The co-existence of volatile chlorinated hydrocarbons (VCHs) and nitrate pollution in groundwater is prominent, but how nitrate exposure affects weak-electrical stimulated bio-dechlorination activity of VCH is largely unknown. Here, by establishing weak-electrical stimulated trichloroethylene (TCE) dechlorination systems, the influence on TCE dechlorination by exposure to the different concentrations (25-100 mg L -1 ) of nitrate was investigated. The existence of nitrate in general decreased TCE dechlorination efficiency to varying degrees, and the higher nitrate concentration, the stronger the inhibitory effects, verified by the gradually decreased transcription levels of tceA. Although the TCE dechlorination kinetic rate constant decreased by 36% the most, under all nitrate concentration ranges, TCE could be completely removed within 32 h and no difference in generated metabolites was found, revealing the well-maintained dechlorination activity. This was due to the quickly enriched bio-denitrification activity, which removed nitrate completely within 9 h, and thus relieved the inhibition on TCE dechlorination. The obvious bacterial community structure succession was also observed, from dominating with dechlorination genera (e.g., Acetobacterium, Eubacterium) to dominating with both dechlorination and denitrification genera (e.g., Acidovorax and Brachymonas). The response of the denitrification community to long-term antibiotic exposure requires further investigation. Here, the significantly altered denitrifying community structure and function were observed by continuous exposure to 1 mg/L sulfamethoxazole (SMZ) or chlortetracycline (CTC) for 180 d in the expanded granular sludge bed reactors. Thaurea, positively correlated with SMZ and NO 3 - removal efficiency (NrE), was highly enriched in the SMZ-added reactor, while, Comamons and Acinetobacter were largely inhibited. The acute inhibited and then gradual-recovered NrE (87.17-90.38 %) was observed with highly expressed narG, indicating the adaptability of Thaurea to SMZ. However, the abundance of Thaurea and Comamonas greatly decreased, while Melioribacter and Acinetobacter were largely enriched in the CTC-added reactor. CTC created more serious and continuous inhibition of NO 3 - reduction (NrE of 64.53-66.95 %), with lowly expressed narG. The accumulation of lipid peroxides is recognized as a determinant of the occurrence of ferroptosis. However, the sensors and amplifying process of lipid peroxidation linked to ferroptosis remain obscure. Here we identify PKCβII as a critical contributor of ferroptosis through independent genome-wide CRISPR-Cas9 and kinase inhibitor library screening. Our results show that PKCβII senses the initial lipid peroxides and amplifies lipid peroxidation linked to ferroptosis through phosphorylation and activation of ACSL4. Lipidomics analysis shows that activated ACSL4 catalyses polyunsaturated fatty acid-containing lipid biosynthesis and promotes the accumulation of lipid peroxidation products, leading to ferroptosis. Attenuation of the PKCβII-ACSL4 pathway effectively blocks ferroptosis in vitro and impairs ferroptosis-associated cancer immunotherapy in vivo. Our results identify PKCβII as a sensor of lipid peroxidation, and the lipid peroxidation-PKCβII-ACSL4 positive-feedback axis may provide potential targets for ferroptosis-associated disease treatment., (© 2022. Met tyrosine kinase, a receptor for a hepatocyte growth factor (HGF), plays a critical role in tumor growth, metastasis, and drug resistance. Mitochondria are highly dynamic and undergo fission and fusion to maintain a functional mitochondrial network. Dysregulated mitochondrial dynamics are responsible for the progression and metastasis of many cancers. Here, using structured illumination microscopy (SIM) and high spatial and temporal resolution live cell imaging, we identified mitochondrial trafficking of receptor tyrosine kinase Met. The contacts between activated Met kinase and mitochondria formed dramatically, and an intact HGF/Met axis was necessary for dysregulated mitochondrial fission and cancer cell movements. Mechanically, we found that Met directly phosphorylated outer mitochondrial membrane protein Fis1 at Tyr38 (Fis1 pY38). Fis1 pY38 promoted mitochondrial fission by recruiting the mitochondrial fission GTPase dynamin-related protein-1 (Drp1) to mitochondria. Fragmented mitochondria fueled actin filament remodeling and lamellipodia or invadopodia formation to facilitate cell metastasis in hepatocellular carcinoma (HCC) cells both in vitro and in vivo. These findings reveal a novel and noncanonical pathway of Met receptor tyrosine kinase in the regulation of mitochondrial activities, which may provide a therapeutic target for metastatic HCC., (© 2021. Tetrabromobisphenol A (TBBPA) has aroused widespread pollution in industrial wastewater. Microbial fuel cell (MFC) was proved powerful in organics degradation and simultaneous resource recovery during wastewater treatment. However, the TBBPA biotransformation potential, pathway and the related molecular mechanism remain poorly understood. In this study, the enhanced degradation and detoxification performance of TBBPA in MFC anode was confirmed, evidenced by the shorter degradation period (2.3 times shorter) and less generation of bisphenol A. UPLC-QTOF-MS analysis verified TBBPA metabolism went through reductive debromination, hydrolytic debromination, oxidative ring cleavage and o-methylation. Accompanied with those biochemical processes, the metabolites underwent dynamic changes. The distinctly decreased abundance and fewer