An atmospheric pressure low-temperature plasma jet has a promising application prospect in biomedical fields due to its low operating temperature and its ability to act in open space. In this paper, a mouse excisional wound splinting (MEWS) model is developed using a BALB/c strain of mice. A pulsed low-temperature plasma jet close to the human temperature is generated to treat the wound of the mouse skin. We have demonstrated that the pulsed low-temperature plasma jet can effectively promote wound healing. The large number of active particles contained in the plasma, such as reactive oxygen species, reactive nitrogen species, etc., may be responsible for its ability to increase the wound healing rate and promote skin tissue regeneration. The MEWS model adopted in this experiment reduces the interference in the evaluation of the wound healing effect caused by skin contraction and wound dressing, and thus is closer to the healing process of human wounds, and the method of wound area acquisition and calculation based on image processing reduces human interference and is more standardized, which can be used to evaluate the effect of the pulsed low-temperature plasma jet on promoting wound healing. In this paper, we introduce a direct method of moving spheres for the fractional Laplacian ( − △ ) α / 2 with 0 < α < 2 , in which a key ingredient is the narrow region maximum principle. As immediate applications, we classify non-negative solutions for semilinear equations involving the fractional Laplacian in R n ; we prove a non-existence result for the prescribing Q α curvature equation on S n ; then by combining the direct method of moving planes and moving spheres, we establish a Liouville type theorem on a half Euclidean space. Bergenin (BER), a natural component of polyphenols, has a variety of pharmacological activities, especially in improving drug metabolism, reducing cholestasis, anti-oxidative stress and inhibiting inflammatory responses. The aim of this study was to investigate the effects of BER on liver injury induced by isonicotinic acid hydrazide (INH) and rifampicin (RIF) in mice. The mice model of liver injury was established with INH (100 mg/kg)+RIF (100 mg/kg), and then different doses of BER were used to intervene. The pathological morphology and biochemical indicators of mice were detected. Meanwhile, RNA sequencing was performed to screen the differentially expressed genes and signaling pathways. Finally, critical differentially expressed genes were verified by qRT-PCR and Western blot. RNA sequencing results showed that 707 genes were significantly changed in the INH+RIF group compared with the Control group, and 496 genes were significantly changed after the BER intervention. These differentially expressed genes were mainly enriched in the drug metabolism, bile acid metabolism, Nrf2 pathway and TLR4 pathway. The validation results of qRT-PCR and Western blot were consistent with the RNA sequencing. Therefore, BER alleviated INH+RIF-induced liver injury in mice. The mammalian brain, with its complexity and intricacy, poses significant challenges for researchers aiming to understand its inner workings. Optical multilayer interference tomography (OMLIT) is a novel, promising imaging technique that enables the mapping and reconstruction of mesoscale all-cell brain atlases and is seamlessly compatible with tape-based serial scanning electron microscopy (SEM) for microscale mapping in the same tissue. However, currently, OMLIT suffers from imperfect coatings, leading to background noise and image contamination. In this study, we introduced a new imaging configuration using carbon spraying to eliminate the tape-coating step, resulting in reduced noise and enhanced imaging quality. We demonstrated the improved imaging quality and validated its applicability through a correlative light–electron imaging workflow. Our method successfully reconstructed all cells and vasculature within a large OMLIT dataset, enabling basic morphological classification and analysis. We also show that this approach can perform effectively on thicker sections, extending its applicability to sub-micron scale slices, saving sample preparation and imaging time, and increasing imaging throughput. Consequently, this method emerges as a promising candidate for high-speed, high-throughput brain tissue reconstruction and analysis. Background: Glioblastoma (GBM) is the most common malignant intracranial tumor with a low survival rate. However, only few drugs responsible for GBM therpies, hence new drug development for it is highly required. The natural product Cudraflavone B (CUB) has been reported to potentially kill a variety of tumor cells. Currently, its anit-cancer effect on GBM still remains unknown. Herein, we investigated whether CUB could affect the proliferation and apoptosis of GBM cells to show anti-GBM potential. Results: CUB selectively inhibited cell viability and induced cell apoptosis by activating the endoplasmic reticulum stress (ER stress) related pathway, as well as harnessing the autophagy-related PI3K/mTOR/LC3B signaling pathway. Typical morphological changes of autophagy were also observed in CUB treated cells by microscope and scanning electron microscope (SEM) examination. 