358. Integrated and Functional Genomics Analysis Validates the Relevance of the Nuclear Variant ErbB380kDa in Prostate Cancer Progression.

Abstract

The EGF-family of tyrosine-kinase receptors activates cytoplasmic pathways involved in cell proliferation, migration and differentiation in response to specific extracellular ligands. Beside these canonical pathways, the nuclear localization of the ErbB receptors in primary tumours and cancer cell lines led to investigate their role as transcriptional regulators of cancer genes. The nuclear localization of ErbB3 has been reported in various cancer tissues and cell lines but the nuclear functions and the putative correlation with tumour progression and resistance to therapy remain unclear. We first assessed ErbB3 expression in normal and tumour prostate tissues. The nuclear staining was mainly due to an isoform matching the C-terminus domain of the full length ErbB3185kDa receptor. Nuclear staining was also restricted to cancer cells and was increased in advanced castration-resistant prostate cancer when compared to localized tumours, suggesting it could be involved in the progression of prostate cancer up to the terminal castration-resistant stage. ChIP-on-chip experiments were performed on immortalized and tumour cell lines selected upon characterization of endogenous nuclear expression of an ErbB380kDa isoform. Among the 1840 target promoters identified, 26 were selected before ErbB380kDa-dependent gene expression was evaluated by real-time quantitative RT-PCR, providing evidence that ErbB380kDa exerted transcriptional control on those genes. Some targets are already known to be involved in prostate cancer progression even though no link was previously established with ErbB3 membrane and/or nuclear signalling. Many others, not yet associated with prostate cancer, could provide new therapeutic possibilities for patients expressing ErbB380kDa. Abstract

Introduction: The objective of the study was to establish the predictive value of prenatal ultrasound markers for complex gastroschisis (GS) in the first 10 days of life. Material and Methods: In this retrospective cohort study over 11 years (2000-2011) of 117 GS cases, the following prenatal ultrasound signs were analyzed at the last second- and third-trimester ultrasounds: intrauterine growth restriction, intra-abdominal bowel dilatation (IABD) adjusted for gestational age, extra-abdominal bowel dilatation (EABD) ≥25 mm, stomach dilatation, stomach herniation, perturbed mesenteric circulation, absence of bowel lumen and echogenic dilated bowel loops (EDBL). Results: Among 114 live births, 16 newborns had complex GS (14.0%). Death was seen in 16 cases (13.7%): 3 intrauterine fetal deaths, 9 complex GS and 4 simple GS. Second-trimester markers had limited predictive value. Third-trimester IABD, EABD, EDBL, absence of intestinal lumen and perturbed mesenteric circulation were statistically associated with complex GS and death. IABD was able to predict complex GS with a sensitivity of 50%, a specificity of 91%, a positive predictive value of 47% and a negative predictive value of 92%. Discussion: Third-trimester IABD adjusted for gestational age appears to be the prenatal ultrasound marker most strongly associated with adverse outcome in GS. [ABSTRACT FROM AUTHOR]

Abstract

The relatively recent use of tire-derived aggregates (TDA) in civil engineering applications makes it necessary to thoroughly understand and characterize its behaviour under static and dynamic loads. Only a limited number of experimental studies have investigated the dynamic behaviour of TDA with large aggregate sizes. This paper aims to investigate the dynamic and cyclic behaviours of TDA and factors that could affect the characterization of TDA using cyclic simple shear tests. A comprehensive series of cyclic direct simple shear tests were performed in this study on TDA under vertical stress and shear strain amplitude ranges of 15–400 kPa and 0.01–10%, respectively. Small-strain shear moduli (G max) of TDA were also determined from shear wave velocities measured at vertical stresses of 15–800 kPa using bender elements. The results were used to study the effects of cyclic loading frequency, drainage condition, fabric anisotropy, shear strain amplitude, and vertical stress on the dynamic behavior of TDA. Shear modulus and damping ratio of TDA ranged from 85 to 4290 kPa and 7–25%, respectively. While the shear modulus of TDA was found to be strongly influenced by the loading frequency, drainage condition, anisotropy, shear strain amplitude and vertical stress, these testing parameters had little or reduced effects on the damping ratios of TDA specimens. Abstract

