Antigens of anti-tick vaccines are more efficacious for homologous challenge with local tick strains. cDNA clones encoding for Bm91 from local Rhipicephalus (Boophilus) microplus strains were developed to immunize Bos indicus cattle under field conditions. Three groups of six animals each were injected with the antigen Bm91, saline, and adjuvant, respectively. Animals were immunized three times at 3-week intervals and a fourth time after six months. The anti-Bm91 antibody level, measured by ELISA, was monitored for 7 months and the reproductive performance of naturally infested R. (B.) microplus was determined. Bm91-immunized animals developed a strong immune response expressed by high anti-Bm91 levels remaining on high levels until the end of the study. Western blot analysis confirmed that Bm91 is immunogenic. Compared to control animals, the reproductive efficiency index and the egg viability were 6% and 8%, respectively, lower in the Bm91 group (P < 0.05). In conclusion, it was demonstrated that Bm91 induced a long-lasting immune response. However, the effect on the tick reproduction was not sufficient for an efficient tick control. Further studies under field conditions are warranted to enhance the effect on the tick reproduction by optimizing the immunization regimen, alone or in combination with other vaccine candidate antigens. Little is known about the induction and modulation of gut-associated immune reactions after nematode infection in the chicken. The objective of the present study was to compare the pathogenesis, induction of immune reactions and electrophysiological changes of the gut after mono-infection with Heterakis gallinarum and after dual infection with H. gallinarum and Histomonas meleagridis in layer chickens. In two experiments 3-week-old chickens were inoculated with embryonated H. gallinarum eggs, which were positive for H. meleagridis. While birds of the first experiment were left untreated, those of the second experiment were treated with dimetridazol to prevent H. meleagridis co-infection. Mild to moderate histological lesions and local immune reactions with a significant increase in CD4+, CD8α+, TCRαβ+ and TCRδγ+ cells in the lamina propria and induction of the T-helper type 2 (Th2) cytokine interleukin-13 dominated the H. gallinarum immune response at 2 weeks post infection. Co-infection with H. gallinarum and H. meleagridis induced an increase in mRNA expression of the T-helper type 1 (Th1) cytokine interferon-γ, a decrease in splenic CD4+ cells and severe destruction of the caecal mucosa in association with strong T-cell infiltration in the caecal lamina propria. There was no obvious effect on the chloride secretion of the caecal epithelium, which was investigated once the mucosa had almost recovered from the infection, in either experiment. These results suggest that the local T-cell reactions to nematode infections in chickens may be comparable with mammals and may be shifted from a Th2-dominated to a Th1-dominated response when accompanied by a protozoan infection. Fast growing broilers are less able to cope with fitness related challenges. As the allocation of metabolic resources may be traded off between performance and defence functions in parasitized hosts, we hypothesized that fast growing broilers are more sensitive to mixed nematode infections compared with slower growing genotypes under the same environmental conditions. Therefore, we compared male birds of genotypes selected for either meat production (Ross-308, R) or egg production (Lohmann Brown Plus, LB) or for both purposes (Lohmann Dual, LD), to assess their resistance and tolerance to mixed nematode infections with Ascaridia galli and Heterakis gallinarum. While infections reduced feed intake in all three genotypes, feed conversion efficiency was not affected. Infections impaired growth performance only in R birds, indicating lower tolerance in the fast growing genotype compared with slower growing LB and LD genotypes. Impaired tolerance in R birds was associated with a relative nutrient scarcity due to an infection-induced lower feed intake. Resistance to experimentally induced infections depended on host genotype as well as on the worm species involved. Overall, the A. galli burden was higher in R than LB, whereas the burden of LD was not different from that of R and LB. In contrast, the H. gallinarum burden of first generation worms was similar in the three genotypes. Susceptibility to re-infection with H. gallinarum was higher in LB than in LD, whereas very low levels of re-infection were observed in R birds. Our data collectively suggest that resistance and tolerance to mixed nematode infections are sensitive to growth rate in chickens. Abstract Worm expulsion is known to occur in mammalian hosts exposed to mono-species helminth infections, whilst this phenomenon is poorly described in avian hosts. Mono-species infections, however, are rather rare under natural circumstances. Therefore, we quantified the extent and duration of worm expulsion by chickens experimentally infected with both Ascaridia galli and Heterakis gallinarum, and investigated the accompanying humoral and cell-mediated host immune responses in association with population dynamics of the worms. Results demonstrated the strong co-expulsion of the two ascarid species in three phases. The expulsion patterns were characterized by non-linear alterations separated by species-specific time thresholds. Ascaridia galli burden decreased at a daily expulsion rate (e) of 4.3 worms up to a threshold of 30.5 days p.i., followed by a much lower second expulsion rate (e = 0.46), which resulted in almost, but not entirely, complete expulsion. Heterakis gallinarum was able to induce reinfection within the experimental period (9 weeks). First generation H. gallinarum worms were expelled at a daily rate of e = 0.8 worms until 36.4 days p.i., and thereafter almost no expulsion occurred. Data on both humoral and tissue-specific cellular immune responses collectively indicated that antibody production in chickens with multispecies ascarid infections is triggered by Th2 polarisation. Local Th2 immune responses and mucin-regulating genes are associated with the regulation of worm expulsion. In conclusion, the chicken host is able to eliminate the vast majority of both A. galli and H. gallinarum in three distinct phases. Worm expulsion was strongly associated with the developmental stages of the worms, where the elimination of juvenile stages was specifically targeted. Factors affecting the development of Ascaridia galli –specific humoral responses and their protective roles are largely unknown. We investigated the effects of time and infection dose on A. galli –specific IgY antibody levels following experimental infection. The acquisition and development of new infections and reinfections were also monitored by using tracer birds. Relationships between the retrospective IgY and the final worm burden of the birds were investigated to determine whether humoral immune responses generated during infection provide protection to the host animal. Young chickens were infected (+) with either 100 or 1000 embryonated eggs of A. galli (100+: n = 45; 1000+: n = 45) or kept as uninfected controls (CON: n = 10). Uninfected birds were also added to each infection group as tracer (T) birds (T100+; n = 5 and T1000+; n = 5). Faecal egg counts and IgY antibody concentrations in plasma and egg yolk were determined at selected intervals. Final worm burdens were quantified at 28 weeks post infection (wpi). The plasma antibody (PAB) and egg yolk antibody (EAB) levels of experimentally infected birds were compared to those of control and tracer birds throughout the study period, and PAB levels were found to depend initially on the infection dose but thereafter mainly on reinfections. Starting at wpi 2, 1000+ had consistently higher PAB levels than CON did (P < 0.05). With exceptions at wpi 0, 2 and 12, PAB levels were also higher (P < 0.05) or tended to be higher (P < 0.10) in 100+ than in CON. An earlier and higher increase was observed in the PAB levels of T1000+ than in those of T100+, implying that (re-)infection occurrence depended on the infection dose. Although 1000+ showed higher (P < 0.05) EAB levels than CON did at both wpi 14 and 18, EAB levels were higher in 100+ than in CON only at wpi 28 (P < 0.05). The total worm burdens in the initial experimentally infected birds were similar (P = 0.257); they were also highly comparable between experimentally and naturally infected birds, indicating that final worm burden was mainly determined by the naturally occurring infections resulting from continuous exposure. When all available information on the retrospective plasma and egg yolk IgY levels was collectively evaluated to estimate the larval or total worm burdens of the experimentally infected birds, both PAB and EAB levels at particular wpi were significantly associated with worm burden, especially with larval count. In conclusion, our data support the hypothesis that the number of larvae, rather than the number of mature worms, affects the antibody levels in both plasma and egg yolk. Despite the increased