The extraction of wheat lodging is of great significance to post-disaster agricultural production management, disaster assessment and insurance subsidies. At present, the recognition of lodging wheat in the actual complex field environment still has low accuracy and poor real-time performance. To overcome this gap, first, four-channel fusion images, including RGB and DSM (digital surface model), as well as RGB and ExG (excess green), were constructed based on the RGB image acquired from unmanned aerial vehicle (UAV). Second, a Mobile U-Net model that combined a lightweight neural network with a depthwise separable convolution and U-Net model was proposed. Finally, three data sets (RGB, RGB + DSM and RGB + ExG) were used to train, verify, test and evaluate the proposed model. The results of the experiment showed that the overall accuracy of lodging recognition based on RGB + DSM reached 88.99%, which is 11.8% higher than that of original RGB and 6.2% higher than that of RGB + ExG. In addition, our proposed model was superior to typical deep learning frameworks in terms of model parameters, processing speed and segmentation accuracy. The optimized Mobile U-Net model reached 9.49 million parameters, which was 27.3% and 33.3% faster than the FCN and U-Net models, respectively. Furthermore, for RGB + DSM wheat lodging extraction, the overall accuracy of Mobile U-Net was improved by 24.3% and 15.3% compared with FCN and U-Net, respectively. Therefore, the Mobile U-Net model using RGB + DSM could extract wheat lodging with higher accuracy, fewer parameters and stronger robustness.

Ottawa sand and Angular sand consist of particles with distinct shapes. The x-ray computed tomography (XCT) image stacks of their in-situ confined compressive testings are provided in this paper. For each image stack, a contact network, a thermal network and a network feature - edge betweenness centrality - of each edge in the networks are also provided. Introduction: The efficacy of memantine for migraine remains controversial. We conduct a systematic review and meta-analysis to explore the influence of memantine versus placebo on treatment in migraine patients., Methods: We search PubMed, EMbase, Web of science, EBSCO, and Cochrane library databases through February 2020 for randomized controlled trials assessing the effect of memantine versus placebo on treatment efficacy in migraine patients. This meta-analysis is performed using the random-effect model., Results: Three randomized controlled trials are included in the meta-analysis. Overall, compared with control group in migraine patients, memantine treatment is associated with substantially reduced monthly attack frequency (mean difference [MD], -2.14; 95% confidence interval [CI], -2.83 to -1.46; P < 0.00001), number of migraine days (MD, -4.17; 95% CI, -6.40 to -1.93; P = 0.0003) and Migraine Disability Assessment (MD, -5.63; 95% CI, -6.46 to -4.79; P < 0.00001), but demonstrates no obvious influence on acute pain medications (MD, -1.23; 95% CI, -4.63 to 2.17; P = 0.48)., Conclusions: Memantine treatment may benefit to the control of migraine., Competing Interests: The authors have no conflicts of interest to declare., (Copyright © 2021 Wolters Kluwer Health, Inc. Nitrogen is an important indicator for monitoring wheat growth. The rapid development and wide application of non-destructive detection provide many approaches for estimating leaf nitrogen content (LNC) in wheat. Previous studies have shown that better results have been obtained in the estimation of LNC in wheat based on spectral features. However, the lack of automatically extracted features leads to poor universality of the estimation model. Therefore, a feature fusion method for estimating LNC in wheat by combining spectral features with deep features (spatial features) was proposed. The deep features were automatically obtained with a convolutional neural network model based on the PyTorch framework. The spectral features were obtained using spectral information including position features (PFs) and vegetation indices (VIs). Different models based on feature combination for evaluating LNC in wheat were constructed: partial least squares regression (PLS), gradient boosting decision tree (GBDT), and support vector regression (SVR). The results indicate that the model based on the fusion feature from near-ground hyperspectral imagery has good estimation effect. In particular, the estimation accuracy of the GBDT model is the best (R 2 = 0.975 for calibration set, R 2 = 0.861 for validation set). These findings demonstrate that the approach proposed in this study improved the estimation performance of LNC in wheat, which could provide technical support in wheat growth monitoring.

