the samples at relatively low concentration and a proportional relationship between the found concentrations and the octanol/water partition coefficients of the drugs has been outlined. This paper presents three experiments to assess the impact of gamifying an audience response system on the perceptions and educational performance of students. An audience response system called SIDRA (Immediate Audience Response System in Spanish) and two audience response systems with gamification features, R-G-SIDRA (gamified SIDRA with ranking) and RB-G-SIDRA (gamified SIDRA with ranking and badges), were used in a General and Descriptive Human Anatomy course. Students participated in an empirical study. In the academic year 2019-2020, a total of 90 students used RB-G-SIDRA, 90 students employed R-G-SIDRA in the academic year 2018-2019, and 92 students used SIDRA in the academic year 2017-2018. Statistically significant differences were found between final exam grades obtained by using RB-G-SIDRA and SIDRA, U = 39.211 adjusted p = 0.001 and RB-G-SIDRA and R-G-SIDRA U = 31.157 adjusted p = 0.015, thus finding strong evidence with respect to the benefit of the badges used in RB-G-SIDRA. Moreover, in the students' SIDRA systems scores, statistically significant differences were found between RB-G-SIDRA and SIDRA, U = -90.521 adjusted p < 0.001, and between R-G-SIDRA and SIDRA, U = -87.998 adjusted p < 0.001. Significant correlations between individual and team scores were also found in all of the tests in RB-G-SIDRA and G-SIDRA. The students expressed satisfaction, engagement, and motivation with SIDRA, R-G-SIDRA, and RB-G-SIDRA, thus obtaining a final average assessment of 4.28, 4.61, and 4.47 out of 5, respectively. Students perform better academically with gamified versus non-gamified audience response systems. Findings can be used to build a gamified adaptive learning system.

In this work, synthetic genes designed from (a), the native amino acid sequence of the class IIa bacteriocins enterocin HF (EntHF) and enterocin CRL35 (EntCRL35), (b) from hybrid bacteriocins derived from fusion of enterocin A (EntA) to itself and to EntHF and EntCRL35 through a tri-glycine peptide linker, and (c) from bacteriocin-derived chimeras devised from fusion of the N-terminal region of EntA and enterocin P (EntP) to the C-terminal end of EntHF and EntCRL35, were cloned in plasmid pPICZαA for expression by P. pastoris X-33. Synthetic genes encoding EntHF and EntCRL35 were also cloned in plasmid pP-αhSUMO3 for expression of the hSUMO3-fused bacteriocins by P. pastoris. Only recombinant P. pastoris expressing the bacteriocin-derived chimeras displayed a direct antimicrobial activity whereas P. pastoris X-33, producer of EntP::EntHF, showed the highest antimicrobial activity in their supernatants and in the multi-step chromatographic purified fractions. The MRM-ESI-LC-MS/MS (QTRAP) analysis of purified fractions from P. pastoris producers of hybrid- and bacteriocin-derived chimeras, permitted detection in the samples of peptides with the expected molecular mass of the bacteriocins produced. The antimicrobial activity of the EntP::EntHF chimera compared to that of the synthetic EntP::EntHF peptide, suggest that the biologically-produced bacteriocin-derived chimera shows a higher specific antimicrobial activity than its synthetic counterpart against different Listeria strains, including L. monocytogenes. More important, the N-terminal region of EntA and EntP seems to drive the production, processing and secretion of hybrid- and bacteriocin-derived chimeras, by P. pastoris X-33., (Copyright © 2019 Elsevier Ltd. Celecoxib is an anti-inflammatory drug with antibacterial activity whose fate in surface water is unknown. Thus, some assays have been conducted under forced biological, photochemical and thermal conditions, and non-forced conditions, to establish its persistence and degradation products in river water. The results suggest that celecoxib dissolved in river water is not biologically degraded while it is minimally altered after its exposure to sunlight or high temperature (70°C). Only the irradiation at 254nm promotes its complete degradation. Celecoxib is degraded about 3%, in 36 weeks, when water was kept at room temperature and the exposure to sunlight was partially limited as it happens inside a body of water. Residues were monitored by ultra-pressure liquid chromatography/quadrupole time-of-flight/mass spectrometry after solid-phase extraction; eleven degradation products were detected and the structures of nine of them were unequivocally proposed from the molecular formulae and fragmentation observed in high-resolution tandem mass spectra. The long-term transformation products under non-forced conditions were 4-[5-(4-methylphenyl)-3-(trifluoromethyl)-1H-pyrazol-1-yl]benzenesulfonic acid, 