364. Analysis of droplet characteristics and cooling lubrication effects in MQL milling of 316L stainless steel

Abstract

The infiltration and cooling effect of lubricant droplets in the cutting zone largely depend on setting minimum quantity lubricant (MQL) system parameters appropriately. The study determines the optimal spray angle and spray distance relying on the geometric position relationship between the nozzle and the overall cutting zone. A numerical model that combines computational fluid dynamics (CFD) and discrete phase model (DPM) is presented to simulate droplet transport process for dynamic milling and one-tooth milling by flow field distribution and droplet characteristics. The influences of different spray distances and spray angles on the infiltration process and cutting tool temperature are analyzed. The experiments aimed at verifying the results of the analysis are conducted by measuring cutting force, cutting temperature, surface roughness, surface microtopography and tool wear. The result indicates that the spray distance has a significant impact on the jet divergence, and insufficient lubrication or excessive lubrication is effectively avoided with the optimal cutting distance. Spraying in the direction of cutting-out promotes the accessibility of the lubricant and greater concentration of droplets in the cutting zone, especially later in the cutting process, which results in increased infiltration, reduced friction, and improved heat dissipation. Moreover, experiments demonstate that the optimal state of jetting in the direction of cutting-out is 46 °C lower than that of dry cutting, the surface roughness is reduced by 19.8%, and the wear amount is reduced by roughly 34%. This study has a better understanding of the setting of spray distance and spray angle in MQL milling and the development of equipment, which provides theoretical guidance for the optimization of MQL system parameters in practical applications.

Abstract

The influence of quench cooling rate on corrosion behaviour of AA6082-T6 Al-Mg-Si alloy extrusion is investigated. It is revealed that different quench cooling rates result in significant difference in grain boundary precipitation and intergranular corrosion (IGC) susceptibility. Transmission electron microscopy is employed to probe corrosion front with the aim of precisely determining the role of each individual grain boundary precipitate in IGC propagation. However, it is observed that Mg2Si precipitate doesn’t significantly affect the process of corrosion propagation along the grain boundary and the presence of Cu-containing precipitates is not a necessary condition for IGC to occur in the alloy.

Abstract

The cross section of the process $e^+e^-\to \pi^+D^0D^{*-}$ for center-of-mass energies from 4.05 to 4.60~GeV is measured precisely using data samples collected with the BESIII detector operating at the BEPCII storage ring. Two enhancements are clearly visible in the cross section around 4.23 and 4.40~GeV. Using several models to describe the dressed cross section yields stable parameters for the first enhancement, which has a mass of $4228.6 \pm 4.1 \pm 6.3 \un{MeV}/c^2$ and a width of $77.0 \pm 6.8 \pm 6.3 \un{MeV}$, where the first uncertainties are statistical and the second ones are systematic. Our resonant mass is consistent with previous observations of the $Y(4220)$ state and the theoretical prediction of a $D\bar{D}_1(2420)$ molecule. This result is the first observation of $Y(4220)$ associated with an open-charm final state. Fits with three resonance functions with additional $Y(4260)$, $Y(4320)$, $Y(4360)$, $\psi(4415)$, or a new resonance, do not show significant contributions from either of these resonances. The second enhancement is not from a single known resonance. It could contain contributions from $\psi(4415)$ and other resonances, and a detailed amplitude analysis is required to better understand this enhancement.

Abstract

Physical review letters 122(10), 102002 (2019). doi:10.1103/PhysRevLett.122.102002, The cross section of the process e+e-→π+D0D*- for center-of-mass energies from 4.05 to 4.60 GeV is measured precisely using data samples collected with the BESIII detector operating at the BEPCII storage ring. Two enhancements are clearly visible in the cross section around 4.23 and 4.40 GeV. Using several models to describe the dressed cross section yields stable parameters for the first enhancement, which has a mass of 4228.6±4.1±6.3 MeV/c2 and a width of 77.0±6.8±6.3 MeV, where the first uncertainties are statistical and the second ones are systematic. Our resonant mass is consistent with previous observations of the Y(4220) state and the theoretical prediction of a DD¯1(2420) molecule. This result is the first observation of Y(4220) associated with an open-charm final state. Fits with three resonance functions with additional Y(4260), Y(4320), Y(4360), ψ(4415), or a new resonance do not show significant contributions from either of these resonances. The second enhancement is not from a single known resonance. It could contain contributions from ψ(4415) and other resonances, and a detailed amplitude analysis is required to better understand this enhancement., Published by APS, College Park, Md.

