The main aim of this research was to explore Parthenium hysterophorus Linn phytochemically and pharmacologically. Phytochemical screening is important for the isolation of active compounds before bulk extraction. The crude extracts and their fractions were screened for enzyme (urease, α-glycosidase, and phosphodiesterase) inhibition assays, in vivo analgesic, anti-inflammatory, and sedative effects. Results indicated the presence of steroids, flavonoids, etc. The crude extracts such as methanol, hexane, aqueous, ethyl acetate, chloroform, and butanol exhibited excellent urease inhibitory activities with IC50 = 43.1 ± 1.24, 31.9 ± 2.21, 31.9 ± 2.21, 57.3 ± 1.27, 49.2 ± 2.16, and 35.3 ± 1.12, respectively, as compared to standard acetohydroxamic acid (20.3 ± 0.43). The extracts (methanol, hexane, aqueous, ethyl acetate, chloroform, and butanol) also showed promising α-glycosidase potency with IC50 = 13.1 ± 0.34, 21.2 ± 1.16, 23.1 ± 0.12, 84.2 ± 2.17, 118.6 ± 3.07, and 840 ± 1.73, respectively against acarbose (840 ± 1.73). The phosphodiesterase activity of the mentioned extracts was also excellent with IC50 = 131.1 ± 2.41, 197.2 ± 3.16, 24.2 ± 0.11, 62.4 ± 2.21, 152.4 ± 1.81, and 55.3 ± 2.15, respectively, against the standard (265.5 ± 2.25). Furthermore, butanol (14.96 ± 1.78), ethyl acetate (18.98 ± 1.71), and methanol (16.87 ± 1.00) showed dose-dependent analgesic effects with a maximum inhibition of acetic acid-induced writhes. Whereas, methanolic and butanol extracts exhibited maximum inhibition of inflammation in the carrageenan paw edema test. The aqueous ( p < 0.01 ) and butanol ( p < 0.01 ) extracts exhibited maximum a sedative effect followed by chloroform ( p < 0.05 ), ethyl acetate ( p < 0.05 ), and methanolic ( p < 0.05 ) fractions as compared to the standard drug. The current research concluded that Parthenium hysterophorus Linn has important phytochemical constituents having inhibitory effects on urease, α-glycosidase, and phosphodiesterase enzymes. Furthermore, the plant has analgesic, anti-inflammatory, and sedative effects. The P. hysterophorus needs to further be explored for the candidate molecules responsible for the abovementioned activities.

Additive manufacturing is a key component of the fourth industrial revolution (IR4.0) that has received increased attention over the last three decades. Metal additive manufacturing is broadly classified into two types: melting-based additive manufacturing and solid-state additive manufacturing. Friction stir additive manufacturing (FSAM) is a subset of solid-state additive manufacturing that produces big area multi-layered components through plate addition fashion using the friction stir welding (FSW) concept. Because of the solid-state process in nature, the part produced has equiaxed grain structure, which leads to better mechanical properties with less residual stresses and solidification defects when compared to existing melting-based additive manufacturing processes. The current review article intends to highlight the working principle and previous research conducted by various research groups using FSAM as an emerging material synthesizing technique. The summary of affecting process parameters and defects claimed for different research materials is discussed in detail based on open access experimental data. Mechanical properties such as microhardness and tensile strength, as well as microstructural properties such as grain refinement and morphology, are summarized in comparison to the base material. Furthermore, the viability and potential application of FSAM, as well as its current academic research status with technology readiness level and future recommendations are discussed meticulously.

Phospholipase A2 (PLA2) promotes inflammation via lipid mediators and releases arachidonic acid (AA), and these enzymes have been found to be elevated in a variety of diseases, including rheumatoid arthritis, sepsis, and atherosclerosis. The mobilization of AA by PLA2 and subsequent synthesis of prostaglandins are regarded as critical events in inflammation. Inflammatory processes may be treated with drugs that inhibit PLA2, thereby blocking the COX and LOX pathways in the AA cascade. To address this issue, we report herein an efficient method for the synthesis of a series of octahydroquinazolinone compounds (4a–h) in the presence of the catalyst Pd-HPW/SiO2 and their phospholipase A2, as well as protease inhibitory activities. Among eight compounds, two of them exhibited overwhelming results against PLA2 and protease. By using FT-IR, Raman, NMR, and mass spectroscopy, two novel compounds were thoroughly studied. After carefully examining the SAR of the investigated compounds against these enzymes, it was found that compounds (4a, 4b) containing both electron-donating and electron-withdrawing groups on the phenyl ring exhibited higher activity than compounds with only one of these groups. DFT studies were employed to study the electronic nature and reactivity properties of the molecules by optimizing at the BLYP/cc-pVDZ. Natural bond orbitals helped to study the various electron delocalizations in the molecules, and the frontier molecular orbitals helped with the reactivity and stability parameters. The nature and extent of the expressed biological activity of the molecule were studied using molecular docking with human non-pancreatic secretory phospholipase A2 (hnps-PLA2) (PDB ID: 1DB4) and protease K (PDB ID: 2PWB). The drug-ability of the molecule has been tested using ADMET, and pharmacodynamics data have been extracted. Both the compounds qualify for ADME properties and follow Lipinski’s rule of five.