In the city of Steyr the Enns River causes frequent inundations in some exposed areas. The endangered areas encompassed by this project are situated on the outer side of a river bend. Next to the settlement area, an estuary of a small creek is also located. Therefore, the residential estate may be overflowed on two sides - by the river Enns and by the creek called Ramingbach. However, the requests of the stakeholders were challenging, because the flood control levees should be avoided. To protect the settlement area, different alternative measures were tested on physical and numerical models: several changes in the riverbed, such as deepening of the riverbed stream up and down, and widening of the river bend on the inner side. The aim of this contribution is to present different alternative ways of flood protection besides levees, to evaluate them, and to discuss them basing on the results of physical and numerical simulations. Abstract

Background: Chlamydiae are intracellular bacteria that cause various severe diseases in humans and animals. The common treatment for chlamydia infections are antibiotics. However, when antibiotics are misused (overuse or self-medication), this may lead to resistance of a number of chlamydia species, causing a real public health problem worldwide. Materials and methods: In the present work, a comprehensive literature search was conducted in the following databases: PubMed, Google Scholar, Cochrane Library, Science direct and Web of Science. The primary purpose is to analyse a set of data describing the genes and mutations involved in Chlamydiae resistance to antibiotic mechanisms. In addition, we proceeded to a filtration process among 704 retrieved articles, then finished by focusing on 24 studies to extract data that met our requirements. Results: The present study revealed that Chlamydia trachomatis may develop resistance to macrolides via mutations in the 23S rRNA, rplD, rplV genes, to rifamycins via mutations in the rpoB gene, to fluoroquinolones via mutations in the gyrA, parC and ygeD genes, to tetracyclines via mutations in the rpoB gene, to fosfomycin via mutations in the murA gene, to MDQA via mutations in the secY gene. Whereas, Chlamydia pneumoniae may develop resistance to rifamycins via mutations in the rpoB gene, to fluoroquinolones via mutations in the gyrA gene. Furthermore, the extracted data revealed that Chlamydia psittaci may develop resistance to aminoglycosides via mutations in the 16S rRNA and rpoB genes, to macrolides via mutations in the 23S rRNA gene. Moreover, Chlamydia suis can become resistance to tetracyclines via mutations in the tet(C) gene. In addition, Chlamydia caviae may develop resistance to macrolides via variations in the 23S rRNA gene. The associated mechanisms of resistance are generally: the inhibition of bacteria's protein synthesis, the inhibition of bacterial enzymes' action and the inhibition of bacterial transcription process. Conclusion: This literature review revealed the existence of diverse mutations associated with resistance to antibiotics using molecular tools and targeting chlamydia species' genes. Furthermore, these mutations were shown to be associated with different mechanisms that led to resistance. In that regards, more mutations and information can be shown by a deep investigation using the whole genome sequencing. Abstract

The application of genomics technologies to medicine and biomedical research is increasing in popularity, made possible by new high-throughput genotyping and sequencing technologies and improved data analysis capabilities. Some of the greatest genetic diversity among humans, animals, plants, and microbiota occurs in Africa, yet genomic research outputs from the continent are limited. The Human Heredity and Health in Africa (H3Africa) initiative was established to drive the development of genomic research for human health in Africa, and through recognition of the critical role of bioinformatics in this process, spurred the establishment of H3ABioNet, a pan-African bioinformatics network for H3Africa. The limitations in bioinformatics capacity on the continent have been a major contributory factor to the lack of notable outputs in high-throughput biology research. Although pockets of high-quality bioinformatics teams have existed previously, the majority of research institutions lack experienced faculty who can train and supervise bioinformatics students. H3ABioNet aims to address this dire need, specifically in the area of human genetics and genomics, but knock-on effects are ensuring this extends to other areas of bioinformatics. Here, we describe the emergence of genomics research and the development of bioinformatics in Africa through H3ABioNet.

Abstract

This article addresses the problem of pervasive microtuning in Musical Instrument Digital Interface (MIDI) networks within the context of the general weaknesses and underusage of MIDI technology. A technique to overcome the bandwidth and network routing limitation in a complex MIDI system is to integrate MIDI communication with a high-speed local area network. A polyphonic aftertouch (PA) or key aftertouch technique can satisfy all the criteria of a good solution for microtuning. An object-oriented program was designed to model each note as an abstract object that encapsulates the note attributes with its retuning using PA. This pervasive microtuning technique was tested on three basic factors: retunability, tone quality and performance. All three tests were passed by the program.

