This study includes modeling and simulation of insulin aspart pharmacokinetics (PK). The authors used PK data of biosimilar insulins—insulin aspart and biphasic insulin aspart 30/70—to develop a predictive population PK model for the insulins. The model was built via Monolix software, taking into account the weight‐based dosing and the dose and body‐weight effects on the parameters. The model‐based simulations were performed using the R package mlxR for various administered doses and various ratios of insulin aspart forms for a better understanding of the insulin behavior. The optimal model was a 1‐compartment model with a combination of zero‐ and first‐order absorptions, with absorption lag for the soluble form of insulin aspart and first‐order absorption for the insulin aspart protamine suspension. The assumption of identical behavior of 2 insulins at the distribution and elimination phases was made. The developed PK model was fitted successfully to the experimental data, and all fitted parameters displayed a moderate coefficient of variation. The PK model allows us to predict PK profiles for various doses and formulations of insulin aspart and can be used to improve the accuracy, safety, and ethics of novel clinical trials of insulin. 404. <SUP>99m</SUP>Tc-Labeled Neuropeptide Y Analogues as Potential Tumor Imaging Agents

- Abstract
The possible use of neuropeptide Y (NPY) as a novel radiopeptide has been investigated. NPY is a 36-amino acid peptide of the pancreatic polypeptide family, which is expressed in the peripheral and central nervous system, and is one of the most abundant neuropeptides in the brain. Its receptors are produced in a number of neuroblastoma and the thereof derived cell lines. As structure−activity relationships of NPY are well-known, we could assume where a radionuclide might be introduced without affecting receptor affinity. We applied the novel [99mTc(OH
2 )3 (CO)3 ]+ aqua complex and PADA (2-picolylamine-N,N-diacetic acid) as bifunctional chelating agent. The peptides were synthesized by solid-phase peptide synthesis, and PADA was coupled to the side chain of Lys4 of the resin-bound peptide. Upon postlabeling of [K4(PADA)]-NPY, 99mTc(CO)3 did not only bind to the desired PADA, but presumably as well to the His in position 26. Since the replacement of His26 by Ala only slightly decreased binding affinity, [K4(PADA),A26]-NPY was specifically postlabeled, and the 185Re surrogate maintained high binding affinity. Furthermore, the prelabeling approach has been applied for the centrally truncated analogue [Ahx5-24]-NPY, which is highly selective for the Y2 receptor. The resulting Ac-[Ahx5-24,K4(99mTc(CO)3 -PADA)]-NPY was produced with a yield of only 16%. Therefore, postlabeling was applied for the short analogue as well, again substituting His26 by Ala. Competitive binding assays using 185Re as a surrogate for 99mTc showed high binding affinity of Ac-[Ahx5-24,K4(185Re(CO)3 -PADA),A26]-NPY. Internalization studies with the corresponding 99mTc-labeled analogue revealed receptor-mediated internalization. Furthermore, biodistribution studies were performed in mice, and stability was tested in human plasma. Our centrally truncated analogue revealed a 6-fold increased stability compared to the natural peptide NPY. We conclude that Ac-[Ahx5-24,K4(99mTc(CO)3 -PADA),A26]-NPY has promising characteristics for future applications in nuclear medicine.- Published

406. Influence of the Denticity of Ligand Systems on the in Vitro and in Vivo Behavior of <SUP>99m</SUP>Tc(I)−Tricarbonyl Complexes: A Hint for the Future Functionalization of Biomolecules