A study on strategies regarding advancement in heat transfer characteristics in two-dimensional closed domains by placing cold cylinders is conducted. This effort is undertaken due to the fact that active and passive control in heat transmission is connected with provision of temperature differences at different locations of enclosures. Based on the experiments, researchers have concluded that placement of cold cylinder in non-uniformly distributed heat in a cavity is the most effective technique to enrich heat transfer rate, along with reducing the the waste of extra heat generation in processes such as polymer and aero dynamical extrusion, glass cooling, refrigeration, heating and cooling systems. Thus, the prime goal of this work is to outline heat and flow characteristics of non-linear fluid occupied in a square enclosure with adjustment of the cold cylinder. Heat transfer attributes are incorporated by accounting buoyancy forces and forming coupling of molecular diffusion of fluid within the flow domain. Formulation of the problem in dimensionless form is attained by encapsulating the aspects of natural convection in view of principal partial differential equations. Parametric study for governing expressions is computed numerically with the finite element method based on COMSOL Multiphysics version 5.6. Quadric interpolating functions are used to obtain information about velocity and temperature on nodes in elements. Hybrid meshing is manifested for discretization of the domain into rectangular and triangular elements. For the optimized variation in flow structures, prospective parameters are varied from 0.5≤n≤1.5, 5 ≤ Pr ≤ 35 and 102 ≤ Ra ≤106. The achieved results are projected graphically through streamlines, isotherms, and local and average Nusselt numbers. Tabular data for kinetic energy and wall heat flux are also calculated. It is inferred through the analysis that, with uplift in the Rayleigh number Ra elevation in the magnitude of kinetic energy and convective heat transfer arises, whereas the reverse pattern is depicted versus the power–law index n.

A method consisting in an optimal combination of conventional topography from a terrestrial acquisition and satellite derived topography is presented. The solution recently implemented in the UAE for the engineering and the construction of a gas export pipeline allows significant cost reduction, time saving, and safety hazard reduction as fewer terrestrial operations are needed. The survey area is split into 2 sub-areas: area with infrastructures requiring a high accuracy is surveyed with terrestrial topographical acquisition methods such as GNSS receivers, the other one with desert conditions is mapped from satellite stereoscopic imagery. Stereoscopic mode refers to when the satellite sensor acquires two images of the same location taken from different angles. Using photogrammetric techniques, it produces a 3D elevation model of the area. The native satellite imagery allows a mapping of the surface features as well. Terrestrial and satellite datasets are finally merged and adjusted to provide engineering and construction contractors with a unique survey dataset. Terrestrial survey methods provide generally 5-10cm horizontal and vertical accuracies whereas satellite topography has accuracy of a few meters, so satellite topography must be controlled and adjusted from terrestrial ground control points which allow to reach an average 50cm absolute accuracy. This is good enough in desert areas with neither particular ground feature nor steep relief requiring complex design. Satellite acquisition has limitations: vegetation masking the ground, steep slopes and dense infrastructures. It is therefore necessary to combine conventional and satellite topography to meet engineering requirements. This is considered when defining the satellite and terrestrial survey areas. Beyond these limitations, this solution has strong advantages. Satellite grid resolution can be better (1-2m versus 5-10m for GNSS surveys). Acquisition and processing are faster (about 2 weeks versus a few weeks or months), and costs are from 10 to 100 times cheaper than conventional methods. No need for personnel and equipment on site, no management of logistics and permitting as well. Finally, it reduces safety hazards such as car accident, harsh weather, manual handling, etc. In addition, limiting the area to be surveyed with conventional equipment may avoid the need to mobilize Airborne photogrammetry or lidar systems usually operated by foreign companies. This limits complex Call for Tender, permitting management and give more opportunity to contract local companies. Satellite topography is widely used for preliminary studies, but the innovation here consists in an optimal combination of terrestrial and satellite datasets for engineering and construction purposes. This solution has however some limitations as it requires suitable conditions for satellite optical imagery acquisitions: no vegetation, limited cloud cover, smooth topography, and limited infrastructures. This is of interest basically in Middle east and North Africa.

