Staphylococcus aureus is the most frequent aetiology of bone and joint infections (BJI) and can cause relapsing and chronic infections. One of the main factors involved in the chronicization of staphylococcal BJIs is the internalization of S. aureus into osteoblasts, the bone-forming cells. Previous studies have shown that S. aureus triggers an impairment of osteoblasts function that could contribute to bone loss. However, these studies focused mainly on the extracellular effects of S. aureus. Our study aimed at understanding the intracellular effects of S. aureus on the early osteoblast differentiation process. In our in vitro model of osteoblast lineage infection, we first observed that internalized S. aureus 8325-4 (a reference lab strain) significantly impacted RUNX2 and COL1A1 expression compared to its non-internalized counterpart 8325-4∆fnbAB (with deletion of fnbA and fnbB). Then, in a murine model of osteomyelitis, we reported no significant effect for S. aureus 8325-4 and 8325-4∆fnbAB on bone parameters at 7 days post-infection whereas S. aureus 8325-4 significantly decreased trabecular bone thickness at 14 days post-infection compared to 8325-4∆fnbAB . When challenged with two clinical isogenic strains isolated from initial and relapse phase of the same BJI, significant impairments of bone parameters were observed for both initial and relapse strain, without differences between the two strains. Finally, in our in vitro osteoblast infection model, both clinical strains impacted alkaline phosphatase activity whereas the expression of bone differentiation genes was significantly decreased only after infection with the relapse strain. Globally, we highlighted that S. aureus internalization into osteoblasts is responsible for an impairment of the early differentiation in vitro and that S. aureus impaired bone parameters in vivo in a strain-dependent manner.

It is common to initiate a long-term corticosteroid therapy for inflammatory diseases. Various specialists are involved in this prescription, and associated measures to prevent side effects are not consensual, with the exception of osteoporosis. The specialty of the prescriber has indeed a significant impact on the attention paid to the adjuvant associated measures. The aim of this review was to draw a summary of the side effects of long-term corticosteroid therapy and of the existing recommendations related to associated measures to prevent them. Unfortunately, it is difficult to give clear recommendations because of the lack of evidence in some fields, especially as they should be adapted to patient's age and comorbidities. We propose a summary table of associated measures to long-term steroid therapy prescription and suggest a monitoring frequency.

We present a database of cerebral PET FDG and anatomical MRI for 37 normal adult human subjects (CERMEP-IDB-MRXFDG). Thirty-nine participants underwent static [18F]FDG PET/CT and MRI, resulting in [18F]FDG PET, T1 MPRAGE MRI, FLAIR MRI, and CT images. Two participants were excluded after visual quality control. We describe the acquisition parameters, the image processing pipeline and provide participants’ individual demographics (mean age 38 ± 11.5 years, range 23–65, 20 women). Volumetric analysis of the 37 T1 MRIs showed results in line with the literature. A leave-one-out assessment of the 37 FDG images using Statistical Parametric Mapping (SPM) yielded a low number of false positives after exclusion of artefacts. The database is stored in three different formats, following the BIDS common specification: (1) DICOM (data not processed), (2) NIFTI (multimodal images coregistered to PET subject space), (3) NIFTI normalized (images normalized to MNI space). At its most basic level, the sense of self is built upon awareness of one's body and the face holds special significance as the individual's most important and distinctive physical feature. Multimodal sensory integration is pivotal to experiencing one's own body as a coherent visual “self” representation is formed and maintained by matching felt and observed sensorimotor experiences in the mirror. While difficulties in individual facial identity recognition and in both self-referential cognition and empathy are frequently reported in individuals with autism spectrum disorder (ASD), studying the effect of multimodal sensory stimulation in this population is of relevant interest. The present study investigates for the first time the specific effect on Interpersonal Multisensory Stimulation (IMS) on face self-recognition in a sample of 30 adults with (n = 15) and without (n = 15) ASD, matched on age and sex. The results demonstrate atypical self-face recognition and absence of IMS effects (enfacement illusion) in adults with ASD compared to controls, indicating that multisensory integration failed in updating cognitive representations of one's own face among persons with this disorder. The results are discussed in the light of other findings indicating alterations in body enfacement illusion and automatic imitation in ASD as well as in the context of the theories of procedural perception and multisensory integration alterations.

We show that by lending to merchants and monitoring them, an e-commerce platform can price-discriminate between merchants with high and low financial constraints: the platform offers credit priced below market rates and designed to select merchants with lower capital or collateral while simultaneously increasing the platform’s access fees. The credit market then becomes endogenously segmented with banks focusing on less financially constrained borrowers. Lending by the platform expands with its monitoring efficiency but can arise even when the platform is less efficient than banks at monitoring. Platform credit benefits more financially constrained merchants as well as buyers, but can hurt less financially constrained merchants if cross-side network effects with buyers are too small. The platform’s propensity to offer credit and the financial inclusion of more constrained merchants depends on the platform’s market power.

We study a cost-sharing mechanism where a content provider contributes to covering the costs incurred by a network operator when delivering content to consumers. The costshare not only boosts the content provider's incentives to moderate trac but also aects the price composition for consumers buying access and content. We show the overall eect on consumer welfare depends on the content provider's ability to monetize users. When that ability is high, introducing a cost-share can lead to lower overall prices and higher consumer welfare. We study the robustness of this result to long-term investments in cost reduction by the operator and to heterogeneity in consumers' taste for content. In extensions with multiple contents and multiple operators, contractual externalities arise that suggest a role for regulation.