4-Phenylbutyric acid (4-PBA), an ER stress inhibitor, restored the CUB-caused alteration in signaling pathway and morphological change. Conclusions: Our finding suggests that CUB impaired cell growth and induced cell apoptosis of glioblastoma through ER stress and autophagy-related signaling pathways, and it might be an attractive drug for treatment of GBM. Serial scanning electron microscopy (sSEM) has recently been developed to reconstruct complex largescale neural connectomes, through learning-based instance segmentation. However, blurry images are inevitable amid prolonged automated data acquisition due to imprecision in autofocusing and autostigmation, which impose a great challenge to accurate segmentation of the massive sSEM image data. Recently, learning-based methods, such as adversarial learning and supervised learning, have been proven to be effective for blind EM image deblurring. However, in practice, these methods suffer from the limited training dataset and the underrepresentation of high-resolution decoded features. Here, we propose a semisupervised learning guided progressive decoding network (SGPN) to exploit unlabeled blurry images for training and progressively enrich high-resolution feature representation. The proposed method outperforms the latest deblurring models on real SEM images with much less ground truth input. The improvement of the PSNR and SSIM is 1.04 dB and 0.086, respectively. We then trained segmentation models with deblurred datasets and demonstrated significant improvement in segmentation accuracy. Glioblastoma is the most lethal intracranial malignant tumor, for which the five-year overall survival rate is approximately 5%. Here we explored the therapeutic combination of vitamin C and plasma-conditioned medium on glioblastoma cells in culture and as subcutaneous or intracranial xenografts in mice. The combination treatment reduced cell viability and proliferation while promoting apoptosis, and the effects were significantly stronger than with either treatment on its own. Similar results were obtained in the two xenograft models. Vitamin C appeared to upregulate aquaporin-3 and enhance the uptake of extracellular H 2 O 2 , while the combination treatment increased intracellular levels of reactive oxygen species including H 2 O 2 and activated the JNK signaling pathway. The cytotoxic effects of the combination treatment were partially reversed by the specific JNK signaling inhibitor SP600125. Our results suggest that the combination of vitamin C and plasma-conditioned medium has therapeutic potential against glioblastoma, and they provide mechanistic insights that may help investigate this and other potential therapies in greater depth. Our results lead us to propose a model in which vitamin C upregulates aquaporin-3, which drives uptake of extracellular H 2 O 2 from vitamin C oxidation and plasma-conditioned medium. Glioblastoma multiforme (GBM) is an aggressive brain tumor with high risks of recurrence and mortality. Chemoradiotherapy resistance has been considered a major factor contributing to the extremely poor prognosis of GBM patients. Therefore, there is an urgent need to develop highly effective therapeutic agents. Here, we demonstrate the anti-tumor effect of morusin, a typical prenylated flavonoid, in GBM through in vivo and in vitro models. Morusin showed selective cytotoxicity toward GBM cell lines without harming normal human astrocytes when the concentration was less than 20 µM. Morusin treatment significantly induced apoptosis of GBM cells, accompanied by the activation of endoplasmic reticulum (ER) stress, and the appearance of cytoplasmic vacuolation and autophagosomes in cells. Then, we found the ER stress activation and cytotoxicity of morusin were rescued by ER stress inhibitor 4-PBA. Furthermore, morusin arrested cell cycle at the G1 phase and inhibited cell proliferation of GBM cells through the Akt–mTOR–p70S6K pathway. Dysregulation of ERs and cell cycle in morusin exposed GBM cells were confirmed by RNA-seq analysis. Finally, we demonstrated the combination of morusin and TMZ remarkably enhanced ER stress and displayed a synergistic effect in GBM cells, and suppressed tumor progression in an orthotopic xenograft model. In conclusion, these findings reveal the toxicity of morusin to GBM cells and its ability to enhance drug sensitivity to TMZ, suggesting the potential application value of morusin in the development of therapeutic strategies for human GBM. The problem of spent fluid catalytic cracking (SFCC) catalyst resource utilization, draws more and more attention to system analysis. SFCC was leached in an oxalic solution for comprehensive utilization. The results showed that for a D50 ≤ 17.34 μm, the catalyst leached for 240 min at 95 °C in the presence of a 2 mol/L oxalic acid solution, and the extent of leaching of V, Ni, Fe, and Al was 73.4%, 32.4%, 48.2%, and 36.8%, respectively. Studies on the