Immunotherapy using rat 7s antibodies against plasmacytoma 5563 in C3H mice has been measured carefully. Prior adsorption of antibodies with normal mouse tissues, and supplements of co-optable non-immunized macrophages and lymphocytes enhanced the scope of immunotherapy against established tumours. It is suggested that available co-optable lymphocytes and macrophages are imporant limiting factors in tumour immunotherapy.

A study of 14 personal patients and 16 others in the literature shows that (1) IgD myelomatosis often presents at a significantly younger age than other forms of myelomatosis, and (2) during life extraosseous tumour can be detected in about two-thirds of these patients.The IgD form represents 1.5% of myelomatosis and shows an increased incidence of osteolytic lesions, hypercalcaemia, and renal failure, together with heavy Bence Jones proteinuria (90% type L).Like only Bence Jones myelomatosis, the IgD form seems to behave clinically in a more vicious manner.

Five patients suffering from alpha chain disease are described. Clinically the patients presented with clubbing and the symptoms of malabsorption. There was a characteristic, predominantly plasma cell infiltrate of the wall of the small intestine. Spread of the plasmacytosis beyond the small intestine to bone marrow (1), peripheral blood (1), and probably the nasopharyngeal lymphoid tissue (1) is described. Fragments of the heavy chain of IgA (alpha chain) were found in serum (5), urine (3), jejunal fluid (2), and saliva (1). The jejunal biopsy of one patient was shown to synthesize free alpha chain in tissue culture. A new and simple immunoselection technique for the identification of free alpha chain is described. Marked clinical remissions were achieved in two patients treated with intermittent cytotoxic and steroid therapy, and in a third patient who received intermittent cytotoxic therapy and tetracycline.

Washed lymphocytes from immunized rats showed no reaction against mouse plasmacytoma 5563 in vivo or in vitro.Cell-free lymph from immunized rats was shown to be cytotoxic to the tumour cells in vitro. This effect we have shown to be solely in the 19s fraction after Sephadex G-200 separation.This 19s fraction conferred partial protection to tumour bearing mice when given shortly after transplantation, but had negligible effect against late well established tumour.
