Abstract

International audience; The weak ferroelectric contribution to the polarization of antiferroelectric lead zirconate (PZO) thin films has been investigated using PUND (Positive Up Negative Down) pulse measurements and hysterons decomposition by First Order Reversal Curves (FORC) technique. The PUND allows decomposing the measured current, obtained from the polarization-electric field loop measurements, into a switching and a non-switching contribution. We show that the weak ferroelectric phase is enhanced when a large electric field has been previously applied to the material, in order to switch from the antiferroelectric to the ferroelectric phase. Using the PUND measurement at fields below the antiferroelectric to ferroelectric phase transition, the polarization loop corresponding only to the ferroelectric switching contribution has been determined revealing that this contribution to the overall polarization is small. FORC measurements, however, indicate that the ferroelectric phase is present at different fields. At low fields, a quite homogeneous distribution of hysterons exists and at large field, a high concentration of hysterons at a field near to the antiferroelectric to ferroelectric phase transition can be seen. Moreover, when changing the delay between pulses of the PUND and the FORC measurements, we show that this weak ferroelectricity contribution is metastable and decreases with time.

402. Global Change Factors Influence Plant-Epichloë Associations

Abstract

There is an increasing interest in determining the influence of global change on plant–microorganism interactions. We review the results of experiments that evaluated the effects of the global change factors carbon dioxide, ozone, temperature, drought, flooding, and salinity on plant symbioses with beneficial Epichloë endophytes. The factors affected the performance of both plants and endophytes as well as the frequency of plants symbiotic with the fungus. Elevated carbon dioxide levels and low temperatures differentially influenced the growth of plants and endophytes, which could compromise the symbioses. Furthermore, we summarise the plant stage in which the effects of the factors were quantified (vegetative, reproductive, or progeny). The factors ozone and drought were studied at all plant stages, but flooding and carbon dioxide were studied in just a few of them. While only studied in response to ozone and drought, evidence showed that the effects of these factors on symbiotic plants persisted trans-generationally. We also identified the putative mechanisms that would explain the effects of the factors on plant–endophyte associations. These mechanisms included the increased contents of reactive oxygen species and defence-related phytohormones, reduced photosynthesis, and altered levels of plant primary metabolites. Finally, we describe the counteracting mechanisms by which endophytes would mitigate the detrimental effects of the factors on plants. In presence of the factors, endophytes increased the contents of antioxidants, reduced the levels of defence-related phytohormones, and enhanced the plant uptake of nutrients and photosynthesis levels. Knowledge gaps regarding the effects of global change on plant–endophyte associations were identified and discussed.

Abstract

Gpr21 KO mice generated with Gpr21 KO ES cells obtained from Deltagen showed improved glucose tolerance and insulin sensitivity when fed a high fat diet. Further mRNA expression analysis revealed changes in Rabgap1 levels and raised the possibility that Rabgap1 gene may have been modified. To assess this hypothesis a new Gpr21 KO mouse line using TALENS technology was generated. Gpr21 gene deletion was confirmed by PCR and Gpr21 and Rabgap1 mRNA expression levels were determined by RT-PCR. The newly generated Gpr21 KO mice when fed a normal or high fat diet chow did not maintain their improved metabolic phenotype. In conclusion, Rabgap1 disturbance mRNA expression levels may have contributed to the phenotype of the originally designed Gpr21 KO mice.

Abstract

Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural clines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale.

Abstract

Im Rahmen der ISAR (“rISk strAtification in end stage Renal disease”) Studie wurde die Assoziation der EKG-LVH-Parameter Sokolow-Lyon, Cornell und Peguero-Lo Presti Index mit der kardiovaskulären Mortalität und Gesamtmortalität bei 308 Hämodialysepatienten untersucht. Sowohl der Cornell als auch der Peguero-Lo Presti Index, nicht aber der Sokolow-Lyon Index, waren auch nach Adjustierung auf verschiedene Risikomodelle signifikant mit der kardiovaskulären Mortalität assoziiert., As part of the ISAR (rISk strAtification in end stage Renal disease) study the association of the ECG-LVH criteria Sokolow-Lyon, Cornell and Peguero-Lo Presti Index with cardiovascular and all-cause mortality in 308 hemodialysis patients was investigated. Both the Cornell and Peguero-Lo Presti Index, but not the Sokolow-Lyon Index, were significantly associated with cardiovascular mortality even after adjustement for different risk models.