interactions with other functional genera for the potential reductive dehalogenators (Pseudomonas, etc.) possibly led to the suppressed reductive debromination (5.1%) in the closed bioanode. Otherwise, the more abundant potential function bacteria with more collaborated interrelations, including hydrolytic dehalogenators (Acinetobacter, etc.), aromatics degrading bacteria (Geobacter, Holophaga, etc.) and electroactive bacteria (Geobacter, Desulfovibrio, etc.) made great sense to the enhanced hydrolytic debromination and detoxification of TBBPA. Naringin exhibited various pharmacological activities. However, its biological function and underlying mechanism in regulating macrophage polarization remain elusive. This study aimed to investigate the regulatory network between naringin and macrophage polarization in sepsis-induced intestinal injury. Cecal ligation and puncture (CLP) was used to establish the animal model of sepsis. Chromatin immunoprecipitation and a luciferase reporter assay were used to determine the interplay between peroxisome proliferator-activated receptor γ (PPARγ) and miR-21 promoter, as well as miR-21 and its target genes. Naringin enhanced the overall survival of septic mice and alleviated the CLP-induced inflammatory response and intestinal damage. This was accompanied by the increased expression of PPARγ in the intestines and the stimulation of ileal macrophages toward the M2 phenotype. Furthermore, in lipopolysaccharide-stimulated bone marrow-derived macrophages, naringin stimulated M2 polarization. Mechanistically, PPARγ inhibition attenuated the promotion of M2 polarization caused by naringin, and the naringin/PPARγ regulatory work was compromised by miR-21 inhibition. The present study suggested that naringin promoted M2 polarization via the PPARγ/miR-21 axis, thus relieving sepsis-induced intestinal injury. Most patients with triple negative breast cancer (TNBC) do not respond to anti-PD1/PDL1 immunotherapy, indicating the necessity to explore immune checkpoint targets. B7H3 is a highly glycosylated protein. However, the mechanisms of B7H3 glycosylation regulation and whether the sugar moiety contributes to immunosuppression are unclear. Here, we identify aberrant B7H3 glycosylation and show that N-glycosylation of B7H3 at NXT motif sites is responsible for its protein stability and immunosuppression in TNBC tumors. The fucosyltransferase FUT8 catalyzes B7H3 core fucosylation at N-glycans to maintain its high expression. Knockdown of FUT8 rescues glycosylated B7H3-mediated immunosuppressive function in TNBC cells. Abnormal B7H3 glycosylation mediated by FUT8 overexpression can be physiologically important and clinically relevant in patients with TNBC. Notably, the combination of core fucosylation inhibitor 2F-Fuc and anti-PDL1 results in enhanced therapeutic efficacy in B7H3-positive TNBC tumors. These findings suggest that targeting the FUT8-B7H3 axis might be a promising strategy for improving anti-tumor immune responses in patients with TNBC.

Background: Although increasing evidence has demonstrated that human dental pulp stem cells (hDPSCs) are efficacious for the clinical treatment of skeletal disorders, the underlying mechanisms remain incompletely understood. Osteoarthritis (OA) is one of the most common degenerative disorders in joints and is characterized by gradual and irreversible cartilaginous tissue damage. Notably, immune factors were newly identified to be closely related to OA development. In this study, we explored the modulatory effects of clinical-grade hDPSCs on osteoarthritic macrophages and their protective effects on cartilaginous tissues in OA joints., Methods: The cell morphology, immunophenotype, and inflammatory factor expression of osteoarthritic macrophages were explored by phase contrast microscope, transmission electron microscopy, immunostaining, flow cytometry, quantitative polymerase chain reaction, and enzyme linked immunosorbent assay, respectively. Additionally, the factors and signaling pathways that suppressed macrophage activation by hDPSCs were determined by enzyme-linked immunosorbent assay and western-blotting. Furthermore, hDPSCs were administered to a rabbit knee OA model via intra-articular injection. Macrophage activation in vivo and cartilaginous tissue damage were also evaluated by pathological analysis., Results: We found that hDPSCs markedly inhibited osteoarthritic macrophage activation in vitro. The cell morphology, immunophenotype, and inflammatory factor expression of osteoarthritic macrophages changed into less inflammatory status in the presence of hDPSCs. Mechanistically, we observed that hDPSC-derived hepatocyte growth factor and transforming growth factor β1 mediated the suppressive effects on osteoarthritic macrophages. Moreover, phosphorylation of MAPK pathway proteins contributed to osteoarthritic macrophage activation, and hDPSCs suppressed their activation by partially inactivating those pathways. Most importantly, injected hDPSCs inhibited macrophage activation in osteochondral tissues in a rabbit knee OA model in vivo. Further histological analysis showed that hDPSCs alleviated cartilaginous damage to knee joints., Conclusions: In summary, our findings reveal a novel function for hDPSCs in suppressing osteoarthritic macrophages and suggest that macrophages are efficient cellular targets of hDPSCs for alleviation of cartilaginous damage in OA. hDPSCs treat OA via an osteoarthritic macrophages-dependent mechanisms. hDPSCs suppress the activation of osteoarthritic macrophages in vitro and in vivo and alleviate cartilaginous lesions in OA models.