Functionalization of biologically relevant molecules for the labeling with the novel fac-[99mTc(OH
2 )3 (CO)3 ]+ precursor has gained considerable attention recently. Therefore, we tested seven different tridentate (histidineL 1 , iminodiacetic acidL 2 , N-2-picolylamineacetic acidL 3 , N,N-2-picolylaminediacetic acidL 4 ) and bidentate (histamineL 5 , 2-picolinic acidL 6 , 2,4-dipicolinic acidL 7 ) ligand systems, with the potential to be bifunctionalized and attached to a biomolecule. The ligands allowed mild radiolabeling conditions with fac-[99mTc(OH2 )3 (CO)3 ]+ (30 min, 75 °C). The ligand concentrations necessary to obtain yields of >95% of the corresponding organometallic complexes1 -7 ranged from 10-6 to 10-4 M. Complexes of the general formula fac-[99mTcL(CO)3 ] (L = tridentate ligand) and fac-[99mTc(OH2 )L(CO)3 ] (L = bidentate ligand), respectively, were produced. Challenge studies with cysteine and histidine revealed significant displacement of the ligands in complexes5 −7 but only little exchange with complexes1 −4 after 24 h at 37 °C in PBS buffer. However, no decomposition to 99mTcO4 - was observed under these conditions. All complexes showed a hydrophilic character (log Po/w values ranging from −2.12 to 0.32). Time-dependent FPLC analyses of compounds1 −7 incubated in human plasma at 37 °C showed again no decomposition to 99mTcO4 - after 24 h at 37 °C. However, the complexes with bidentate ligands (5 −7 ) became almost completely protein bound after 60 min, whereas the complexes with tridentate coordinated ligands (1 −4 ) showed no reaction with serum proteins. The compounds were tested for their in vivo stability and the biodistribution characteristics in BALB/c mice. The complexes with tridentate coordinated ligand systems (1 −4 ) revealed generally a good and fast clearance from all organs and tissues. On the other hand, the complexes with only bidentate coordinated ligands (5 −7 ) showed a significantly higher retention of activity in the liver, the kidneys, and the blood pool. Detailed radiometric analyses of murine plasma samples, 30 min p.i. of complex fac-[99mTcL1(CO)3 ],1 , revealed almost no reaction of the radioactive complex with the plasma proteins. By contrast, in plasma samples of mice, which were injected with complex fac-[99mTc(OH2 )L5(CO)3 ]+,5 , the entire radioactivity coeluded with the proteins. On the basis of these in vitro and in vivo experiments, it appears that functionalization of biomolecules with tridentate-chelating ligand systems is preferable for the labeling with fac-[99mTc(OH2 )3 (CO)3 ]+, since this will presumably result in radioactive bioconjugates with better pharmacokinetic profiles.- Published

Introducción y objetivos: Actualmente, son escasos los estudios en la literatura que comparen los resultados de las técnicas de resección y de implante de neocuerdas para la corrección del prolapso de velo posterior en la insuficiencia mitral degenerativa y en su mayoría analizan los resultados a corto plazo exclusivamente. Este estudio se ha realizado para comparar los resultados a medio plazo entre estas 2 técnicas en nuestro medio, haciendo especial hincapié en el análisis del índice de coaptación de los velos. Métodos: Se ha incluido en el estudio a 98 pacientes con insuficiencia mitral degenerativa por prolapso de velo posterior como causa principal, corregido mediante implante de neocuerdas o resección cuadrangular, comparando los resultados clínicos y ecocardiográficos en el seguimiento. Resultados: A nivel perioperatorio, no se observaron diferencias en relación con los tiempos quirúrgicos, el riesgo de movimiento anterior sistólico o la insuficiencia mitral residual intraoperatoria. En cuanto al seguimiento posterior, no se observaron diferencias en la evolución clínica y ecocardiográfica, salvo en la reducción del diámetro telediastólico del ventrículo izquierdo, que fue superior en el grupo de resección (–12,3 ± 8,7 vs. –8,1 ± 8,5 mm, p

Limited research has focused on exploring the collaboration between home and school to address cyberbullying. In this qualitative study, we interviewed 17 school counselors to explore their experiences working with parents and caregivers in addressing cyberbullying. We identified four themes: (a) connections with parents and families, (b) digital citizenship, (c) blurred boundaries, and (d) family/counselor support. We discuss the themes and offer implications for school counselors to address cyberbullying in schools, including highlighting strategies for bridging the gap between families and schools through policies and communication about cyberbullying.

