Abstract

The authors describe a fluorescence based assay for determination of the macrolid antibiotic erythromycin (ERY). It is based in the use of fluorescent gold nanoclusters (AuNCs) coated first with silica (to obtain AuNC@SiO2) and then with a molecularly imprinted polymer (MIP). The MIP was synthesized from 3-aminopropyltriethoxysilane as the functional monomer, ERY as the template, and tetraethoxysilane as the cross-linker via a sol-gel process that leads to surface imprinting. By using this method, the strong fluorescence of AuNCs was maintained in the resultant MIPs. After optimization of the experimental conditions, the signal (measured at excitation/emission wavelengths of 396/585 nm) decreases linearly with increasing concentration of ERY in the 0.1 to 11.9 μM ERY concentration range. The limit of detection is 12 nM, and the imprinting factor is 4.0. The method was successfuly applied to the determination of ERY in spiked human urine and saliva.

Abstract

Random effects in longitudinal multilevel models represent individuals' deviations from population means and are indicators of individual differences. Researchers are often interested in examining how these random effects predict outcome variables that vary across individuals. This can be done via a two‐step approach in which empirical Bayes (EB) estimates of the random effects are extracted and then treated as observed predictor variables in follow‐up regression analyses. This approach ignores the unreliability of EB estimates, leading to underestimation of regression coefficients. As such, previous studies have recommended a multilevel structural equation modeling (ML‐SEM) approach that treats random effects as latent variables. The current study uses simulation and empirical data to show that a bias–variance tradeoff exists when selecting between the two approaches. ML‐SEM produces generally unbiased regression coefficient estimates but also larger standard errors, which can lead to lower power than the two‐step approach. Abstract

Two of the most common and well-characterized epigenetic changes, DNA methylation and histone modifications, occur in leukemia. Decitabine (5-aza-2’-deoxycytidine, DAC), as a hypomethylating agent (HMA), and chidamide (CS055), as a histone deacetylase inhibitor (HDACi), each demonstrate effects against leukemia. However, whether the combination of low-dose DAC with chidamide constitutes an effective epigenetic regimen for the treatment of myeloid leukemia is currently unknown. In this study, the combination of DAC at low doses and chidamide showed enhanced inhibition of myeloid leukemia cell (K562, THP-1) growth. As a novel HDACi, chidamide increased the level of ace-H3K18 expression. Combined use of low-dose DAC and chidamide arrested the cell cycle at the G0/G1 phase by upregulating p21 expression, and the combination also suppressed PI3K/AKT/mTOR signaling pathway. Furthermore, chidamide enhanced the apoptotic effect of DAC by downregulating expression of Bcl-2 and pro-caspase-3 and upregulating that of Bax, cleaved PARP-1, and caspase-9. Moreover, the mitochondrial transmembrane potential was significantly decreased in DAC-, chidamide-, or combination-treated leukemia cells. These results suggest that targeting the leukemia epigenome through the combination of low-dose DAC and chidamide is a promising approach.

Abstract

To understand new fathers' experiences and well-being, we examine links between fathers and their partners' replenishing and stressful daily experiences-exercise, sleep, work, chores, general stress, and parenting stress-and their own and their partners' well-being and family relations. Fathers and mothers of ten-month old infants (N=143/140 mothers/fathers) in the U.S. reported on daily experiences for eight consecutive days. Results of multilevel models indicated that more replenishing and fewer stressful daily experiences were generally linked to more parent happiness, better couple relations, and greater closeness with the infant. Several gender differences also emerged that may reflect different stress and coping processes or different social roles for mothers and fathers; most striking was that on days that fathers spent more time on chores, mothers reported greater couple closeness but fathers reported more arguments. This exploration of new parents' daily experiences demonstrates the value of the method to generate intervention-relevant insights, as well as the importance of examining fathers' (and mothers') experiences in the context of couple-level dynamics.

Abstract

Mixture modeling is commonly used to model sample heterogeneity by identifying unobserved classes of individuals with similar characteristics. Despite abundance of evidence in the literature suggesting that individuals are often characterized by different dynamic processes underlying their physiological, cognitive, psychological, and behavioral states, applications of dynamic mixture modeling are surprisingly lacking. We present here a proof-of-concept example of dynamic mixture modeling, where latent groups of individuals were identified based on different dynamic patterns in their time series. Our sample consists of 192 men who were in a heterosexual relationship. They were asked to complete a daily questionnaire involving emotions related to their relationship. Two latent groups were identified based on the strength of association between positive (e.g., loving) and negative (e.g., doubtful) affect. Men in the group characterized by a strong negative association ( β = −.67 ) tended to be younger and had higher levels of anxiety toward their relationship than men in the other group, which was characterized by a weaker negative association ( β = −.31 ). We illustrate the specification and estimation of dynamic mixture model using "dynr," an R package capable of handling a broad class of linear and nonlinear discrete- and continuous-time models with regime-switching properties. Abstract

Background and Objectives Scholars argue that volunteering enhances social, physical, and cognitive activities that are increasingly valued as people age, which in turn improves older adults' well-being via a host of psychosocial and neurobiological mechanisms. This study explicitly tested older adults' self-perceptions of aging as a mechanism underlying the mental health benefits of volunteering. Research Design and Methods Using 2-wave data from the Health and Retirement Study (2008/2010 for Wave 1 and 2012/2014 for Wave 2), we analyzed reports from a pooled sample of older adults aged 65 or older (N = 9,017). Participants reported on demographic characteristics, volunteer work (did not volunteer, 1–99 h/year, 100+ h/year), self-perceptions of aging, and depressive symptoms. We estimated an autoregressive cross-lagged panel model. Results Volunteering for 100 h or more per year was associated with older adults' more positive and less negative self-perceptions of aging in the subsequent wave (i.e. 4 years later), which in turn predicted fewer depressive symptoms. Discussion and Implications This study suggests the promising role of volunteering in shaping older adults' self-perceptions of aging on a sustained basis and refines our understanding of the benefits volunteering brings.
