Abstract

The mammalian ubiquitin ligase Hrd1 is the central component of a complex facilitating degradation of misfolded proteins during the ubiquitin-proteasome-dependent process of ER-associated degradation (ERAD). Hrd1 associates with cofactors to execute ERAD, but their roles and how they assemble with Hrd1 are not well understood. Here, we identify crucial cofactor interaction domains within Hrd1 and report a previously unrecognised evolutionarily conserved segment within the intrinsically disordered cytoplasmic domain of Hrd1 (termed the HAF-H domain), which engages complementary segments in the cofactors FAM8A1 and Herp (also known as HERPUD1). This domain is required by Hrd1 to interact with both FAM8A1 and Herp, as well as to assemble higher-order Hrd1 complexes. FAM8A1 enhances binding of Herp to Hrd1, an interaction that is required for ERAD. Our findings support a model of Hrd1 complex formation, where the Hrd1 cytoplasmic domain and FAM8A1 have a central role in the assembly and activity of this ERAD machinery.

Abstract

The title dispiro hydrocarbon 1 was designed as a new electrochromic material. This multiply clamped hexaphenylethane-type electron donor was prepared from 2,2'-diiodobiphenyl via biphenyl-2,2'-diylbis(dibenzotropylium) 2(2+) salt. X-ray analysis of 1 revealed a highly strained structure as reflected by an elongated ethane bond [bond length: 1.6665(17) angstrom] and nearly eclipsed conformation. The weakened bond was cleaved upon two-electron oxidation to regenerate the deeply colored dication 2(2+). The reversible interconversion between 1 and 2(2+) is accompanied not only by a drastic color change but also by C-C bond formation/cleavage. Thus, the voltammogram showed a pair of well-separated redox waves, which is characteristic of "dynamic redox (dyrex)" behavior. The tetrahydro derivative of 1 with two units of spiro(dibenzocycloheptadiene), which suffers from more severe steric congestion, was also prepared. The crystallographically determined bond length for the central C-C bond [1.705(4) angstrom] is greatest among the values reported for 9,9,10,10-tetraaryl-9,10-dihydrophenanthrene derivatives.

377. The Recovery Process of Nerve after Inferior Alveolar Nerve Scission in Rats : Analysis by Quantative Morphology and HRP Study

Abstract

P(論文), To evaluate the general recovery process of sensory neural tracts of the inferior alveolar nerve, temporal changes in the recovery were histologically examined on rat at 3, 7, 14, 28, 42, 70 and 91 days after the nerve cutting. The scission of the nerve in the buccal cortical bone of a mandibula was made on an eight-week-old Wistar rat by a disk cutter. The results are as follows. 1. The nerve degeneration occurred by degrees until 14 days after nerve injury, in which myelin sheaths were flat compared with the control (the intact contralateral nerve), and the number of axons decreased. Few HRP-labeled cells within the Gasserian ganglion were observed. 2. At twenty-eight days after the nerve scission, morphological recovery of myelin sheaths were evident with a tendency for axons to become more hypertrophied. In addition, more HRP-labeled cells in the ganglion tended to be observed, which suggest that antidromic transport of HRP in the cutting portion occured. After 42 days, the experimental group exhibited about one and a half times the number of axons as the control. On the other hand, the number of HRP-labeled cells were half compared with that of the control. 3. At ninety-one days after nerve cutting, axon densities and their appearance in both experimental and control groups were very similar, and the numbers of axons were about one point three times higher than the control. The numbers of HRP-labeled cells in the ganglion had not changed from those observed at 42 days. In conclusion, these results suggest that the regeneration of the inferior alveolar nerve occurred within 91 days after the nerve scission in the rat.

