Three-dimensional turbulent magnetoconvection at a Rayleigh number of Ra = 107 in liquid gallium at a Prandtl number Pr = 0.025 is studied in a closed square cell for very strong external vertical magnetic fields B0 in direct numerical simulations which apply the quasistatic approximation. As B0, or equivalently the Hartmann number Ha, are increased, the convection flow, which is highly turbulent in the absence of magnetic fields, crosses the Chandrasekhar linear stability limit for which thermal convection ceases in an infinitely extended layer and which can be assigned a critical Hartmann number Hac. Similar to rotating Rayleigh-Bénard convection, our simulations reveal subcritical sidewall modes that maintain a small but finite convective heat transfer for Ha > Hac. We report a detailed analysis of the complex two-layer structure of these wall modes, their extension into the cell interior, and a resulting sidewall boundary layer composition that is found to scale with the Shercliff layer thickness. Aims: Dianthus caryophyllus is a commercially important ornamental flower. Plant growth promoting rhizobacteria are increasingly applied as bio-fertilisers and bio-fortifiers. We studied the effect of a rhizospheric isolate Klebsiella SGM 81 strain to promote D. caryophyllus growth under sterile and non-sterile conditions, to colonise its root system endophytically and its impact on the cultivatable microbial community. We identified the auxin indole-3-acetic acid (IAA) production of Klebsiella SGM 81 as major bacterial trait most likely to enhance growth of D. caryophyllus.Methods: ipdC dependent IAA production of SGM 81 was quantified using LC-MS/MS and localised proximal to D. caryophyllus roots and correlated to root growth promotion and characteristic morphological changes. SGM 81 cells were localised on and within the plant root using 3D rendering confocal microscopy of gfp expressing SGM 81. Using Salkowski reagent IAA production was quantified and localised proximal to roots in situ. The effect of different bacterial titres on rhizosphere bacterial population was CFU enumerated on nutrient agar. The genome sequence of Klebsiella SGM 81 (accession number PRJEB21197) was determined to validate PGP traits and phylogenic relationships.Results: Inoculation of D. caryophyllus roots with Klebsiella SGM 81 drastically promoted plant growth when grown in agar and soil, concomitant with a burst in root hair formation, suggesting an increase in root auxin activity. We sequenced the Klebsiella SGM 81 genome, identified the presence of a canonical ipdC gene in Klebsiella SGM 81, confirmed bacterial production and secretion of IAA in batch culture using LC-MS/MS and localised plant dependent IAA production by SGM 81 proximal to roots. We found Klebsiella SGM 81 to be a rhizoplane and endophytic coloniser of D. caryophyllus roots in a dose dependent manner. We found no adverse effects of SGM 81 on the overall rhizospheric microbial population unless supplied to soil in very high titres.Conclusion: Klebsiella SGM 81 effectively improves root traits of D. caryophyllus in a dose dependent manner, likely through tryptophan dependent IAA production in the rhizoplane and potentially within the intercellular spaces of root tissue. Under optimal plant growth promoting conditions in non-sterile soil, the high total microbial titre in the rhizosphere supports a mutualistic relationship between Klebsiella SGM 81 and carnation that potentially extends to the wider rhizosphere microbiota. We report velocity measurements in a vertical turbulent convection flow cell that is filled with the eutectic liquid metal alloy gallium-indium-tin by the use of local Lorentz force velocimetry (LLFV) and ultrasound Doppler velocimetry. We demonstrate the applicability of LLFV for a thermal convection flow and reproduce a linear dependence of the measured force in the range of micronewtons on the local flow velocity magnitude. Furthermore, the presented experiment is used to explore scaling laws of the global turbulent transport of heat and momentum in this low-Prandtl-number convection flow. We explore the mechanisms of heat transfer in a turbulent constant heat flux-driven Rayleigh–Bénard convection flow, which exhibits a hierarchy of flow structures from granules to supergranules. Our computational framework makes use of time-dependent flow networks. These are based on trajectories of Lagrangian tracer particles that are advected in the flow. We identify coherent sets in the Lagrangian frame of reference as those sets of trajectories that stay closely together for an extended time span under the action of the turbulent flow. Depending on the choice of the measure of coherence, sets with different characteristics are detected. First, the application of a recently proposed evolutionary spectral clustering scheme allows us to extract granular coherent features that are shown to contribute significantly less to the global heat transfer than their spatial complements. Moreover, splits and mergers of these (leaking) coherent sets leave spectral footprints. Second, trajectories which exhibit a small node degree in the corresponding network represent objectively highly coherent flow structures and can be related to supergranules as the other stage of the present flow hierarchy. We demonstrate that the supergranular flow structures play a key role in the vertical heat transport and that they exhibit a greater spatial extension than the granular structures obtained from spectral clustering. European mistletoe (Viscum album L.) has largely infested Central European forests and causes high mortality probability particularly in dry years. However, little information is available about the consequences of mistletoe infestation for metabolic processes in bark and wood of its host, despite their important roles in infestation defense. We analyzed the tissue hydration, carbohydrate composition, phytohormone profile, reactive oxygen species and anti-oxidant levels in bark and wood of Scots pines (Pinus sylvestris L.), as dependent on mistletoe infestation. As a consequence of mistletoe infestation, host bark and wood showed impaired hydration and reduced total carbon content. In the bark, soluble sugar and lignin contents increased, apparently at the expense of holo-cellulose. Hydrogen peroxide accumulation was accompanied by increased glutathione and decreased reduced ascorbic acid levels. Mistletoe infestation mediated alteration of the phytohormone profile in bark and wood of its host. Cytokinins, jasmonic acid and abscisic acid levels increased in both tissues, whereas salicylic acid and indole-3-acetic acid, which were only detected in the bark, declined. The present results show that mistletoe infestation affects both the host's anti-oxidative defense system and the phytohormone profile after establishment of the xylem tapping haustorium. The significance of these processes for the development of the woody mistletoe stem and the haustorium is discussed. [ABSTRACT FROM AUTHOR]