The mode of transportation that travels from Kupang City to Rote Ndao Regency is currently a mode of sea transportation and air transportation. This study aims to find out user information based on its mode choices and model it in the multinomial log function, besides that it's hoped that this result can be used as a reference in determining policies that improve functions and facilities for mobilizing people, goods and services carried out on the Kupang to Rote.Data analysis was carried out in two ways, namely with descriptive statistical analysis techniques to explain the characteristics of respondents and multinomial logistic regression analysis to create a model of mode transportation selection for Kupang-Rote service. The results of the study showed that users of transportation services from Kupang to Rote, generally men prefer to use ferry boats while women prefer to use fast boats. The characteristics of respondents is age 20-39 years. While the factors that influence the choice of transportation are the influence of luggage, weather and comfort. For the selection model, the probability of a speedboat is 34.65%,the probability of choosing a ferry is 42.96%, and the probability of choosing an aircraft is 22.3%.

- Abstract
Tilt data obtained from July 1983-January 1986 at three sites in the Central Apennines (Italy) have been examined. A filtering of raw recordings indicated the existence of residual anomalies. Attempting a correlation between such anomalies and regional seismicity (3.0 magnitudeM 5.4) has allowed us to interpret the anomalies as possible preseismic variations. The results seem to confirm the existence of a relation between precursor time T, earthquake magnitude Mand epicentral distance R. Finally, the presence of a time delay between the onsets of the anomalies detected at different sites for the same earthquake gives further credit to the possibility of the existence of their propagation.

We present an unusual case of hepatocellular carcinoma presenting as massive portal vein thrombosis with progression to the intrahepatic bile ducts without demonstrable primary hepatic tumor. Ultrasound, magnetic resonance, and percutaneous transhepatic cholangiography findings are described. Introducción y objetivos: En la sustitución valvular aórtica se considera que las prótesis no soportadas (PNS) revierten la hipertrofia ventricular izquierda en mayor cuantía que las prótesis soportadas (PS). No existe una comparativa en una población no anglosajona. Pretendemos detectar diferencias significativas analizando la supervivencia, la clase funcional de la New York Heart Association y la regresión del índice de masa ventricular izquierda, entre otros. Método: Estudio analítico retrospectivo, pareado mediante puntuación de propensión, de 120 pacientes intervenidos de forma programada de sustitución valvular aórtica aislada por estenosis aórtica severa desde abril de 2002 a marzo de 2009 con prótesis biológicas de pericardio bovino (60 PNS Freedom, 60 PS Mitroflow, Sorin Group) sobre un total de 138. Resultados: El seguimiento medio fue de 48,4 ± 22 meses. No se detectaron diferencias en la clase funcional New York Heart Association (media 1,78 ± 0,5 PNS, 1,72 ± 0, 6 PS, p = 0,616) ni en la regresión media del índice de masa ventricular izquierda, que fue de 42,2 g/m2 (25,3%) en PS y de 36,3 g/m2 (22,6%) en PNS (p = 0,467). Las PNS presentaron menores gradientes medios transaórticos (PS 14,4 ± 6,8 mmHg, PNS 9,3 ± 3,2 mmHg, p = 0,05) y mayores índices de áreas de orificio efectivo (PS 0,76 ± 0,1 cm2/m2, PNS 0,83 ± 0,1 cm2/m2, p = 0,04) dentro de los primeros 24 meses, igualándose con posterioridad. Tampoco existieron diferencias en la supervivencia (Mantel-Cox, p = 0,185). Conclusiones: Las PS de nueva generación son tan efectivas como las PNS, sin presentar diferencias estadísticamente significativas en supervivencia, clase funcional New York Heart Association ni regresión del índice de masa ventricular izquierda durante el seguimiento, incluso en pacientes con anillo aórtico de 19–21 mm.