occurrence state of the main ions (V, Ni, Fe, and Al) in the leaching solution were presented. Additionally, the separation of the main ions from such a solution by the "solvent extraction-stripping-hydrothermal precipitation-comprehensive recovery of valuable metal" process was studied. The immobilization rates of vanadium and nickel in geopolymers can be obtained using the toxicity characteristic leaching procedure (TCLP) test, and the geopolymers prepared by SFCC leaching residues can be considered a non-hazardous material. • Water content has a significant impact on the strength of geopolymers. • As the water content increases, the porosity and pore size of geopolymer increases. • Changes in water content lead to gel structure changes of geopolymer. • Compared with changes in pores, changes in microstructure have a greater impact on the strength of geopolymers. This study synthesized geopolymers with different content of water from 34.35% (10 mol) to 43.97% (15 mol), and the effect of initial water content in pore behavior and gel structure of geopolymer was investigated. The microstructure of geopolymer was characterized through MIP analysis, SEM, TGA-MS, XRD, FTIR and NMR measurement. As the water content in synthesis increasing, the compressive strength of geopolymer significantly decreased from 40.3 MPa to 10.2 MPa. The high initial water content results in a large increase in the diameter of pore in geopolymer, which lead to the transformation of harmless pores to harmful pores. In addition, excess water will also introduce more water to participate in the geopolymer network, so that the content of Q3 structures increased 2.94% from 19.24%, which leads to a decrease in the degree of polymerization. Both of pore behavior and gel structure changes reduce the mechanical property of geopolymers. When the initial water content is low (10–11 mol), the change of geopolymer gel structure has a greater impact on the strength of the geopolymer, while when it is high (12–15 mol), the compressive strength is dominated by the pore structure. The vasculature of the central nervous system is a 3D lattice composed of laminar vascular beds interconnected by penetrating vessels. The mechanisms controlling 3D lattice network formation remain largely unknown. Combining viral labeling, genetic marking, and single-cell profiling in the mouse retina, we discovered a perivascular neuronal subset, annotated as Fam19a4/Nts-positive retinal ganglion cells (Fam19a4/Nts-RGCs), directly contacting the vasculature with perisomatic endfeet. Developmental ablation of Fam19a4/Nts-RGCs led to disoriented growth of penetrating vessels near the ganglion cell layer (GCL), leading to a disorganized 3D vascular lattice. We identified enriched PIEZO2 expression in Fam19a4/Nts-RGCs. Piezo2 loss from all retinal neurons or Fam19a4/Nts-RGCs abolished the direct neurovascular contacts and phenocopied the Fam19a4/Nts-RGC ablation deficits. The defective vascular structure led to reduced capillary perfusion and sensitized the retina to ischemic insults. Furthermore, we uncovered a Piezo2-dependent perivascular granule cell subset for cerebellar vascular patterning, indicating neuronal Piezo2-dependent 3D vascular patterning in the brain. The present work synthesized geopolymer with quartz mechanical activated at different time, and the strengthening mechanism of geopolymer by activated quartz was investigated. The changes of quartz before and after activation and the microstructure of geopolymer was explained by the determinations of particle size, specific surface area and the concentration of reactive Si, as well as XRD, XPS and NMR measurement. With the activation time of quartz increasing from 0 to 60 min, the compressive strength of geopolymer increased from 31.5 to 55.2 MPa. The results showed that increase of quartz reactivity is mainly due to the formation of the Si-centered radicals and non-bridging oxygen on the surface rather than the changes in physical property or crystal structure of quartz. The high activity tetrahedra Si with non-bridge oxygen on the surface of quartz replace the tetrahedra Al in geopolymer gel, which lead to a Si-rich gel formed around quartz and the improvement of compressive strength. Effective treatment of liver fibrosis remains a challenging medical problem. Taraxasterol (TAR) has anti-inflammatory, anti-tumor and hepatoprotective effects. Studies have shown that TAR has good biological activity against liver injury induced by various factors. However, the anti-fibrotic effect of TAR and its mechanism are never clarified. The purpose of this study was to investigate the effects of TAR in liver fibrosis and to reveal its possible mechanism by RNA sequencing. Our results suggested that TAR attenuated CCl 4 -induced hepatocyte necrosis, inflammatory infiltration and ECM deposition. TAR inhibited the levels of ALT, AST, ALP, γ-GT, LN, HA, PC III and IV-C in serum and TNF-α, IL-6, IL-1β and MDA in liver. In addition, TAR increased the activities of SOD and GSH-Px in liver. RNA sequencing analysis of liver tissues revealed that