Multi-ovary wheat (three pistil) is a unique germplasm for the seed production of hybrid wheat. The purpose of the present study was to transfer the multi-ovary trait to semi-dwarf plants to increase the production of grains in wheat crops. Therefore, tall, semi-dwarf, and dwarf plants were crossed with plants with the three-pistil trait. A three-pistil tall plant was used as the female parent, while tall (Synthetic hexaploid), semi-dwarf, and dwarf plants were used as male parents. F1 and F2 progenies with parents were planted in 2015-16 using RCBD. The outcome of the crosses showed that multi-ovary tall plants gave significant difference for all five traits (days to maturity, plant height, number of seeds per spike, grain weight per spike, and grain yield per unit area) in both generations. The greatest number of grains per spike and grain yield per unit area were obtained from the cross of three-pistil tall and dwarf parent (P1/P6) in the F1 and F2 generations. The cross also resulted in a significant reduction in height (96 cm). Further heterosis studies conducted with crosses between three-pistil tall and dwarf parent (P1/P6) showed the greatest heterosis and heterobeltiosis for the number of grains per spike (60.0 and 26.19%, respectively) and grain yield per m2 (27.68 and 2.85%, respectively). In the case of grain weight per spike, the heterosis value was also positive and significant (17.7). Meanwhile, for other traits, their values were negative for heterosis and heterobeltiosis. High numbers of grains and grain weight were found to be associated with positive heterobeltiosis and in turn the grain yield per m2, but plant height and maturity had negative affirmation with heterobeltiosis. Heterosis studies also indicated the dominant gene action for the three-pistil trait. Thus, the study clearly signified that grain yield can be increased by using the multi-ovary genotype with the semi-dwarf height. This new germplasm will be helpful for breeders to increase the production of wheat crops in the southern climate of Pakistan.

Acetyl-CoA carboxylase (ACC) is crucial for polyketides biosynthesis and acts as an essential metabolic checkpoint. It is also an attractive drug target against obesity, cancer, microbial infections, and diabetes. However, the lack of knowledge, particularly sequence-structure function relationship to narrate ligand-enzyme binding, has hindered the progress of ACC-specific therapeutics and unnatural "natural" polyketides. Structural characterization of such enzymes will boost the opportunity to understand the substrate binding, designing new inhibitors and information regarding the molecular rules which control the substrate specificity of ACCs. To understand the substrate specificity, we determined the crystal structure of AccB (Carboxyl-transferase, CT) from Streptomyces antibioticus with a resolution of 2.3 Å and molecular modeling approaches were employed to unveil the molecular mechanism of acetyl-CoA recognition and processing. The CT domain of S. antibioticus shares a similar structural organization with the previous structures and the two steps reaction was confirmed by enzymatic assay. Furthermore, to reveal the key hotspots required for the substrate recognition and processing, in silico mutagenesis validated only three key residues (V223, Q346, and Q514) that help in the fixation of the substrate. Moreover, we also presented atomic level knowledge on the mechanism of the substrate binding, which unveiled the terminal loop (500-514) function as an opening and closing switch and pushes the substrate inside the cavity for stable binding. A significant decline in the hydrogen bonding half-life was observed upon the alanine substitution. Consequently, the presented structural data highlighted the potential key interacting residues for substrate recognition and will also help to re-design ACCs active site for proficient substrate specificity to produce diverse polyketides.

This study investigates the efficacy of a prepared Ni/θ-Al2O3 catalyst during the pyrolytic conversion of Parachlorella kessleri HY-6 and compares the results with non-catalytic conversion. The catalyst was characterized by techniques such as Brunauer–Emmett–Teller (BET) for surface area, acidity, and X-ray powder diffraction (XRD). Isoconversional and combined kinetic methods were used to study the pyrolytic kinetics of the process. Ni/θ-Al2O3 was used at 10, 20, and 30% of the algal biomass. The addition of Ni/θ-Al2O3 facilitated the conversion by lowering the mean activation energy during pyrolysis. The catalytic effect was more pronounced at lower and higher conversions. The presence of the catalyst facilitated the pyrolysis as indicated by the lower value of activation energy and ∆H, and ∆G. Gases evolved during pyrolysis were qualitatively analyzed by FTIR to see the effect of catalyst on evolved gas composition during the pyrolysis process.