Recent advances in image classification methods, along with the availability of associated tools, has seen their use become widespread in many domains. This paper presents a novel application of current image classification approaches in the area of emergency situation awareness. We discuss image classification based on low level features as well as methods built on top of pre-trained classifiers. The performance of the classifiers are assessed in terms of accuracy along with consideration to computational aspects given the size of the image database. Specifically, we investigate image classification in the context of a bush fire emergency in the Australian state of NSW where images associated with Tweets during the emergency were used to train and test classification approaches. Emergency service operators are interested in having images relevant to such fires reported as extra information to help manage evolving emergencies. We show that these methodologies can classify images into fire and not fire related classes with an accuracy of 86%.

Detailed QRS wave analysis from the limb leads of the surface high resolution electrocardiograms recorded in five cyclosporine-treated heart transplant recipients gave good correlation with the corresponding results of endomyocardial biopsy. That is when the result of the biopsies has identified the presence of rejection, a concomitant (p = 0.001) variation in some parameters of the QRS has been observed. The reproducibility of such parameters (established by means of the correlation coefficient r) gave r values ranging from 0.6 up to 0.93. The correlation regarded parameters analyzed both in time and frequency domain. Biopsy results were divided into two classes on the basis of the presence or absence of rejection. The most significant parameters obtained from high-frequency (25-300 Hz) ECGs within a few hours of each biopsy were: a) the total high-frequency voltage amplitude Vt of the QRS and the voltage amplitude of its initial Vi and middle thirds Vm; b) the peak voltage amplitude Vp of the QRS; c) the QRS duration L; d) the integrated voltage time product I of the QRS; e) three mean voltage amplitudes V5, V6, V7, of the power spectral density constructed on the basis of the 512-point fast Fourier transform applied on each recording. The above mentioned parameters appear to be useful in predicting the biopsy result in terms of the presence or absence of rejection.

Evidence is presented relating to the impossibility of micropore formation in the cellular substructure by a microshrinkage mechanism. A study is made of the detailed relationship between solidification substructure and microsegregation in dilute tin alloys. A method of investigating the instability of interfaces when Constitutional Supercooling exists in ternary alloys is developed. Interaction among the solutes under non-ideality conditions is considered. A cold microscope stage is developed for the study of predendritic growth in organic compounds such as ciclohexanol and carbon tetrabromide. The effect of scale up the ingot's size on structure and substructure is studied. Three sets of ingots of approximately 50 cc., 500 cc. and 5000 cc., is investigated.

The rapid Golgi method, combined with current optical and electronmicroscopica1 techniques, was used in three central gangliogliomas and in one dysplastic gangliocytoma of the cerebellum to study the morphology of ganglionic cells. Gangliogliomas were composed of bipolar, fusiform and radiate cells with dense core and clear vesicles in the perikaryon and cellular processes, the number of each cellular type varying from one case to another. These features, together with the fact that isodendritic neurons are considered to be phylogenetically old neurons, suggest that these tumours are composed of «primitive» neurons that are not homogeneous with regard to their morphology. In contrast, ganglionic cells in dysplastic gangliocytoma are huge cells with long, stereotyped neurites that establish unique asymmetric contacts with neighbouring perikarya and neurites by means of claw-shaped processes covered with synaptic buttons. These morphological characteristics are different from those of any other neuron of the CNS.

The age and size structure of a population of Pachyiulusfoetidissimus L. (Diplopoda) was studied in October 1973 in a broadleaved forest in the Caucasus. P. foetidissimus is the dominating mesofaunal species in soil animal communities in the region. Its population density reaches 30 ind mf2. The male-female ratio in the autumn population was 1: 1.6. Instars 9 and 10 dominated in the population and made up more than half of the total number. Representatives of instars 11 and 7-8 were also numerous. Males dominated among specimens of instar 11 and females among instars 7-8. Males of P. foetidissimus showed an increase of individuals of older instars compared to females. Insignificant sexual weight differences occurred in medium instars. The mean weight of females in instar 11 was double than that of males. Individual weights of animals varied from 360 to 1191 mg. Most numerous were individuals with body weights 150-350 mg represented by instars 8-9. Medium instars dominated in the autumn population, which was characterized by a high level of feeding activity and growth rates. Those groups dominated the influence of P. foetidissimus populations on decomposition of leaf litter and energy flow in the habitats studied.
