Abstract

A quantitative approximation of the cortical surface potential from measured scalp surface potential data is developed. The derivation is based on a local Taylor series expansion (TSE) in the surface normal coordinate. Analytical and numerical results for the four shell spherical head model show that the TSE method improves the spatial deblurring of the surface Laplacian method. The inclusion of the biharmonic term, the extension to other geometries, and the application to electrocorticogram measurements are discussed. Abstract

The incretin hormone glucagon-like peptide 1 (GLP-I) promotes glucose homeostasis and enhances B-cell function. GLP-1 receptor agonists (GLP-1 RAs) and dipeptidyl peptidase-4 (DPP-4) inhibitors, which inhibit the physiological inactivation of endogenous GLP-1, are used for the treatment of type 2 diabetes. Using the Metabochip, we identified three novel genetic loci with large effects (30-40%) on GLP-1-stimulated insulin secretion during hyperglycemic clamps in nondiabetic Caucasian individuals (TMEM114; CHST3 and CTRB1/2; n = 232; all P ≤ 8.8 x [10.sup.-7]). rs7202877 near CTRB1/2, a known diabetes risk locus, also associated with an absolute 0.51 ± 0.16% (5.6 ± 1.7 mmo/mol) lower A1C response to DPP-4 inhibitor treatment in G-allele carriers, but there was no effect on GLP-1 RA treatment in type 2 diabetic patients (n = 527). Furthermore, in pancreatic tissue, we show that rs7202877 acts as expression quantitative trait locus for CTRB1 and CTRB2, encoding chymotrypsinogen, and increases fecal chymotrypsin activity in healthy carriers. Chymotrypsin is one of the most abundant digestive enzymes in the gut where it cleaves food proteins into smaller peptide fragments. Abstract

Most soil organic matter (SOM) derives from plant material, but there are substantial gaps in our understanding of its transformation mechanisms. Alterations that occur as SOM decays and is stabilized have proved difficult to study owing largely to its diverse initial chemical composition and stable isotope values. We examined SOM stable isotope ratios in relation to composition using solid-state [sup.13]C Cross Polarization Magic Angle Spin (CPMAS) nuclear magnetic resonance (NMR) in sequentially deeper organic and mineral horizon soil samples collected from an unpolluted ecosystem in southeast Alaska, USA, dominated by [C.sub.3] vegetation. We found that, as humified carbon (C not respired as C[O.sub.2] during decomposition) increased in aliphaticity (ratio of unsubstituted aliphatics to carbohydrates), it also increased in [sup.15]N but showed no clear trend in [sup.13]C. These results imply that humification (and the concomitant stabilization of soil C) at our site resulted from microbial alteration of organics rather than from accumulation of recalcitrant compounds. The strong, and previously unreported, relation between [sup.15]N and SOM composition found at our study site suggests that degree of SOM humification may be correlated with an increase in [delta] [sup.15]N SOM values relative to [delta] [sup.15]N for fresh litter and other source material. Abstract

Background: Sodium-glucose cotransporter-2 inhibitors (SGLT2i) and glucagon-like peptide-1 receptor agonists (GLP-1RA) lower blood pressure (BP). When SGLT2i and GLP-1RA are combined, synergistic effects on BP have been observed. The mechanisms underlying these BP reductions are incompletely understood. The aim of this study was to assess the mechanisms underlying the BP reduction with the SGLT2i dapagliflozin, GLP-1RA exenatide, and dapagliflozin-exenatide compared with placebo in people with obesity and type 2 diabetes. Methods: Sixty-six people with type 2 diabetes were randomized to 16 weeks of dapagliflozin 10 mg/day, exenatide 10 µg twice daily, dapagliflozin-exenatide, or placebo treatment. The effect of treatments on estimates of: (1) plasma volume (calculated by Strauss formula, bioimpedance spectroscopy, hematocrit, (2) autonomic nervous system activity (heart rate variability), (3) arterial stiffness (pulse wave applanometry), (4) systemic hemodynamic parameters including peripheral vascular resistance, cardiac output and stroke volume (all derived from non-invasively systemic hemodynamic monitoring), and (5) natriuresis (24-hour urine collection) were assessed after 10 days and 16 weeks of treatment. Results: After 10 days, dapagliflozin reduced systolic BP (SBP) by − 4.7 mmHg, and reduced plasma volume. After 16 weeks, dapagliflozin reduced SBP by − 4.4 mmHg, and reduced sympathetic nervous system (SNS) activity. Exenatide had no effect on SBP, but reduced parasympathetic nervous system activity after 10 days and 16 weeks. After 10 days, dapagliflozin-exenatide reduced SBP by − 4.2 mmHg, and reduced plasma volume. After 16 weeks, dapagliflozin-exenatide reduced SBP by − 6.8 mmHg, and the reduction in plasma volume was still observed, but SNS activity was unaffected. Conclusions: The dapagliflozin-induced plasma volume contraction may contribute to the initial SBP reduction, while a reduction in SNS activity may contribute to the persistent SBP reduction. Dapagliflozin-exenatide resulted in the largest decrease in SBP. The effect on plasma volume was comparable to dapagliflozin monotherapy, and SNS activity was not reduced, therefore other mechanisms are likely to contribute to the blood pressure lowering effect of this combination, which need further investigation. 379. Official Responses to Ethnic Unrest in the USSR, 1985–1991.

Abstract

The Soviet Union, like the large, multiethnic land empires in Europe that came to an end in the early 20th century (Habsburg, Imperial Russian, Ottoman), consisted of a central government ruling over far-flung regions in which particular ethnic and cultural groups were predominant. For many years, Soviet leaders were able to maintain the internal stability of the multiethnic Soviet state by relying on a mix of extreme coercion and occasional concessions to local demands. Soon after Mikhail Gorbachev became the General Secretary of the Soviet Communist Party in 1985, he adopted sweeping political liberalization and democratization, including the first free elections ever held in the USSR. The loosening of political control in a state that had long been known for brutal repression had far-reaching consequences for social stability. The political opportunities that opened for ethnic groups in the Soviet Union to push for far-reaching change, including independence, created great difficulty for Gorbachev's attempts to hold the Soviet Union together. Although he could have resorted to the use of large-scale violence as previous Soviet leaders had repeatedly done, he was deeply reluctant to cause mass bloodshed. His aversion to the use of mass repression was one of the key factors that precipitated the unraveling of the USSR.