CCl 4 and TAR significantly altered 4,155 genes and 2,675 genes, respectively. TAR reversed changes in ECM-related genes. More specifically, TAR mediated the expression of genes related to the activation of the Hippo pathway, while inhibiting the expression of genes related to the activation of HIF-1α, TGF-β/Smad, and Wnt pathways. In the validation experiments, the qRT-PCR results showed that the expression levels of Yap1, Tead3, Hif1α, Vegfa, Tgfβ1, Want3a, and Ctnnb1 mRNA were consistent with the RNA sequencing results. The Western blot results showed that TAR inhibited the levels of TGF-β1 and p-Smad2. In addition, the results in vitro were consistent with those in vivo. Vanadium slag (VS) is a by-product of vanadium titanium magnetite BOF steelmaking process and becomes an important material for vanadium products preparation. Sodium roasting-water leaching is a mainstream process for industrial production, but the water leaching residue (WLR) usually remains 1.0 wt% to 3.0 wt% V 2 O 5 , which still has value for recycling extraction of vanadium. In this research, a two-stage leaching process was proposed for high efficient vanadium extraction. The first stage leaching was sodium roasting-water leaching for vS and the second stage leaching was sulfuric acid leaching for WLR. As a result, the total leaching efficiency of vanadium can reach as high as 98.52%, which was 7.72% higher than that of conventional water leaching efficiency of vanadium (90.80%). The mechanism analysis indicates that the CaNaFe 2 Si 4 O 12 and NaFeSi 2 O 6 water-insoluble phases formed and covered on (Mn,Fe)(V,Cr) 2 O 4 spinel surface during roasting, hindering vanadium oxidation and dissolution. The structures of wrapping phases and spinel can be effectively destroyed by acid leaching, releasing vanadium in WLR. By using hydrolytic precipitation of iron ions in acid leachate, 99.78% of vanadium can be recovered as V-Fe precipitation slag, which contains 6.83 wt% V 2 O 5 and can be cycled back to sodium roasting process. The V-Fe slag belongs to amorphous precipitation, illustrating that residual vanadium in WLR can be changed to unstable compound by acid leaching and hydrolysis precipitation, which makes vanadium easier to extract during sodium roasting-water leaching process. This research has the advantages of enhanced reuse of V resource in the WLR, achieving a high V recovery. Background: Triple-negative breast cancer (TNBC) is prone to relapse due to the lack of effective therapeutic targets. Macrophages are the most abundant immune cells in the tumor microenvironment (TME) of breast cancer. Targeting the cross-talk between macrophages and cancer cells provides a more efficient strategy for anti-tumor therapy. Toll-like receptors (TLRs) are important players involved in macrophage activation, and TLR agonists are known to play roles in cancer therapy. However, the combination strategy of TLR agonists with chemotherapy drugs is still not well characterized., Methods: RT-PCR and Western blot were used to detect the expression of TLRs. The communication between breast cancer cells and macrophages were determined by co-culture in vitro. Tumor cells proliferation and migration were investigated by MTT assay and scratch wound assay. The effects of drug combinations and toxic side effects were assessed by immunohistochemistry and Hematoxylin & Eosin staining., Results: Expression of TLR3 and TLR4 were lower in breast tumor tissues compared with adjacent normal tissues. Patients with higher TLR3 or TLR4 expression levels had a better prognosis than those with lower expression levels. TLR3/4 expression was significantly inhibited when breast cancer cells MDA-MB-231 and E0771 were conditioned-cultured with macrophages in vitro and was also inhibited by pirarubicin (THP). However, the combination of TLR agonists and THP could reverse this response and inhibit the proliferation and migration of breast cancer cells. Background: There is an unmet need for stand-alone digital therapeutics for cognitive impairment in schizophrenia. This study aimed to evaluate the efficacy and acceptability of a novel digital therapeutic, IBT-SC02, for cognitive impairment in stable schizophrenia patients., Methods: A randomized, parallel-group trial was conducted at the Sichuan Province Institute of Mental Health, China. Participants aged 18-50 diagnosed with schizophrenia were randomized to either the IBT-SC02 intervention or a wait-list control. The primary outcome was cognitive performance measured using the MATRICS Consensus Cognitive Battery (MCCB) composite score., Results: A total of 80 patients were randomized (40 intervention, 40 control). The dropout rate was 5 %. The intervention group exhibited significant improvements in the MCCB composite score compared to the control, though the improvement lessened after excluding data collected by unmasked raters. Post-hoc analyses revealed that participants in the intervention group improved in four out of the seven MCCB domains (speed of processing, verbal learning, visual learning and reasoning and problem