Soluble solids content (SSC) is one of the important components for evaluating fruit quality. The rapid development of hyperspectral imagery provides an efficient method for non-destructive detection of SSC. Previous studies have shown that the internal quality evaluation of fruits based on spectral information features achieves better results. However, the lack of comprehensive features limits the accurate estimation of fruit quality. Therefore, the deep learning theory is applied to the estimation of the soluble solid content of peaches, a method for estimating the SSC of fresh peaches based on the deep features of the hyperspectral image fusion information is proposed, and the estimation models of different neural network structures are designed based on the stack autoencoder-random forest (SAE-RF). The results show that the accuracy of the model based on the deep features of the fusion information of hyperspectral imagery is higher than that of the model based on spectral features or image features alone. In addition, the SAE-RF model based on the 1237-650-310-130 network structure has the best prediction effect (R 2 = 0.9184, RMSE = 0.6693). Our research shows that the proposed method can improve the estimation accuracy of the soluble solid content of fresh peaches, which provides a theoretical basis for the non-destructive detection of other components of fresh peaches.

Objectives: Over the past years, growing attention has been paid to deciphering the pivotal role of long non-coding RNAs (lncRNAs) in regulating the occurrence and development of human malignancies, cervical cancer (CC) included. Nonetheless, the regulatory role of lncRNA BBOX1 antisense RNA 1 (BBOX1-AS1) has not been explored as yet., Material and Methods: The expression of BBOX1-AS1 was detected by reverse transcription real-time quantitative polymerase chain reaction (RT-qPCR). Cell Counting Kit-8 (CCK-8), colony formation, TUNEL, Western blot, transwell and immunofluorescence assays testified the critical role of BBOX1-AS1 in CC. The relationship between RNAs (BBOX1-AS1, miR-361-3p, HOXC6 and HuR) was analysed by luciferase reporter, RNA Immunoprecipitation (RIP) and RNA pull-down assays., Results: BBOX1 antisense RNA 1 antisense RNA 1 was revealed to be highly expressed in CC. Decreased expression of BBOX1-AS1 had suppressive effects on CC cell growth and migration. Molecular mechanism assays verified that BBOX1-AS1 had negative interaction with miR-361-3p in CC. Additionally, homeobox C6 (HOXC6) was validated to be a downstream target of miR-361-3p in CC. Furthermore, ELAV-like RNA-binding protein 1, also known as HuR, was uncovered to be capable of regulating the mRNA stability of HOXC6 in CC. More importantly, rescue assays delineated that knockdown of HuR after overexpressing miR-361-3p could reverse BBOX1-AS1 upregulation-mediated effect on CC progression. Similarly, the function induced by BBOX1-AS1 upregulation on CC progression could be countervailed by HOXC6 depletion., Conclusions: BBOX1 antisense RNA 1 facilitates CC progression by upregulating HOXC6 expression via miR-361-3p and HuR., (© 2020 The Authors. Background: The efficacy of dexketoprofen for migraine attack remains controversial. We conduct a systematic review and meta-analysis to explore the influence of dexketoprofen supplementation versus placebo on pain control in migraine attack patients., Methods: We search PubMed, EMbase, Web of science, EBSCO, and Cochrane library databases through March 2019 for randomized controlled trials (RCTs) assessing the effect of dexketoprofen supplementation versus placebo on pain control for migraine attack patients. This meta-analysis is performed using the random-effect model., Results: Five RCTs involving 794 patients are included in the meta-analysis. Overall, compared with control group for migraine attack, dexketoprofen supplementation is associated with substantially increased pain free at 2 hours (RR = 1.90; 95% CI = 1.43-2.53; P < .0001), pain free at 48 hours (RR = 1.63; 95% CI = 1.07-2.49; P = .02), good or excellent treatment (RR = 1.48; 95% CI = 1.24-1.78; P < .0001) and pain relief at 2 hours (RR = 1.80; 95% CI = 1.17-2.77; P = .007), as well as reduced need for rescue drug (RR = 0.64; 95% CI = 0.43-0.94; P = .02), with no significant increase in adverse events (RR = 1.51; 95% CI = 0.87-2.62; P = .14)., Conclusion: Dexketoprofen supplementation benefits to improve pain control at 48 hours and reduce the need for rescue drug in migraine attack patients.