The excess organic carbon is often added to meet denitrification requirements during municipal wastewater treatment, resulting in the carbon waste and increased risk of secondary pollution. In this study, microbial fuel cell (MFC) was coupled with an up-flow denitrification biofilter (BF), and the long-term performances of denitrification and power output were investigated under the different carbon source concentration. With sodium acetate (NaAc) of 600 mg/L and 300 mg/L, the favorable denitrification efficiencies were obtained (98.60%) and the stable current output was maintained (0.44 mÃ0.48 mA). By supplying NaAc of 150 mg/L, the high denitrification efficiency remained in a high range (89.31%) and the current output maintained at 0.12 mA, while, the denitrification efficiency dropped to 71.34% without coupling MFC. Electron balance analysis indicated that both nitrate removal and electron recovery efficiencies were higher in MFC-BF than that in BF, verifying the improved denitrification and carbon utilization performance. Coupling MFC significantly altered the bacterial community structure and composition, and while, the diversified abundance and distribution of bacterial genera were observed at the different locations. Compared with BF, the more exoelectrogenic genera (Desulfobacterium, Trichococcus) and genera holding both denitrifying and electrogenic functions (Dechloromonas, Geobacter) were found dominated in MFC-BF. Instead, the dominating genera in BF were Dechloromonas, Desulfomicrobium, Acidovorax and etc. By coupling MFC, the more complex and diversified network and the closer interaction relationships between the dominant potential functional genera were found. Reducing the use of liquid organic carbon electron donors during biostimulation of heterotrophic denitrification is critical for sustainable groundwater remediation. Solid-phase humin isolated from natural sources can provide a cost-effective alternative to classical electron donors. In this study, the low-temperature denitrification capacity of an acetate-fed microbial community was enhanced using humin at 20 °C and 10 °C. These enhancements were not caused by faster acetate consumption and greater bacterial growth with the addition of humin. Estimation of the electron balance and first-order kinetics suggested that the enhancement in denitrification occurred mainly after acetate exhaustion. Humin may therefore have acted as an additional electron donor for the denitrifying microbial community, with the reduced quinone group in humin potentially responsible for electron donation. The addition of humin increased the richness and diversity of the denitrifying microbial community, in which Dechloromonas spp. played a critical role. Polycomb repressor complex 1 (PRC1) is linked to the regulation of gene expression and histone ubiquitylation conformation, which contributes to carcinogenesis. However, the upstream regulators of PRC1 biogenesis machinery remain obscure. Here, we report that the polycomb group-related mammalian gene Mel18 is a target of the protein kinase AKT. AKT phosphorylates Mel18 at T334 to disrupt the interaction between Mel18 and other PRC1 members, leading to attenuated PRC1-dependent ubiquitylation of histone H2A at Lys119. As such, PRC1 target genes, many of which are known oncogenes, are derepressed upon T334-Mel18 phosphorylation, which promotes malignant behaviours, including cell proliferation, tumour formation, migration and invasion, bone and brain metastatic lesion formation. Notably, a positive correlation between AKT activity and pT334-Mel18 is observed, and prognostic models based on p-AKT and pT334-Mel18 that predicted overall survival and distant metastasis-free survival in breast cancer patients are established. These findings have implications for understanding the role of AKT and its associated proteins in chromatin ubiquitylation, and also indicate the AKT-Mel18-H2AK119ub axis as a novel prognostic biomarker and therapeutic target for cancer patients.

To characterize the impact of reactor configuration and influent loading on elemental sulphur (S 0 ) recovery during denitrification desulfurization, a laboratory-scale expanded granular sludge bed (EGSB) reactor was established under two influent acetate/nitrate/sulphide loadings; the water flow velocity, microbial community, and functional genes at different heights were investigated. There was no S 0 generated when acetate/nitrate/sulphide loadings were set to 0.95/0.60/1.05 kg/m 3 .d (low-loading). Furthermore, there were no typical denitrifying sulphide oxidizing bacteria under this condition, and Syntrophobacter, Anaerolineaceae genera were predominant in the reactor. As the influent loading was doubled (high-loading), S 0 recovery increased to 87%; the bacterial distribution was relatively homogeneous with sulphide oxidation genera (Thauera) being predominant. Neither nirK nor sqr genes were detected in the low-loading sample at a height of 50 cm. The sqr/sox ratios of low-loading stage were 2.50 (10 cm), 0.94 (30 cm), and 0 (50 cm), and the ratios of the high-loading stage were 1.38 (10 cm), 1.33 (30 cm), and 1.08 (50 cm). A hydrodynamics analysis indicated that the water flow velocity was homogenous throughout the reactor. Antibiotics commonly exist in municipal, livestock and industrial wastewaters. However, the response of key microbiota performance in wastewater treatment plants to antibiotic exposure lacks systematic research. In this study, the short-term acute stress of four commonly used antibiotics (sulfamethoxazole, chlortetracycline, ciprofloxacin, and amoxicillin) on microbial denitrification performance was systematically investigated. All tested antibiotics exhibited the inhibitory effects in varying degrees by repeated addition for six cycles. The nitrate removal efficiencies (NrE) decreased to 7.98-26.80%, accompanied by the significant decrease of the expressed narG gene, by exposure to sulfamethoxazole, chlortetracycline or