Background and aims Iron (Fe) deficiency in plants is a common problem affecting agricultural production. Cadmium (Cd) is a toxic metal that can be taken up and transported within plants by transporters for divalent metals including Fe(II). The present study aims to investigate the functions of OsNRAMP2 (Natural Resistance-Associated Macrophage Protein 2) in the remobilization and distribution of Fe and Cd in rice. Methods The expression pattern of OsNRAMP2 was determined by quantitative real-time PCR and pOsNRAMP2:GUS assay. Knockout mutants of OsNRAMP2 were generated by using CRISPR/Cas9 gene editing. Localization of Fe in the vacuolar globoids of germinating seeds was imaged by high-resolution transmission electron microscopy coupled with energy-dispersive X-ray spectroscopy. Distributions of Fe and Cd between different plant tissues were investigated in hydroponic and soil pot experiments. Results OsNRAMP2 was mainly expressed in the embryo of germinating seeds, roots, leaf sheaths and leaf blades. OsNRAMP2 was localized at the tonoplast. Knockout of OsNRAMP2 delayed seed germination and produced chlorotic seedling leaves. Remobilization of Fe stored in the protein storage vacuoles in the scutellum of germinating seeds was restricted in osnramp2 mutants compared with wild type. Expression of genes related to Fe uptake was enhanced in the seedlings of osnramp2 mutants. Knockout of OsNRAMP2 significantly decreased the distribution of Cd, but not Fe, from leaves and straws to rice grains. Conclusions OsNRAMP2 plays an important role in remobilizing vacuolar Fe during seed germination and affects translocation of Cd from vegetative tissues to rice grains.

ACT001, derived from traditional herbal medicine, is a novel compound with effective anticancer activity in clinical trials. However, little is known regarding its role in pituitary adenomas. Here, we demonstrated that ACT001 suppressed cell proliferation and induced cell death of pituitary tumor cells in vitro and in vivo. ACT001 was also effective in suppressing the growth of different subtypes of human pituitary adenomas. The cytotoxic mechanism ACT001 employed was mainly related to autophagic cell death (ACD), indicated by autophagosome formation and LC3-II accumulation. In addition, ACT001-mediated inhibitory effect decreased when either ATG7 was downregulated or cells were cotreated with autophagy inhibitor 3-methyladenine (3-MA). RNA-seq analysis showed that mitogen-activated protein kinase (MAPK) pathway was a putative target of ACT001. Specifically, ACT001 treatment promoted the phosphorylation of JNK and P38 by binding to mitogen-activated protein kinase kinase 4 (MEK4). Our study indicated that ACT001-induced ACD of pituitary tumor cells via activating JNK and P38 phosphorylation by binding with MEK4, and it might be a novel and effective anticancer drug for pituitary adenomas.

Seed deterioration during rice seed storage can lead to seed vigor loss, which adversely affects agricultural production, the long-term preservation of germplasm resources, and the conservation of species diversity. However, the mechanisms underlying seed vigor maintenance remain largely unknown. In this study, 16 hybrid rice combinations were created using four sterile lines and four restorer lines that have been widely planted in southern China. Following artificial aging and natural aging treatments, germination percentage and metabolomics analysis by gas chromatography–mass spectrometry was used to identify the metabolite markers that could accurately reflect the degree of aging of the hybrid rice seeds. Significant differences in the degree of seed deterioration were observed among the 16 hybrid rice combinations tested, with each hybrid combination having a different germination percentage after storage. The hybrid rice combination with the storage-resistant restorer line Guanghui122 exhibited the highest germination percentage under both natural and artificial storage. A total of 89 metabolic peaks and 56 metabolites were identified, most of which were related to primary metabolism. Interestingly, the content of galactose, gluconic acid, fructose and glycerol in the seeds increased significantly during the aging process. Absolute quantification indicated that galactose and gluconic acid were highly significantly negatively correlated with the germination percentage of the seeds under the different aging treatments. The galactose content was significantly positively correlated with gluconic acid content. Additionally, glycerol showed a significant negative correlation with the germination percentage in most hybrid combinations. Based on the metabolomics analysis, metabolite markers that could accurately reflect the aging degree of hybrid rice seeds were identified. Galactose and gluconic acid were highly significantly negatively correlated with the germination percentage of the seeds, which suggested that these metabolites could constitute potential metabolic markers of seed vigor and aging. These findings are of great significance for the rapid and accurate evaluation of seed aging degree, the determination of seed quality, and the development of molecular breeding approaches for high-vigor rice seeds. Interleukin-37b (hereafter called IL-37) was identified as fundamental inhibitor of natural and acquired immunity. The molecular mechanism and function of IL-37 in colorectal cancer (CRC) has been elusive. Here, we found that IL-37 transgenic (IL-37tg) mice were highly susceptible to colitis-associated colorectal cancer (CAC) and suffered from dramatically increased tumor burdens in colon. Nevertheless, IL-37 is dispensable for intestinal mutagenesis, and CRC cell proliferation, apoptosis, and migration. Notably, IL-37 dampened protective cytotoxic T cell-mediated immunity in CAC and B16-OVA models. CD8+ T cell dysfunction is defined by reduced retention and activation as well as failure to proliferate and produce cytotoxic cytokines in IL-37tg mice, enabling tumor evasion of immune surveillance. The dysfunction led by IL-37 antagonizes IL-18–induced proliferation and effector function of CD8+ T cells, which was dependent on SIGIRR (single immunoglobulin interleukin-1 receptor-related protein). Finally, we observed that IL-37 levels were significantly increased in CRC patients, and positively correlated with serum CRC biomarker CEA levels, but negatively correlated with the CD8+ T cell infiltration in CRC patients. Our findings highlight the role of IL-37 in harnessing antitumor immunity by inactivation of cytotoxic T cells and establish a new defined inhibitory factor IL-37/SIGIRR in cancer-immunity cycle as therapeutic targets in CRC.

Background Growth arrest-specific 6 (GAS6) is a vitamin K-dependent protein related to inflammation, fibrosis, as well as platelet function. Genetic ablation of GAS6 in mice protects against cardiac hypertrophy and dysfunction. Nonetheless, the association between plasma GAS6 levels and acute heart failure (AHF) patients is still unknown. Methods We measured plasma GAS6 concentrations in 1039 patients with AHF who were enrolled in the DRAGON-HF trial (NCT03727828). Mean follow-up of the study was 889 days. The primary endpoint is all-cause death. Results In total, there were 195 primary endpoints of all-cause death and 135 secondary endpoints of cardiovascular death during the mean follow-up duration of 889 days. Represented herein is a new manner to form fluorenones via direct excitation of substrate instead of photocatalyst to activate sp2 C-H bond under redox-neutral condition. Our design relies on the photoexcited triplet state of aromatic aldehyde intermediates with higher reactivity that can be intercepted by cobaloxime catalyst through single electron transfer for following β-H elimination. The simple generation of acyl radical and successful interception by a metal catalyst cobaloxime avoid the use of both photocatalyst and stoichiometric external oxidants, affording a series of highly substituted fluorenones, even for six-membered ketones, such as xanthone and thioxanthone derivatives in good to excellent yields, and H2 is as the only byproduct. This catalytic system features the readily available metal catalyst, mild reaction conditions and broad substrate scope, in which sunlight reaction and scale-up experiment by the continuous-flow approach make the developed methodology sustainable and amenable for potentially operational procedures.