Objective: To compare the reduction of hepatocellular carcinoma (HCC) risk between long-term treatment of entecavir and low genetic barrier antiviral agents in hepatitis B virus (HBV)-related cirrhotic patients., Study Design: An observational study., Place and Duration of Study: Department of Infectious Diseases and Hepatology, the Second Hospital of Shandong University, Jinan, China, from October 2008 to October 2016., Methodology: HBV-related cirrhotic patients with antiviral treatment for at least 12 months were consecutively included. Propensity score matching analysis was performed to improve comparability of the data from both entecavir group and the control group. Log-rank test was used to compare influence of various nucleos(t)ide analogs (NAs) for incidence of HCC. Independent risk factors were estimated by multivariable Cox proportional hazards models., Results: The total cohort included 207 HBV-related cirrhotic patients, of which 83 patients were treated with entecavir initially. The present study found no statistical difference for the incidence of HCC between entecavir group and the control group in the total cohort (p=0.525). However, the difference became statistically significant (p=0.014) after propensity score matching. Number needed to treat (NNT) were 8 patients, 6 patients and 3 patients at years 2, 3 and 4, respectively. Multivariable Cox regression in propensity score matching cohort revealed older age (HR: 1.066, p=0.041), NAs of low generic barrier (HR: 6.944, p=0.016), NAs resistance (HR: 3.648, p=0.041), and lower platelet counts (<80x10 ⁹/L) (HR: 6.718, p=0.009) as independent risk factors for HCC incidence., Conclusion: Entecavir is more efficient in reducing the incident HCC risk for HBV-related cirrhotic patients in comparison to low genetic barrier NAs.

Free amino acids are an important indicator of the freshness of yellow tea. This study investigated a novel procedure for predicting the free amino acid (FAA) concentration of yellow tea. It was developed based on the combined spectral and textural features from hyperspectral images. For the purposes of exploration and comparison, hyperspectral images of yellow tea (150 samples) were captured and analyzed. The raw spectra were preprocessed with Savitzky-Golay (SG) smoothing. To reduce the dimension of spectral data, five feature wavelengths were extracted using the successive projections algorithm (SPA). Five textural features (angular second moment, entropy, contrast, correlation, and homogeneity) were extracted as textural variables from the characteristic grayscale images of the five characteristic wavelengths using the gray-level co-occurrence matrix (GLCM). The FAA content prediction model with different variables was established by a genetic algorithm-support vector regression (GA-SVR) algorithm. The results showed that better prediction results were obtained by combining the feature wavelengths and textural variables. Compared with other data, this prediction result was still very satisfactory in the GA-SVR model, indicating that data fusion was an effective way to enhance hyperspectral imaging ability for the determination of free amino acid values in yellow tea., Competing Interests: The authors have declared that no competing interests exist.

The effects of Staphylococcal enterotoxin B (SEB) on regulation of immune response have been recognized; whether SEB can enhance the effects of immunotherapy on glioma remains to be investigated. This study tests a hypothesis that administration with SEB enhances the effects of specific immunotherapy on glioma growth in mice. In this study, a glioma-bearing mouse model was developed by adoptive transfer with GL261 cells (a mouse glioma cell line). The mice were treated with the GL261 cell extracts (used as an Ag) with or without administration of SEB. We observed that treating glioma-bearing mice with the glioma Ag and SEB induced glioma-specific Th9 cells in both glioma tissue and the spleen. Treating CD4+ CD25- T cells with SEB increased p300 phosphorylation, histone H3K4 acetylation at the interleukin (IL)-9 promoter locus, and increased the IL-9 transcriptional factor binding to the IL-9 promoter. Treating CD4+ CD25- T cells with both SEB and glioma Ag induced glioma-specific Th9 cells. The glioma-specific Th9 cells induced glioma cell apoptosis in the culture. Treating the glioma-bearing mice with SEB and glioma Ag significantly inhibited the glioma growth. In conclusion, SEB plus glioma Ag immunotherapy inhibits the experimental glioma growth, which may be a novel therapeutic remedy for the treatment of glioma.