Abstract

Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural dines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale.

Abstract

ObjectiveA major challenge in multiple sclerosis (MS) research is the understanding of silent progression and Progressive MS. Using a novel method to accurately capture upper cervical cord area from legacy brain MRI scans we aimed to study the role of spinal cord and brain atrophy for silent progression and conversion to secondary progressive disease (SPMS).MethodsFrom a single-center observational study, all RRMS (n = 360) and SPMS (n = 47) patients and 80 matched controls were evaluated. RRMS patient subsets who converted to SPMS (n = 54) or silently progressed (n = 159), respectively, during the 12-year observation period were compared to clinically matched RRMS patients remaining RRMS (n = 54) or stable (n = 147), respectively. From brain MRI, we assessed the value of brain and spinal cord measures to predict silent progression and SPMS conversion.ResultsPatients who developed SPMS showed faster cord atrophy rates (-2.19%/yr) at least 4 years before conversion compared to their RRMS matches (-0.88%/yr, p < 0.001). Spinal cord atrophy rates decelerated after conversion (-1.63%/yr, p = 0.010) towards those of SPMS patients from study entry (-1.04%). Each 1% faster spinal cord atrophy rate was associated with 69% (p < 0.0001) and 53% (p < 0.0001) shorter time to silent progression and SPMS conversion, respectively.InterpretationSilent progression and conversion to secondary progressive disease are predominantly related to cervical cord atrophy. This atrophy is often present from the earliest disease stages and predicts the speed of silent progression and conversion to Progressive MS. Abstract

Tobacco-specific nitrosamines (TSNAs) are emitted during smoking and form indoors by nitrosation of nicotine. Two of them, N'-nitrosonornicotine (NNN) and 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), are human carcinogens with No Significant Risk Levels (NSRLs) of 500 and 14 ng day-1, respectively. Another TSNA, 4-(methylnitrosamino)-4-(3-pyridyl) butanal (NNA), shows genotoxic and mutagenic activity in vitro. Here, we present additional evidence of genotoxicity of NNA, an assessment of TSNA dermal uptake, and predicted exposure risks through different pathways. Dermal uptake was investigated by evaluating the penetration of NNK and nicotine through mice skin. Comparable mouse urine metabolite profiles suggested that both compounds were absorbed and metabolized via similar mechanisms. We then investigated the effects of skin constituents on the reaction of adsorbed nicotine with nitrous acid (epidermal chemistry). Higher TSNA concentrations were formed on cellulose and cotton substrates that were precoated with human skin oils and sweat compared to clean substrates. These results were combined with reported air, dust, and surface concentrations to assess NNK intake. Five different exposure pathways exceeded the NSRL under realistic scenarios, including inhalation, dust ingestion, direct dermal contact, gas-to-skin deposition, and epidermal nitrosation of nicotine. These results illustrate potential long-term health risks for nonsmokers in homes contaminated with thirdhand tobacco smoke.

Abstract

Objectives: Cutaneous leishmaniasis (CL) in Asia, Northern, and Sub-Saharan Africa is mainly caused by Leishmania major and Leishmania tropica. We describe and evaluate the treatment outcome of CL among travelers and migrants in Europe.Methods: We conducted a retrospective study of parasitological confirmed CL cases caused by L. major and L. tropica during 2013-2019 in Europe. Data were collected from medical records and databases within the LeishMan network.Results: Of 206 included cases of CL, 75 were identified as L. major and 131 as L. tropica. Of patients with L. tropica infection, 80% were migrants, whereas 53% of patients with L. major infection had been visiting friends and relatives. Among patients with L. tropica, 48% were younger than 15 years. Pentavalent antimony cured 73% ( L. major ) and 78% ( L. tropica) of patients. The cure rate for intralesional administration was 86% and 67% for systemic, on L. tropica. Liposomal amphotericin B had a cure rate of 44-63%. Conclusion: L. major infections were mostly found in individuals visiting friends and relatives, whereas L. tropica were mainly identified in migrants. No patients with L. major relapsed. Pentavalent antimony, liposomal amphotericin B, and cryotherapy had cure rates in accordance with previous studies.(c) 2022 The Author(s). Abstract