solving). Nucleophosmin 1 (NPM1) is commonly mutated in myelodysplastic syndrome (MDS) and acute myeloid leukemia. Concurrent inflammatory bowel diseases (IBD) and MDS are common, indicating a close relationship between IBD and MDS. Here we examined the function of NPM1 in IBD and colitis-associated colorectal cancer (CAC). NPM1 expression was reduced in patients with IBD. Npm1 +/- mice were more susceptible to acute colitis and experimentally induced CAC than littermate controls. Npm1 deficiency impaired the function of interleukin-22 (IL-22)-producing group three innate lymphoid cells (ILC3s). Mice lacking Npm1 in ILC3s exhibited decreased IL-22 production and accelerated development of colitis. NPM1 was important for mitochondrial biogenesis and metabolism by oxidative phosphorylation in ILC3s. Further experiments revealed that NPM1 cooperates with p65 to promote mitochondrial transcription factor A (TFAM) transcription in ILC3s. Overexpression of Npm1 in mice enhanced ILC3 function and reduced the severity of dextran sulfate sodium-induced colitis. Large-scale electron microscopy (EM) has enabled the reconstruction of brain connectomes at the synaptic level by serially scanning over massive areas of sample sections. The acquired big EM data sets raise the great challenge of image mosaicking at high accuracy. Currently, it simply follows the conventional algorithms designed for natural images, which are usually composed of only a few tiles, using a single type of keypoint feature that would sacrifice speed for stronger performance. Even so, in the process of stitching hundreds of thousands of tiles for large EM data, errors are still inevitable and diverse. Moreover, there has not yet been an appropriate metric to quantitatively evaluate the stitching of biomedical EM images. Here we propose a two-stage error detection method to improve the EM image mosaicking. It firstly uses point-based error detection in combination with a hybrid feature framework to expedite the stitching computation while maintaining high accuracy. Following is the second detection of unresolved errors with a newly designed metric of EM stitched image quality assessment (EMSIQA). Electron microscopy (EM) image denoising is critical for visualization and subsequent analysis. Despite the remarkable achievements of deep learning-based non-blind denoising methods, their performance drops significantly when domain shifts exist between the training and testing data. To address this issue, unpaired blind denoising methods have been proposed. However, these methods heavily rely on image-to-image translation and neglect the inherent characteristics of EM images, limiting their overall denoising performance. In this paper, we propose the first unsupervised domain adaptive EM image denoising method, which is grounded in the observation that EM images from similar samples share common content characteristics. Specifically, we first disentangle the content representations and the noise components from noisy images and establish a shared domain-agnostic content space via domain alignment to bridge the synthetic images (source domain) and the real images (target domain). To ensure precise domain alignment, we further incorporate domain regularization by enforcing that: the pseudo-noisy images, reconstructed using both content representations and noise components, accurately capture the characteristics of the noisy images from which the noise components originate, all while maintaining semantic consistency with the noisy images from which the content representations originate. To guarantee lossless representation decomposition and image reconstruction, we introduce disentanglement-reconstruction invertible networks. Finally, the reconstructed pseudo-noisy images, paired with their corresponding clean counterparts, serve as valuable training data for the denoising network. Extensive experiments on synthetic and real EM datasets demonstrate the superiority of our method in terms of image restoration quality and downstream neuron segmentation accuracy. Objective: To investigate the effects of recombinant human type III collagen on atrophic scars and its impact on the p38 mitogen-activated protein kinase (p38MAPK) signaling pathway., Methods: A total of 94 patients with atrophic scars admitted to our hospital from March 2020 to October 2022 were selected as subjects and evenly divided into a control group and an observation group. The control group (n = 47) received carbon dioxide fractional laser treatment, while the observation group (n = 47) was treated with recombinant human type III collagen dressings in addition to the laser treatment. Clinical efficacy, scar conditions, skin physiological parameters, serum levels of p38MAPK pathway-related proteins, and inflammatory markers were compared between the two groups., Results: The overall effective rate in the observation group was 95.74%, significantly higher than 74.47% in the control group (P < .05). Before treatment, there was no significant difference in Vancouver Scar Scale (VSS) scores, stratum corneum hydration, and