Schismotherium fractum, the type species for the genus, is a basal Megatherioidea (Xenarthra, Folivora) from the Santa Cruz Formation (Early Miocene) of Patagonia. The type specimen, currently lost, was described as a fragmented left mandible. Several years later, Ameghino refined his description based on additional material that included the skull and mandible, and several postcranial elements, all deposited in the Museo Argentino de Ciencias Naturales “Bernardino Rivadavia” (Buenos Aires, Argentina; MACN-A 6445-70). A neotype for this species is required to permit further systematic and taxonomic work on several sloth taxa, and we designate MACN-A 6445-70 as neotype for Schismotherium fractum.

The reproductive consequences of global warming are not currently understood. In order to address this issue, we have examined the reproductive consequences of exposing male mice to a mild heat stress. For this purpose, adult male mice were exposed to an elevated ambient temperature of 35°C under two exposure models. The first involved acute exposure for 24 h, followed by recovery periods between 1 day and 6 weeks. The alternative heating regimen involved a daily exposure of 8 h for periods of 1 or 2 weeks. In our acute model, we identified elevated sperm mitochondrial ROS generation (P 0.05), increased sperm membrane fluidity (P 0.05), DNA damage in the form of single-strand breaks (P 0.001), and oxidative DNA damage (P 0.05), characteristic of an oxidative stress cascade. This DNA damage was detected in pachytene spermatocytes (P 0.001) and round spermatids (P 0.001) isolated from testes after 1 day heat recovery. Despite these lesions, the spermatozoa of heat-treated mice exhibited no differences in their ability to achieve hallmarks of capacitation or to fertilize the oocyte and support development of embryos to the blastocyst stage (all P 0.05). Collectively, our acute heat stress model supports the existence of heat susceptible stages of germ cell development, with the round spermatids being most perturbed and spermatogonial stem cells exhibiting resistance to this insult. Such findings were complemented by our chronic heat stress model, which further supported the vulnerability of the round spermatid population.

We have previously reported the synthesis and breast cancer selectivity of (Z)-2-(3,4-dichlorophenyl)-3-(1H-pyrrol-2-yl)acrylonitrile (ANI-7) in cancer cell lines. To further evaluate the selectivity of ANI-7, we have expanded upon the initial cell line panel to now include the breast cancer cell lines (MCF7, MCF7/VP16, BT474, T47D, ZR-75-1, SKBR3, MDA-MB-468, BT20, MDA-MB-231); normal breast cells (MCF-10A); and cell lines derived from colon (HT29), ovarian (A2780), lung (H460), skin (A431), neuronal (BE2C), glial (U87, SJG2), and pancreatic (MIA) cancers. We now show that ANI-7 is up to 263-fold more potent at inhibiting the growth of breast cancer cell lines (MCF7, MCF7/VP16, BT474, T47D, ZR-75-1, SKBR3, MDA-MB-468) than normal breast cells (MCF-10A) or cell lines derived from other tumor types. Measures of growth inhibition, cell cycle analysis, morphologic assessment, Western blotting, receptor binding, gene expression, small interfering RNA technology, reporter activity, and enzyme inhibition assays were exploited to define the mechanism of action of ANI-7. In this work, we report that ANI-7 mediates its effects via the activation of the aryl hydrocarbon receptor (AhR) pathway and the subsequent induction of CYP1-metabolizing mono-oxygenases. The metabolic conversion of ANI-7 induces DNA damage, checkpoint activation, S-phase cell cycle arrest, and cell death in sensitive breast cancer cell lines. Basal expression of AhR, the AhR nuclear translocator, and the CYP1 family members do not predict for sensitivity; however, inherent expression of the phase II-metabolizing enzyme sulfur transferase 1A1 does. For the first time, we identify (Z)-2-(3,4-dichlorophenyl)-3-(1H-pyrrol-2-yl)acrylonitrile as a new AhR ligand.