levels of A. galli –specific antibodies in plasma and egg yolk throughout the study period, only a weak indication was found that antibodies might be directly associated with protection. The aim of this study was to document the effectiveness of the implementation of a cosynch protocol and fixed time artificial insemination in suckled beef cows kept under range conditions in Germany. In addition, the suitability of hCG administration as an alternative to the GnRH analog buserelin (Receptal®, Intervet, Germany) as ovulation inducing agent was investigated. The 120 Simmental and 120 German Angus cows were randomly divided into four groups. Cows of Group 1 were subjected to the classical cosynch protocol, constituting administration of GnRH, followed, 7 days later, by prostaglandin F 2α and, after 2 days, another GnRH administration and simultaneous AI. In cows of Group 2, hCG was administered instead of GnRH at the beginning of the protocol, in Group 3 at the end and in Group 4 at the beginning and at the end. Inseminations with frozen-thawed semen from Limousin, Charolais, Simmental and German Angus sires were carried out by the same skilled inseminator. Blood samples collected 11 days before and on days 0, 7 and 9 of the cosynch protocol were analyzed for progesterone concentration. The study extended over three consecutive years. Of the total of 489 inseminations, 53% resulted in pregnancies (Simmental: 60%; German Angus: 48%). When GnRH was administered at the beginning and at the end of the cosynch protocol (Group 1), pregnancy rate across breeds was 65% (Simmental: 61%; German Angus: 68%); likewise when hCG was substituted for GnRH at the end of the protocol (Group 2). Pregnancy rate across breeds reached 48% when hCG was given at the beginning (Group 3) and was lowest (37%) when hCG was given at the beginning and at the end. German Angus cows responded particularly poorly to hCG treatment. In cows that were cycling, as indicated by serum progesterone levels 11 and 0 days before treatment, pregnancy rate across breeds and treatments was highest (57%) if the cosynch protocol was initiated at a stage of the cycle when peripheral progesterone levels were on the rise. In non-cycling cows, representing only 13% of the herd, contrary to expectation pregnancy rate reached 65%. In conclusion, implementation of fixed-time artificial insemination in suckled beef cows kept under local range conditions led to satisfactory pregnancy results, especially if the cosynch protocol was initiated at a stage when progesterone levels were on the rise. Breeding of cattle with docile temperament can simplify routine practices and enhance welfare of cattle in common production systems. The aim of the study was to compare and validate some aspects of assessment methods for temperament traits in cattle for their potential use in breeding programmes. Calves of different beef and dairy cattle breeds were video-recorded during a 2 min tethering (n = 160) and a 2 min crush test (n = 185) at the age of 38 ± 25 days. Subsequently, two observers blinded to the identity of records each analysed the video clips three times with the software Interact ® in intervals of 1–2 days between viewings. Frequency and duration of behaviour patterns such as head movement, tail flicking or defecation were recorded. In addition, each behaviour pattern was assessed on a 10 cm visual analogue scale (VAS) and in the end of each observation an overall behaviour score on a numerical scale from 1 to 5 and on a VAS was assigned to each animal. Using variance components from mixed model analysis, results revealed that both inter- and intra-observer reliability (OR) were similar or superior for the majority of behaviour patterns when assessed with the VAS compared to the more time-consuming assessment of exact frequencies and durations (e.g. Intra-OR ‘tail flicking’: Observer I: r VAS = 0.83 and r Freq = 0.26; Observer II: r VAS = 0.91, r Freq = 0.37 and r Dur = 0.87 for the tethering test; Inter-OR ‘body movement’: r VAS = 0.81 and r Freq = 0.33 for the crush test). The overall behaviour score showed a high intra-observer reliability when assessed with the VAS as well as with the numerical scale (Observer I: r VAS = 0.97 and r Num = 0.77; Observer II: r VAS = 0.82 and r Num = 0.84 for the crush test). However, not all traits, and particularly including those assessing rarely occurring behaviour patterns, proved to be appropriate for recording with a VAS. Seasonality in Boer goats in Northern Europe and attempts to overcome it is addressed in three studies. In Experiment 1, monitoring of estrus and serum progesterone concentration in sexually mature nulliparous Boer goat crosses indicated that ovarian activity ceased in February/March and was resumed from August onward. In 63.7% of 22 does the first estrus of the season was preceded by one or two ovulations, indicated by an increase in progesterone; in 22.7% first estrus and first ovulation coincided and in the remaining 13.6% the first estrus was not succeeded by formation of a functional corpus luteum. In Experiment 2, 31 Boer goats, weaned between July and September, were treated with progestogen-containing intravaginal sponges and 250 IU eCG either one (n = 11), seven (n = 10) or fourteen (n = 10) days after weaning. All but two does showed estrous symptoms and were naturally mated. Transrectal ultrasonography 45 days after mating revealed a pregnancy rate of 42%. All of these carried to term (36% treated immediately after weaning, 40% treated one week later and 50% treated two weeks later) and gave birth to, on average, 2.1 kids. In Experiment 3, of 18 nulliparous does, 9–12 months of age, subjected to progestogen-eCG treatment out-of-season, 15 (83%) responded by showing estrous symptoms; 14 were naturally mated and six were diagnosed as pregnant five weeks later. One of the pregnant does aborted two weeks before parturition was due; in the others pregnancy had ceased at an earlier stage without visible symptoms. In conclusion, in northern Europe, Boer goats undergo a period of reduced ovarian activity, although to a lesser degree than dairy breeds. In most cases acyclicity was overcome by hormonal stimulation and, if mated, part of the weaned does carried to term, whereas in young nulliparous does pregnancy was not maintained. Palearctic species of Culicoides (Diptera, Ceratopogonidae), in particular of the Obsoletus and Pulicaris complexes, were identified as putative vectors of bluetongue virus serotype 8 (BTV-8) on ruminant farms during the epizootic in Germany from 2006 to 2009. BTV may cause severe morbidity and mortality in ruminants and sporadically in South American camelids (SAC). However, the fauna of Culicoides spp. on SAC farms has not been investigated. Therefore, the ceratopogonid fauna was monitored on three farms with BTV-seropositive SAC in Germany. Black–light traps were set up on pastures and in stables from summer 2008 to autumn 2009. Additionally, ceratopogonids were caught in emergence traps mounted on llama dung and dung-free pasture from spring to autumn 2009. After morphological identification, selected Culicoides samples were analysed for BTV-RNA by real-time RT-PCR. The effects of the variables ‘location’, ‘temperature’ and ‘humidity’ on the number of Culicoides caught in black–light traps were modelled using multivariable Poisson regression. In total, 26 species of Culicoides and six other genera of biting midges were identified. The most abundant Culicoides spp. collected both outdoors and indoors with black-light traps belonged to the Obsoletus (77.4%) and Pulicaris (16.0%) complexes. The number of Culicoides peaked in summer, while no biting midges were caught during the winter months. Daily collections of Culicoides were mainly influenced by the location and depended on the interaction of temperature and humidity. In the emergence traps, species of the Obsoletus complex predominated the collections. In summary, the absence of BTV-RNA in any of the analysed Culicoides midges and in the BTV-seropositive SAC on the three farms together with the differences in the pathogenesis of BTV-8 in SAC compared to ruminants suggests a negligible role of SAC in the spread of the virus. Although SAC farms may provide similar suitable habitats for putative Culicoides vectors than ruminant farms, the results suggest that geographic and meteorological factors had a stronger influence on Culicoides abundance than the animal species. In beef cows, docility is important for animal welfare reasons, the farmer's and veterinarian's safety as well as for economic reasons. The hormone testosterone is often mentioned in the context with aggressions and docility. The present study tested the hypothesis that higher testosterone levels in beef cows are associated with more aggressive behavior and lower docility during behavior tests. German Simmental (beef) (n = 21) and Charolais heifers (n = 20) were subjected with one repetition to a chute test as well as a separation and restraint test. Heart rate, thermal images of the eye, saliva cortisol as well as saliva testosterone were collected along with behavioral parameters during the tests. For all physiological and most of the behavioral parameters significant breed differences were detected. Charolais heifers (0.10 ± 0.01 ng/ml saliva) had considerably higher testosterone levels compared with Simmental heifers (0.04 ± 0.01 ng/ml; P = 0.0001). Interestingly, the Charolais heifers expressed a higher docility, so across breeds higher levels of testosterone are associated with higher docility. However, all the parameters presumably linked to stress (heart rate, eye temperature, cortisol levels) generally showed stronger correlations to behavior traits than testosterone (e.g., trait “time in corner” — testosterone: r = 0.25 vs. cortisol: r = − 0.43 (Fisher's r-to-z: P = 0.0985); eye temperature: r = − 0.65; (P = 0.0005); heart rate: r = − 0.60 (P = 0.003)). Eye temperature correlated with both heart rate (r = 0.68) and cortisol levels (r = 0.62; both P < 0.0001). Therefore, eye temperature more so than testosterone appears to be a suitable indicator of stress, and as such also an, albeit incomplete, indicator of docility in cattle. [ABSTRACT FROM AUTHOR]