transepidermal water loss between the two groups (P > .05). After treatment, the VSS score in the observation group was significantly lower than in the control group (P < .05). Similarly, prior to treatment, there were no significant differences in serum levels of mitogen-activated protein kinase 1 (MEK1), mitogen-activated protein kinase 2 (MEK2), extracellular signal-regulated kinase 1 (ERK1), and extracellular signal-regulated kinase 2 (ERK2), interleukin-10 (IL-10) and tumor necrosis factor-alpha (TNF-α) between the two groups (P > .05). After treatment, levels of MEK1, MEK2, ERK1, ERK2, IL-10, and TNF-α in the observation group were significantly lower than those in the control group (P < .05)., Conclusion: Recombinant human type III collagen significantly improves the treatment of atrophic scars, effectively ameliorating scar conditions and skin physiology. It also regulates the p38MAPK signaling pathway and reduces inflammation.

Since the outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its resultant pneumonia in December 2019, the cumulative number of infected people worldwide has exceeded 670 million, with over 6.8 million deaths. Despite the marketing of multiple series of vaccines and the implementation of strict prevention and control measures in many countries, the spread and prevalence of SARS-CoV-2 have not been completely and effectively controlled. The latest research shows that in addition to angiotensin converting enzyme II (ACE2), dozens of protein molecules, including AXL, can act as host receptors for SARS-CoV-2 infecting human cells, and virus mutation and immune evasion never seem to stop. To sum up, this review summarizes and organizes the latest relevant literature, comprehensively reviews the genome characteristics of SARS-CoV-2 as well as receptor-based pathogenesis (including ACE2 and other new receptors), mutation and immune evasion, vaccine development and other aspects, and proposes a series of prevention and treatment opinions. Introduction: The exorbitant cost of accurately annotating the large-scale serial scanning electron microscope (SEM) images as the ground truth for training has always been a great challenge for brain map reconstruction by deep learning methods in neural connectome studies. The representation ability of the model is strongly correlated with the number of such high-quality labels. Recently, the masked autoencoder (MAE) has been shown to effectively pre-train Vision Transformers (ViT) to improve their representational capabilities., Methods: In this paper, we investigated a self-pre-training paradigm for serial SEM images with MAE to implement downstream segmentation tasks. We randomly masked voxels in three-dimensional brain image patches and trained an autoencoder to reconstruct the neuronal structures., Results and Discussion: We tested different pre-training and fine-tuning configurations on three different serial SEM datasets of mouse brains, including two public ones, SNEMI3D and MitoEM-R, and one acquired in our lab. A series of masking ratios were examined and the optimal ratio for pre-training efficiency was spotted for 3D segmentation. The MAE pre-training strategy significantly outperformed the supervised learning from scratch. The J-domain proteins (JDP) form the largest protein family among cellular chaperones. In cooperation with the Hsp70 chaperone system, these co-chaperones orchestrate a plethora of distinct functions, including those that help maintain cellular proteostasis and development. JDPs evolved largely through the fusion of a J-domain with other protein subdomains. The highly conserved J-domain facilitates the binding and activation of Hsp70s. How JDPs (re)wire Hsp70 chaperone circuits and promote functional diversity remains insufficiently explained. The spent caustic with strong alkali first replaced the alkali activator to prepare the geopolymer. The influence of spent caustic to the geopolymer was characterized through compressive strength measurement, XRD, MIP analysis and NMR, and the immobilization efficiency of organic in geopolymer was evaluated through the measurement of total organic carbon (TOC). The results show that the spent caustic can partially replace the alkali activator to prepare the geopolymer, and it shows a better performance than that which was activated with pure NaOH solution when the alkalinity is between 4 mol and 14 mol. The organic matter in the spent alkali can be effectively fixed in the geopolymer, which will hinder the geopolymerization in the initial stage of the polymerization reaction but has little effect on the chemical structure and mechanical properties of the final product. With the degree of alkalinity increasing, the immobilization efficiency is improved, and the maximum can reach 84.5%. The organics in the spent caustic will hinder geopolymerization at the initial stage but has little effect on the chemical structure and mechanical property of the final product. This study proposes a new method for the recycling of spent caustic, which also reduces the preparation cost of geopolymers.