Objective: Previous studies have demonstrated that levels of hypermethylation of paired boxed gene 1 in cervical tissues are associated with the grades of severities of cervical neoplasia in women, which suggests that testing for DNA methylation has a potential role in neoplasma screening. In this study, by testing methylation levels of PAX1 genes in cervical scrapings and cervical tissues of different lesion levels, aims to evaluate the diagnostic value of DNA methylation testing as a biomarker for early detecting cancerous changes in cervical tissues and to compare the efficacy between PAX1 methylation test and HPV test in detecting of cervical cancer., Methods: A total of 121 cervical scrapings were analyzed, including normal (n = 28), cervical intraepithelial neoplasm 1 (CIN1; n = 32), CIN2/3 (n = 34), and invasive cancer (n = 27), which were all diagnosed by pathologic examination., Results: The values of PAX1 methylation reference in invasive cancer (mean [SE], 26.3 [3.5]) was significantly higher than CIN2/3 (13. 2 [2.2]) and the CIN1 (4.5 [0.45]; P < 0.001). The PAX1 promoter was hypermethylated in 100% of invasive cancer tissue compared with 0% of normal tissue, 9% of CIN1, 44% of CIN2/3 (P < 0.01). Methylation levels of cervical scrapings and cervical tissues represent strong consistency within each group. In contrast, the HPV test result was positive in 17% of normal tissue, 81% of CIN1, 91% of CIN2/CIN3, and 92% of invasive cancer. Based on receiver operating characteristic (ROC) analysis, hypermethylation of PAX1 was a significant candidate in segregating cervical cancer from normal/cervical neoplasia cases (P < 0.001). At an optimal cutoff value, sensitivity and specificity between 80% and 93% were obtained. In conclusion, the current results indicated that the methylation density of PAX1 by pyrosequencing in cervical scrapings held a great promise for cervical cancer screening.

There is now a large body of evidence demonstrating that fluid mechanical forces generated by blood flowing through the vasculature play a direct role in regulating endothelial cell structure and function. Integrin receptors that localize to the basal surface of the endothelium participate in both outside-in and inside-out signaling events that influence endothelial gene expression and morphology in response to flow. Our analyses of apical plasma membranes derived from cultured bovine aortic endothelial cells revealed that integrins are also expressed on this cell surface. Here, we tested whether these integrins participate in mechanotransduction events that are known to occur on the endothelial cell luminal/apical membrane. We found that apically expressed β 1 integrins are rapidly activated in response to acute shear stress. Blockade of β 1 integrin activation attenuated a shear-induced signaling cascade involving Src-family kinase, PI3-kinase, Akt and eNOS on this cell surface. In addition, β 1 integrin activation and associated signaling events were dependent on the structural integrity of caveolae but not the actin cytoskeleton. Taken together, these data indicate that endothelial responses to shear stress are mediated by spatially distinct pools of integrins.

Excessive estrogenic influence is known to be associated with initiation/promotion of endometrial cancer (EC). Common variants among genes coding for enzymes in sex steroid biosynthetic pathways may influence the risk of EC. Cytochrome P450c17α (CYP17), a gene that codes for a key enzyme (cytochrome P450c17α) in a rate-limiting step of estrogen biosynthesis has attracted considerable attention as a candidate gene for EC. The relationship between CYP17 and EC has been reported in various ethnic groups; however, these studies have yielded contradictory results. To investigate this inconsistency, we performed a meta-analysis of 13 studies involving a total of 3,258 cases and 4,614 controls for -34T>C (rs743572) polymorphism of the CYP17 gene to evaluate the effect of CYP17 on genetic susceptibility for EC. An overall random effects odds ratio of 0.71 (95% confidence interval 0.58-0.88, P=0.001) was found under recessive genetic model. Stratified analysis based on ethnicity, sample size and Hardy-Weinberg equilibrium status was conducted to explore potential heterogeneity. This meta-analysis demonstrated that the C allele of -34T>C in CYP17 is a protective factor associated with decreased EC susceptibility, but these associations vary in different ethnic populations.