amoxicillin. Nitrite reduction was inhibited more severely than nitrate reduction, which was further verified by the low- or non-expressed nirS and nosZ genes. Furthermore, a higher antibiotic concentration made stronger inhibitory effect. Except for chlortetracycline, 2.09-6.80 times decrease of k value was commonly observed as concentration increased from 10 to 50 or 100 mg L -1 . Even in a short period (24 h), antibiotics largely decreased the abundance of the dominant denitrifying bacterial genera (Thauera, Comamonas, etc.), while, some unclassified populations (Labrenzia, Longilinea, etc.) were enriched. The denitrification desulfurization system can be used to remediate wastewater containing carbon, nitrogen, and sulfur. However, the relationship between autotrophic and heterotrophic bacteria remains poorly understood. To better understand the roles and relations of core bacteria, an expanded granular sludge bed (EGSB) reactor was continuously operated under autotrophic (stage I), heterotrophic (stage II) and mixotrophic (stages III-VII) conditions with a 490-day period. Stage IV represented the excellent S 0 recovery rate (69.5%). The different trophic conditions caused the obvious succession of dominant bacterial genera. Autotrophic environment (stage I) enriched mostly Thiobacillus, and heterotrophic environment (stage II) was dominated with Azoarcus and Pseudomonas. Thauera, Arcobacter and Azoarcus became the predominant genera under mixotrophic conditions (stage III-VII). Strains belonged to these core genera were further isolated, and all seven isolates were confirmed with denitrifying sulfur oxidation capacity. Heterotrophic strain HDD1 (genus of Thauera) possessed both the highest sulfide degradation and S 0 recovery rates. Expression levels of cbbM and gltA genes were positively related with the autotrophic and heterotrophic conditions, respectively. NirK gene was highly expressed between log 3.7-log 4.3 during the entire run. Expression of both sqr and soxB genes were closely related with sulfur conversion. More than 57.5% of S 0 recovery rate could be obtained as sqr gene expression was greater than log 3.2, and while, sulfate was the primary form as soxB gene expression higher than log 3.9. The correlation between core microbial genera was very low from network, indicating a complex and non-specific mutualistic network between bacterial functional groups under each nutrient condition, and a stable coexistence state was possibly formed through utilizing each the secondary or waste metabolites in the mixotrophic conditions. Tetrabromobisphenol A (TBBPA) has aroused serious pollution in surface sediment. To date, whether and how iron-based nanoparticles could stimulate TBBPA in situ anaerobic biodegradation in sediment remains poorly understood. In this study, the distinctly enhanced TBBPA degradation activity with the rate constant k improved 4.7 times by fed with Pd/Fe nanoparticles (0.412 g L -1 dosage, 0.5 wt% Pd loading) was observed. TBBPA degradation first went through reductive dehalogenation with bisphenol A (BPA) as the metabolites, and after the addition of Pd/Fe nanoparticles, BPA was further degraded to 4-(allene)phenol and 2,2-bis(4-hydroxyphenyl) propanoic acid via UPLC-QTOF-MS analysis, suggesting the complete detoxification potential. By the addition of Pd/Fe nanoparticles, the large amount of H 2 production (560 times higher) and the significant inhibition of methane generation facilitated the metabolism of potential reductive dehalogenators (Desulfovibrio, Clostridium, etc.), demonstrated by their increased ecological abundance and the tighter cooperative interrelations between each other. Meanwhile, the addition of Pd/Fe nanoparticles largely promoted the ecological abundance of Fe(III) reducing and aromatics degrading bacteria (Bacillus, Cryptanaerobacter, etc.), resulting in BPA further degradation. The bacterial ecological network further revealed that the potential BPA degrading bacteria shared the more positive interactions with the potential dehalogenators in the presence of Pd/Fe nanoparticles. The study firstly revealed the addition of Pd/Fe nanoparticles obviously enhanced the respiratory metabolic activities and cooperative interrelations of reductive dehalogenators and BPA degraders, which gives suggestions for in situ remediation and detoxification of BFRs in contaminated sediment., (Copyright © 2019. Expanded granular sludge blanket (EGSB) is regarded as a promising reactor to carry out denitrifying sulfide removal (DSR) and elemental sulfur (S 0 ) recovery. Although the recirculation ratio is an essential parameter for EGSB reactors, how it impacts the DSR process remains poorly understood. Here, three lab-scale DSR-EGSB reactors were established with the different recirculation ratios (3:1, 6:1 and 9:1) to evaluate the corresponding variations in pollutant removal, S 0 recovery, anaerobic granular sludge (AGS) characteristics and microbial community composition. It was found that an intermediate recirculation ratio (6:1) could facilitate long-term reactor stability. Adequate recirculation ratio could enhance S 0 recovery, but an excessive recirculation ratio (9:1) was likely to cause AGS fragmentation and biomass loss. The S 0 desorbed more from sludge at higher recirculation ratios, probably due to the enhanced hydraulic disturbance caused by the increased recirculation ratios. At the low recirculation ratio (3:1), S 0 accumulation as inorganic suspended solids in AGS led to a decrease in VSS/TSS ratio and mass transfer efficiency. Although typical denitrifying and sulfide-oxidizing bacteria (e.g., Azoarcus, Thauera and Arcobacter) were predominant in all conditions, facultative and heterotrophic functional bacteria (e.g., Azoarcus and Thauera) were more adaptable to higher recirculation ratios than autotrophs (e.g., Arcobacter, Thiobacillus and Vulcanibacillus), which was conducive to the formation of bacterial aggregates to response to the increased recirculation