Abstract

The parent-child relationship, especially between mothers and daughters, is repeatedly proven in previous research as a crucial factor in the development and symptom maintenance of bulimia nervosa. This thesis attempts to answer mainly three questions: 1. Do patients with bulimia nervosa have more interpersonal problems, especially in the relationship with their mothers, than controls? 2. Compared to controls, what are the most distinctive relationship patterns of the daughters with bulimia nervosa based on their descriptions of their interactions with their mothers? 3. How are the associations between those relationship conflicts and the psychological distress and eating disorder symptoms of bulimic patients? This study has explored the mother-daughter relationship conflicts with a combination of qualitative and quantitative methods. 30 female patients with diagnosis of bulimia nervosa, and 30 controls, who are free from any eating disorders and comparable in the aspects of age and education level were recruited in the study. The participants in both groups were interviewed with a semi-structured qualitative Interview, the Adjective Interview on Relationships, which was later coded with the Core Conflictual Relationship Theme-LU (CCRT-LU). A set of questionnaires were applied to both groups: the Symptom checklist-90 Revised (SCL-90-R), the Eating Disorder Inventory-2 (EDI-2), the Family Environment Scale and the Experience of Close Relationships-Relationship Structures Questionnaire (ECR-RS). Additionally, in order to evaluate the severity of the eating pathology, a clinical diagnostic questionnaire, Eating Disorder Examination-Questionnaire (EDE-Q) was applied to the patient group with bulimia nervosa. The patients with bulimia nervosa showed significant higher psychological distress such as anxiety, depression, and hostility in SCL-90-R, which are significantly correlated with the relationship experiences in the mother-daughter relationship (superclusters ABC: loving, EFG: weak, and KL: fighting in the R dimension: dimension of reaction of CCRT-LU analysis). Those superclusters in different dimensions correlated as well significantly with the characteristic features of bulimia nervosa in EDI-2 in both groups, and with core symptoms measured with EDE-Q in the patient group. In respect to the family environment, the patients reported in the FES a rigid, full of control but lacking emotional cohesiveness environment. The experiences of the parental relationship indicate that a greater percentage of the daughters with bulimia nervosa may have an insecure attachment pattern with significant higher scores in both anxious and avoidance dimensions in ECR-RS than their counterparts in the control group. In the CCRT-LU analysis, patients with bulimia nervosa generally used fewer words from harmonious categories in their descriptions of the mother-daughter relationship interactions than controls. Fathers are perceived by the patients with bulimia nervosa as cold and distancing in the analysis of adjectives, while mothers as unsupportive, anxious, rejecting and distancing. For wishes (W), the most common wishes from all the participants were “to be loved by the mother”. The discrepancy of wishes between the two groups suggested the pseudo-autonomic feature of the daughters with bulimia nervosa. For reactions (R), the central relationship conflictual theme of the mother-daughter relationship can be summarized as, the weakness and anxiety of the mother not only limit their capability to support and love their daughter, but also calls for the reverse care from their daughters. The daughters receive through this reverse care of the mothers both emotional stress and benefits. To cope with the stress, they developed pseudo-autonomic behaviors and emotional suppressions as their coping strategies, but it is not enough. The psychological decompensation in the stress situation may be highly associated with their impulsive eating symptoms, such as bingeing. Later, by applying a discriminant analysis with jackknife resampling procedure, the mothers’ personality features (loving and self-determined in the control group vs. weak and anxious in the patient group), as well as the daughters’ responses (reverse care with stress as well as benefit in the patient group and the loving, but also with anger and impatience response in the control group) were identified as the most crucial factors to separate the patients with bulimia nervosa from the controls. The discriminant function for this model with four factors reached 83.3%. Moreover, in the comparison of the additional classifications, mothers’ interferences, especially about appearance or eating differed significantly between the two groups. Daughters in the patient group show a distinctive reverse care tendency towards their mothers.