The objectives of the present study were to evaluate the suitability of potential biomarkers for maternal ability in cattle, and in addition to test the hypothesis that dairy cows have a less pronounced motherliness than beef cows. Therefore, maternal behavior of 20 Simmental beef-type (S) and 20 German Black Pied (dairy-type) Cattle (BP) was assessed on the 2nd and again on the 3rd day of the calf's life. Measurements included the frequency of interactions between cow and calf, the cow's willingness to defend her calf, the overall maternal behavior, saliva cortisol, saliva oxytocin, heart rate, and thermal images of the eye (ET). Mixed model analysis revealed that BP had significantly (P < 0.05) higher oxytocin (88.6 ± 9.2 vs. 62.8 ± 9.2 pg/ml saliva) and cortisol (1.3 ± 0.1 vs. 1.0 ± 0.1 ng/ml saliva) levels, but lower heart rates (80.0 ± 2.0 vs. 95.8 ± 2.0 bpm) than S cows. Simmental (beef) cows showed more defensive behavior (3.5 ± 0.2 vs. 2.7 ± 0.2 scores), but fewer total interactions between cow and calf (8.1 ± 1.4 vs. 13.8 ± 1.4), compared to BP (dairy). However, with the exception of heart rate and overall maternal behavior, breed differences tended to diminish from the 2nd to the 3rd day of the calf's life. Repeatabilities ranged from 9 ± 23% (ET) to 77 ± 7% (maternal behavior measured on a visual analogue scale), and correlations between physiological parameters and behavior differed between breeds and were generally at a low level. The objective of the present study was to evaluate the behavioural effects of Burdizzo castration of crossbred beef calves when weaned and castrated separately (castrated 8 weeks after weaning) or in combination. Over two consecutive years a total of 111 Limousin × Simmental male calves were included in the study. At an age of 7 months calves were randomly assigned to 1 of 4 treatment groups and either weaned in week-8 (Bull-8; N = 27); weaned in week-8 and castrated in week 0 (Cas-8; N = 26); weaned in week 0 (Bull-0; N = 29); or weaned and castrated in week 0 (Cas-0; N = 29). The behaviour of the calves including the number of vocalisations and time spent feeding, lying and standing/walking was recorded during the first 3 days of week 0. Bull-8 and Cas-8 did not show any vocalisation activity during the observation period. In contrast, Bull-0 and Cas-0, showed more than 3 calls/10 min period on the first day, which decreased steadily until the third day in both groups. In all groups the time spent feeding increased from day 1 to 3. Bull-8 animals did not vary their standing/walking and lying behaviour, but Cas-8 increased the time spent standing/walking and decreased lying. Bull-0 decreased the proportion of standing/walking from approximately 60 to 40% and increased lying from 23 to more than 30%. A similar rate of change with standing/walking decreasing from 70 to 50% and lying increasing from 12 to 20% was noted in Cas-0. Compared to bulls, castrates spent more time standing/walking and less lying. From week-8 to 0, Bull-8 and Cas-8 gained about 700 g/d, whereas the 2 other groups gained more than 1.000 g/d. Average daily gains from week 0–3 of Cas-8 were lower than in the other groups ( P < 0.05). In conclusion, Burdizzo castration did not cause considerable effects on the behaviour, whereas weaning had a pronounced impact. The combination of both management procedures did not intensify the behavioural responses. In modern husbandry systems, feed rations for laying hens may contain high levels of dietary fibre, which may only be fermented intestinally. Depending on fermentation properties, the type of dietary fibre may influence the intra-caecal environment constituting the predilection site of Heterakis gallinarum. We investigated egg production dynamics and fecundity of H. gallinarum residing in different caecal environments induced through different types of dietary fibre. Growing layers were fed a standard (CON) or an insoluble- (I-) or soluble- (S-) non-starch polysaccharides-(NSP) supplemented diet for the first 11 weeks (wk) of life in a twice-replicated experiment. At 3 wk of age, the birds were infected with 200 embryonated eggs of H. gallinarum . Starting from 3 wk post-infection (p.i.), individual daily total excreta were collected. The number of eggs per gram of faeces (EPG) was determined ( N = 2240), and the number of eggs per day (EPD) were estimated. The birds were necropsied 8 wk p.i. and the worm burdens were quantified. The nematode began to lay eggs as early as 23 d.p.i. and thereafter laid on average 436 eggs/d. I-NSP- and S-SNP-supplemented diets expedited the onset of patency by approximately 5 days, and increased total egg excretion by 110% and 185%, respectively, due to higher worm counts. The latter diet (S-SNP) additionally increased total egg excretion by 94% due to enhanced fecundity. Longitudinally collected data showed that fecundity of the nematode (EPD/female) was relatively stable starting from the second week of the patent phase. Results suggest that NSP-induced enlargement of caeca favoured establishment of the nematode while effects of an intensified microbial fermentation through S-NSP were crucial for an enhancement in the worm fecundity. Based on the long-term data obtained from daily collections of total excreta in experimentally H. gallinarum- infected chickens, it is concluded that a diet enriched with pea bran as source of insoluble NSP increased worm egg production merely through elevated worm counts, whereas a diet supplemented with chicory root meal as source of soluble NSP elevated worm egg production through both increased worm counts, and more profoundly through enhanced fecundity. Abstract: Group housing of pigs leads inevitably to more or less serious agonistic interactions during the establishment of the social rank order of the group. In order to reduce the number of severe agonistic interactions and thus the negative effects on well-being and performance, the use of genetic selection of calm sows maybe a possible strategy. Therefore, in this study the behaviour of 112 German Landrace sows was observed after the animals were brought together in a group of 10–20 sows. After this initial period, the sows were integrated into a large dynamic group in the dry sow area where the animals were housed for 71 days. Before moving the sows into the farrowing area, skin lesions scores for three body regions (front/middle/rear) on both sides were recorded using a scoring system from 1 (no lesions) to 4 (wounds, lesions all over the body area). After farrowing, sows’ reaction towards the separation from their litter was recorded to analyse relationships between aggressiveness and handling. Earlier research suggests that skin lesions recorded shortly after mixing are associated with agonistic interactions at mixing and might therefore be a useful indicator for the evaluation of recent aggressiveness of animals. However, results of the present study show that an individuals’ frequency of being initiator of agonistic interactions post mixing do not affect (p >0.1) the extent of skin lesion recorded 10 weeks later. Conversely, animals being attacked frequently were evaluated with higher scores in the anterior region (p =0.0435). These findings indicate that a higher skin lesion score does not represent generally more aggressive sows under commercial housing systems, but it is still an indicator for overall aggressiveness within pens or groups. Between the different groups significant differences in the extent of skin lesions were found (e.g. skin lesion score front: p =0.0228). A negative relationship was found between skin lesion score in caudal region and sows’ reaction towards stockperson when handling their piglets (r =−0.28, p <0.01). Furthermore, behaviour traits related to aggressiveness correlated with later reproductive performance. While sows recorded frequently as aggressors post mixing gave birth to more total and live born piglets, sows with higher skin lesion scores had a lower reproductive performance (e.g. skin lesion score front vs. total born piglets: r =−0.28, p <0.01). Taken together, these results suggest that more severe skin lesions are indicative of low-ranking and less vital sows, but skin lesions are not useful to identify the generally more aggressive individuals. In mountain areas such as South Tyrol, Italy, in addition to extensively managed farms with dual-purpose breeds, there is an increasing number of farms raising high-yielding breeds, that depend on large quantities of concentrated feed imported from other regions. A large supply of nutrients not only leads to unbalanced nutrient budgets but also to a lower contribution to net human food supply by using large amounts of human edible feedstuff. 