Purpose: The aim of this study was to evaluate the clinical response and safety of immediately loaded single-tooth implants placed in the posterior region of the maxilla and mandible. Materials and Methods: Single-tooth implants were placed in healed extraction sites in 20 adult patients. Temporary prefabricated acrylic resin crowns were prepared and adjusted. The crown occlusion was adjusted to obtain minimal contacts in maximum intercuspation. After 6 weeks a ceramometal or all-ceramic crown was cemented. Radiographic and clinical examinations were made at baseline and at 3, 6, and 12 months. Cortical bone response and peri-implant mucosal responses were evaluated. Results: The marginal bone level at the time of implant placement was preserved. The mean change in marginal bone level was 0.01 mm at 12 months. The mean Periotest value after 360 days was -4. The peri-implant mucosal adaptation to the anatomic form of the provisional crown resulted in a natural esthetic outcome, and a gain in papilla length was observed. One implant failure was recorded because of provisional luting cement impaction. Discussion: Clinical research has shown that immediate loading is a possible treatment modality. The immediate functional loading of implants placed in this study resulted in bone adaptation to loading. A satisfactory success rate with positive tissue responses was achieved. Conclusions: The results of this limited investigation indicated that immediate loading of unsplinted single-tooth implants in the posterior region may be a viable treatment option with an esthetic outcome.—Reprinted with permission of Quintessence Publishing. We measured the static error rate of a high-T[sub c] superconductor dc superconducting quantum interference device (SQUID), which, in the form as a storage loop for single flux quanta, is a basic element of rapid single flux quantum circuits. Using high-T[sub c] multilayer bicrystal technology, we fabricated a stacked dc SQUID pair, one SQUID serving as the storage loop, the other one as the readout device. The escape rate of a stored flux quantum was measured as a function of the bias current at a temperature of 28 K. Thick (up to 20 μm) coatings of yttria-stabilized zirconia with columnar structure have been grown by thermal MOCVD at 630–820 °C using acetylacetonates of Zr and Y as precursors. The samples have been characterized by Scanning Electron Microscopy (SEM), Wave Dispersion X-ray Analysis (WDX) and X-ray Diffraction (XRD) to find out the interplay of deposition conditions with coating microstructure, yttria content and phase composition. In particular, it has been found that homogenous nucleation reactions in gas phase at high temperature lead to the decrease of total growth rate and enrichment of resulting coating with yttrium. It has also been found that heterovalent substitution in ZrO2 leads to remarkable increase of a column width and a coating integrity. MOCVD is an economic method for the production of coated conductors. A single source MOCVD-system for continuous deposition on metal tapes was developed. Single crystals, IBAD buffered tapes and textured Ni-tapes were transported through the deposition zone and coated with YBCO. The velocity for the substrate transport was 4 m/h. On single crystals and IBAD buffered substrates the best critical current density of the 300–350 nm thick YBCO films were 2.5 and 4.8 MA cm−2 at 77 K. On CeO2/YSZ/CeO2 buffered textured nickel tapes the jc of the 500 μm thick YBCO film was 0.25 MA cm−2 at 77 K. Textured oxide buffer layers were deposited by MOCVD on single crystals and textured Ni in reducing atmosphere. [Copyright &y& Elsevier]