In this work, laser-induced breakdown spectroscopy (LIBS) is applied for quantitative measurements of Pb in aerosols. In order to investigate the carrier gas role and, in particular, the effect of O2 addition to the gas itself, measurements are carried out in nitrogen and air atmosphere. Aerosol particles are produced by nebulizing Pb(CH3COO)2 * 3H2O aqueous solutions of known concentration and the atomic line of 405.8 nm is detected as Pb signature. The plasma generated with the laser pulse is characterized in terms of plasma temperature and electron density, showing no substantial differences with the two carrier gases used. The behavior of the LIBS signal as a function of the delay time with respect to the laser pulse is investigated changing the environmental conditions and, in particular, the Pb concentration values. The different trends registered in the case of relatively short (up to 20 μs) and long delay time, resulting to be the same whatever the Pb concentration value, could have a significant effect on the calibration curve performed in different experimental conditions.

Critical infrastructures are an integral part of our society and economy. Services like gas supply or water networks are expected to be available at all times since a service failure may incur catastrophic consequences to the public health, safety, and financial capacity of the society. Several resilience strategies have been examined to reduce disaster risk and evaluate the downtime of infrastructures following destructive events. This paper introduces an indicator-based downtime estimation model for buried infrastructures (i.e., water and gas networks). The model distinguishes the important aspects that contribute to determining the downtime of buried infrastructure following a hazardous event. The proposed downtime model relies on two inference methods for its computation, Fuzzy Logic (FL) and Bayesian Network (BN), which are adapted for the current application. Finally, through a case scenario, a comparison of the two inference methods, in terms of results and limitations, is presented. Results show that both methods incorporate intuitive knowledge and/or historical data for defining fuzzy rules (in FL) and estimating conditional probabilities (in BN). The difference stands in the interpretation of the outcome. The output of the FL is a membership that defines how well the downtime fits the fuzzy levels while the BN output is a probability distribution that represents how likely the downtime is in a certain state. Nevertheless, both approaches can be utilized by decision-makers to easily estimate the time to restore the functionality of buried infrastructures and plan preventive safety measures accordingly.

The aims of this study were to investigate the proteome of koala spermatozoa and that of the prostatic bodies with which they interact during ejaculation. For this purpose, spermatozoa and prostatic bodies were fractionated from the semen of four male koalas and analysed by HPLC MS/MS. This strategy identified 744 sperm and 1297 prostatic body proteins, which were subsequently attributed to 482 and 776 unique gene products, respectively. Gene ontology curation of the sperm proteome revealed an abundance of proteins mapping to the canonical sirtuin and 14-3-3 signalling pathways. By contrast, protein ubiquitination and unfolded protein response pathways dominated the equivalent analysis of proteins uniquely identified in prostatic bodies. Koala sperm proteins featured an enrichment of those mapping to the functional categories of cellular compromise/inflammatory response, whilst those of the prostatic body revealed an over-representation of molecular chaperone and stress-related proteins. Cross-species comparisons demonstrated that the koala sperm proteome displays greater conservation with that of eutherians (human; 93%) as opposed to reptile (crocodile; 39%) and avian (rooster; 27%) spermatozoa. Together, this work contributes to our overall understanding of the core sperm proteome and has identified biomarkers that may contribute to the exceptional longevity of koala spermatozoa during ex vivo storage.

In spite of the advances in laser diagnostics in combustion, the effect of rapid laser irradiation on the physical/chemical properties of soot particles is far from being comprehensively understood. Optical properties, particle nanostructure and aggregate size of laser-irradiated soot particles are analyzed in this paper. Carbonaceous particles sampled from nitrogen-quenched diffusion flames of ethylene and methane are irradiated on-line by a 1064-nm short laser pulse. Wavelength-resolved extinction measurements in the visible range are used to follow their transformation by varying the laser energy density. A variation of the extinction coefficient of the irradiated particles compared to the extinction coefficient of the pristine ones is observed, especially for ethylene soot. The particle nanostructures are analyzed by Raman spectroscopy and the effect of laser irradiation on aggregate structure is evaluated by measuring particle size distributions. The results indicate that both soot nanostructure and optical properties are strongly dependent on the laser energy density when irradiated by a laser source. This is significant for ethylene soot, while for methane soot the degree of variation of such properties is less pronounced, at least in the investigated heating conditions. © 2020 Elsevier Inc.