64 dairy farms managed at differing concentrated feed intensity levels in South Tyrol were assessed. The farms were divided into four groups according to the amount of concentrated feed used (high [H] vs. low [L]) and the breed (Tyrolean Grey [TG] vs. Brown Swiss [BS]). The following groups were analyzed: H-BS with ⌀ 8.7, H-TG with ⌀ 6.1, L-BS with ⌀ 4.0 and L-TG with ⌀2.8 kg concentrated feed per cow and day. Farms cultivated only permanent grassland without any cash crops. N budgets for H-BS, H-TG, L-BS and L-TG were 79.54, 50.81, 37.85 and 34.16 kg ha−1, respectively, with significantly higher values for H-BS compared to the other groups. P and K budgets were also significantly higher for H-BS (12.8 and 43.96 kg ha−1, respectively) when compared to L-BS (3.10 and 8.86 kg ha−1) and L-TG (1.50 and 11.91 kg ha−1). The nutrient use efficiency, calculated as the proportion of output to input, for N and K was similar across all groups, while the P-efficiency was significantly higher in L-BS and L-TG (91.3 and 98.5%, respectively) compared to H-BS and H-TG (73.0 and 72.4%, respectively). HeFCE for protein and energy was lowest for H-TG (0.99 and 2.38, respectively) compared to L-TG (1.54 and 3.73, respectively), with the decisive factor being the amount of concentrated feed per kg ECM produced (g DM/kg ECM). • N, P, K budget and net human food supply were calculated. • High input dairy farms are at higher risk for nutrient surpluses. • High-input farms with Tyrolean Grey contribute less to human food supply. • Contribution to human food supply and nutrient surpluses are linked. Agricultural livestock production ranks among the most environmental impactful industry sectors at the global level, and within the livestock sector, beef production accounts for a large proportion of environmental damage. Beef production in Alpine mountain regions, such as in South Tyrol (Italy), is a small, but increasing agricultural sector. Thus, the aim of this study was to examine the environmental impact of different organic and conventional beef production systems in South Tyrol and to compare their environmental impact and effect on biodiversity under Alpine production conditions. Live cycle assessment (LCA) approach was used and 1 kg of live weight (LW) was chosen as functional unit (FU). Global warming potential (GWP, kg CO 2 -eq), acidification potential (AP, g SO 2 -eq), eutrophication potential (EP, g PO 4 -eq), non-renewable energy use (NRE, MJ-eq), land occupation (LO, m2 organic land/year) and biodiversity damage potential (BDP) expressed in potential disappeared fraction (PDF) were investigated. The study involved 18 beef cattle farms in the South Tyrolean region: Conventional calf-fattening farms (CCF = 6), organic suckler cow farms (SCF = 6), and conventional heifer/ox fattening farms (HOF = 6). The CCF system showed a higher environmental impact compared to SCF and HOF systems for all impact categories (P < 0.05). Between the organic and the conventional system (SCF and HOF), no significant differences (P > 0.05) were found for most of the considered impact categories (means ± SEM per FU): GWP: 19.8 vs 17.1 ± 4.2 kg CO 2- eq, AP: 11.4 vs 9.3 ± 4.7 g SO 2 -eq, EP: 4.1 vs 2.8 ± 1.2, NRE: 21.9 vs 13.8 ± 7 MJ-eq, SCF and HOF respectively. Only for LO (70.8 vs 44.1 ± 17.7 m2 organic/y, P < 0.01, SCF and HOF respectively) and the effect on BDP (−1.93 vs −0.85 ± 0.35, PDF, P < 0.01, SCF and HOF respectively) differences between organic and conventional production methods could be revealed. The study showed that beef cattle husbandry in the Alpine area has a satisfactory environmental performance. Modern chickens have been genetically developed to perform high under optimal conditions. We hypothesized that high-performance is associated with a higher sensitivity to environmental challenges in laying hens. By using nematode infections as an environmental stressor, we assessed performance-level associated host responses in a high (i.e. Lohmann Brown Plus, LB) and in a lower performing, a so-called dual-purpose chicken genotype (i.e. Lohmann Dual, LD). The hens were infected with 1000 eggs of Ascaridia galli and Heterakis gallinarum at 24 weeks of age. Hen performance parameters, humoral immune responses in plasma and egg yolks and worm burdens were assessed at several occasions over a period of 18 weeks post infection (wpi). While infections had no significant effect on feed intake (P = 0.130) and body weight in both genotypes (P = 0.392), feed conversion efficiency was negatively affected by infections (P = 0.017). Infections reduced both laying rate and egg weight and thereby per capita egg mass in both genotypes (P < 0.05). While laying rate in infected LB hens decreased significantly (P < 0.05) in the early infection period (i.e. by 3 wpi), the decrease in LD hens appeared much later (i.e. by 14 wpi). Worm burdens resulting from the experimental infection were not different between the genotypes for both worm species (P > 0.05), whereas LB hens were more susceptible (P < 0.05) to re-infections than LD hens. Changes in humoral immune responses (i.e. ascarid-specific IgY antibodies in plasma and egg yolks) of the two genotypes over time reflected closely the corresponding changes in larval counts of the hens, descending from both experimental and subsequent natural infections in both genotypes. Infections caused a shift in egg size classes, leading to smaller frequency of larger eggs in both genotypes. Infections reduced egg weight (P = 0.018) and led to a reduced fat content in the egg yolks (P = 0.045). The proportion of poly-unsaturated fatty acids (PUFA), especially n -6-PUFA, was also lower in egg yolks of the infected hens (P = 0.032). We conclude that tolerance to nematode infections in laying hens is dependent on host-performance level. The impairment in host tolerance was both genotype and time dependent, likely due to differences in genetic programming for production peak and persistency of the two genotypes. The two genotypes exhibited similar levels of resistance after a fully controlled experimental infection, but the high performing hens were more susceptible to subsequent natural infections. Mountain dairy cattle farming systems are pivotal for the economy, as well as for social and environmental aspects. They significantly contribute to rural development, which is currently strongly prioritized in the common European Union agricultural policy; at the same time, they are also increasingly criticized for having a relatively high environmental impact (such as greenhouse gas emissions) per kilogram of product. Consequently, the aim of this study was to assess and compare the environmental efficiency of 2 common alpine dairy farming systems, with a focus on the effects of grazing, considering the seasonal variability in feeding at the individual cow level and farm management over a 3-yr period. This study focuses on alpine farming systems, but can also be considered to effectively represent other topographically disadvantaged mountain areas. We compared an intensively managed and globally dominating production system (high-input) aimed at high milk yield through relatively intensive feeding and the use of the high-yielding dual-purpose Simmental cattle permanently confined in stables, with a forage-based production system (low-input) based on seasonal grazing and the use of the autochthonous dual-purpose breed Tyrolean Grey. For the present analysis, we used a dataset with information on feed intake and diet composition, as well as animal productivity at the individual cow level and farm management data based on multiyear data recording. We quantified 4 impact categories for 3 consecutive years: global warming potential (GWP 100 ), acidification potential (AP), marine eutrophication potential (MEP), and land use (LU; in square meters per year and eco points [Pt], with the latter additionally considering the soil quality index). In addition to being attributed to 1 kg of fat- and protein-corrected milk (FPCM), these impact categories were also related to 1 m 2 of on-farm area. Due to limited agronomic options beyond forage production and pasture use in alpine regions, net provision of protein was calculated for both farming systems to assess food supply and quantify the respective food-feed competition. Overall, the low-input farming system had greater environmental efficiency in terms of MEP per kilogram of FPCM, as well as MEP and AP per square meter than the high-input system. Land use was found to be