Sensitive and convenient methodologies for detection of influenza viruses are essential for further disease control. Loop-mediated isothermal amplification (LAMP) is the most commonly used method of nucleic acid isothermal amplification. However, with regard to multiplex LAMP, differentiating the ladder-like LAMP products derived from multiple targets is still challenging today. The requirement of specialized instruments has further hindered the on-site application of multiplex LAMP. We have developed an integrated assay coupling multiplex reverse transcription LAMP with cascade invasive reaction using nanoparticles (mRT-LAMP-CIRN) as a sensor for the detection of three subtypes of influenza viruses: A/H1N1pdm09, A/H3 and influenzaB. The analytic sensitivities of the mRT-LAMP-CIRN assay were 101 copies of RNA for both A/H1N1pdm09 and A/H3, and 102 copies of RNA for influenza B. This assay demonstrated highly specific detection of target viruses and could differentiatethem from other genetically or clinically related viruses. Clinical specimen analysis showed the mRT-LAMP-CIRN assay had an overall sensitivity and specificity of 98.3% and 100%, respectively. In summary, the mRT-LAMP-CIRN assay is highly sensitive and specific, and can be used as a cost-saving and instrument-free method for the detection of influenza viruses, especially for on-site use. Comets hold the key to the understanding of our solar system, its formation and its evolution, and to the fundamental plasma processes at work both in it and beyond it. A comet nucleus emits gas as it is heated by the sunlight. The gas forms the coma, where it is ionised, becomes a plasma and eventually interacts with the solar wind. Besides these neutral and ionised gases, the coma also contains dust grains, released from the comet nucleus. As a cometary atmosphere develops when the comet travels through the solar system, large-scale structures, such as the plasma boundaries, develop and disappear, while at planets such large-scale structures are only accessible in their fully grown, quasi-steady state. In situ measurements at comets enable us to learn both how such large-scale structures are formed or reformed and how small-scale processes in the plasma affect the formation and properties of these large scale structures. Furthermore, a comet goes through a wide range of parameter regimes during its life cycle, where either collisional processes, involving neutrals and charged particles, or collisionless processes are at play, and might even compete in complicated transitional regimes. Thus a comet presents a unique opportunity to study this parameter space, from an asteroid-like to a Mars- and Venus-like interaction. Fast flybys of comets have made many new discoveries, setting the stage for a multi-spacecraft mission to accompany a comet on its journey through the solar system. This white paper reviews the present-day knowledge of cometary plasmas, discusses the many questions that remain unanswered, and outlines a multi-spacecraft ESA mission to accompany a comet that will answer these questions by combining both multi-spacecraft observations and a rendezvous mission, and at the same time advance our understanding of fundamental plasma physics and its role in planetary systems.

© 2019 Elsevier B.V. Assessing and comparing global river ecosystem health in an objective and quantitative way remains a major challenge. In this study the widely-used semi-quantitative methods Rapid Biological assessment Protocols (RBPs) was used to determine the health of rivers. The findings were then compared to the results derived from our new UAV (Unmanned aerial vehicles) orthophotographic imagery method. This method quantitatively and objectively assesses river ecosystem health. As a comparison, our method was used to quantitatively measure distance and areas of a range of hydrological and biological attributes thus improving the accuracy of distance- and area-related indices, consequently avoiding subjective errors in these estimations that is fraught in methods like the RBPs. To strengthen the objectivity of the assessment the weights of these indices were objectively determined using the entropy weighting method. This new method was then tested using 9551 UAV orthophotographs taken over six field campaigns. It performed satisfactorily, showing that in our study area the health status of mountain rivers was the best with the highest score of 0.94 out of 1.0. Temporally, the health of the river was better in summer (0.65) compared with that in autumn (0.40). Changes in river ecosystem health were driven by variations in biology and water quality. In contrast the outputs of RBPs, especially in relation to distance and area indices, had ~ 20% uncertainty due to visual errors and subjectivity in estimations by observers. The UAV orthophotographic imaging method proposed in this study can improve the ability to compare the health of rivers across different periods and regions throughout the globe.

Cancer cells can switch between signaling pathways to regulate growth under different conditions. In the tumor microenvironment, this likely helps them evade therapies that target specific pathways. We must identify all possible states and utilize them in drug screening programs. One such state is characterized by expression of the transcription factor Hairy and Enhancer of Split 3 (HES3) and sensitivity to HES3 knockdown, and it can be modeled in vitro. Here, we cultured 3 primary human brain cancer cell lines under 3 different culture conditions that maintain low, medium, and high HES3 expression and characterized gene regulation and mechanical phenotype in these states. We assessed gene expression regulation following HES3 knockdown in the HES3-high conditions. We then employed a commonly used human brain tumor cell line to screen Food and Drug Administration (FDA)-approved compounds that specifically target the HES3-high state. We report that cells from multiple patients behave similarly when placed under distinct culture conditions. We identified 37 FDA-approved compounds that specifically kill cancer cells in the high-HES3–expression conditions. Our work reveals a novel signaling state in cancer, biomarkers, a strategy to identify treatments against it, and a set of putative drugs for potential repurposing.—Poser, S. In this paper, we propose a novel framework for performance optimization in Internet of Things (IoT)-based next-generation wireless sensor networks. In particular, a computationally-convenient system is presented to combat two major research problems in sensor networks. First is the conventionally-tackled resource optimization problem which triggers the drainage of battery at a faster rate within a network. Such drainage promotes inefficient resource usage thereby causing sudden death of the network. The second main bottleneck for such networks is the data degradation. This is because the nodes in such networks communicate via a wireless channel, where the inevitable presence of noise corrupts the data making it unsuitable for practical applications. Therefore, we present a layer-adaptive method via 3-tier communication mechanism to ensure the efficient use of resources. This is supported with a mathematical coverage model that deals with the formation of coverage holes. We also present a transform-domain based robust algorithm to effectively remove the unwanted components from the data. Our proposed framework offers a handy algorithm that enjoys desirable complexity for real-time applications as shown by the extensive simulation results.

cited By 0; The kinetics of crystallization of Zn38Mg12Ca32Yb18 bulk metallic glass was studied by differential scanning calorimetry (DSC) in both non-isothermal and isothermal (at different heating rates) modes. Under isochronal process, theoretical models were adopted to analyze the apparent activation energies for characteristic temperatures. The results indicate that the apparent activation energies for characteristic temperatures in the Zn38Mg12Ca32Yb18 bulk metallic glass by Kissinger model, Flynn-Wall-Ozawa (FWO) model and Augis-Bennett (AB) model are in good agreement with each other. Furthermore, crystallization transformation kinetics during isothermal process was analyzed by the Johnson-Mehl-Avrami (JMA) model. The Avrami exponent n ranges from 3.25 to 4.12 in the isothermal mode. It is noted that the activation energy corresponding to isothermal conditions calculated using Arrhenius equation is larger than the value calculated by the Kissinger method in isochronal conditions, because the energy barrier in isothermal annealing mode is higher than that of isochronal conditions.