Objectives: Cold atmospheric plasma (CAP), a room temperate ionized gas, seems to be a possible way to enhance tissue recovery. An in vitro study was conducted to investigate the influence of medical CAP on the regenerative capacity of human periodontal ligament (PDL) cells.Material and methods: Human PDL cells were subjected to CAP at various intensities, distances, and durations. The effects of CAP on a number of specific markers were studied at transcriptional level using real-time PCR. Additionally, an in vitro wound healing assay was applied to PDL cell monolayers either in the presence or absence of CAP by using JuLI™ Br Live Cell Analyzer and software. Finally, cell viability of CAP-treated cells was analyzed by an XTT assay.Results: CAP treatment enhanced significantly the expression of the cytokines tumor necrosis factor (TNF)α, cyclooxygenase (COX)2, interleukin (IL)-1β, IL-6, IL-8, collagen (COL)1α, and matrix metalloproteinase (MMP)1, as well as the proliferation markers Ki67 and proliferating cell nuclear antigen (PCNA), but downregulated apoptotic markers Apaf1 and p53. Additionally, the in vitro wound healing rate was significantly enhanced after CAP application. Moreover, CAP treatment resulted in a significantly increased cell viability in the XTT assay.Conclusion: This in vitro study shows that CAP has regulatable effects on markers of periodontal wound healing thereby underlining the potential use of CAP as a benefit treatment strategy.Clinical relevance: Our study demonstrates the application of CAP in the treatment of oral pathologies suggesting a promising future treatment approach. Mdm2 is the major negative regulator of p53 tumor-suppressor activity. This oncoprotein is overexpressed in many human tumors that retain the wild-type p53 allele. As such, targeted inhibition of Mdm2 is being considered as a therapeutic anticancer strategy. The N-terminal hydrophobic pocket of Mdm2 binds to p53 and thereby inhibits the transcription of p53 target genes. Additionally, the C-terminus of Mdm2 contains a RING domain with intrinsic ubiquitin E3 ligase activity. By recruiting E2 ubiquitin-conjugating enzyme(s), Mdm2 acts as a molecular scaffold to facilitate p53 ubiquitination and proteasome-dependent degradation. Mdmx (Mdm4), an Mdm2 homolog, also has a RING domain and hetero-oligomerizes with Mdm2 to stimulate its E3 ligase activity. Recent studies have shown that C-terminal residues adjacent to the RING domain of both Mdm2 and Mdmx contribute to Mdm2 E3 ligase activity. However, the molecular mechanisms mediating this process remain unclear, and the biological consequences of inhibiting Mdm2/Mdmx co-operation or blocking Mdm2 ligase function are relatively unexplored. This study presents biochemical and cell biological data that further elucidate the mechanisms by which Mdm2 and Mdmx co-operate to regulate p53 level and activity. We use chemical and genetic approaches to demonstrate that functional inhibition of Mdm2 ubiquitin ligase activity is insufficient for p53 activation. This unexpected result suggests that concomitant treatment with Mdm2/Mdmx antagonists may be needed to achieve therapeutic benefit. [ABSTRACT FROM AUTHOR]

A RTR MOCYD was used for the deposition of YBCO and various oxides. A new MOCVD technique enable us to coat the metal tape directly without formation of NiO. At a low oxygen partial pressure the buffer layers were grown on the metal tape with good in plane and out of plane texture. The texture of the MgO buffer layers depend on the surface of the alloyed Ni-tape. YBCO layers exhibit excellent out of plane texture with FWHM(005) = 1.4 – 3.5° and good in plane texture of FWHM(103) = 5 – 7°. On MgO buffered Ni5at%W the inductive measured Jc was 0.3–0.7 MA/cm2 at 77 K. [ABSTRACT FROM AUTHOR]

The article focuses on keeping p53 in check under functions of Mdm2 and Mdm4 proteins. In the absence of stress signals, the p53 protein is kept in check to allow normal cell proliferation and maintenance of cell viability. In this process, the critical importance are the two structurally related proteins, the Mdm2 and Mdm4 proteins. Germline inactivation of either mdm2 and mdm4 leads to embryonic lethal phenotypes that are completely overcome by noncomitant inactivation of the p53 gene.

Summary. Fungal infections of the maxillary sinus are frequently caused by Aspergillus species, particularly A. fumigatus . In otherwise healthy persons there is an association with overfilling of dental root canals, when zinc-containing filling materials were used. Biaxially textured buffer layers and YBa2Cu3O7 (YBCO) films were grown on NiO/Ni based substrates by pulsed injection MOCVD. High quality biaxially textured NiO layers have been prepared following an optimised recrystallisation–oxidation process. The yttria stabilized zirconia (YSZ) and its combination with CeO2 and/or Y2O3 were investigated in the role of buffer layers for YBCO. Thin biaxially textured buffer layers were deposited in a temperature range from 600 to 800 °C. The best superconducting properties correspond to the Ni/NiO/YSZ/Y2O3/YBCO architecture: YBCO layers were grown epitaxially and exhibited a critical temperature Tc∼90 K (ΔTc⩽1 K) and critical current densities Jc⩾5×105 A/cm2 at 77 K.