consistently higher for the high-input than for the low-input system, the GWP 100 per kilogram of FPCM was lower for the high-input system. Additionally, pasture access had a significant effect on the reduction of environmental impacts. Lastly, the net protein provision was slightly negative for the high-input system and marginally positive for the low-input system, indicating a lower food-feed competition for the latter. Future studies should also address the social and economic aspects of the farming systems in order to offer a comprehensive overview of the 3 key factors necessary for achieving more sustainable farming systems, particularly in disadvantaged marginal regions such as mountain areas., (The Authors. Cow milk microbiota has received increased attention in recent years, not only because of its importance for human health but also because of its effect on the quality and technological properties of milk. Several studies, therefore, have investigated the effect of various production factors on the microbial composition of milk. However, most of the previous studies considered a limited number of animals from experimental or single farm, which could have biased the results. Therefore, this study aimed to understand the effect of different alpine production systems in Italy on the compositional and microbiological quality of milk, considering commercial herds with different feeding intensities and cattle breeds. The results obtained in this work indicated that the month and season of sampling (July for summer or February for winter) more than farm, breed, and cow diet exerted significant effects on cow milk parameters and microbiota. In particular, significant differences were observed for urea content in milk between sampling seasons. Differences in milk fat were mainly related to breed specific effects. From a microbiological point of view, statistically significant differences were found in presumptive lactic acid bacteria counts. Based on a culture-independent method, milk obtained in February harbored the highest number of Firmicutes (e.g., Lactobacillus) and the lowest number of Actinobacteria (e.g., Corynebacterium). Moreover, bacterial richness and diversity were higher in July during alpine pasture season indicating a significant effect of pasture feed on the growth of bacterial communities. This study estimates the association between the level of biosecurity, animal welfare, milk quality, and economic performance on 2291 mountain dairy farms which largely differs in management and structure from large dairy operations in the lowlands mainly due to climatic and topographic constraints in mountain areas. The dairy industry's increasing emphasis on biosecurity is crucial for ensuring animal health, productivity, and disease mitigation. Therefore, in the present study the biosecurity and animal welfare status of mountain dairy farms were assessed considering the official welfare protocol for dairy cows of the Italian ClassyFarm system. Our findings reveal a suboptimal adoption of biosecurity measures, attributed to structural limitations in mountain farms and farmers' awareness gaps. Despite these challenges, the economic significance of biosecurity adoption is evident, emphasizing potential benefits for farm viability and animal health. Conversely, the study indicates a moderate to good welfare status in assessed farms, reflecting farmers' awareness of the importance of high welfare standards for dairy cows. Improved welfare positively correlates with milk sales and productivity, highlighting the economic advantages of prioritizing animal well-being. Challenges persist, particularly regarding animal housing deficiencies and related consumer concerns about animal welfare in dairy production. Therefore, targeted interventions and educational initiatives are crucial to empower farmers and promote best practices in biosecurity and animal welfare management. Reports of Ascaridia galli in laying hens in Europe have increased since the ban on conventional battery cages in 2012. As this parasite is transmitted directly via the faecal-oral route by parasite eggs containing a larva, it is reasonable to assume that the escalating problem is related to the increased exposure now occurring in modern welfare-friendly cage-free housing systems. On many farms, A. galli reappears in subsequent flocks, even though the birds have no access to the outdoors, biosecurity is high and empty houses are cleaned and disinfected during downtime. Since the egg production cycle lasts only ≈80 weeks and recombinant antigen production for helminth vaccines has not yet been solved, the development of a vaccine seems to be an unrealistic option. Therefore, disrupting the life cycle of the parasite by other means, including the strategic use of dewormers, appears to be the key to controlling infection. Of concern is that only one class of anthelmintics is licenced for poultry in Europe and that are usually administered indiscriminately through the birds' drinking water and often too late when the parasite is already established. If current calendar-based parasite control strategies are not changed, there is a risk that resistance to anthelmintics may develop, as has already been demonstrated with nematodes in livestock. We insist that treatments can be more effective and the risk of developing drug resistance can be mitigated if we invest in a better understanding of A. galli responses to more prudent and judicious use of anthelmintics. Cicerbita alpina (L.) Wallr, is a perennial alpine plant and a member of the Asteraceae family, typically found at altitudes above 1000 meters in the Italian Alps. Although previously utilized primarily as a local delicacy, recent studies have revealed strong antiparasitic activity through in vitro experiments. In Europe, numerous chemical drugs employed to combat nematodes - helminths that infest the digestive tract of livestock - are banned due to their environmental harm or show only reduced efficiency because of the development of resistance. Consequently, there is a growing demand for new alternative anthelmintic treatments in agricultural practices. Specialized metabolites found in the extracts of C. alpina could offer a sustainable and biological alternative to chemical drugs, specifically for nematode control. For this purpose, a unique germplasm collection originating from eight distinct natural populations in the Italian Alps was analyzed for its chemical diversity using state-of-the-art targeted LC-MS/MS spectrometry, including quantification based on multiple reaction monitoring. The predominant metabolites identified from the species were the caffeic acid derivatives chicoric acid, chlorogenic acid, and 3. 5-dicaffeoylquinic acid, the sesquiterpene lactone derivative 8- O -acetyl-15- ß -D-glucopyranosyl lactucin and the flavone glycosides, apigenin-7- O -glucoside and luteolin-7- O -glucoside, alongside their precursors apigenin and luteolin, respectively. Additionally, the genetic diversity of eighty individual plants within the germplasm collection was evaluated using ten DNA molecular markers (Simple Sequence Repeats), successfully transferred from two closely related species ( Cichorium intybus and Tanacetum parthenium ). This investigation unveiled a significant range of genetic diversity within the examined populations, resulting in the establishment of three distinct genetic groups. The findings were further correlated with the original ecological environment and local climate conditions spanning a biennial period, indicating substantial variations among the different accessions and the intricate interplay between genetic background and environmental factors. The aim of the present study was to assess the impact of altitude on the quality of milk from dairy cows housed in small-scale farms in alpine areas. Therefore, a dataset comprising 5,680 bulk milk samples from 32 farms located at different geographical altitudes was considered. Milk traits such as fat, protein, lactose, free fatty acids, casein, milk urea nitrogen content, pH value, and somatic cell score were examined in the laboratories of the South Tyrolean Dairy Association using mid-infrared spectroscopy. The data were analyzed using a statistical model that considered altitude category, grazing practice, housing system, and season of milk analysis as fixed effects. The findings revealed a positive association between altitude and milk fat, free fatty acids, and somatic cell count. Conversely, lactose content, milk urea nitrogen, and pH value in milk from cows kept on farms at higher altitude (>1,200 m above sea level) showed a negative relationship with altitude. Farms located at altitudes above 1,200 m exhibited higher fat, protein, urea, and somatic cell content than farms located at lower levels. The results provide new insights into a production effect that has so far received little attention and should be considered in farm management (e.g., feeding management, breed decision) to ensure animal health and the associated animal welfare as well as the productivity of dairy cows reared in traditional small-scale mountain dairy farms., (© 2023.)

