In 2010, the LGI1 protein was identified as the target of autoantibodies in a subgroup of autoimmune encephalitis. The main symptoms are tonic-dystonic motor seizures (TDS), which are atypical and pathognomonic of this disorder, and limbic seizures, which are common to other encephalitis. Several studies have shown a pathogenic role of anti-LGI1 antibodies, via an alteration of Kv1 channels and AMPA receptors. However, the link between these disturbances and the seizures has never been demonstrated. The main objectives of my PhD work were to determine whether anti-LGI1 antibodies were responsible for seizures and to investigate the underlying pathophysiological mechanisms. 1/ I first performed acute and chronic injections of patients’ anti-LGI1 antibodies into mice and rats brains in vivo. The electrophysiological recordings with surface EEG or intracranial electrodes did not show any epileptic-like activity.2/ Then, I sought to indirectly reproduce in vivo the disturbances induced by anti-LGI1 antibodies in vitro. For this purpose, I injected dendrotoxin-K (DTX), a specific Kv1.1 inhibitor, into the primary motor cortex of rats. All injected rats exhibited seizures, which characteristics were found similar as those observed in 12 patients suffered from anti-LGI1 encephalitis. 3/ Extracellular and intracellular neuronal recordings in vivo in sedated rats after injection of DTX allowed me to describe the neuronal activities associated with TDS, and to propose a neuronal scenario that could explain the high frequency of these seizures and their regularity., En 2010, la protéine LGI1 a été identifiée comme la cible d’anticorps dans un sous-groupe d’encéphalites auto-immunes. Les principaux symptômes sont des crises motrices toniques-dystoniques (TDS) pathognomoniques et atypiques, et des crises limbiques communes aux autres encéphalites. Plusieurs études ont montré un rôle pathogène des anticorps anti-LGI1, par une altération des canaux Kv1 et des récepteurs AMPA, mais le lien entre ces perturbations et les crises n’a jamais été établi. L’objectif de ma thèse a été de déterminer si les anticorps anti-LGI1 étaient responsables des crises d'épilepsie, et par quels mécanismes. 1/ J’ai d’abord réalisé des injections aiguës ou chroniques d’anticorps anti-LGI1 de patients, chez la souris et le rat in vivo. Les enregistrements électrophysiologiques, par des électrodes EEG de surface ou intracrâniennes, n’ont pas permis de mettre en évidence d’activité épileptique. 2/ J’ai ensuite cherché à reproduire in vivo les perturbations induites par les anticorps anti-LGI1, observées jusqu’alors in vitro. J’ai réalisé des injections de dendrotoxine-K (DTX), un inhibiteur spécifique de Kv1.1, dans le cortex moteur primaire de rats. L’ensemble des rats injectés ont présenté des crises, qui avaient les caractéristiques pathognomoniques des TDS observées dans une cohorte de 12 patients atteints d’encéphalite à anticorps anti-LGI1.3/ Par des enregistrements neuronaux extracellulaires et intracellulaires in vivo chez le rat sédaté après injection de DTX, j’ai pu décrire les activités neuronales associées aux TDS, et mettre en évidence un scénario neuronal qui pourrait expliquer la fréquence élevée de ces crises et leur survenue régulière.

We developed a new non-invasive integrated microsystem for electrophysiological measurements on Xenopus laevis oocytes. The Xenopus oocyte is a well-known expression system for various kinds of ion channels that are of potential interest for drug screening. In the traditional "Two Electrode Voltage Clamp" (TEVC) method, delicate micromanipulation is required to impale an oocyte with two microelectrodes. In our system, a non-invasive electrical access to the cytoplasm is provided by permeabilizing the cell membrane with an ionophore (e.g. nystatin). Unlike for the classical patch-clamp or "macropatch" techniques, this method does not require removal of the fibrous vitelline membrane. Cell handling is significantly simplified, resulting in more robust recordings with increased throughput. Moreover, because only part of the oocyte surface is exposed to reagents, the required volume of reagent solutions could be reduced by an order of magnitude compared to the TEVC method. In a second part of this work, a rapid fluidic exchange system was implemented on-chip to allow recording of fast kinetic events of exogenous ion channels expressed in the cell membrane. Reducing fluidic exchange times of extracellular reagent solutions is a great challenge with these large millimeter-size cells. Fluidic switching is obtained by shifting the laminar flow interface in a perfusion channel under the cell by means of integrated poly-dimethylsiloxane (PDMS) microvalves. Reagent solution exchange times down to 20 ms have been achieved. An on-chip purging system allows to perform complex pharmacological protocols, making the system suitable for screening of ion channel ligand libraries. The fabrication process for this disposable microchip, based on PDMS micromolding, is cost-efficient and simple. Moreover, an innovative integration of agar gel-based reference electrodes was developed. A conductive liquid junction is injected by capillary force filling of suitable microchannels, overcoming the problems encountered with the integration of Ag/AgCl thin film or wire reference electrodes. We tested this new microdevice by performing electrophysiological measurements on oocytes expressing the human Epithelial Sodium Channel (hENaC). Transmembrane currents could be recorded with a large signal-to-noise ratio. The performance of the integrated rapid fluidic exchange system was demonstrated by investigating the self-inhibition of hENaC. Our results show that the response time of this ion channel to a specific reactant is about one order of magnitude faster than estimated with the traditional TEVC technique. We conclude that this new microdevice has high potential for improved electrophysiological investigations with oocytes in the field of pharmacology and toxicology.

