In this study, a novel Z-scheme heterojunction photocatalyst was developed by integrating g-C3N4 nanoplates into ZnIn2S4 microspheres. X-ray photoelectron spectroscopy analysis revealed a directional electron transfer from g-C3N4 to ZnIn2S4 upon heterojunction formation. Under irradiation, electrochemical tests and electron paramagnetic resonance spectroscopy demonstrated significantly enhanced charge generation and separation efficiencies in the ZnIn2S4/g-C3N4 composite, accompanied by reduced charge transfer resistance. In photocatalytic CO2 reduction, the ZnIn2S4/g-C3N4 composite achieved the highest CO yield, 1.92 and 5.83 times higher than those of pristine g-C3N4 and ZnIn2S4, respectively, with a notable CO selectivity of 91.3% compared to H2 (8.7%). The Z-scheme heterojunction mechanism, confirmed in this work, effectively preserved the strong redox capabilities of the photoinduced charge carriers, leading to superior photocatalytic performance and excellent long-term stability. The periaqueductal gray (PAG) plays a well-established pivotal role in the descending pain modulatory circuit. The objective of this study was to investigate morphological changes in the astroglia in models that are commonly used in pain and itch studies. Five different mouse models of pain, as well as two models of chronic itch, were established using complete Freund's adjuvant (CFA), spared nerve injury (SNI), bone cancer pain (BCP), cisplatin (CIS), and paclitaxel (PTX) for pain, and diphenylcyclopropenone (DCP) and acetone and diethyl ether followed by water (AEW) for chronic itch. von Frey tests and video recordings were employed to assess pain and itching behaviors. The immunofluorescence of S100β, pSTAT3, and glial fibrillary acidic protein (GFAP) was examined. Two- and three-dimensional studies were used to evaluate changes in astrocyte morphology. Significant scratching was caused by DCP and AEW, whereas the administration of CFA, SNI, BCP, CIS, and PTX produced clear mechanical allodynia. The expression of GFAP in the lPAG/vlPAG was upregulated in CFA, SNI, BCP, CIS, PTX, and DCP mice but decreased in AEW mice. According to Sholl analysis, CFA, SNI, PTX, and BCP mice showed substantially higher astrocyte intersections in the vlPAG, whereas CFA, SNI, BCP, CIS, and DCP mice presented longer peak lengths. In three-dimensional analysis, CFA, SNI, PTX, and DCP mice showed increased astrocyte surface areas, while CIS and AEW mice showed both reduced surface areas and/or volumes of astrocytes. The findings showed that different pain and itching conditions have different astrocyte morphologies, and these variations in morphological changes help to explain the pathophysiology of these conditions. Cerebral ischemia-reperfusion injury involves a series of pathophysiological processes that occur when blood supply is restored after cerebral vascular obstruction, leading to neuronal damage. The AMPK/ERK1/2 signaling pathway has been identified as crucial in this process, although the exact mechanisms underlying the induction of ischemia-reperfusion injury remain unclear. In this study, we investigated the involvement of the AMPK/ERK1/2 signaling pathway in neuronal oxidative stress damage following cerebral ischemia-reperfusion by establishing animal and cell models. Our experimental results demonstrated that cerebral ischemia-reperfusion leads to oxidative stress damage, including cell apoptosis and mitochondrial dysfunction. Moreover, further experiments showed that inhibition of AMPK and ERK1/2 activity, using U0126 and Compound C respectively, could alleviate oxidative stress-induced cellular injury, improve mitochondrial morphology and function, reduce reactive oxygen species levels, increase superoxide dismutase levels, and suppress apoptosis. These findings clearly indicate the critical role of the AMPK/ERK1/2 signaling pathway in regulating oxidative stress damage and cerebral ischemia-reperfusion injury. Two-dimensional helical nanowire and nanosphere superstructures of PS- b -P2VP/S-mandelic acid with homochirality have been fabricated at the liquid/liquid and air/liquid interfaces via two kinds of approaches, respectively. Chiral supramolecules have attracted great research interest due to their unique properties and functions. How to assemble them into the ordered superstructures, however, remains a challenge. In this paper, two methods have been developed to fabricate such superstructures. In method I, polystyrene- b -poly (2-vinylpyridine) (PS- b -P2VP) and S-mandelic acid (S-MA) diffused from the oil and aqueous phases respectively to the oil/water interface, combined and self-assembled into helical nanowires and nanospheres with monochirality, and