The objective of this study was to determine the risk factors for skin alterations at herd and cow level on dairy farms with different housing systems in South Tyrol (Northern Italy). A cross-sectional study was conducted on 204 farms (111 free stalls and 93 tie stalls) from March to October 2019 assessing the level of animal welfare using resource-based and animal-based indicators. A total number of 1,891 dairy cows were evaluated, of which 43.5% were reared in tie stalls and 56.5% in free stalls. A logistic regression model identified the herd and cow level risks factors for neck and leg skin alterations in the two different systems. There was a higher prevalence for skin lesions on the neck (Odd Ratio (OR) = 2.36) and hock (OR = 2.82) for tie stalls. Irrespective of the housing system the soft-based stall mattresses had a lower prevalence for knee and hock lesions of 0.48 and 0.54, respectively, compared to wood base stalls for both knee (OR = 2.19) and hock (OR = 2.47) consecutively. The prevalence of skin alterations on the knee (OR = 0.42) and hock (OR = 0.33) decreased by the presence of sawdust as bedding material. Similarly, straw (OR = 0.61) and lime-straw-water bedding (OR = 0.59) reduced the prevalence for skin alterations on the hock. Access to pasture reduced the prevalence of skin alterations on the neck (OR = 0.34), the knee (OR = 0.77), and on the hock (OR = 0.46) regardless of the housing system. Livestock production systems with ruminants play a relevant role in the emission of the greenhouse gas CH 4 , which is known to significantly contribute to global warming. Consequently, it is a major societal concern to develop strategies in mitigating such emissions. In addition to breeding toward low-emitting cows, management strategies could also help in reducing greenhouse gas emissions from dairy farms. However, information is required for appropriate decision making. To the best of our knowledge, this is the first study that considers different, already available equations to estimate CH 4 emissions of small-scale dairy farms in the mountain region, which largely differ from large dairy farms in the lowlands concerning management and production. For this study, 2 different production systems, both typical for small-scale dairy farming in mountain regions, were simultaneously run over 3 yr at an experimental farm as follows: (1) a high-input production system, characterized by intensive feeding with high amounts of external concentrates and maize silage, year-round housing, and high yielding Simmental cattle breed, and (2) a low-input production system, characterized by prevailing hay and pasture feeding and silage ban, thus covering most of the energy requirements by forage harvested on-farm and the use of the local Tyrolean Grey cattle breed. Results reveal that feeding management has a significant effect on the amount of CH 4 emissions. The low-input production system produced less CH 4 per cow and per day compared with the high-input production system. However, if calculated per kilogram of milk, the high-input scenario produced proportionally less CH 4 than the low-input one. Findings of this study highlight the potential to assess in a fast and cost-effective way the CH 4 emission in different dairy production systems. Background: A coproantigen enzyme-linked immunosorbent assay (ELISA) has recently been proposed for detecting ascarid infections in chickens. The excretion pattern of ascarid antigens through chicken faeces and the consistency of measurements over the course of infections are currently unknown. This study evaluates the pattern and repeatability of worm antigen per gram of faeces (APG) and compares the diagnostic performance of the coproantigen ELISA with a plasma and egg yolk antibody ELISA and McMaster faecal egg counts (M-FEC) at different weeks post-infection (wpi)., Methods: Faecal, blood and egg yolk samples were collected from laying hens that were orally infected with a mix of Ascaridia galli and Heterakis gallinarum eggs (N = 108) or kept as uninfected controls (N = 71). Measurements including (a) APG using a coproantigen ELISA, (b) eggs per gram of faeces (EPG) using the McMaster technique and (c) ascarid-specific IgY in plasma and in egg yolks using an ascarid-specific antibody ELISA) were performed between wpi 2 and 18., Results: Time-dependent significant differences in APG between infected and non-infected laying hens were quantified. At wpi 2 (t (164)  = 0.66, P = 1.00) and 4 (t (164)  = -3.09, P = 0.094) no significant differences were observed between the groups, whereas infected hens had significantly higher levels of APG than controls by wpi 6 (t (164)  =  -6.74, P < 0.001). As indicated by a high overall repeatability estimate of 0.91 (CI = 0.89-0.93), APG could be measured consistently from the same individual. Compared to McMaster and antibody ELISA, coproantigen ELISA showed the highest overall diagnostic performance (area under curve, AUC = 0.93), although the differences were time-dependent. From wpi 6 to 18 coproantigen ELISA had an AUC > 0.95, while plasma IgY ELISA showed the highest diagnostic performance in wpi 2 (AUC = 0.95). M-FEC had the highest correlation with total worm burden, while APG had highest correlations with weights and lengths of A. galli., Conclusion: Ascarid antigen excretion through chicken faeces can be measured with high accuracy and repeatability using a coproantigen ELISA. The antigen excretion increases over time, and is associated with worm maturation, particularly with the size of A. galli. The present study aims to find efficient alternatives to synthetic anthelmintics among ethno-veterinary herbs. Ascaridia galli eggs isolated from the worm uterus were exposed in vitro to methanolic extracts (ME) of nine plant species such as Achillea millefolium (AM), Artemisia absinthium (AA), Artemisia vulgaris (AV), Cicerbita alpina (CA), Cichorium intybus (CI), Inula helenium (IH), Origanum vulgare (OV), Tanacetum vulgare (TV), Tanacetum parthenium (TP). Flubendazole (FL), 0.5% formalin with dimethylsulfoxide and Petri dishes without the addition of reagents were used as positive, negative and untreated control respectively. The effects of the different ME at concentrations 0.500, 0.325, 0.200 mg/ml were assessed on the embryonic development (ED) of the eggs in duplicate. Logit analysis was used to calculate EC 50 values. A generalized linear mixed model, having plant species and concentration as fixed effect and day as repeated measure, was used to determine differences in ED. Estimated EC 50 was the lowest for FL at 0.11 mg/ml. CA and TV followed with 0.27 mg/ml and 0.32 mg/ml. ED for FL was significantly lower (25%) than that of CA (47%). The analysis showed 0.5 mg/ml of the ME of CA and TV significantly affected the ED at 35% and 42% inhibitions respectively. The ED for all ME showed similar pattern i.e., relatively higher efficacy in the first experimental week compared to the rest of the experimental period. The effect from all multicomponent extracts is time and dose dependent. A reliable method of diagnosing the most prevalent helminth infections in chickens is vital for developing effective control strategies. Ascaridia galli and Heterakisgallinarum are phylogenetically close nematode species that can elicit the development of cross-reactive antibodies in chickens. Therefore, an enzyme-linked immunoassay (ELISA) based on Ascaridia galli antigens in faeces of chickens to detect and quantify infections with both A. galli and H. gallinarum was developed. The ELISA utilised polyclonal antibodies that were obtained from rabbits immunised with soluble antigens isolated from A. galli. In two separate experiments, chickens were kept as uninfected controls or were orally infected with either 100 or 1000 of embryonated eggs of A. galli or H.gallinarum. Faecal samples were collected after 28-30 weeks post-infection. The ELISA was then used to quantify the concentration of soluble worm antigens in faecal samples, i.e., the amount of antigen per gram faeces, APG. The APG from infected chickens was significantly higher than non-infected groups in both experiments (P 0.001). Both 100 and 1000 infection dose groups were not significantly different (P = 0.999) in the experiment with H. gallinarum, whereas in the experiment with A. galli, APG was significantly higher in the 1000 infection group (P 0.001). A receiver operation characteristics (ROC) analysis that evaluates the qualitative performance of diagnostics tests was used to calculate the assay parameters within each mono-infection experiment. The result showed that the assay had a high diagnostics accuracy with an area-under-curve (AUC) of 0.99 in detecting infection in A. galli infected chickens and a moderate-high accuracy (AUC = 0.89) in birds infected with H. gallinarum. The diagnostic sensitivity and specificity of the assay at the optimal cut-off point equivalent to Youden index were 93% and 100% for detecting infections in A. galli experiment and 85% and 92% in H. gallinarum experiment, respectively. The correlation between faecal antigen concentration and all worm burden parameters was positive but generally low (r < 0.33), which provided less information about infection intensities. The objective of this study was to examine the association of herd size with animal welfare in dairy cattle herds. Therefore, 80 conventional dairy cattle farms were classified by the number of cows into 4 herd size classes, C1 (<100 cows), C2 (100-299 cows), C3 (300-499 cows), and C4 (≥500 cows), and assessed using multiple animal-based measures of the Welfare Quality Assessment protocol for dairy cattle. Data were recorded from April 2014 to September 2016 by an experienced single assessor in northern Germany. Each farm was visited 2 times at an interval of 6 mo (summer period and winter period) to avoid seasonal effects on the outcome. The average herd size was 383 ± 356 Holstein-Friesian cows (range 45 to 1,629). Only farms with freestall (cubicle) housing and a maximum of 6 h access to pasture per day were included