Background Invasive plants may displace native species. This is the case of Poa annua, the only non-native plant species successfully established in Maritime Antarctica. Nonetheless, it is uncertain which factors drive the competitive success of P. annua in the harsh environmental conditions of the region. The ability of this plant species to establish novel mutualistic interactions with resident soil fungi may be crucial for its invasiveness. Such ability may be linked to the vertical transmission of the fungal endophytes via seeds. Aims We undertook a study to assess the role of seed fungal endophytes as promoters of the establishment and invasion of Poa annua in Maritime Antarctica. Methods We explored the composition and diversity of fungal communities associated with different P. annua tissues (seeds, leaves and roots) and the soil. We also measured parameters including germination rate, above-ground biomass, reproductive structures, and the survival of invasive P. annua as well as of the native Colobanthus quitensis and Deschampsia antarctica grown from seeds with and without endophytes. Furthermore, we conducted inter- and intraspecific competition experiments among native and invasive plants, where chemically-mediated plant-to-plant interference (allelopathy) and plant growth rate were measured to calculate a relative competition index. Results We found that fungal endophyte taxa associated with P. annua tissues were very different from those in the soil. Fungal endophytes in P. annua differed among seed, root and shoot tissues, which suggests low transmission among different organs. The removal of endophytes from P. annua seeds was associated with reduced seed germination, plant growth and survivorship, while the competitive ability of P. annua (assessed by accumulated biomass) relative to native species, as well as levels of allelochemicals in soils, were higher in the presence of seed fungal endophytes. Abstract

Seeds commonly harbour diverse bacterial communities that can enhance the fitness of future plants. The bacterial microbiota associated with mother plant’s foliar tissues is one of the main sources of bacteria for seeds. Therefore, any ecological factor influencing the mother plant’s microbiota may also affect the diversity of the seed’s bacterial community. Grasses form associations with beneficial vertically transmitted fungal endophytes of genus Epichloë. The interaction of plants with Epichloë endophytes and insect herbivores can influence the plant foliar microbiota. However, it is unknown whether these interactions (alone or in concert) can affect the assembly of bacterial communities in the produced seed. We subjected Lolium multiflorum plants with and without its common endophyte Epichloë occultans (E+, E-, respectively) to an herbivory treatment with Rhopalosiphum padi aphids and assessed the diversity and composition of the bacterial communities in the produced seed. The presence of Epichloë endophytes influenced the seed bacterial microbiota by increasing the diversity and affecting the composition of the communities. The relative abundances of the bacterial taxa were more similarly distributed in communities associated with E+ than E- seeds with the latter being dominated by just a few bacterial groups. Contrary to our expectations, seed bacterial communities were not affected by the aphid herbivory experienced by mother plants. We speculate that the enhanced seed/seedling performance documented for Epichloë-host associations may be explained, at least in part, by the Epichloë-mediated increment in the seed-bacterial diversity, and that this phenomenon may be applicable to other plant-endophyte associations.

Das Jahr 2021 hat in Deutschland neuen Schwung in die Bemühungen zum Klimaschutz gebracht. Die Novelle des Klimaschutzgesetzes schreibt eine Reduktion der Treibhausgasemissionen um 65 % bis zum Jahr 2030 gegenüber 1990 fest. Maßgeblich dazu beitragen soll ein massiver Ausbau der Stromerzeugung aus erneuerbaren Energien. Nach Verschärfung der Klimaschutzmaßnahmen stellt sich die Frage, ob das bisherige Strommarktdesign und die damit einhergehenden Vergütungsregeln für die Stromerzeugung auch bei einem von erneuerbaren Energien dominierten Strommarkt weiterhin geeignet sind, um die im Energiewirtschaftsgesetz festgeschriebenen Ziele der Versorgungssicherheit und Preisgünstigkeit bestmöglich zu erreichen. The year 2021 has brought new momentum to climate protection efforts in Germany. The amendment to the Climate Protection Act stipulates a 65 % reduction in greenhouse gas emissions by 2030 compared to 1990. A major contribution to this will come from a massive expansion of electricity generation from renewable energies. Following the tightening of the climate protection measures, the question arises as to whether the current electricity market design and the associated remuneration rules for electricity generation are still suitable for achieving the goals of security of supply and low prices stipulated in the Energy Industry Act, even with an electricity market dominated by renewable energies.