ratio. Microbial reductive dechlorination of chlorinated aromatics frequently suffers from the long dechlorination period and the generation of toxic metabolites. Biocathode bioelectrochemical systems were verified to be effective in the degradation of various refractory pollutants. However, the electrochemical and microbial related working mechanisms for bio-dechlorination by electro-stimulation remain poorly understood. In this study, we reported the significantly improved 2,4,6-trichlorophenol dechlorination activity through the weak electro-stimulation (cathode potential of -0.36 V vs. SHE), as evidenced by the 3.1 times higher dechlorination rate and the complete dechlorination ability with phenol as the end dechlorination product. The high reductive dechlorination rate (20.8 μM/d) could be maintained by utilizing electrode as an effective electron donor (coulombic efficiency of 82.3 ± 4.8%). Cyclic voltammetry analysis of the cathodic biofilm gave the direct evidences of the cathodic respiration with the improved and positive-shifted reduction peaks of 2,4,6-TCP, 2,4-DCP and 4-CP. The optimal 2,4,6-TCP reductive dechlorination rate (24.2 μM/d) was obtained when a small amount of lactate (2 mM) was added, and the generation of H 2 and CH 4 were accompanied due to the biological fermentation and methanogenesis. The electrical stimulation significantly altered the cathodic biofilm structure and composition with some potential dechlorinators (like Acetobacterium) predominated. The microbial interactions in the ecological network of cathodic biofilm were more simplified than the planktonic community. However, some potential dechlorinators (Acetobacterium, Desulfovibrio, etc.) shared more positive interactions. The co-existence and possible cooperative relationships between potential dechlorinators and fermenters (Sedimentibacter, etc.) were revealed. Meanwhile, the competitive interrelations between potential dechlorinators and methanogens (Methanomassiliicoccus) were found. In the network of plankton, the fermenters and methanogens possessed the more positive interrelations. Electro-stimulation at the cathodic potential of -0.36 V selectively enhanced the dechlorination function, while it showed little influence on either fermentation or methanogenesis process. Volatile chlorinated hydrocarbons (VCHs) are often found as a type of persistent and ubiquitous contaminant in groundwater. The feasibility, characteristics and microbial mechanism of acclimation of biodiversity-rich inoculation source for bioelectrochemical stimulated VCH dechlorination remain poorly understood. Here, the superior bioelectrochemical catalytic activities were observed for tetrachloroethylene (0.26 mM d -1 ) and 1,2-dichloroethane (2.20 mM d -1 ) dechlorination in anaerobic sludge-acclimated biocathodes with an optimal potential of -0.5 V, averaging 1.60-2.71 times higher than those reported in previous works on biocathodes. When the cathode was applied as the sole electron donor for dechlorination, columbic efficiencies reached the values greater than 80%. Tetrachloroethylene dechlorination showed a metabolic pathway with cis-1,2-dichloroethene as the main product, whereas 1,2-dichloroethane was dechlorinated entirely to the nontoxic ethene. The cathodic biofilms were highly abundant with the dechlorination and electro-active genera, while significant bacterial consortium variation was observed in response to the different VCH types and changes in cathodic potential. Bacillus, Pseudomonas and Lactococcus were mostly enriched for tetrachloroethylene dechlorination, and pceA, tceA and omcX were highly expressed. Geobacter was the most predominant during 1,2-dichloroethane dechlorination with rdhA, tceA and omcX highly expressed. In addition, although the impact of cathodic potentials was weaker than that of VCH types, the lower cathodic potentials, the more abundant of the electrode respiring populations and the higher expression of extracellular electron transfer related gene. This study demonstrated the great potential of acclimation of anaerobic sludge by electrical stimulation for accelerating VCH remediations and gave insights into its working molecular mechanisms., (Copyright © 2019. Improving anode configuration with polymer or nanomaterial modification is promising for enhancing microbial fuel cell performance. However, how anode modification affects biofilm development and electrogenic function remains poorly understood. In this study, the carbon cloth anode modified with polyaniline and reduced graphene oxide was successfully fabricated which obtained the highest power output. Accelerated electrogenic biofilm formation and the better electrogenic bacterial colonization based on the superior material properties (preferable electrochemical characteristics, the film-like structure and the more activated sites) were observed with the in situ biofilm development monitoring. The acclimation time was 2.4 times shorter with graphene and polyaniline modified anode than the bare one when inoculated with wastewater. Biofilm structure and function analysis show that Geobacter is the most predominant with the abundance as high as 81.4%, and meanwhile, electrogenesis related outer-surface octaheme c-type cytochrome omcZ is highly expressed in the modified anode. The anode modified with graphene and polyaniline favors Geobacter colonization, accelerates electrogenic biofilm formation and improves omcZ expression level, eventually leading to the improved performance of microbial fuel cell. The study for the first time reveals the impacts on biofilm development and microbial function by anode modification, which will better guide the potential application of microbial fuel cell for wastewater recovery., (Copyright © 2018. The hostile environment of an injured spinal cord makes it challenging to achieve higher viability in a grafted tissue-engineered neural network used to reconstruct the spinal cord circuit. Here, we investigate whether cell