402. Individual Coal Mine Methane Emissions Constrained by Eddy-Covariance Measurements: Low Bias and Missing Sources

Abstract

China’s Shanxi Province accounts for 12 % of global coal output, and therefore is responsible for a very large fraction of the total global methane (CH4) emissions, as well as being a large source of uncertainty due to the lack of in-situ and field measurements. This work introduces the first comprehensive attempt to compute the coal mine methane emissions (CMM) throughout Shanxi, using a mixture of bottom-up and top-down approaches. First, public and private data from 636 individual coal mines in Shanxi Province were analyzed following the IPCC Tier 2 approach, using three to five sets of observed emission factors, and rank information based on methods issued by the National Coal Mine Safety Administration and the National Energy Administration, to compile a range of bottom-up CMM on a mine-by-mine basis. An eddy-covariance tower is set up near the output flue of a well-characterized high rank coal mine in Changzhi, and used to produce an average observed CH4 flux over two two-month long periods (Winter 2021 and Autumn 2022). The observed half-hourly CH4 flux variability is found to be roughly stable over the entire observed time, and is subsequently used to produce a set of scaling factors (RATIO correction) to updating the preliminary bottom-up coal mine methane emissions to account for both bias and high-frequency temporal variabiliy. The resulting emissions dataset have been compared against commonly used global CMM datasets including EDGAR and GFEI v2, and yield three unique scientific conclusions. First, their total CH4 emissions over Shanxi lie in between this work’s 50th percentile and 70th percentile range, meaning they are slightly high. Second, both datasets have a very large amount of emissions which occur where there are no coal mines and no CH4 emitting industry, indicating that there are significant spatial disparities, with the overlapped portion of CMM emissions where mines exist consistently close to the 30th percentile of this work’s emissions, meaning they underestimate CMM in general on a mine-by-mine basis. Third, some of the mines have average emissions values which are more than the 90th percentile of the computed mine-by-mine emissions, while many are far below the 10th percentile, showing that there is a significant issue with the sampling not capturing the observed temporal variability. It is hoped that this mine-by-mine and high frequency approximation of CMM emissions can improve both top-down observation campaigns as well as provide quantitative support and identification of mitigation opportunities.

Abstract

Background: High-level nursing talents play an important role in solving nursing scientific problems and leading nursing innovation, so the construction of high-level nursing talents has become a necessary guarantee for the health strategy in the new era. Managers have high expectations for master’s degree in nursing, hoping that they will develop in all aspects of research, teaching, and professionalism, etc. However, the high expectations of managers have led to unclear job orientation, high clinical and scientific research pressure, and low recognition of nurses’ sense of professional value from the outside world, which have led to the lowering of master’s degree of professional identity and the tendency to leave the profession, which is not conducive to the stabilization of the nursing workforce. Purpose: To explore the influence of adversity quotient, perceived humanistic care of nurse leaders on professional identity of nursing masters, and to clarify the relationship model between the three. Methods: A convenience sampling method was used to conduct an online questionnaire survey on 160 master’s degree nursing students in Ningbo and Shanghai from July to September 2022 using the general information questionnaire, adversity quotient, career identity and perceived humanistic care of nurse leaders scale. Results: Adversity quotient, occupational identity and perceived humanistic care of nurse leaders were all at a moderate level. Adversity quotient (r = 0.430, P < 0.01), perceived humanistic care of nurse leaders (r = 0.443, P < 0.01) and occupational identity were positively correlated, and perceived humanistic care of nurse leaders could affect occupational identity through the mediating effect of adversity quotient, with the mediating effect accounting for 31.8%. Conclusion: Nursing managers should create a good organizational atmosphere, build a training system for adversity quotient courses, conduct regular assessment and humanistic care to improve the ability of nursing masters to face adversity and enhance the awareness of their own professional values.

The aim of this study was to assess the cost-effectiveness of TIO/OLO FDC in patients with moderate to very severe COPD in China.Methods: A Markov model was developed to estimate the cost-effectiveness of TIO/OLO FDC versus TIO in the treatment of COPD from Chinese health system perspective. Four health states were based on 2021 Global Initiative for Chronic Obstructive Lung Disease (GOLD 2021), which included moderate (GOLD II, 50% ≤ FEV1 ≤ 80% of predicted), severe (GOLD III, 30% ≤ FEV1 ≤ 50% of predicted) and very severe (GOLD IV, FEV1 > 30% of predicted) COPD and death. The model simulated in cycles yearly. The indicators of total costs, number of COPD exacerbations, life years (LYs) and quality-adjusted life-years (QALYs) were used as the model output. Costs and outcomes were discounted at a 5% annual rate. A cost-effectiveness analysis was conducted over a 10-year time horizon. The threshold of incremental total cost per unit effectiveness gained (ICER) was 1.5 times of GDP per capita. Uncertainty was assessed by one-way and probabilistic sensitivity analysis.Results: TIO/OLO was 0.007 QALYs more than TIO but 0.012 LYs lower, which increased the total cost by &dollar;2268.17 per patient, but the total exacerbations number was less. Abstract