Rationale: The source of Ca(2+) to activate pathological cardiac hypertrophy is not clearly defined. Ca(2+) influx through the L-type Ca(2+) channels (LTCCs) determines "contractile" Ca(2+), which is not thought to be the source of "hypertrophic" Ca(2+). However, some LTCCs are housed in caveolin-3 (Cav-3)-enriched signaling microdomains and are not directly involved in contraction. The function of these LTCCs is unknown., Objective: To test the idea that LTCCs in Cav-3-containing signaling domains are a source of Ca(2+) to activate the calcineurin-nuclear factor of activated T-cell signaling cascade that promotes pathological hypertrophy., Methods and Results: We developed reagents that targeted Ca(2+) channel-blocking Rem proteins to Cav-3-containing membranes, which house a small fraction of cardiac LTCCs. Blocking LTCCs within this Cav-3 membrane domain eliminated a small fraction of the LTCC current and almost all of the Ca(2+) influx-induced NFAT nuclear translocation, but it did not reduce myocyte contractility., Conclusions: We provide proof of concept that Ca(2+) influx through LTCCs within caveolae signaling domains can activate "hypertrophic" signaling, and this Ca(2+) influx can be selectively blocked without reducing cardiac contractility.

A major source of reactive oxygen species (ROS) in endothelial cells is the NADPH oxidase enzyme complex. The selective distributions of any enzyme within cells have important implications in regulating enzyme effectiveness through facilitation of access to local substrates and/or product targets. Because membrane rafts provide a spatially preferable environment for a variety of enzyme systems, we sought to determine whether NADPH oxidase is present and functional in this plasma membrane compartment in endothelial cells. We found that, in resting endothelial cells, NADPH oxidase subunits were preassembled and the enzyme functional in membrane rafts, specifically in caveolae. Stimulation with TNF-alpha induced additional recruitment of the p47(phox) regulatory subunit to raft-localized NADPH oxidase and enhanced ROS production within raft domains. TNF-alpha also induced nitric oxide production through activation of endothelial nitric oxide synthase (eNOS) present in the same membrane compartment. The dual activation of superoxide and nitric oxide-generating systems provided a spatially favorable environment for nitration of tyrosine-containing proteins localized to rafts. Perturbation of membrane raft structural integrity with cholesterol-sequestering compounds caused the delocalization of NADPH oxidase subunits and eNOS from the rafts and inhibited TNF-alpha-induced ROS production and protein tyrosine nitration. Together, these data provide evidence that membrane rafts and caveolae play a role in the spatial regulation of NADPH oxidase and subsequent ROS/reactive nitrogen species in endothelial cells.

Reactive oxygen species (ROS) generated during pathological events, such as inflammation and ischemia-reperfusion, activates both proapoptotic and antiapoptotic signaling programs in endothelial cells. Because cholesterol-rich, plasma membrane rafts serve as platforms for organizing and integrating signaling transduction processes, we asked whether these membrane regions play a mechanistic role in H2O2-induced responses. Bovine aortic endothelial cell cultures exposed to a 500-microM bolus of H2O2 showed progressive activation of caspase 3 and an increase in the number of TUNEL-positive cells. Pretreatment with either wortmannin or PD 098059 heightened these apoptotic responses, demonstrating that both PI3 kinase/Akt and ERK1/2 serve as signaling mediators to alleviate H2O2 cytotoxic effects. To investigate the role of lipid rafts in these signaling processes, endothelial cells were pretreated with methyl-beta-cyclodextrin (CD) or filipin to ablate raft structures. H2O2-induced phosphorylation of Akt and ERK 1/2 was attenuated, while caspase 3 and the number of TUNEL positive cells was enhanced in CD-pretreated cells exposed to H2O2. Reconstitution of raft domains restored H2O2-induced Akt and ERK1/2 phosphorylation, which was concomitant with reduction of caspase 3 activation and DNA fragmentation. Taken together, our findings suggest that plasma membrane compartments rich in cholesterol participate in signal transduction pathways activated by oxidative stress.
