423. Determination of Photocyanine in Human Serum by HPLC and Application to Pharmacokinetic Study

Abstract

Photocyanine, a novel amphoteric phthalocyanine drug, showed favorable anticancer activity in vivo. Pharmacokinetic study in cancer patients is an important component in dose administration choice. In this study, a rapid, sensitive analytical method based on high-performance liquid chromatography with ultraviolet detection was developed and validated for the determination of four isomers of photocyanine (FD1-4) in cancer patients. Sample preparation involved liquid-liquid extraction with a combination of ultrasound and N,N-dimethyl formamide. Calibration curves (1/x(2)) offered satisfactory linearity for the four isomers of photocyanine. The lower limit of quantification (LLOQ) for FD1-3 isomers was 30 ng/mL, and LLOQ for FD-4 was 5 ng/mL. Inter- and intra-day accuracies for four isomers ranged from 96.6 to 105.5%, and 95.0 to 103.6%, respectively. Inter- and intra-day precision ranged from 4.8 to 8.9%, and 3.6 to 12.2%, respectively. Stability studies showed that photocyanine was stable. This method was successfully used to quantify photocyanine in a pharmacokinetic study in which a single-dose of photocyanine (0.1 mg/kg) was intravenously administered to patients with cancer. On the basis of the discovery that photocyanine has a half-life of 57.5 h in vivo, we suggest that avoiding light for a longer period is essential for patients undergoing photocyanine therapy.

Abstract

Objective We conducted a study to understand the genetic effect of ERCC1 and RRM1 on the chemotherapy response and clinical outcome of Non-Small Cell Lung Cancer (NSCLC). Methods The relative cDNA quantification for ERCC1 and RRM1 was conducted using a fluorescence-based real-time detection method among 294 NSCLC patients. Results Compared with the internal reference gene β-action, the median levels of ERCC1 and RRM1 expression were 2.43×10(-2) and 0.11×10(-2), respectively. Our study showed response to platinum-containing regimen chemotherapy was high in those with high ERCC1 expression, and the OR (95% CI) were 1.73(1.06-2.81). An apparently high response to chemotherapy was decreased when patients were carrying both high expression of ERCC1 and RRM1, with OR (95% CI) of 2.57(1.21-4.90). Patients with high expression of ERCC1 were associated with a longer OS by multivariate Cox proportional hazards model. Moreover, those carrying both high levels of ERCC1 and RRM1 were seem to have a longer OS when compared with those with low expression (HR=0.31, 95% CI=0.13-0.62 for OS). Conclusion This observation could be used in personalized chemotherapy, increase the response rate and prolonged survival time, and could encourage to explore the predictive value of other genes.

425. Atmospheric evidence for a global secular increase in carbon isotopic discrimination of land photosynthesis.

Abstract

A decrease in the 13C/12C ratio of atmospheric CO2 has been documented by direct observations since 1978 and from ice core measurements since the industrial revolution. This decrease, known as the 13C-Suess effect, is driven primarily by the input of fossil fuel-derived CO2 but is also sensitive to land and ocean carbon cycling and uptake. Using updated records, we show that no plausible combination of sources and sinks of CO2 from fossil fuel, land, and oceans can explain the observed 13C-Suess effect unless an increase has occurred in the 13C/12C isotopic discrimination of land photosynthesis. A trend toward greater discrimination under higher CO2 levels is broadly consistent with tree ring studies over the past century, with field and chamber experiments, and with geological records of C3 plants at times of altered atmospheric CO2, but increasing discrimination has not previously been included in studies of long-term atmospheric 13C/12C measurements. We further show that the inferred discrimination increase of 0.014 ± 0.007‰ ppm-1 is largely explained by photorespiratory and mesophyll effects. This result implies that, at the global scale, land plants have regulated their stomatal conductance so as to allow the CO2 partial pressure within stomatal cavities and their intrinsic water use efficiency to increase in nearly constant proportion to the rise in atmospheric CO2 concentration.

Table tennis, as one of the most popular sports in China, has grown considerably since its developed in the 19th century in England. The biomechanics of the research methods in many sports has been widely used, for understanding of sports and technology and improve sports played an important role Fast break and curving ball technology is this game’s core technology. In this study, we based on fast break and curving ball features of kinematic to reveal the table tennis forehand techniques. Eight male volunteers were participated in this tests, the speed of the racket during the playing was recorded through Vicon Motion Capture System. The action was divided into three major phases: back swing, attack and follow through. At the end of back swing stage, break and curl technologies, the speed parameter shows some differences. While the peak speed in ball contact frame, the speed of curling ball was significantly higher than the fast break. Further study could be carried out in detailing analysis at sub-stage of the action for integral considering.

429. Micro-Pulse Lidar Measurements of Aerosol Vertical Structure over the Loess Plateau

Abstract

Knowledge of the vertical distribution of aerosols in the free troposphere is important for estimating their impact on climate. In this study, direct observations of the vertical distribution of aerosols in the free tropo- sphere are made using surface Micro-Pulse Lidar (MPL) measurements. The MPL measurements were made at the Loess Plateau (35.95°N, 104.1°E), which is near the major dust source regions of the Taklimakan and Gobi deserts. The vertical distribution of the MPL backscattering sug- gested that non-dust aerosols floated from ground level to an altitude of approximately 9 km around the source re- gions. Early morning hours are characterized by a shallow aerosol layer of a few hundred meters thick. As the day progresses, strong convective eddies transport the aero- sols vertically to more than 1500 m.