373. Enhancing the Therapeutic Efficacy of KRASG12C Inhibitors in Lung Adenocarcinoma Cell Models by Cotargeting the MAPK Pathway or HSP90.

Abstract

Background. KRASG12C inhibitors have shown promising efficacy in early clinical trials, but drug resistance compromises their long-term benefits. Therefore, it is critical to understand the mechanisms of drug resistance and to design appropriate combinatory treatments to improve efficacy. Methods. To understand the comprehensive mechanisms of drug resistance, we treated lung cancer cells with KRASG12C inhibitors for different periods and performed transcriptional profiling and signaling analysis to identify critical factors and pathways that drive drug tolerance and resistance. We also evaluated several drug combinations in vitro and in vivo to identify potentially effective therapeutics. Results. We found that the feedback activation of multiple receptor tyrosine kinases (RTKs) may have cooperatively induced intrinsic and adaptive resistance to KRASG12C inhibitors. Notably, continuous KRAS inhibition induced a multidrug-resistant phenotype, implying that upfront combinatory treatment might be required to treat this group of patients. We also demonstrated that concurrently targeting multiple nodes in the RTK/RAS/RAF/MEK/ERK axis improved the efficacy of KRASG12C inhibitors, mainly by suppressing the reactivation of the mitogen-activated protein kinase (MAPK) pathway. Moreover, the combined use of HSP90 and KRASG12C inhibitors effectively induced tumor regression in lung adenocarcinoma models in vitro and in vivo. Conclusion. 375. Pattern and optimal enzymolysis conditions of non-starch polysaccharide (NSP) enzymes in corn-soybean feed.

Abstract

[Objective] This study aimed to increase content of oligosaccharides, obtain enzymolysis products with antibacterial properties, and improve in vitro dry matter digestibility (IVDMD) of corn-soybean feed by screening enzyme patterns and optimizing enzymolysis conditions of non-starch polysaccharide (NSP). [Method] The content of reducing sugar was used as the evaluation index. Five NSP enzymes with different contents (cellulose 110, 220 and 330 U/kg, xylanase 176, 352 and 528 U/kg, pectinase 331, 662 and 993 U/kg, β-glucanase 124, 248 and 372 U/kg, (3-mannase 83, 166 and 249 U/kg) were added into corn-soybean feed to screen optimum enzyme pattern using Li8 (37) orthogonal design. The effects of temperature (35, 45, 55 and 65 °C) on reducing sugar content, and time (0, 3, 6, 9, 12, 15, 18, 21 and 24 h) on carbohydrate composition, antibacterial effect and IVDMD of feed were studied to obtain optimal enzymatic hydrolysis conditions. The enzymatic hydrolysis feed was prepared according to above optimized conditions. A storage test was carried out at 25 °C to compare antibacterial effects among ordinary feed, enzymatic hydrolysis feed and the feed with antiseptic (0. 05% and 0. 10% mass fraction). [Result] The optimum enzyme combinations were cellulose 110 U/kg, xylanase 528 U/kg, pectinase 993 U/kg, (3-glucanase 372 U/kg and p-mannanase 249 U/kg in corn-soybean feed. The optimum temperature for enzymatic hydrolysis was 55 °C and highest reduction sugar yield (17. 98 mg/g) was obtained. The optimum time was 15 h, at which the oligosaccharide content in enzymatic hydrolysis feed was 78. 65 mg/g and enzymolysis products had the strongest inhibitory effect on Escherichia coli. Furthermore, it also showed inhibitory effects on foodborne pathogenic bacteria such as Bacillus cereus, Salmonella and Lactobacillus plantarum. The IVDMD of feed increased over enzymolysis time and reached almost equilibrium of 54. 25% - 56. 25% at 15 h. Feed storage test showed that no spoilage was found in enzymatic feed and the feed with 0. 10% mildew inhibitor, while ordinary feed and feed with 0. 05% mildew inhibitor appeared rot after 2 days storage under certain conditions. The total bacterial plate counts and coliform counts in enzymatic feed were well below the counts in ordinary feed and feed with mildew inhibitor. Enzymatic feed was ineffective against blue mold. [Conclusion] The NSP enzyme pattern and optimal enzymolysis conditions for corn-soybean feed were obtained. 377. MiR-137-3p Inhibits Colorectal Cancer Cell Migration by Regulating a KDM1A-Dependent Epithelial-Mesenchymal Transition.