Background: Glioblastoma stem cells (GSCs) and their interplay with tumor-associated macrophages (TAMs) are responsible for malignant growth and tumor recurrence of glioblastoma multiforme (GBM), but the underlying mechanisms are largely unknown., Methods: Cell viability, stemness, migration, and invasion were measured in GSCs after the knockdown of upstream stimulating factor 1 (USF1). Luciferase assay and chromatin immunoprecipitation qPCR were performed to determine the regulation of CD90 by USF1. Immunohistochemistry and immunofluorescent staining were used to examine the expression of USF1 and GSC markers, as well as the crosstalk between GSCs and TAMs. In addition, the interaction between GSCs and TAMs was confirmed using in vivo GBM models., Results: We show that USF1 promotes malignant glioblastoma phenotypes and GSCs-TAMs physical interaction by inducing CD90 expression. USF1 predicts a poor prognosis for glioma patients and is upregulated in patient-derived GSCs and glioblastoma cell lines. USF1 overexpression increases the proliferation, invasion, and neurosphere formation of GSCs and glioblastoma cell lines, while USF1 knockdown exerts an opposite effect. Further mechanistic studies reveal that USF1 promotes GSC stemness by directly regulating CD90 expression. Importantly, CD90 of GSCs functions as an anchor for physical interaction with macrophages. Additionally, the USF1/CD90 signaling axis supports the GSCs and TAMs adhesion and immunosuppressive feature of TAMs, which in turn enhance the stemness of GSCs. Moreover, the overexpression of CD90 restores the stemness property in USF1 knockdown GSCs and its immunosuppressive microenvironment., Conclusions: Our findings indicate that the USF1/CD90 axis might be a potential therapeutic target for the treatment of glioblastoma., (© The Author(s) 2022. As a common industrial by-product, the spend fluid catalytic-cracking (SFCC) catalyst was used to prepare phosphate-based geopolymer for the first time. The structure and property of geopolymer with phosphoric acid concentration ranging from 6 to 14 mol/L was characterized by compressive strength measurements, X-ray powder diffraction (XRD), Fourier Transform Infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and 27 Al and 29 Si nuclear magnetic resonance (NMR). A stable binder was formed with the compressive strength in the range of 9.8 to 30.2 MPa when the acid concentration was between 6 and 12 mol/L. The higher concentration of acid can promote the dissolution of raw materials and formation of geopolymer gels. The coordination of silicon and aluminum in geopolymer gel synthesized with the SFCC catalyst and metakaolin is similar. Compared with the geopolymer with metakaolin, which forms more Si-O-Al bonds, in the networks of geopolymer with the SFCC catalyst, more Si(Al)-O-P bonds were formed. These results indicate that the SFCC catalyst can be an excellent raw material for the synthesis of phosphate-based geopolymer.