in the study. Data were statistically analyzed using a generalized linear mixed model. None of the farms reached the highest overall rating of "excellent." The majority of the farms were classified as "enhanced" (30%) or "acceptable" (66%), and at 6 assessments the farms were rated as "not classified" (4%). Regarding single indicators, mean trough length per cow, percentage of cows with nasal discharge, and vulvar discharge increased with increasing herd size, whereas it was vice versa for displacements of cows. Percentage of lean cows, percentage of dirty lower legs, and duration of the process of lying down showed a curvilinear relationship with the number of cows per farm. Herd size was not associated with any other measures of the Welfare Quality protocol. In conclusion, herd size effects were small, and consequently herd size cannot be considered as a feasible indicator of the on-farm animal welfare level. The aim of this study was to examine effects of pasturing in dairy cows on claw condition (claw length, hardness) and on the prevalence of claw diseases. At claw trimming, a total of 240 Holstein-Friesian or Red-Holstein cows from 20 German farms were examined twice, at the end of the pasture and barn season. Each individual claw was trimmed at both assessments. Farms were classified based on animals' pasture access during pasture season into: group 1 (G1) >10 hours pasture access per day, group 2 (G2) 6-10 hours, group 3 (G3) <6 hours and group 4 (G4) without pasture access. Greater values for hardness were associated with lower scores (=prevalence×severity level) of sole ulcers, white line disease, sole haemorrhage, heel horn erosion and interdigital hyperplasia. In pasture groups, heel horn erosion showed lower frequencies in summer compared with winter, while it was vice versa in G4. In G1 and G3, lower frequencies of white line disease were found in summer compared with winter. Overall, pasture access had positive effects in particular for claw diseases that are related to moist environments. Maternally derived antibodies can provide partial protection against certain bacterial and viral infections. We investigated whether chicks descending from nematode-infected hens are more resistant against Ascaridia galli, a prevalent gastrointestinal nematode, than chicks from nematode-free mothers. One-day-old chicks (N=153) from infected (mab+; maternal antibody+) or uninfected control dams (mab-) were experimentally infected with A. galli at two different levels (100 or 1000 eggs/chick). The worm burdens of the chicks were determined at 6 weeks post infection. There was a high correlation (r=0.89) between A. galli-specific antibody concentrations in dam plasma and egg yolk. There was no difference between worm burdens of chicks descending from infected or uninfected dams (P=0.892), indicating no maternally derived protection against A. galli. Chicks receiving the higher infection dose had higher worm burdens (P<0.05). Although there was no difference (P>0.05) between worm counts of female and male chicks infected with 100 eggs, females chicks infected with 100 eggs harboured longer and heavier female worms. Experimental infection models for Ascaridia galli rely on the use of eggs isolated either directly from worm uteri or from host faeces. We investigated whether A. galli eggs isolated from the two sources differ in their embryonation ability. A. galli eggs originating from 12 worm infrapopulations were isolated both from faeces of the living host (faecal eggs) and directly from worm uteri after host necropsy (uterine eggs). The isolated eggs from each infrapopulation and source were incubated in Petri dishes (n=24) containing a potassium-dichromate (0.1%) medium for 28 days (d) at room temperature. Starting from the day of egg isolation (d0), in ovo larval development was evaluated every second day by examining morphological characteristics of 200 eggs/petri dish. A total of 72,000 eggs were classified into undeveloped, early development, vermiform or fully embryonated stages. Isolation procedures caused similar damage to uterine and faecal eggs (2.2% and 0.5%, respectively; P=0.180). The first sign of in ovo embryonic development in faecal eggs (7%) was observed during the 24-h period when faeces were collected. On d28, a higher percentage of uterine eggs remained undeveloped when compared with faecal eggs (58.6% vs 11.0%; P<0.001). Although a higher (P<0.001) percentage of faecal eggs entered both the early developmental and vermiform stages, which took place primarily within the first two weeks of incubation, there was no time-shift between the development of faecal and uterine eggs. Starting from day 10, higher (P<0.05) percentages of faecal eggs completed embryonation compared with uterine equivalents. Eggs from both sources reached a plateau of embryonation by the end of 2nd week of incubation, with faecal eggs having a greater than two-fold higher embryonation ability. Cumulative mortality was higher in uterine eggs (14.3%) than in faecal eggs (0.2%). The aim of the present study was to assess the behaviour of llamas (Lama glama) and German blackhead mutton sheep (Ovis orientalis forma aries) when kept under Central European grazing conditions. In total, six adult female sheep and six adult female llamas were observed by direct observation during one week, in which each group was observed for a total time of 24 h. The animals were kept on the same pasture, but the species were raised in separate plots. Forage height before and after the experimental period were determined using a rising plate meter to calculate the average daily dry matter intake (DMI). Llamas had a daily DMI of 0.85%/BW and sheep of 1.04%/BW, respectively. The following behaviours were recorded by direct observation: grazing standing up, grazing lying down, ruminating standing up, ruminating lying down, lying down, lying down lateral and standing. Both species grazed for more than 50% of the time. Ruminating was predominantly performed while standing and lying by sheep (about 50% of the night and 12% of the day) and while lying by llamas (54% of the night and 10% of the day). In conclusion, sheep and llamas differed in grazing behaviour and daily biorhythm. These differences indicate that sheep and llamas may not synchronize their behaviour when co-grazed, though particularly in co-grazing studies the observation period should be extended.

We investigated whether nematode egg excretion through feces of naturally or experimentally infected chickens follow certain patterns within a day, which may allow determining the most appropriate sampling time for the highest parasite egg concentration. Feces samples (n=864) from chickens (n=36) with naturally occurring mixed nematode infections (trials N1, N2) or with an experimental Ascaridia galli infection (E) were collected quantitatively every 4h for four consecutive days. Number of eggs per gram of feces (EPG) was determined, and accumulative egg output (AEO) at each sampling time as well as total number of eggs excreted within 24h (eggs per day, EPD) were then estimated. At the end of the collection period, the hens were necropsied and their worm burdens determined. Naturally infected hens harbored Heterakis gallinarum (100%), Capillaria spp. (95.7%) and A. galli (91.3%). The experimental A. galli infection produced patent infections in all the birds. In general, both fecal egg concentration (EPG) and the amount of feces increased (P<0.05) sharply from the early morning to early-noon (10:00 a.m.) and remained at a high level until evenings which thereafter decreased to their initial levels during the night both in naturally and experimentally infected birds. This resulted in a more apparent increase or a decrease in AEO at the corresponding time points, respectively, and led to much higher egg excretions during the daytime than the nights. Despite the apparent within day fluctuations in egg excretion, neither EPG (P=0.704) nor AEO (P=0.499) nor EPD (P=0.149) was significantly different among the four collection days. Similarly, there was no significant interaction (P>0.05) between effects of sampling hours and days on EPG and AEO, suggesting the existence of repeatable diurnal fluctuations within each day. Although an association between climatic parameters (e.g., ambient temperature and relative humidity) and the nematode egg excretion was quantified, a causal relationship could not be demonstrated. We conclude that nematode egg excretion through chicken feces in both natural and experimental infections shows repeatable diurnal fluctuations, which may indicate adaptive strategies by nematodes and eventually favor parasite spread. The number of South American camelids (SAC) is increasing in Germany since decades. Due to a lack of scientifically based publications the knowledge about feeding SACs is still poor. Therefore, the aim of this study was to estimate the dry matter intake (DMI) of SACs as a basis for calculations of feed rations. Previous studies proposed a DMI of up to 3% of the body weight (BW) (Vaughan und Gauly, 2011). In the present study, eight llamas (Llama glama) were allocated to two groups of four animals each. The two groups were fed with hay of different qualities over a total period often weeks, which was divided into two runs of five weeks each. During the first run, group 1 was fed with hay 1 (15.1% crude protein; 8.5% crude ash; 3.1% crude fat; 52.6% NDF per kg DM) and group 2 with hay 2 (6.6% crude protein; 6.2% crude ash; 2.1% crude fat; 64.3% NDF per kg DM). After five weeks the groups were changed and group 1 received hay 2 and group 2 received hay 1. BW was measured at the start and end of each run (week zero, five and ten). The hay quality affected the DMI, but the animals did not compensate a lower feed quality with an increased DMI. The total DMI was 1.26% and 0.89% of the BW for hay 1 and hay 2, respectively, which was lower than expected in both groups. In conclusion, calculations of feed rations for SACs should be adjusted to the present findings of a lower DMI capability.