The potato's most devastating disease is late blight, which is caused by Phytophthora infestans. Whereas various resistance (R) genes are known, most are typically defeated by this fast-evolving oomycete pathogen. However, the broad-spectrum and durable R8 is a vital gene resource for potato resistance breeding. To support an educated deployment of R8, we embarked on a study on the corresponding avirulence gene Avr8. We overexpressed Avr8 by transient and stable transformation, and found that Avr8 promotes colonization of P. infestans in Nicotiana benthamiana and potato, respectively. A yeast-two-hybrid (Y2H) screen showed that AVR8 interacts with a desumoylating isopeptidase (StDeSI2) of potato. We overexpressed DeSI2 and found that DeSI2 positively regulates resistance to P. infestans, while silencing StDeSI2 downregulated the expression of a set of defense-related genes. By using a specific proteasome inhibitor, we found that AVR8 destabilized StDeSI2 through the 26S proteasome and attenuated early PTI responses. Altogether, these results indicate that AVR8 manipulates desumoylation, which is a new strategy that adds to the plethora of mechanisms that Phytophthora exploits to modulate host immunity, and StDeSI2 provides a new target for durable resistance breeding against P. infestans in potato.

Abstract

Background: Studies have confirmed that Infectious bovine rhinotracheitis virus (IBRV) infection induces mitochondrial damage. MicroRNAs (miRNAs) are a class of noncoding RNA molecules, which are involved in various biological processes and pathological changes associated with mitochondrial damage. It is currently unclear whether miRNAs participate in IBRV-induced mitochondrial damage in Madin-Darby bovine kidney (MDBK) cells. Results: In the present study, we used high-throughput sequencing technology, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis to screen for mitochondria-related miRNAs and messenger RNAs (mRNAs). In total, 279 differentially expressed miRNAs and 832 differentially expressed mRNAs were identified in 6 hours (IBRV1) versus 24 hours (IBRV2) after IBRV infection in MDBK cells. GO and KEGG enrichment analysis revealed that 42 differentially expressed mRNAs and 348 target genes of differentially expressed miRNAs were correlated with mitochondrial damage, and the miRNA-mitochondria-related target genes regulatory network was constructed to elucidate their potential regulatory relationships. Among the 10 differentially expressed miRNAs, 8 showed expression patterns consistent with the high-throughput sequencing results. Functional validation results showed that overexpression of miR-10a and miR-182 aggravated mitochondrial damage, while inhibition of miR-10a and miR-182 alleviated mitochondrial damage. Conclusions: This study not only revealed the expression changes of miRNAs and mRNAs in IBRV-infected MDBK cells, but also revealed possible biological regulatory relationship between them. Abstract

Glomerulonephritis (GN) is the most common cause of end-stage renal failure worldwide; in most cases, it cannot be cured and can only delay the progression of the disease. At present, the main treatment methods include symptomatic therapy, immunosuppressive therapy, and renal replacement therapy. However, effective treatment of GN is hindered by issues such as steroid resistance, serious side effects, low bioavailability, and lack of precise targeting. With the widespread application of nanoparticles in medical treatment, novel methods have emerged for the treatment of kidney diseases. Targeted transportation of drugs, nucleic acids, and other substances to kidney tissues and even kidney cells through nanodrug delivery systems can reduce the systemic effects and adverse reactions of drugs and improve treatment effectiveness. The high specificity of nanoparticles enables them to bind to ion channels and block or enhance channel gating, thus improving inflammation. This review briefly introduces the characteristics of GN, describes the treatment status of GN, systematically summarizes the research achievements of nanoparticles in the treatment of primary GN, diabetic nephropathy and lupus nephritis, analyzes recent therapeutic developments, and outlines promising research directions, such as gas signaling molecule nanodrug delivery systems and ultrasmall nanoparticles. Abstract

Intramuscular fat (IMF) is a crucial determinant of meat quality and is influenced by various regulatory factors. Despite the growing recognition of the important role of long noncoding RNAs (lncRNAs) in IMF deposition, the mechanisms underlying buffalo IMF deposition remain poorly understood. In this study, we identified and characterized a lncRNA, lncFABP4, which is transcribed from the antisense strand of fatty acid‐binding protein 4 (FABP4). lncFABP4 inhibited cell proliferation in buffalo intramuscular preadipocytes. Moreover, lncFABP4 significantly increased intramuscular preadipocyte differentiation, as indicated by an increase in the expression of the adipogenic markers peroxisome proliferator‐activated receptor gamma (PPARG), CCAAT enhancer binding protein alpha (C/EBPα), and FABP4. Mechanistically, lncFABP4 was found to have the potential to regulate downstream gene expression by participating in protein–protein interaction pathways. Abstract

To effectively suppress the longitudinal vibration of the car under the conditions of high-speed operation and emergency braking, and improve the ride comfort of the car system, this paper proposes an adaptive fuzzy inverse optimal output feedback control strategy. Firstly, the dynamic model of the high-speed elevator system is established and the nonlinear dynamic model is approximated by the fuzzy logic system, and the auxiliary system model is established. Fuzzy state observer is designed to estimate the unmeasurable state. Furthermore, an adaptive inverse optimal output feedback controller based on fuzzy observer is designed by using adaptive backstepping technology and inverse optimal control principle. The stability analysis shows that the proposed adaptive fuzzy inverse optimal output feedback control strategy not only ensures the stability of the car attitude of high-speed elevator but also realizes the inverse optimization of the target cost function. Finally, the acceleration time–frequency response analysis of the two typical stages of high-speed elevator uniform running and emergency braking is carried out, and the numerical results are compared with the linear quadratic controller optimized by stepping quantum genetic algorithm (GA-LQR) and controller based on the state-dependent Ricatti equation (SDRE).