Abstract

Background Multisystem inflammatory syndrome in children (MIS-C), a hyperinflammatory condition associated with SARS-CoV-2 infection, has emerged as a serious illness in children worldwide. Immunoglobulin or glucocorticoids, or both, are currently recommended treatments. Methods The Best Available Treatment Study evaluated immunomodulatory treatments for MIS-C in an international observational cohort. Analysis of the first 614 patients was previously reported. In this propensity-weighted cohort study, clinical and outcome data from children with suspected or proven MIS-C were collected onto a web-based Research Electronic Data Capture database. After excluding neonates and incomplete or duplicate records, inverse probability weighting was used to compare primary treatments with intravenous immunoglobulin, intravenous immunoglobulin plus glucocorticoids, or glucocorticoids alone, using intravenous immunoglobulin as the reference treatment. Primary outcomes were a composite of inotropic or ventilator support from the second day after treatment initiation, or death, and time to improvement on an ordinal clinical severity scale. Secondary outcomes included treatment escalation, clinical deterioration, fever, and coronary artery aneurysm occurrence and resolution. This study is registered with the ISRCTN registry, ISRCTN69546370. Findings We enrolled 2101 children (aged 0 months to 19 years) with clinically diagnosed MIS-C from 39 countries between June 14, 2020, and April 25, 2022, and, following exclusions, 2009 patients were included for analysis (median age 8·0 years [IQR 4·2–11·4], 1191 [59·3%] male and 818 [40·7%] female, and 825 [41·1%] White). 680 (33·8%) patients received primary treatment with intravenous immunoglobulin, 698 (34·7%) with intravenous immunoglobulin plus glucocorticoids, 487 (24·2%) with glucocorticoids alone; 59 (2·9%) patients received other combinations, including biologicals, and 85 (4·2%) patients received no immunomodulators. There were no significant differences between treatments for primary outcomes for the 1586 patients with complete baseline and outcome data that were considered for primary analysis. Adjusted odds ratios for ventilation, inotropic support, or death were 1·09 (95% CI 0·75–1·58; corrected p value=1·00) for intravenous immunoglobulin plus glucocorticoids and 0·93 (0·58–1·47; corrected p value=1·00) for glucocorticoids alone, versus intravenous immunoglobulin alone. Adjusted average hazard ratios for time to improvement were 1·04 (95% CI 0·91–1·20; corrected p value=1·00) for intravenous immunoglobulin plus glucocorticoids, and 0·84 (0·70–1·00; corrected p value=0·22) for glucocorticoids alone, versus intravenous immunoglobulin alone. Abstract

Aseptic loosening and mechanical failure of acetabular reinforcement components are among the main causes of their reduced service life. Current acetabular implants typically feature a structural solid layer that provides load bearing capacity, coated with a foam of uniform porosity to reduce stress shielding and implant loosening. This paper presents an alternative concept for a 3D printed cage that consists of a multifunctional fully porous layer with graded attributes that integrate both structural function and bone in-growth properties. The design comprises a hemispherical cup affixed to a superior flange with architecture featuring an optimally graded porosity. The methodology here presented combines an upscaling mechanics scheme of lattice materials with density-based topology optimization, and includes additive manufacturing constraints and bone ingrowth requirements in the problem formulation. The numerical results indicate a 21.4% reduction in the maximum contact stress on the bone surface, and a 26% decrease in the bone-implant interface peak micromotion, values that are indicative of enhanced bone ingrowth and implant long-term stability. 395. Impacts of climate change on water quantity, water salinity, food security, and socioeconomy in Egypt

Abstract

Climate change might have direct impacts on water quantity in Egypt and lead to indirect effects on Mediterranean saltwater intrusion to groundwater, which exposes agriculture to vulnerability. This study investigated impacts of climate change on agriculture, with particular regard to food security and socioeconomy, and quantified the effectiveness of cropping pattern adaptation measures by integrating three mathematical models. The BlueM model was used for hydrological simulations of Nasser Lake under flooding scenarios to predict the water supply from the High Aswan Dam. The water and salinity balance (WB-SAL) model was adopted to estimate the water salinity in the Nile Delta. The simulated results from the BlueM and WB-SAL models were integrated with the agricultural simulation model for Egypt (ASME) to project cropping patterns, food security, and socioeconomy throughout the country. The results showed that future climate change will directly affect the total crop area; crop areas for 13 crop types; the self-sufficiency of wheat, rice, cereal, and maize supplies; and socioeconomic indicators. The proposed cropping pattern adaptation measures focus on fixing the crop areas of rice and orchards and providing half of the population with lentils, maize, onion, vegetables, milk, and meat. The adaptation measures have the potential to promote food security without causing deterioration of the socioeconomic situation. However, water availability has much more significant effects on food security and socioeconomy than cropping pattern adaptation measures do. Accordingly, the country should rationalize water use efficiency and increase water supply.