survival and synaptic transmission within an NT-3 and TRKC gene-overexpressing neural stem cell-derived neural network scaffold (NN) transplanted into transected spinal cord could be promoted by electroacupuncture (EA) through improving the microenvironment. Our results showed that EA facilitated the cell survival, neuronal differentiation, and synapse formation of a transplanted NN. Pseudorabies virus tracing demonstrated that EA strengthened synaptic integration of the transplanted NN with the host neural circuit. The combination therapy also promoted axonal regeneration, spinal conductivity, and functional recovery. The findings highlight EA as a potential and safe supplementary therapeutic strategy to reinforce the survival and synaptogenesis of a transplanted NN as a neuronal relay to bridge the two severed ends of an injured spinal cord., (Copyright © 2018 The Authors. Background: To develop and validate a nomogram based on log of odds between the number of positive lymph node and the number of negative lymph node (LODDS) in predicting the overall survival (OS) and cancer specific survival (CSS) for epithelial ovarian cancer (EOC) patients., Materials and Methods: A total of 10,692 post-operative EOC patients diagnosed between 2004 and 2013 were obtained from the Surveillance, Epidemiology, and End Results (SEER) database and randomly divided into training (n = 7,021) and validation (n = 3,671) cohorts. Multiple clinical pathological parameters were assessed and compared with outcomes. Parameters significantly correlating with outcomes were used to build a nomogram. Bootstrap validation was subsequently used to assess the predictive value of the model., Results: In the training set, age at diagnosis, race, marital status, tumor location, stage, grade and LODDS were correlated significantly with outcome in both the univariate and multivariate analyses and were used to develop a nomogram. The nomogram demonstrated good accuracy in predicting OS and CSS, with a bootstrap-corrected concordance index of 0.757 (95% CI, 0.746-0.768) for OS and 0.770 (95% CI, 0.759-0.782) for CSS. Notably, in this population our model performed favorably compared to the currently utilized Federation of Gynecology and Obstetrics (FIGO) model, with concordance indices of 0.699 (95% CI, 0.688-0.710, P < 0.05) and 0.719 (95% CI, 0.709- 0.730, P < 0.05) for OS and CSS, respectively. Using our nomogram in the validation cohort, the C-indices were 0.757 (95% CI, 0.741-0.773, P < 0.05, compared to FIGO) for OS and 0.762 (95% CI, 0.746-0.779, P < 0.05, compared to FIGO) for CSS., Conclusions: LODDS works as an independent prognostic factor for predicting survival in patients with EOC regardless of the tumor stage. By incorporating LODDS, our nomogram may be superior to the currently utilized FIGO staging system in predicting OS and CSS among post-operative EOC patients.

Objective: To explore the effect of Rhodiola on the expression of iNOS mRNA in severe acute pancreatitis (SAP) associated renal injury rats., Methods: A total of 72 healthy rats were randomly divided into the sham-operated group (S), the SAP associated renal injury group (M), and the Rhodiola-treated group (RHO), 24 in each group. Rats in S and M groups were peritoneally injected with 10 mL/kg saline 3h before modeling, while rats in the RHO group were peritoneally injected with 10 mL/kg Rhodiola Injection 3 h before modeling. The peripheral ligament of pancreas was bluntly dissociated in rats of M and RHO groups. The head of pancreas was occlused by nontraumatic blood vessel forceps 3 h later to establish the model. Eight rats were randomly selected from each group at 12, 24, and 36 h after modeling to detect levels of serum amylase, creatinine, and blood urea nitrogen. Serum levels of interleukin 1β (IL-1β) and interleukin 10 (IL-10) were detected by enzyme-linked immunosorbent assay (ELISA). Pathological changes of the left kidney were observed under light microscope. The expression of inducible nitric oxide synthase (iNOS) mRNA in the right kidney was detected with real time polymerase chain reaction (RT-PCR)., Results: Compared with the S group, serum levels of amylase, creatinine (Cr), blood urea nitrogen (BUN), IL-1β, IL-10, and iNOS mRNA expression significantly increased in the M group (P < 0.01). The function of kidney and pancreas were obviously improved in the RHO group than in the M group. Levels of IL-1β and iNOS significantly decreased, but IL-10 levels significantly increased in the RHO group with statistical difference (P < 0.05)., Conclusion: Rhodiola had better protective effect on SAP associated renal injury, which might be achieved through inhibiting the expression of IL-1β, stimulating the expression of IL-10, down-regulating iNOS mRNA expression, reducing the generation of oxygen free radicals and NO damage to cells, and improving hypoxia tolerance capabilities of the kidney.

Semen cryopreservation is an important method in assisted reproductive technology, but meanwhile it is complicated by cryodamage to spermatozoa. Mechanisms behind the cryodamage to spermatozoa are thought to be multifactorial, and the excessive generation of reactive oxygen species (ROS) has been suggested as a major contributing factor. DNA-damaged sperm is also capable of fertilizing oocytes, significantly affecting the outcome of reproduction. As a sensor of DNA damage responses, protein kinase ATM (ataxia telangiectasia mutated) can be initiated through rapid intermolecular autophosphorylation induced by DNA damage, phosphorylate various proteins, and amplify the responses to DNA damage. After DNA damage, histone H2AX is activated by ATM, which contributes to the repair of sperm-DNA damage after fertilization and regulates the cell cycle during embryo development. Given the important role of the ATM/H2AX signaling pathway in the response to and repair of DNA damage induced by oxidative stress, it may mediate the repair of sperm-DNA damage resulting from cryopreservation.