The present study aimed to investigate the effects of monoaminergic system lesionson the motor and non-motor functions, including anxiety, depression and circadian rhythmswithin the context of Parkinson’s disease. First, we developed appropriate animal models usingcombined paraquat/maneb (PQ/MB) intoxication or using selective lesions of monoaminergicsystems; second, we characterized the models using behavioral, biochemical andelectrophysiological approaches.In the first part of the study, we investigated the relevance of the PQ/MB model by studyingthe effects of combined PQ/MB on: (1) locomotor activity and motor coordination using theopen field and the rotarod test respectively, (2) anxiety and “depressive-like” behaviors usingthe elevated plus maze and the forced swim test respectively, (3) subthalamic nucleus neuronalactivity using extracellular single unit recordings and (4) tissue level of dopamine,noradrenaline and serotonin in the striatum and frontal cortex.Our data provide evidence that male Sprague Dawley rats are not equally sensitive to PQ/MBand that the observed motor deficits in vulnerable animals are not only a result of dopamineneuron degeneration, but may also be a consequence of peripheral disabilities. Nevertheless,the parkinsonian-like non-motor impairments may be a direct consequence of the bilateraldopamine depletion.Based on the results of the first part, we used the 6-OHDA rat model to investigate the effectsof DA cell degeneration, alone or combined with the noradrenaline (NA) and/or serotonin (5-HT) depletions, on the electrical activity of suprachiasmatic nucleus (SCN) neurons usingextracellular electrophysiological recordings. SCN is a key structure involved in the control ofcircadian rhythms. Our data provide the first evidence that monoamine depletions are at theorigin of changes in the firing activity of SCN neurons, suggesting new insight into theinvolvement of these electrical changes in the pathophysiology of circadian rhythms disruptionin PD., La présente étude a investi les effets des lésions des systèmes monoaminergiques surles fonctions motrices et non-motrices, y compris l’évaluation des comportements anxieux etdépressifs ainsi que les rythmes circadiens dans le contexte de la maladie de Parkinson (MP).D’abord, nous avons mis en place un modèle animal approprié en utilisant une intoxication auParaquat/Maneb (PQ/MB) ou avec des lésions sélectives des systèmes monoaminergiques.Ensuite, nous avons caractérisé les modèles en utilisant des approches comportementale,biochimique et électrophysiologique.Dans la première partie de cette étude, nous avons investi la validité du modèle PQ/MB parévaluer les effets du PQ/MB sur : (1) l’activité locomotrice et la coordination motrice enutilisant les tests de l’Open Field et du Rotarod, (2) les comportements anxieux et dépressif enutilisant les tests du labyrinthe en croix surélevé et de la nage forcée respectivement, (3)l’activité neuronale du noyau sous-thalamique en utilisant des enregistrements extracellulairesunitaires et (4) les concentrations tissulaires en dopamine, noradrénaline et sérotonine dans lestriatum et le cortex frontal.Nos résultats montrent que les rats Sprague Dawley mâles ne sont pas sensibles de la mêmefaçon au PQ/MB et que les déficits moteurs observés chez les animaux vulnérables ne sont passeulement le résultat d’une dégénérescence des neurones dopaminergiques, mais pourraient êtreaussi une conséquence des problèmes périphériques. Néanmoins, les troubles non-moteursobservés chez les animaux de tous les groupes traités pourraient constituer une conséquencedirecte de la déplétion dopaminergique bilatérale.En se basant sur les résultats de la première partie, nous avons utilisé le modèle du rat 6-OHDApour investir les effets de dégénérescence des cellules dopaminergique seule ou combinée avecles déplétions noradrénergique et sérotoninergique sur l’activité électrique des neurones dunoyau suprachiasmatique (SCN) en utilisant des enregistrements électrophysiologiquesextracellulaires. Le SCN est une structure clé impliquée dans le contrôle des rythmescircadiens. Nos résultats montrent pour la première fois que les déplétions monoaminergiquessont à l’origine des changements dans l’activité électrique des neurones du SCN, apportant desrésultats nouveaux qui suggèrent que les changements dans la décharge électrique des SCN quiperturbent les rythmes circadiens font partie de la physiopathologie de la MP.