Lysosome plays important roles in cellular homeostasis, and its dysregulation contributes to tumor growth and survival. However, the understanding of regulation and the underlying mechanism of lysosome in cancer survival is incomplete. Here, we reveal a role for a histone acetylation-regulated long noncoding RNA termed lysosome cell death regulator ( LCDR ) in lung cancer cell survival, in which its knockdown promotes apoptosis. Mechanistically, LCDR binds to heterogenous nuclear ribonucleoprotein K (hnRNP K) to regulate the stability of the lysosomal-associated protein transmembrane 5 ( LAPTM5 ) transcript that maintains the integrity of the lysosomal membrane. Knockdown of LCDR , hnRNP K, or LAPTM5 promotes lysosomal membrane permeabilization and lysosomal cell death, thus consequently resulting in apoptosis. LAPTM5 overexpression or cathepsin B inhibitor partially restores the effects of this axis on lysosomal cell death in vitro and in vivo. Similarly, targeting LCDR significantly decreased tumor growth of patient-derived xenografts of lung adenocarcinoma (LUAD) and had significant cell death using nanoparticles (NPs)-mediated systematic short interfering RNA delivery. Moreover, LCDR /hnRNP K/LAPTM5 are up-regulated in LUAD tissues, and coexpression of this axis shows the increased diagnostic value for LUAD. Collectively, we identified a long noncoding RNA that regulates lysosome function at the posttranscriptional level. Glioblastoma multiforme (GBM) is the most aggressive brain tumor, with a 5-year survival ratio <5%. Invasive growth is a major determinant of the poor prognosis in GBM. In this study, we demonstrate that high expression of PPFIA binding protein 1 (PPFIBP1) correlates with remarkable invasion and poor prognosis of GBM patients. Using scratch and transwell assay, we find that the invasion and migration of GBM cells are promoted by overexpression of PPFIBP1, while inhibited by knockdown of PPFIBP1. Then, we illustrate that overexpression of PPFIBP1 facilitates glioma cell infiltration and reduces survival in xenograft models. Next, RNA-Seq and GO enrichment analysis reveal that PPFIBP1 regulates differentially expressed gene clusters involved in the Wnt and adhesion-related signaling pathways. Furthermore, we demonstrate that PPFIBP1 activates focal adhesion kinase (FAK), Src, c-Jun N-terminal kinase (JNK), and c-Jun, thereby enhancing Matrix metalloproteinase (MMP)-2 expression probably through interacting with SRCIN1 (p140Cap). Finally, inhibition of phosphorylation of Src and FAK significantly reversed the augmentation of invasion and migration caused by PPFIBP1 overexpression in GBM cells. In order to solve the increasingly serious problems of energy and environment, microalgae are used as a raw material for extracting lipids to produce biodiesel. Prior to the extraction of lipids, microalgae were treated with high-voltage pulsed electric field (PEF) to break the cell membrane. It was found that the lipid extraction yield depends on the electric field strength (E) and the specific energy input (Wsp), and has a certain relationship with the cell disintegration rate of Chlorella. The perforation degree of the Chlorella's cell membrane by PEF treatment is controllable, moderate perforation can be ensured by controlling the power parameters. PEF treatment significantly improved the extraction yield of lipids. Compared with the test samples without PEF treatment, PEF treatment increased the lipid extraction yields by up to 166.67%. The widespread human exposure to perfluoroalkyl acids (PFAAs) has led to increasing public concern. In this study, we present a comprehensive measurement of total fluorine (TF), extractable organic fluorine (EOF), identified organic fluorine (IOF, total concentration of identified PFAAs quantified as fluorine) and 11 target PFAAs in human serum (n = 60), hair (n = 49) and nails (n = 39) collected from non-occupation exposed volunteers in 10 cities of Guangdong Province, China. The results indicated that EOF was the major form of fluorine in serum, accounting for 70-80% of TF. The levels of IOF contributed less than 10% of EOF. Perfluorooctane sulfonic acid (PFOS) was found to be the dominant PFAA with mean concentration of 23 ng·mL -1 in serum, 35 ng·g -1 in hair and 33 ng·g -1 in nail, respectively. Short-chain PFAAs (C ≤ 10) were the predominant PFAAs in three matrices. Levels of PFOS, perfluorohexane sulfonic acid (PFHxS), perfluoroundecanoic acid (PFUdA) and perfluorotridecanoic acid (PFTrDA) in males are significantly higher than those in females (p < 0.01). Significant positive correlations were observed between nail and serum for PFOS (p < 0.01), perfluorooctanoic acid (PFOA) (p < 0.05) and PFHxS (p < 0.01), suggesting that human nails, a noninvasive sample, are a promising bio-indicator for PFAA risk assessment.

Objectives: This study investigated whether variations in GSTT1 and GSTM1 gene are associated with the DNA damage level of p53 gene., Methods: We quantified urinary 1-hydroxypyrene using high-performance liquid chromatography, and examined the DNA damage level of p53 gene by real-time quantitative PCR in 756 coke-oven workers. Multiplex PCR was used to detect the presence or absence of genes., Results: DNA damage levels of p53 gene in the high exposure group and intermediate exposure group were significantly higher than that of p53 gene in the low exposure group (P < 0.01). In coke-oven workers, the DNA damage levels of subjects with non-null genotype in GSTT1 or GSTM1 gene were significantly higher than that of those with the null genotype (P < 0.01)., Conclusions: GSTT1 and GSTM1 may modulate DNA damage levels of p53 gene when exposed to polycyclic aromatic hydrocarbons.