Male germ cells are particularly susceptible to DNA damage by genotoxic agents during spermiogenesis and spermatozoal maturation, and meanwhile lack an effective repair system to eliminate the lesions. Because the DNA damaged sperm still has fertilizability and developmental potentiality, damage repair may occur after fertilization, but its mechanism remains unknown. Histone H2AX phosphorylation (gammaH2AX) is reportedly involved in the repair of damaged sperm DNA after fertilization. This review aims to summarize the present knowledge on the mechanism of gammaH2AX-mediated repair of DNA damaged sperm in the zygote.

Objective: To establish the rat model of estradiol valerate medication in early pregnancy, and to investigate the effects of estradiol valerate on the development of the reproductive system of the first filial generation (F1) male rats by evaluating the anogenital distance (AGD) and the development of the testis and epididymis., Methods: Pregnant SD rats were divided at random into a blank control group and a low dose, a medium dose and a high dose medication group to receive intragastric estradiol valerate at 0.2 mg/kg, 0.5 mg/kg and 0.8 mg/kg, respectively. The newborn F1 male rats were normally fed. Their anogenital distances were measured on postnatal day (PND) 3 and 21, the organ coefficients of the testis and epididymis (testicular and epididymal weight g/body weight 100 g) were obtained on PND 60, the morphological changes of spermatogenic cells were observed by testis biopsy, and the diameter of the seminiferous tubule and epithelial height were measured., Results: There was no significant difference between the control and medicated F1 male rats in AGD on PND 3 and 21 (P > 0.05), nor in the organ coefficients of the testis and epididymis on PND 60 (P > 0.05), nor in the diameter of the seminiferous tubule and epithelial height., Conclusion: Medication of estradiol valerate (0.2 -0.8 mg/kg) to rats in early pregnancy neither significantly affects the reproductive system development, nor induces obvious histological changes of the testis in the sexual maturation period of their F1 males.

Influencing factors, adsorption isotherm, adsorption kinetics and preliminary discussion on the mechanism of sulfide adsorption by nanoscale iron prepared in laboratory were studied using manual simulation sulfide wastewater. Experimental results indicate that the removal efficiency of S2- increases with increasing iron dosage and decreases with increasing initial S2- concentration and pH values. The removal efficiency of S2- is 100% when initial concentration is less than 100 mg x L(-1) and are 87.34%, 65.80% and 44.61% at pH 2, 7 and 13. The temperature at 25 degrees C favors the maximum adsorption of S2- with 19.17 mg x g(-1) of equilibrium adsorption quantity and the adsorption capacity decreas at higher or lower temperature. The adsorption data fit well to the Langmuir equation and the Freundlich equation. The sulfide adsorption follows the pseudo second order equation with the maximum initial sorption rate(h) is 1.575 3 mg x (g x mg)(-1) at 25 degrees C and the adsorption rate constant increases with the increasing of temperature. The activation energy(Ea) is 8.22 kJ x mol(-1). The mechanism of sulfide removal is being sorbed onto the iron nanoparticles via formation of surface compleses, FeOSH and iron sulfides (FeS, FeS2, FeSn).

Some volatile chlorinated hydrocarbons such as VC, 1,1-DCE, TCE, PCE, gamma-HCH, were dechlorinated by synthesized supported nanoscale Pd/Fe. The dechlorination reactions of PCE, TCE, 1,1-DCE, VC, gamma-HCH follow the pseudo-frist order kinetics equations with the k(obs) of 2.79 h(-1), 2.35 h(-1), 1.12 h(-1), 2.14 h(-1) and 4.02 h(-1) respectively. Little or no medial products were detected and the main end products were C2H6 and C2H4 during the dechlorination of VC, 1,1-DCE, TCE, PCE. The total carbon ratio of C2H6 and C2H4 were 70% and 10% respectively during the dechlorination of TCE. The supported nanoscale Pd/Fe particles after exposed to air for 24 h were used for 8 cycle experiments and the results indicate that the particles have favorable stability. The reactivity has no obvious decrease after 200 hours' successive dechlorination experiments of gamma-HCH which indicates that the particles represent good durability. The reaction activation energy of all the chlorinated hydrocarbons are bigger than 29 kJ x mol(-1) which shows that the surface-chemical reaction rather than diffusion is the rate-limiting step in the metal-mediated dechlorination process. A consistence between the experimental data and simulated curves indicates that the muti-step reaction pathways proposed offer a better explanation of the reaction mechanisms than sequential hydrogenolysis reactions in the transformation of chlorinated hydrocarbons by supported nanoscale Pd/Fe.