Abstract

This paper describes an embodied conversational agent enhanced with specific conversational strategies aiming to foster learners' readiness towards communication in a second language (L2). Willingness to communicate (WTC) in a second language is believed to have a direct and sustained influence on learners' actual usage frequency of the target language. To help overcome the lack of suitable environments for increasing L2 learners' WTC, our approach is to build embodied conversational agents that can help learners surmount their apprehension towards communication in L2. Here, we focus on the dialogue management aspects of our approach and propose a model based on a set of communication strategies (CS) and affective backchannels (AB) to foster such agents' ability to carry on natural and WTC-friendly conversations with learners. We examined learners' expected WTC after interacting with one of the following versions of the system: an agent featuring both CS and AB; an agent featuring only CS; and an agent featuring only AB. The results suggested that combining CS and AB empowers the conversational agent and leads to higher expected WTC among L2 learners. We also found that even the AB-only version of the system had the potential to enhance WTC to some extent. These findings are evidence of the feasibility of enhancing L2 learners' engagement towards communication using a computer-based environment coupled with appropriate conversational strategies. 379. Synthesis of Thieno[3,2-b]indoles via Halogen Dance and Ligand-Controlled One-Pot Sequential Coupling Reaction

Abstract

A two-pot synthesis of thieno[3,2-b]indole from 2,5-dibromothiophene is described. A halogen dance of 2,5-dibromothiophene was performed with LDA, and subsequent Negishi coupling was performed with 2-iodoaniline derivatives to provide the corresponding coupling products. The resulting two bromo groups have different reactivities, which were utilized for the one-pot Suzuki–Miyaura coupling/intramolecular Buchwald–Hartwig amination to produce thieno[3,2-b]indole via an assisted tandem catalysis that involved in situ ligand exchange.

症例は58歳，男性．倦怠感と腹痛を主訴に近医を受診．胸腹部CT検査にて両側肺野に粒状影・空洞性病変を認め，腹部にガス像を伴う巨大な腹腔内膿瘍の所見を認めた．喀痰よりガフキー1号，PCRにて結核菌が検出され，胃液よりガフキー6号を認めた．活動性肺結核および腸結核による消化管穿孔の診断で，腹腔内膿瘍に対して経皮的にドレナージチューブを挿入後，加療目的に当院へ転院となった．腹部症状および腹部所見が軽度であったことや，低栄養状態で結核性腹膜炎による腹腔内の強い炎症および癒着が危惧されたため，先に保存的加療を行い，栄養状態の改善後に手術を施行する方針とした．抗結核薬の投与を開始し，腹腔内膿瘍に対してはドレナージを継続し，抗生剤の投与を行った．腹腔内膿瘍は徐々に縮小・瘻孔化し，第115病日に退院となった．今回，われわれは保存的に治療しえた腸結核による空腸穿孔の1例を経験したので，文献的考察を含めて報告する．

Abstract

P(論文), Platelet rich plasma (PRP) has beneficial effects on wound healing and bone regeneration. Therefore, PRP is applied in various clinical treatments hereafter, for example, in oral implants and in bone grafts. But the method of centrifugal separation for preparation of PRP is not established, and it varies from each hospital. So, we investigated an efficient one. We prepared PRP by centrifugal separations at various revolutions per minute and various spin times, and measured concentrations of platelets. Concentrations of platelets of PRP separated at higher revolutions with longer spin times are significantly lower than those separated at a moderate number of revolutions with moderate spin times. It is made clear that the most efficient centrifugal separation is at about 1000 rpm, 5 minutes. And it is suggested that a double spin technique is far more efficient than a single spin technique.

Abstract

In recent years, changes in lifestyle have caused increases in insulin resistance as well as development of diabetes. For this reason, the clinical importance of the multiple risk factors syndrome, which is the main pathogenesis of arteriosclerotic diseases, has increased. Therefore, a precise evaluation of insulin resistance has become noticeable. Although there were many evaluation methods of insulin resistance in the past, nowadays the hyperinsulinemic euglycemic clamp method is supposed to be the most precise and reliable means for assessing the level of insulin action. However, as for its relatively complicated procedures, the technique has been rarely used. In this report, we summarized our experience and originality in rat studies that have been carried out for several years. Additionally, we tried to establish a simplified insulin clamp method in conscious animals.