The current communication is designed by keeping in the mind high heat transfer capabilities of nanoliquids with the dispersion of diversified-natured nanoparticles in poorly conducting base liquids. Here, an amalgamation of metallic (Cu) and hybridization of metallic and non-metallic oxide (Cu-TiO2) nanoparticles to uplift thermophysical attributes of water is deliberated. The magnetically affected flow between rotating disks under the impact and permeability aspect is assumed. Empirical relations for effective dynamic viscosity, density, and heat capacitance to show mesmerizing features of obliged nanoparticles are also expressed. In addition, mathematical relations also depend on morphological factors like shape, size, and diameter of inducted nanoparticles. The mathematical formulation of the problem is conceded in the form of a system of ODEs after using similarity transformation on dimensional PDEs. Simulations of the complex coupled differential structure are solved by using a numerical approach by employing shooting and Runge–Kutta procedures jointly. The impact of flow concerning variables on associated distributions is revealed through tabular and graphical manner. Quantities of engineering interest associated with work like wall friction and thermal flux coefficients at walls of the disk are also calculated. It is deduced from an examination that the addition of metallic particles raises heat transfer more than non-metallic particles. A significant impression of magnetic field on shear stress is executed by hybrid nanoparticles along the surface of disks. In addition, elevation in Nusselt number and depreciation in skin friction coefficient is revealed against increasing magnitude of nanoparticle volume fraction. A positive trend in skin friction coefficient is manifested against the increasing magnitude of Reynold number. Multiple morphological abnormalities of the flagella (MMAF) can lead to male infertility due to impaired sperm motility and morphology. Calmodulin- and spoke-associated complex (CSC) are known for their roles in radial spoke (RS) assembly and ciliary motility in Chlamydomonas, while the role of cilia- and flagella-associated protein 61 (CFAP61), a mammalian ortholog of the CSC subunits, in humans is yet unknown. Here, we recruited three unrelated Pakistani families comprising of 11 infertile male patients diagnosed with MMAF.

Lubricants operate as antifriction media, preserving machine reliability, facilitating smooth operation, and reducing the likelihood of frequent breakdowns. The petroleum-based reserves are decreasing globally, leading to price increases and raising concerns about environmental degradation. The researchers are concentrating their efforts on developing and commercializing an environmentally friendly lubricant produced from renewable resources. Biolubricants derived from nonedible vegetable oils are environmentally favorable because of their non-toxicity, biodegradability, and close to net zero greenhouse gas emissions. The demand for bio lubricants in industry and other sectors is increasing due to their non-toxic, renewable, and environmentally friendly nature. Good lubrication, anti-corrosion, and high flammability are characteristic properties of vegetable oils due to their unique structure. This study presents several key properties of nonedible oils that are used to produce lubricants via the transesterification process. Bibliometric analysis is also performed, which provides us with a better understanding of previous studies related to the production of bio lubricants from the transesterification process. Only 371 published documents in the Scopus database were found to relate to the production of bio lubricants using the transesterification process. The published work was mostly dominated by research articles (286; 77.088%). Significant development can be seen in recent years, with the highest occurrence in 2021, reaching 68 publications accounting for 18.38% of the total documents. In the second step, (i) the authors with the most number of publications; (ii) journals with the most productions; (iii) most productive countries; and (iv) the authors' most frequently used keywords were evaluated. These results will provide a pathway for researchers interested in this field. Lastly, recommendation is made on research gaps to device possible strategies for its commercialization.

The co-torrefaction of several biomasses may be a viable solution in the study area, as it produces biofuels and addresses waste-treatment concerns. This review evaluates biomass through ultimate, proximate, and FTIR analyses, and the mechanism of the co-torrefaction process is observed for product quality with a synergistic effect. Furthermore, the parameters of co-torrefaction, including temperature, reaction time, mass yield, energy yield, and the composition of the H/C and O/C ratio of the co-torrefied materials, are similar to those for coal composition. Different reactor types, such as fixed-bed, fluidized-bed, microwave, and batch reactors, are used for co-torrefaction, in which biomass blends with optimized blend ratios. The co-torrefaction process increases the bio-solid yield and heating value, the capacity to adsorb carbon dioxide, and the renewable fuel used for gasification. One of the objectives of this study is to adopt a process that must be viable, green, and sustainable without generating pollution. For this reason, microwave co-torrefaction (MCT) has been used in many recent studies to transform waste and biomass materials into an alternative fuel using a microwave reactor.