Abstract

Background: In colorectal cancer (CRC), miR-137-3p downregulation is associated with disease progression, but the mechanism is not fully understood. KDM1A, also known as LSD1, is upregulated in various cancer and promotes tumor metastasis. Interestingly, miR-137-3p is downregulated by hypoxia, which plays critical roles in tumor metastasis, and KDM1A is a miR-137-3p target gene in brain tumors.Aims: To study if CRC metastasis is regulated by a hypoxia/miR-137-3p/KDM1A axis and if the epithelial-mesenchymal transition (EMT) process is involved.Methods: We measured the levels of miR-137-3p, KDM1A, and some EMT markers in CRC biopsy tissues and cell lines. We also investigated the regulation of KDM1A by miR-137-3p and the effects of KDM1A inhibition on the EMT process and cell migration.Results: We verified the low miR-137-3p and high KDM1A levels in CRC tumors. Inhibiting miR-137-3p upregulated KDM1A expression and promoted the invasiveness of CRC cells. KDM1A knockdown, or treatment with tranylcypromine, a specific KDM1A inhibitor, reduced the migration and invasion of CRC cells by inhibiting the EMT process. CRC cells cultured under hypoxic conditions expressed less miR-137-3p but more KDM1A than cells cultured under normal conditions, implying the involvement of miR-137-3p and KDM1A in hypoxia-induced tumor metastasis.Conclusions: We conclude that MiR-137-3p inhibits CRC cell migration by regulating a KDM1A-dependent EMT process. The phosphate and tensin homology (PTEN) protein, often mutated or absent in tumor cells, compromises its role as a tumor suppressor. Addressing this deficit in PTEN functionality within tumor cells holds immense promise for effective anti-tumor strategies. Notably, the high expression of glycoprotein 60 (Gp60, also named as albondin) in vascular endothelial cells, serving as a prevalent albumin receptor, and the overexpression of secreted protein acidic and rich in cysteine (SPARC, also called as osteonectin or BM40) in various tumor types as an albumin-binding protein, underscore a potential avenue for targeted therapy. Building upon our prior investigations, we posit a hypothesis: leveraging active human serum albumin could facilitate the delivery of PTEN protein into tumor cells. This proposed approach capitalizes on the ability of active albumin to target tumor cells via Gp60- and SPARC-mediated pathways. Abstract

In the present study, the corrosion behaviour of AA6082 Al-Mg-Si alloy extrusion, consisting of a recrystallized surface region and non-recrystallized fibre structure core, is investigated. It is found that the corrosion penetration rate and propagation path are closely related to grain structure and local plastic deformation. Corrosion morphology changes from intergranular corrosion in recrystallized region to both intergranular and intragranular corrosion of the heavily deformed subgrains in non-recrystallized region. Further, local plastic deformation is quantified by electron backscatter diffraction and transmission Kikuchi diffraction and correlated to corrosion propagation path.