The microstructural evolutions occurring during the thermo-hydro-glycol ageing of an injection molded PA66 were studied. They were correlated to the evolutions of its mechanical properties. The aged samples were immersed in an antifreeze fluid—mainly composed of water and ethylene glycol—at varying times and temperatures. The aim was to combine an as exhaustive as possible microstructural investigation and a rigorous mechanical analysis. Consequently, the microstructure of the aged and unaged PA66 was assessed through the average molar mass, the diameter of the spherulites, the lamellae thickness, the crystallite’s apparent size, a crystal perfection index, and a crystallinity index. Moreover, a core-skin approach was set up. The mechanical consequences of the microstructural changes were investigated by DMA and tensile testing. The local true strain fields were measured with a digital image correlation system. The temperatures and strain rates of the tests were chosen by referring to the time-temperature superposition principle. It is concluded that the water and ethylene glycol intake resulted in an intense plasticization, the loss of the molar mass resulted in the embrittling of the polymer, and finally, it was identified that the changes of the crystalline structure have an influence on the stiffness of PA66.

(1) Background: Autism spectrum disorder (ASD) is a neurodevelopmental disorder that is highly associated with various somatic conditions that can be masked by the core symptoms of ASD and thus complicate the diagnosis. Identifying co-occurring somatic disorders is critical for providing effective healthcare and social services for ASD populations and influences their long-term outcomes. A systematic assessment of co-occurring somatic conditions is essential during this ASD diagnostic process. Therefore, this study aimed to identify the organization and content of the initial somatic assessment (ISA). (2) Methods: We conducted a systematic review of the clinical practice guidelines (CPG) for the ASD diagnostic process published between January 2005 and December 2019 in English and French and performed an appraisal following the Appraisal of Guidelines Research and Evaluation, second edition (AGREE-II). (3) Results: We selected 14 CPGs that were heterogeneous in quality, with methodological scores between 32.3 and 91.9. Clinical examinations are the first step in the ISA, and the participation of pediatric, neuropediatric, and genetic specialists was highly recommended by the majority of the CPGs. The recommendations included hearing screening tests (10/14), visual examinations (8/14), and systematic genetic investigations (4/14). The CPGs also described additional investigations that should be conducted based on numerous warning signs. (4) Conclusions: Screening for consensual international warning signs is necessary to perform a comprehensive and systematic ISA during the ASD diagnostic process. A “referral form” could be used to guide clinicians and improve the coordination process. This tool may reinforce epidemiological data on co-occurring somatic disorders in patients with ASD.

Ventricular preexcitation secondary to anterograde conduction through an accessory pathway was diagnosed in two Golden Retriever dogs. Both dogs demonstrated similar segmental myocardial thinning and systolic dyskinesia of the basal interventricular wall on echocardiography. These changes are widely recognised in people with ventricular preexcitation but have not been previously described in dogs. Ventricular preexcitation should be considered as a potential cause for segmental wall motion abnormalities in these two dogs.

Resume L’incidence des fractures de fragilite augmente progressivement apres 50 ans et la proportion de la population mondiale souffrant d’osteoporose et de fractures augmentera inevitablement sous l’effet du vieillissement general. Les consequences des fractures sont plus graves chez les personnes âgees : la plupart des fractures a faible energie cinetique ont ete associees a un risque de mortalite accru et le risque de nouvelle fracture osteoporotique majeure apres une premiere fracture augmente egalement avec le vieillissement. De meme que la diminution de la densite minerale osseuse, les chutes jouent un role majeur dans la survenue de fractures de fragilite chez les personnes âgees et l’evaluation du risque de chute fait partie integrante de l’evaluation du risque de fracture. Malgre les progres realises dans le diagnostic de l’osteoporose et l’evaluation du risque de fracture, et malgre la disponibilite de nombreux traitements anti-osteoporotiques efficaces, dont des medicaments administres par voie parenterale qui peuvent ameliorer l’observance, de nombreuses donnees indiquent que l’osteoporose des personnes âgees est insuffisamment prise en charge.

International audience; This paper presents a thorough analysis of direct microwave heating as a sintering process of ceramic materials. It questions why susceptor-assisted microwave heating is used in most experimental works, although direct microwave coupling is preferable for taking advantage of the possible beneficial effects of the microwave field on the sintering phenomena. This issue was investigated by conducting both experiments and numerical simulations. The experiments consisted of sintering alumina and yttria doped zirconia powder samples in a 2.45 GHz resonant cavity with automatic thermal monitoring, whereas the numerical simulations coupled electromagnetics, thermal transfer and sample deformation. Alumina and yttria doped zirconia are widely used materials and they exhibit different microwave field behaviours (transparent and absorbent, respectively), which are representative of most ceramic materials. The influence of the insulating material was discussed by considering different sintering cell designs. The very low coupling capacity of alumina made its direct heating very difficult. It was therefore necessary to apply a strong electric field to heat it. This situation promoted the absorption of microwave energy by other elements such as the insulating material, leading to heating instabilities and degradation of the insulation cell. In the case of zirconia, its coupling properties change abruptly with increasing temperature. It is poorly absorbent at low temperature, highly absorbent at intermediate temperature and it finally becomes reflective at the end of the sintering process. The consequences of this behaviour are (i) a very difficult control of direct heating (ii) a propensity to form damaging hot spots and (iii) the impossibility to reach high temperatures without forming plasma. Therefore, this experimental and numerical study showed that direct microwave heating is not suitable for conducting reliable and homogeneous sintering of classical ceramics. This explains why susceptor-assisted heating is most of time preferred.