4-[1-(4-sulfoaminephenyl)-3-(trifluoromethyl)-1H-pyrazol-5-yl]benzoic acid and a hydroxylated derivative. The degradation over time in presence of sediment was monitored, being slightly higher, about 4%. The fate of the pharmaceutical drugs tenoxicam, piroxicam and meloxicam in river water is evaluated here for first time. So, biological, photochemical and thermal degradation assays have been conducted to estimate their degradation rates and know their degradation products. Results indicated that the direct sunlight irradiation, without any protection, promoted a fast degradation of the oxicams while the chemical reactions in solution were less important. The biological degradation in water was negligible except for tenoxicam in whose case its influence was scarce. When the exposition of river water to sunlight was partially limited and kept under the natural day-night cycle, as occurs inside a body of water, tenoxicam, piroxicam and meloxicam (at 2 μg L -1 ) were detected during a period of 15, 27 and 45 days, respectively. Residues were monitored by ultra-pressure liquid chromatography/quadrupole time-of-flight/mass spectrometry after solid-phase extraction and several degradation products were found (10 for tenoxicam, 9 for piroxicam and 7 for meloxicam) and monitored over time. Their structures were proposed from the molecular formulae and fragmentation observed in high-resolution tandem mass spectra; the nature of the transformation products found in the long-term resulted to be very variable for each oxicam. Furthermore, the degradation in presence of river sediment was also monitored over time, with some differences being noted; the adsorption coefficients of the compounds on sediment were calculated, meloxicam exhibited a higher sorption capacity. Toxic effects of the non-biodegradable drug chlorpromazine and its degradation products have been reported on microorganisms in aqueous media. Here, chlorpromazine degradation assays in forced and non-forced conditions have been done to know its persistence and degradation products in river water. Sunlight irradiation promotes the complete degradation of chlorpromazine (2 μg L(-1)) in less than 4 h, but if the exposure to sunlight is limited chlorpromazine is detected during 4 weeks in river water. Sixteen degradation products in surface water are described for first time after solid-phase extraction and analysis by ultra-pressure liquid chromatography/quadrupole time-of-flight/mass spectrometry; their structures are proposed from the molecular formulae of the fragment-ions observed in high-resolution tandem mass spectra. Hydroxylation and oxidation products such as chlorpromazine sulfoxide, 2-hydroxypromazine and 2-hydroxypromazine sulfoxide were predominant degradation products in the early stages; some benzo[1,4]thiazin-6-ol derivatives resulting from the breakdown of the phenothiazine core were the major and relatively stable products after 20 weeks under non-forced conditions. A degradation pathway of chlorpromazine in water is outlined. Moreover, it is shown that chlorpromazine is very strongly adsorbed on sediment while the degradation products that kept the promazine core have a notable capacity of sorption, too; sorption coefficients are calculated. Finally, a prediction about the toxicity of the degradation products in aquatic ecosystems suggests that some of them have toxicities similar, or even higher, than chlorpromazine., (Copyright © 2016 Elsevier Ltd. The use of synthetic genes may constitute a successful approach for the heterologous production and functional expression of bacterial antimicrobial peptides (bacteriocins) by recombinant yeasts. In this work, synthetic genes with adapted codon usage designed from the mature amino acid sequence of the bacteriocin enterocin A (EntA), produced by Enterococcus faecium T136, and the mature bacteriocin E 50-52 (BacE50-52), produced by E. faecium NRRL B-32746, were synthesized. The synthetic entA and bacE50-52 were cloned into the protein expression vectors pPICZαA and pKLAC2 for transformation of derived vectors into Pichia pastoris X-33 and Kluyveromyces lactis GG799, respectively. The recombinant vectors were linearized and transformed into competent cells selecting for P. pastoris X-33EAS (entA), P. pastoris X-33BE50-52S (bacE50-52), K. lactis GG799EAS (entA), and K. lactis GG799BE50-52S (bacE50-52). P. pastoris X-33EAS and K. lactis GG799EAS, but not P. pastoris X-33BE50-52S and K. lactis GG799BE50-52S, showed antimicrobial activity in their supernatants. However, purification of the supernatants of the producer yeasts permitted recovery of the bacteriocins EntA and BacE50-52. Both purified bacteriocins were active against Gram-positive bacteria such as Listeria monocytogenes but not against Gram-negative bacteria, including Campylobacter jejuni.