The objective of this study was to monitor establishment and development of gastro-intestinal helminth infections in chickens over two production years (PY) on a free-range farm in Lower Saxony, Germany. The data were collected between July 2010 and June 2011 (PY1) and July 2011 and January 2013 (PY2), respectively. During PY1, Lohmann Brown classic (LB classic, N = 450) was tested, while in PY2 two different genotypes (230 LB classic, 230 LB plus) were used. The hens were kept in two mobile stalls that were moved to a new position at regular intervals. In both PY1 and PY2, 20 individual faecal samples per stall were randomly collected at monthly intervals in order to calculate the number of internal parasite eggs per gram of faeces (EPG). At the end of the laying periods, approximately 10% (N = 42) or more than 50% (N = 265) of hens were subjected to post-mortem parasitological examinations in PY1 and PY2, respectively. No parasite eggs were found in the faecal samples during PY1, whereas almost all of the hens (97.6%) were infected with Heterakis gallinarum (36 worms/hen) at the end of the period. In PY2, nematode eggs in faeces were found from the third month onwards at a low level, increasing considerably towards the final three months. There was no significant difference between the two genotypes of brown hens neither for EPG (P = 0.456) or for overall prevalence (P = 0.177). Mortality rate ranged from 18.3 to 27.4% but did not differ significantly between genotypes or production years. Average worm burden was 207 worms/hen in PY2. The most prevalent species were H. gallinarum (98.5%) followed by Ascaridia galli (96.2%) and Capillaria spp. (86.1%). Furthermore, three Capillaria species, C. obsignata, C. bursata and C. caudinflata were differentiated. In conclusion chickens kept on free-range farms are exposed to high risks of nematode infections and have high mortality rates with no obvious link to parasite infections. Once the farm environment is contaminated with the nematode eggs, establishment and further spread of nematodes to the hens is a matter of time. This will latest be the case in the second production period, even if the hens are kept in a rotation system and in small herd sizes. This underlines the importance of nematode infections in all free-range systems.

Isoflurane-anaesthesia combined with an analgesic represents a welfare-friendly method of pain mitigation for castration of piglets. However, it requires an inhaler device, which is uneconomic for small farms. Sharing a device among farms may be an economical option if the shared use does not increase labour time and the resulting costs. This study aimed to investigate the amount and components of labour time required for piglet castration with isoflurane anaesthesia performed with stationary and shared devices. Piglets (N = 1579) were anaesthetised with isoflurane (using either stationary or shared devices) and castrated.The stationary devices were used in a group (n = 5) of larger farms (84 sows/farm on an average), whereas smaller farms (n = 7; 32 sows/farm on an average) shared one device. Each farm was visited four times and labour time for each process-step was recorded. The complete process included machine set-up, anaesthesia and castration by a practitioner, and preparation, collection and transport of piglets by a farmer. Labour time of the complete process was increased (P = 0.012) on farms sharing a device (266 s/piglet) compared to farms using stationary devices (177 s/ piglet), due to increased time for preparation (P = 0.055), castration (P = 0.026) and packing (P = 0.010) when sharing a device. However, components of the time budget of farms using stationary or shared devices did not differ significantly (P > 0.05). Cost arising from time spent by farmers did not differ considerably between the use of stationary (0.28 Euro per piglet) and shared (0.26 Euro) devices. It is concluded that costs arising from the increased labour time due to sharing a device can be considered marginal, since the high expenses originating from purchasing an inhaler device are shared among several farms.

Castration of piglets with the use of isoflurane-anaesthesia in combination with an analgesic has been proven to be a welfare-friendly approach. However, castration is performed with an equipped anaesthetic device which is not profitable for small farms. Thus, this study aimed at investigating whether sharing the anaesthetic device among farms results in an elevated risk of bacteriological contamination and further spreading in spite of a thorough disinfection. The study included two groups of organic farms. Piglets (n = 1579) were anaesthetised with isoflurane supplied from an equipped device and castrated. Stationary anaesthetic devices were used in the first group of farms, whereas farms in the second group shared one device. Each farm was visited four times and the colony forming units (CFU) of total mesophilic bacteria, Staphylococcus spp., Methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli were determined. Sampling took place before castration and after disinfection, and included snout masks, retaining fixtures, pedals and wheels of the device (n = 376). The results indicated presence of Staphylococcus species in 56.5% and 40.3% of samples obtained from farms using stationary and shared devices, respectively. MRSA was detected in 2.4% of the samples and only one pathogenic Shiga toxin-producing E. coli (Stx2e, F18) could be detected. Bacterial counts were lower on the shared device than on the stationary devices (p = 0.038), particularly on restraining fixtures (p < 0.05). In both groups wheels were the most and pedals the least contaminated materials (p < 0.05). It is concluded that sharing an anaesthetic device on several farms does not increase bacteriological contamination after a thorough disinfection and thus imposing no additional hygiene risk to farms with similar hygienic conditions.

During the fattening period of a broiler flock four different litter materials (peat, chopped straw, shavings, silage) were tested simultaneously.The separated sections were tested for the presence of Salmonella bacteria using the sock-sampling method as described in the regulation EC No. 646/2007 with slight modifications in the sampling technique and the laboratory protocol. In addition, some chemical and physical parameters regarding litter quality and house climate were analysed. Samples were taken at day 0 (housing of animals), day 14, and day 30. At the end of the fattening period Salmonella Paratyphi B (d-tartrate +), representing a common strain in German broiler flocks, were isolated from culture. According to the various types of bedding materials some differences in the frequency of Salmonella detection were found. With three samples out of six samples being positive, wood shavings showed the highest Salmonella prevalence, followed by peat with two positive results. Corn silage was found to be a suitable alternative for common litter materials and revealed only one positive sample. Chopped straw was found to be free of Salmonella in our study. However, the latter had a higher risk for negative side effects concerning animal health and house climate. The study showed that the choice of an appropriate litter material might be of considerable importance in order to decrease the Salmonella burden within poultry flocks.

The aim of this study was to estimate the effects of flying insects (Order Diptera) on the behaviour of grazing horses in relation to the use of insecticide and repellent substances. The investigations were done between June and August in 2008 in 3 periods of 7 days each. As insecticide and repellent two substances were used: "Well-care emulsion" (Co. Essex Tierarznei, München, GER) containing Permethrin and "Bremsen-Frei-Plus" (Co. Dr. Schaette AG, Bad Waldsee, GER) based on etheric oils. Both groups were compared with a non treated control group in a crossover-design. Each group (n = 3-5) was used alternately as control and treatment group. Several climate parameters were taken during the study. Furthermore, the number of insects per animal was estimated at certain times. Once per observation period insects were caught using Malaise traps and differentiated by species. The proportion of horse relevant species of the genera Diptera, especially Culex pipiens and Musca autumnalis, caught was 9% on an average. There was no correlation between the number of Tabanidae caught in the Malaise traps and the number observed near by the horses. Behaviour parameters like tailswishing, headshaking, stamping, skintwitching, snapping at the body, and moving were observed more frequently with increasing insect infestation. When horses were infested with a high number of flying insects, feeding activity was significant lower, whereas locomotion activity was significant higher. Both substances had positive effects for about 50 hours after application with no apparent difference between the substances. However, a lower frequency of headshaking and tailswishing could be observed in the Permethrin treated horses.