Abstract

In the present study, the microstructure and corrosion behaviour of laser beam welded 2A97 alloy are investigated. Welding process results in segregation of alloying elements within the fusion zone and, consequently, formation of T1 (Al2CuLi), TB (Al7Cu4Li) and R (Al5CuLi3) phase interdendritic intermetallics. The distribution of the interdendritic intermetallics is non-uniform across the weld. In NaCl solution, corrosion initiates at interdendritic intermetallic with selective dissolution of Al and Li, resulting in a copper-rich remnant, which subsequently acts as effective cathode supporting the dissolution of alloy matrix. The equiaxed dendrite zone at the centre of the weld is most susceptible to corrosion.

398. Reverse identification of constitutive parameters of Inconel 718 alloy based on analytical model and thermo-mechanical loads analysis of machined surface

Abstract

Finite element simulation is able to predict cutting force, temperature, stress distribution and chip formation. Constructing a constitutive model suitable for cutting simulation can accurately describe the state of the cutting process, which plays an important role in improving surface quality as well as optimizing cutting parameters and tool geometric design. Nickel-based superalloys are widely used in aerospace due to their excellent physical and mechanical properties at high temperatures. In this study, J-C parameters reverse identification of Inconel 718 alloy was carried out in both the solution annealed state and precipitation hardened state. The average cutting force and chip thickness were obtained by conducting orthogonal cutting experiments, after which the shear angle was calculated. Through quasi-static compression experiments, the yield strength, strain hardening modulus and hardening index of the material were then obtained. Considering the influence of the cutting-edge radius on cutting force during the cutting process, the Waldorf model was introduced into the analytical model in order to describe the deformation characteristics (strain, temperature, strain rate and stress) of the primary shear zone, after which the shear force was modified. The J-C constitutive parameters of the solution annealed state and precipitation hardened state were inversely identified by the intelligent optimization algorithm. According to the given range of cutting conditions, the accuracy of the method was verified through comparison with the cutting force and chip morphology of the finite element simulation. The differences in quasi-static and cutting dynamic mechanical responses of the two states were subsequently expounded. The effects of tool wear types and cutting parameters on the material flow characteristics at the tool tip as well as the thermal-mechanical load on the machined surface of precipitation hardened state Inconel 718 alloy were analyzed via numerical simulation. Accordingly, this study may provide insight into the identification of constitutive parameters of superalloys as well as the control of machined surface quality.

Abstract

Recently, the dual beam Xe+ plasma focused ion beam (Xe+pFIB) instrument has attracted increasing interest for site‐specific transmission electron microscopy (TEM) sample preparation for a local region of interest as it shows several potential benefits compared to conventional Ga+FIB milling. Nevertheless, challenges and questions remain especially in terms of FIB‐induced artefacts, which hinder reliable S/TEM microstructural and compositional analysis. Here we examine the efficacy of using Xe+ pFIB as compared with conventional Ga+ FIB for TEM sample preparation of Al alloys. Three potential source of specimen preparation artefacts were examined, namely: (1) implantation‐induced defects such as amophisation, dislocations, or 'bubble' formation in the near‐surface region resulting from ion bombardment of the sample by the incident beam; (2) compositional artefacts due to implantation of the source ions and (3) material redeposition due to the milling process. It is shown that Xe+pFIB milling is able to produce improved STEM/TEM samples compared to those produced by Ga+ milling, and is therefore the preferred specimen preparation route. Strategies for minimising the artefacts induced by Xe+pFIB and Ga+FIB are also proposed. Lay Description: FIB (focused ion beam) instruments have become one of the most important systems in the preparation of site‐specific TEM specimens, which are typically 50‐100 nm in thickness. TEM specimen preparation of Al alloys is particularly challenging, as convention Ga‐ion FIB produces artefacts in these materials that make microstructural analysis difficult or impossible. Recently, the use of noble gas ion sources, such as Xe, has markedly improved milling speeds and is being used for the preparation of various materials. Hence, it is necessary to investigate the structural defects formed during FIB milling and assess the ion‐induced chemical contamination in these TEM samples.