A sample preparation procedure that combines a liquid-liquid extraction (LLE) with a dispersive solid-phase extraction (DSPE) has been devised to determine residues of four phenoxyacid herbicides, two aminopolycarboxylic acids and five acidic anti-inflammatory drugs in small volumes of river water samples. Two aliquots of acetone (3 and 0.5 mL) were used to extract the analytes from a 10 mL water sample at pH 2 containing 5 mg of octadecylsilane (ODS) sorbent and NaCl at a 5.5 M concentration. Acetone was isolated by the salting-out effect, collected, evaporated and the extract was treated with BF3/methanol to obtain the methyl esters of the analytes and determine them by GC with mass spectrometric detection. Recoveries were comprised between 82% and 114% with relative standard deviations about 5-15% (n=5) within a concentration range about 0.03-44 µg L(-1). The amount of ODS added to sample resulted to be a significant factor in the recovery for most of the analytes as deduced from an experimental design; the sample pH was not a so critical factor. A robustness study of the proposed sample preparation was carried out as defined by Youden and Steiner and an estimation of the uncertainties of the measured concentrations was made following the EURACHEM/CITAC guidelines, too., (Copyright © 2013 Elsevier B.V. Mature sakacin A (SakA, encoded by sapA) and its cognate immunity protein (SakI, encoded by sapiA), and two SakA-derived chimeras mimicking the N-terminal end of mature enterocin P (EntP/SakA) and mature enterocin A (EntA/SakA) together with SakI, were fused to different signal peptides (SP) and cloned into the protein expression vectors pNZ8048 and pMG36c for evaluation of their production and functional expression by different lactic acid bacteria. The amount, antimicrobial activity, and specific antimicrobial activity of SakA and its chimeras produced by Lactococcus lactis subsp. cremoris NZ9000 depended on the SP and the expression vector. Only L. lactis NZ9000 (pNUPS), producing EntP/SakA, showed higher bacteriocin production and antimicrobial activity than the natural SakA-producer Lactobacillus sakei Lb706. The lower antimicrobial activity of the SakA-producer L. lactis NZ9000 (pNUS) and that of the EntA/SakA-producer L. lactis NZ9000 (pNUAS) could be ascribed to secretion of truncated bacteriocins. On the other hand, of the Lb. sakei Lb706 cultures transformed with the pMG36c-derived vectors only Lb. sakei Lb706 (pGUS) overproducing SakA showed a higher antimicrobial activity than Lb. sakei Lb706. Finally, cloning of SakA and EntP/SakA into pPICZαA and pKLAC2 permitted the production of SakA and EntP/SakA by recombinant Pichia pastoris X-33 and Kluyveromyces lactis GG799 derivatives although their antimicrobial activity was lower than expected from their production.

A new sample preparation procedure to determine aminopolycarboxylic acids (ethylenediaminetetraacetic acid, EDTA, nitrilotriacetic acid, NTA, diethylenetriaminepentaacetic acid, DTPA, and cyclohexanediaminetetraacetic acid, CDTA) in river water is described. The procedure consists of the solid-phase extraction of the aminopolycaroxyllic acids on activated charcoal cartridges after increasing the ionic strength and acidifying the sample. The extract was eluted with methanol and the analytes were methylated in presence of BF3/methanol to determine them by GC with mass spectrometric detection. Recoveries were higher than 90% with good repeatabilities and inter-day precision for concentrations close to quantification limits (about 10 μg L(-1)) and higher. It has been verified that the proposed method is robust according to the Youden and Steiner test and free of matrix effects arisen from the presence of organic matter and iron(III) as deduced from statistical tests. A bottom-up approach was followed to estimate the uncertainty of the measured concentration. At concentrations close to 10 μg L(-1) the most relevant step of the method is the calculus of the interpolated concentration which has a high value of relative standard uncertainty., (Copyright © 2013 Elsevier B.V. Replacement of the leader sequence (LS) of the bacteriocin enterocin A (LS(entA)) by the signal peptides (SP) of the protein Usp45 (SP(usp45)), and the bacteriocins enterocin P (SP(entP)), and hiracin JM79 (SP(hirJM79)) permits the production, secretion, and functional expression of EntA by different lactic acid bacteria (LAB). Chimeric genes encoding the SP(usp45), the SP(entP), and the SP(hirJM79) fused to mature EntA plus the EntA immunity genes (entA+entiA) were cloned into the expression vectors pNZ8048 and pMSP3545, under control of the inducible P(nisA) promoter, and in pMG36c, under control of the constitutive P(32) promoter. The amount, antimicrobial activity, and specific antimicrobial activity of the EntA produced by the recombinant Lactococcus lactis, Enterococcus faecium, E. faecalis, Lactobacillus sakei and Pediococcus