Les condensateurs sont dimensionnés pour fonctionner jusqu'à 65 GHz, en offrant des accordabilités de l'ordre de 40% sous des tensions de polarisation de 100 V et présentant des pertes relativement faibles. Abstract

Background Surveillance of human leishmaniasis in Europe is mostly limited to country-specific information from autochthonous infections in the southern part. As at the end of 2021, no integrated analysis has been performed for cases seen across centres in different European countries. Aim To provide a broad perspective on autochthonous and imported leishmaniasis cases in endemic and non-endemic countries in Europe. Methods We retrospectively collected records from cutaneous, mucosal and visceral leishmaniasis cases diagnosed in 15 centres between 2014 and 2019. Centres were located in 11 countries: Belgium, France, Germany, Italy, the Netherlands, Norway, Portugal, Spain, Sweden, Switzerland and the United Kingdom. Data on country of infection, reason for travelling, infecting species, age and sex were analysed. Results We obtained diagnostic files from 1,142 cases, of which 76%, 21% and 3% had cutaneous, visceral, and mucosal disease, respectively. Of these, 68% were men, and 32% women, with the median age of 37 years (range: 0–90) at diagnosis. Visceral leishmaniasis was mainly acquired in Europe (88%; 167/190), while cutaneous leishmaniasis was primarily imported from outside Europe (77%; 575/749). Sixty-two percent of cutaneous leishmaniasis cases from outside Europe were from the Old World, and 38% from the New World. Geographic species distribution largely confirmed known epidemiology, with notable exceptions. Conclusions Our study confirms previous reports regarding geographic origin, species, and traveller subgroups importing leishmaniasis into Europe. We demonstrate the importance of pooling species typing data from many centres, even from areas where the aetiology is presumably known, to monitor changing epidemiology.

Abstract

International audience; Domains of antiferroelectric PbZrO3 have been studied. The (111) PbZrO3 has lower field transitions due to the reduction of the angle between the ferroelectric polar axis and the applied electric field. The study of the permittivity reveals also that the (111) PbZrO3 has a higher correlation of dipoles (|Shf| = 0.007) due to the energy gain associated with their orientation. Therefore, the (111) crystallographic orientation is a better choice for obtaining easily switching domain. The dielectric response of the domain walls in the (100) and (111) PbZrO3 are identical because they have the same environment (same grain size and similar defects) and interact in the same way. Only the domain wall density is higher in the (111) PbZrO3 due to its lower crystallographic orientation factor. Different crystallographic directions have more obstacles due to the inhomogeneity of the crystallization and consequently has more nucleation site for domain walls.

International audience; OBJECTIVES: Cutaneous leishmaniasis (CL) in Asia, Northern, and Sub-Saharan Africa is mainly caused by Leishmania major and Leishmania tropica. We describe and evaluate the treatment outcome of CL among travelers and migrants in Europe.METHODS: We conducted a retrospective study of parasitological confirmed CL cases caused by L. major and L. tropica during 2013-2019 in Europe. Data were collected from medical records and databases within the LeishMan network.RESULTS: Of 206 included cases of CL, 75 were identified as L. major and 131 as L. tropica. Of patients with L. tropica infection, 80% were migrants, whereas 53% of patients with L. major infection had been visiting friends and relatives. Among patients with L. tropica, 48% were younger than 15 years. Pentavalent antimony cured 73% (L. major) and 78% (L. tropica) of patients. The cure rate for intralesional administration was 86% and 67% for systemic, on L. tropica. Liposomal amphotericin B had a cure rate of 44-63%. CONCLUSION: L. major infections were mostly found in individuals visiting friends and relatives, whereas L. tropica were mainly identified in migrants. No patients with L. major relapsed. Pentavalent antimony, liposomal amphotericin B, and cryotherapy had cure rates in accordance with previous studies.

443. Seed fungal endophytes promote the establishment of invasive Poa annua in maritime Antarctica

Abstract

Invasive plants may displace native species. This is the case for Poa annua, the only non-native plant species successfully established in Maritime Antarctica. Nonetheless, it is uncertain which factors drive the competitive success of P. annua in the harsh environmental conditions of the region. The ability of this plant species to establish novel mutualistic interactions with resident soil fungi may be crucial for its invasiveness. Such ability may be linked to the vertical transmission of fungal endophytes via seeds. We undertook a study to assess the role of seed fungal endophytes as promoters of the establishment and invasion of Poa annua in Maritime Antarctica. We explored the composition and diversity of fungal communities associated with different P. annua tissues (seeds, leaves and roots) and the soil. We also measured parameters including germination rate, above-ground biomass, reproductive structures, and the survival of invasive P. annua as well as of the native Colobanthus quitensis and Deschampsia antarctica grown from seeds with and without endophytes. Furthermore, we conducted inter- and intraspecific competition experiments among native and invasive plants, where chemically mediated plant-to-plant interference (allelopathy) and plant growth rate were measured to calculate a relative competition index. We found that fungal endophyte taxa associated with P. annua tissues were very different from those in the soil. Fungal endophytes in P. annua differed among seed, root and shoot tissues, which suggests low transmission among different organs. The removal of endophytes from P. annua seeds was associated with reduced seed germination, plant growth and survivorship, while the competitive ability of P. annua (assessed by accumulated biomass) relative to native species, as well as levels of allelochemicals in soils, were higher in the presence of seed fungal endophytes. Our results suggest that fungal endophytes, maternally inherited through seeds, improve host fitness and may contribute to the invasive success of P. annua in Antarctica.