formed parallel-aligned nanowire arrays and close-packed arrays at the liquid/liquid interface. In method II, PS- b -P2VP in DMF droplets was introduced into the aqueous solution of S-MA through a liquid/liquid interfacial phase transfer process. Then the polymer molecules self-assembled into helical nanowires and nanospheres with monochirality in the aqueous phase, which adsorbed at the air/liquid interface and organized into superstructures. The formation of chiral aggregates should be attributed to the induction of S-MA, while the superstructures at the interfaces were considered to result from the modulation by the interfacial pressures. This study has developed one-step techniques to assemble the chiral aggregates and superstructures, which is expected to become universal for fabrication of functional superstructures. The cone penetration test (CPT) is a major in-situ testing with advantages in efficiency, reliability, and continuous measurement. However, it is often limited in penetration depth in soils such as dense sand and gravel due to high resistance and insufficient pushing force. Studies have shown that rotation can reduce penetration resistance, leading to the development of the rotary CPT. In addition, the torque measurement in rotary CPT could potentially provide extra information for soil behavior. This study simulates regular and rotary CPT tests with varied confining pressure and initial relative density using the discrete element method (DEM). Macroscale results verify that rotation can improve the penetrability of CPT by reducing vertical soil resistance. The reduction mechanism was explained by the microscopic observations and theoretical calculations that rotation can reduce the magnitude of the contact force and tilt its main direction horizontally. To advance the application of rotary CPT, a strong linear correlation between the measurements in rotary and regular CPTs was established, enabling interpretation of the rotary CPT measurements via the vast knowledge accumulated for regular CPT soundings. Furthermore, a theoretical derivation was developed to calculate the soil-probe interface friction coefficient directly by using the measurements of rotary CPT. The influence of the confining pressure, probe friction coefficient, and rotational speed ratio on the calculation accuracy are discussed and explained via particle-scale observations. Nicotine, a naturally occurring alkaloid found in tobacco, is a potent neurotoxin extensively used to control Nilaparvata lugens (Stål), a destructive insect pest of rice crops. The insect gut harbors a wide array of resident microorganisms that profoundly influence several biological processes, including host immunity. Maintaining an optimal gut microbiota and immune homeostasis requires a complex network of reciprocal regulatory interactions. However, the underlying molecular mechanisms driving these symbiotic exchanges, particularly between specific gut microbe and immunity, remain largely unknown in insects. Our previous investigations identified and isolated a nicotine-degrading Burkholderia cepacia strain (BsNLG8) with antifungal properties. Building on those findings, we found that nicotine intake significantly increased the abundance of a symbiotic bacteria BsNLG8, induced a stronger bacteriostatic effect in hemolymph, and enhanced the nicotine tolerance of N. lugens. Additionally, nicotine-induced antimicrobial peptides (AMPs) exhibited significant antibacterial effects against Staphylococcus aureus. We adopted RNA-seq to explore the underlying immunological mechanisms in nicotine-stressed N. lugens. Bioinformatic analyses identified numerous differentially expressed immune genes, including recognition/immune activation (GRPs and Toll) and AMPs (i.e., Defensin, Lugensin , lysozyme). Temporal expression profiling (12, 24, and 48 hours) of immune genes revealed pattern recognition proteins and immune effectors as primary responders to nicotine-induced stress. Defensin A , a broad-spectrum immunomodulatory cationic peptide, exhibited significantly high expression. RNA interference-mediated silencing of Defensin A reduced the survival, enhanced nicotine sensitivity of N. lugens to nicotine, and decreased the abundance of BsNLG8. The reintroduction of BsNLG8 improved the expression of immune genes, aiding nicotine resistance of N. lugens. Our findings indicate a potential reciprocal immunomodulatory interaction between Defensin A and BsNLG8 under nicotine stress. Moreover, this study offers novel and valuable insights for future research into enhancing nicotine-based pest management programs and developing alternative biocontrol methods involving the implication of insect symbionts.