acidilactici hosts varied depending on the signal peptide, the expression vector, and the host strain. However, the antimicrobial activity and the specific antimicrobial activity of the EntA produced by most of the LAB transformants was lower than expected from their production. The supernatants of the recombinant L. lactis NZ9000 (pNZUAI) and L. lactis NZ9000 (pNZHAI), overproducers of EntA, showed a 1.2- to 5.1-fold higher antimicrobial activity than that of the natural producer E. faecium T136 against different Listeria spp., (Copyright © 2011 Elsevier B.V. Introduction and Objectives: Our aim was to study the radiological anatomy of the ethmoidal arteries., Methods: A descriptive study was performed including CT images of 20 cadaver heads. The specimens were perfused with a radiopaque material and various anatomical parameters were analysed., Results: The anterior ethmoidal artery was found in 95% (38/40) of cases. It originated from the ophthalmic artery in 87.5% (34/40) of nasal cavities. In six cases, normal variants were found. The mean length of the anterior ethmoidal canal was 8.43 ± 0.74 mm. The angle performed into the skull base was 37.3 ± 5.48°. In 90% of cases (36/40), it was located between the second and third lamella. The posterior ethmoidal artery was localised only in 14/40 cases, with 28.5% (4/14) of them showing normal variants. The mean length of the posterior ethmoidal canal was 7.1 ± 1.02 mm. The angle performed into the skull base was 7.11 ± 4.07°. The distance from sill to the anterior ethmoid artery was 55.51 ± 5.52 mm. The angle between the nasal spine and the anterior ethmoidal canal was 57.67 ± 1.68°. The distance between the nasion and the anterior ethmoidal canal was 29.31 ± 2.53 mm, the distance was 11.24 ± 2.14 mm from the anterior ethmoid artery to the posterior ethmoid artery and from the posterior ethmoid artery to the optic nerve, 7.26 ± 1.33 mm. Replacement of the signal peptide (SP) of the bacteriocins enterocin P (EntP) and hiracin JM79 (HirJM79), produced by Enterococcus faecium P13 and Enterococcus hirae DCH5, respectively, by the signal peptide of Usp45 (SP(usp45)), the major Sec-dependent protein secreted by Lactococcus lactis, permits the production, secretion, and functional expression of EntP and HirJM79 by L. lactis. Chimeric genes encoding the SP(usp45) fused to either mature EntP (entP), with or without the immunity gene (entiP) or to mature HirJM79 (hirJM79), with or without the immunity gene (hiriJM79), were cloned into the expression vector pMG36c, carrying the P(32) constitutive promoter, and into pNZ8048 under control of the inducible PnisA promoter. The production of EntP and HirJM79 by most of the L. lactis recombinant strains was 1.5- to 3.7-fold higher and up to 3.6-fold higher than by the E. faecium P13 and E. hirae DCH5 control strains, respectively. However, the specific antimicrobial activity of the recombinant EntP was 1.1- to 6.2-fold higher than that produced by E. faecium P13, while that of the HirJM79 was a 40% to an 89% of that produced by E. hirae DCH5. Chimeras of SP(usp45) fused to mature EntP or HirJM79 drive the production and secretion of these bacteriocins in L. lactis in the absence of specific immunity and secretion proteins. The supernatants of the recombinant L. lactis NZ9000 strains, producers of EntP, showed a much higher antimicrobial activity against Listeria spp. than that of the recombinant L. lactis NZ9000 derivatives, producers of HirJM79.

Introduction and Objectives: Postoperative atrial fibrillation is a common complication of carrying out cardiac surgery with extracorporeal circulation (ECC). The aim of this study was to determine whether preoperative left atrial contractile dysfunction, as assessed by tissue Doppler echocardiography, is associated with the development of postoperative new-onset atrial fibrillation (PAF)., Methods: Transthoracic Doppler echocardiography was performed preoperatively in patients undergoing elective cardiac surgery. Left atrial contractile function was evaluated by tissue Doppler imaging (TDI) of the mitral annulus., Results: The study included 92 patients in sinus rhythm preoperatively who underwent elective cardiac surgery with ECC: 73 (79%) were male and 19 (21%) were female, and their mean age was 67 (10) years. Of these, 19 (20.6%) developed PAF 34 (12) h postoperatively. Bivariate analysis showed that PAF was associated with older age (71 [7] years vs 66 [10] years; P=.034), a large left atrial diameter (LAD), and a low peak atrial systolic mitral annular velocity (A velocity) and a high mitral E/A ratio on TDI. Logistic regression analysis showed that PAF was independently associated with a large LAD (odds ratio [OR] =2.23; 95% confidence interval [CI], 1.05-4.76; P=.033) and a low A velocity (OR=0.70; 95% CI, 0.55-0.99; P=.034)., Conclusions: Preoperative left atrial dysfunction, as assessed by TDI, was associated with an increased risk of PAF.