449. Geographic Variation in Festuca rubra L. Ploidy Levels and Systemic Fungal Endophyte Frequencies.

Abstract

Polyploidy and symbiotic Epichloë fungal endophytes are common and heritable characteristics that can facilitate environmental range expansion in grasses. Here we examined geographic patterns of polyploidy and the frequency of fungal endophyte colonized plants in 29 Festuca rubra L. populations from eight geographic sites across latitudes from Spain to northernmost Finland and Greenland. Ploidy seemed to be positively and negatively correlated with latitude and productivity, respectively. However, the correlations were nonlinear; 84% of the plants were hexaploids (2n = 6x = 42), and the positive correlation between ploidy level and latitude is the result of only four populations skewing the data. In the southernmost end of the gradient 86% of the plants were tetraploids (2n = 4x = 28), whereas in the northernmost end of the gradient one population had only octoploid plants (2n = 8x = 56). Endophytes were detected in 22 out of the 29 populations. Endophyte frequencies varied among geographic sites, and populations and habitats within geographic sites irrespective of ploidy, latitude or productivity. The highest overall endophyte frequencies were found in the southernmost end of the gradient, Spain, where 69% of plants harbored endophytes. In northern Finland, endophytes were detected in 30% of grasses but endophyte frequencies varied among populations from 0% to 75%, being higher in meadows compared to riverbanks. The endophytes were detected in 36%, 30% and 27% of the plants in Faroe Islands, Iceland and Switzerland, respectively. Practically all examined plants collected from southern Finland and Greenland were endophyte-free, whereas in other geographic sites endophyte frequencies were highly variable among populations. Common to all populations with high endophyte frequencies is heavy vertebrate grazing. We propose that the detected endophyte frequencies and ploidy levels mirror past distribution history of F. rubra after the last glaciation period, and local adaptations to past or prevailing selection forces such as vertebrate grazing.

Abstract

Introduction We describe a protocol for FIRE CORAL, an observational cohort study that examines the recovery from COVID-19 disease following acute hospitalization with an emphasis on functional, imaging, and respiratory evaluation. Methods and analysis FIRE CORAL is a multicenter prospective cohort study of participants recovering from COVID-19 disease with in-person follow-up for functional and pulmonary phenotyping conducted by the National Heart, Lung and Blood Institute (NHLBI) Prevention and Early Treatment of Acute Lung Injury (PETAL) Network. FIRE CORAL will include a subset of participants enrolled in Biology and Longitudinal Epidemiology of PETAL COVID-19 Observational Study (BLUE CORAL), an NHLBI-funded prospective cohort study describing the clinical characteristics, treatments, biology, and outcomes of hospitalized patients with COVID-19 across the PETAL Network. FIRE CORAL consists of a battery of in-person assessments objectively measuring pulmonary function, abnormalities on lung imaging, physical functional status, and biospecimen analyses. Participants will attend and perform initial in-person testing at 3 to 9 months after hospitalization. The primary objective of the study is to determine the feasibility of longitudinal assessments investigating multiple domains of recovery from COVID-19. Secondarily, we will perform descriptive statistics, including the prevalence and characterization of abnormalities on pulmonary function, chest imaging, and functional status. We will also identify potential clinical and biologic factors that predict recovery or the occurrence of persistent impairment of pulmonary function, chest imaging, and functional status. Ethics and dissemination FIRE CORAL is approved via the Vanderbilt University central institutional review board (IRB) and via reliance agreement with the site IRBs. Results will be disseminated via the writing group for the protocol committee and reviewed by the PETAL Network publications committee prior to publication. Data obtained via the study will subsequently be made publicly available via NHLBI’s biorepository.