Objective: To evaluate the protective effects of oxidative DNA damage by adding antioxidants: ascorbate, catalase (CAT), and superoxide dismutase (SOD) in human semen samples undergoing cryopreservation procedure., Methods: Semen sample form 30 fertile men were mixed with modified cryoprotectant and divided into six groups according to the category and concentration of antioxidants: ascorbate 300 micromol/L, ascorbate 600 micromol/L, CAT 200 U/ml, CAT 400 U/ml, SOD 200 U/ml, and SOD 400 U/ml. Comet assay was conducted to measure the percentage of comet cells, and the nuclear DNA damaged parameters: tail DNA percentage (TD%) and Olive tail moment (OTM). Flow cytometry was used to detect the reactive oxidative species (ROS). The motility (a + b grade), viable recovery rate, nuclear DNA integrity and reactive oxidative species (ROS) of all groups were analyzed before and/or after freeze-thawing., Results: (After cryopreservation, compared with the control group, the a + b grade sperm rates of the ascorbate 300 micromol/L, CAT 200 U, and CAT 400 U groups were all higher than that of the control group (all P < 0.05), however, the levels of reactive oxygen species (ROS) of the ascorbate 300 micromol/L, CAT 200 U, and CAT 400 U groups were 30 +/- 13, 30 +/- 11, and 30 +/- 11 respectively, all significantly lower than that of the control group (37 +/- 17 , all P < 0.05). The viable recovery rates of the ascorbate 300 micromol/L , CAT 200 U, and CAT 400 U groups were 67% +/- 14%, 68% +/- 14%, and 69% -/+ 15% respectively, all significantly higher than that of the control group (59% +/- 10%, all P < 0.05). (2) The TD% levels of the ascorbate 300 micromol/L, CAT 200 U, and CAT 400 U groups were 41% +/- 4%, 40% +/- 7%, 40% +/- 6%, all similar to that of the raw semen (all P > 0.05), but significantly lower than that of the control group (46% +/- 6%, all P < 0.01). The OTM levels of the ascorbate 300 micromol/ L, CAT 200 U, and CAT 400 U groups were 7.7 +/- 1.2, 7.5 +/- 1.6, and 7.8 +/- 1.9, all similar to that of the raw semen (all P > 0.05), but significantly lower than that of the control group (10.1 +/- 3.1, all P < 0.01) too. The TD% and OTM levels of the other groups were all significantly higher than that of the raw semen (all P < 0.01), but not significantly different from those of the control group (all P > 0.05). (3) ROS was significantly negatively correlated with the motility in all groups (P < 0.05 or P < 0.01). Apart from the ascorbate 600 micromol/L group, the TD% and OTM of the other groups were all significantly positively correlated with the ROS (P < 0.05 or P < 0.01)., Conclusion: Supplementation of ascorbate or CAT reduces the level of ROS that induces sperm nuclear DNA damage, and improves the human sperm quality in the process of freeze-thawing.

Objective: To investigate the relationship between microalbuminuria and endothelial-dependent relaxing function and atherosclerosis of common carotid artery (CCA) in aged patients with essential hypertension (EH)., Methods: Sixty-four aged EH patients were recruited. According to the albumin excretion rate (AER) in the urine measured by immunoturbidimetry, patients were divided into 2 groups: normoalbuminuria group (NAU group) and microalbuminuria group (MAU group). Thirty aged persons without EH were served as the control group. The endothelium-dependent relaxing function of blood vessels, intima-media thickness (IMT) and the plaque of CCA were measured by color Doppler ultrasound., Results: The flow-mediated dilation in the MAU group [(4.98+/-1.35)%] and that in the NAU group [(6.31+/-1.14)%] were significantly lower than that in the control group [(9.09+/-1.83)%, P<0.05, respectively], especially lower in the MAU group. The IMT of CCA in the MAU group [(0.97+/-0.19)mm] and that in the NAU group [(0.86+/-0.10)mm] were significantly thicker than that in the control group [(0.78+/-0.13)mm] (P<0.05, respectively), especially thicker in the MAU group. The analysis of multiple stepwise regression showed that the microalbuminuria was successively related to EDF, the IMT of CCA, the plaque index of CCA, systolic blood pressure, etc., Conclusion: EDF is impaired, and there is the atherosclerosis of CCA in aged patients with EH. Microalbuminuria correlates with the decrease of endothelium-dependent relaxing function and the IMT of CCA in aged patients with EH.

Aim: To evaluate the effect of intracytoplasmic sperm injection (ICSI) in the management of cases with a history of conventional in vitro fertilization (IVF) failure., Methods: Two groups of patients, 19 with normal semen parameters and a history of IVF failure (metaphase II oocytes: 0-30 %) and 28 with severe male factor infertility received ICSI technology during the same period. Ovarian stimulation was achieved by conventional procedure. Transvaginal ultrasound-guided oocyte collection was done 35-37 h after human chorionic gonadotrophin (hCG) injection. Only metaphase II oocytes were selected for microinjection., Results: Fertilization was achieved with ICSI in all the patients. The fertilization rate (75.6 % +/-21.1 % vs. 73.9 % +/-19.2 %), cleavage rate (85.1 % +/-19.3 % vs. 82.7 % +/-22.1 %), clinical pregnancy rate per embryo transfer cycle (31.6 % vs. 28.6 %) and implantation rate per embryo (15.3 % vs. 14.4 %) did not differ significantly between the two groups., Conclusion: ICSI is a valuable method for couples with a history of IVF failure. These patients may have a similar ICSI result as in severe male infertility.
