364. Transcriptome and phytochemical analyses reveal the roles of characteristic metabolites in the taste formation of white tea during the withering process

Abstract

In the postharvest processing of tea leaves, withering is the first indispensable manufacturing process which produces the mellow, umami and sweet taste of white tea. In this study, we aimed to determine the dynamic changes of the main metabolites and clarify the key differentially expressed genes (DEGs) involved in forming the characteristic taste of white tea during withering. Phytochemical analyses revealed that the contents of total catechins and starch decreased continuously, whereas the contents of theaflavin, γ-aminobutyric acid (GABA), maltose, and soluble sugars increased significantly during withering (from 0–48 h). Meanwhile, the elevation of α-amylase (AMY), β-amylase (BAM), total amylase, and glutamate decarboxylase (GAD) activities may be correlated with the accumulation of GABA and maltose. By transcriptome sequencing, we detected 9 707, 15 921, 17 353, and 17 538 DEGs at 12, 24, 36, and 48 h of the withering process, respectively, compared with 0 h sample (fresh leaves). The transcript levels of most of the DEGs involved in catechin biosynthesis were significantly inhibited, whereas those involved in catechin oxidation were significantly up-regulated, which could be correlated to a decrease in catechin content and an increase in theaflavin content. The DEGs involved in GABA biosynthesis were considerably up-regulated, and the down-regulation of SPMS could reduce the competition for converting spermidine to GABA. The up-regulation of the AMY and BAM genes could trigger starch degradation, resulting in the increase of soluble sugar content. These results provide new insights into the importance of the withering process to the characteristic taste of white tea.

Abstract

We present a series of TCAD analysis of gallium nitride (GaN) heterojunction bipolar transistors (HBTs) that investigates the impact of various key parameters on the gain characteristics, output characteristics, and breakdown characteristics. It has been observed that the DC gain of the AlGaN/GaN HBTs exhibits a non-linear relationship with the increase in the Al fraction. Specifically, the DC gain initially rises, then declines after reaching its peak value at approximately 7%. By optimizing the concentration of the base and the concentration and thickness of the collector epitaxial layer, it is possible to achieve devices with breakdown voltages of 1270 V (with a collector thickness of 6 μm and a carrier concentration of 2 × 1016 cm−3), specific on-resistance of 0.88 mΩ·cm2, and a current gain of 73. In addition, an investigation on breakdown characteristics is conducted for HBTs with two types of substrates, namely QV-HBTs and FV-HBTs, at different inclinations of the ramp. We propose that critical angles are 79° and 69° to prevent the surface breakdown of the device, which helps to achieve an avalanche in GaN HBTs. We anticipate that the aforementioned findings will offer valuable insights for designing GaN-based power HBTs with elevated breakdown thresholds, heightened current densities, and increased power capabilities. Abstract

Soft and stretchable electronics have emerged as highly promising tools for biomedical diagnosis and biological studies, as they interface intimately with the human body and other biological systems. Most stretchable electronic materials and devices, however, still have Young's moduli orders of magnitude higher than soft bio-tissues, which limit their conformability and long-term biocompatibility. Here, we present a design strategy of soft interlayer for allowing the use of existing stretchable materials of relatively high moduli to versatilely realize stretchable devices with ultralow tissue-level moduli. We have demonstrated stretchable transistor arrays and active-matrix circuits with moduli below 10 kPa—over two orders of magnitude lower than the current state of the art. Benefiting from the increased conformability to irregular and dynamic surfaces, the ultrasoft device created with the soft interlayer design realizes electrophysiological recording on an isolated heart with high adaptability, spatial stability, and minimal influence on ventricle pressure. In vivo biocompatibility tests also demonstrate the benefit of suppressing foreign-body responses for long-term implantation. With its general applicability to diverse materials and devices, this soft-interlayer design overcomes the material-level limitation for imparting tissue-level softness to a variety of bioelectronic devices. Stretchable electronics are attractive for a range of biomedical applications, but are challenging to prepare with suitable mechanical properties. 373. Anti-CD47 Antibodies: A Potential New Cancer Immunotherapy.

Abstract

Objective: Immune checkpoint inhibitors, particularly anti-programmed cell death protein 1 (PDCD1/PD-1), anti-CD274 molecule (CD274/PD-L1), or anti-cytotoxic T-lymphocyte-associated protein 4 inhibitors, have shown notable anticancer efficacy in 10%-40% of cancer patients and >10 different cancer types. However, they have also reached an impasse. Therefore, targeting tumor-associated myeloid immune cells has become another viable approach. The CD47 molecule (CD47) is a "do not eat me" signal to phagocytes widely expressed in various tumor cells as a potential immune surveillance evasion mechanism. CD47 receptor signal-regulating protein alpha (SIRPa) is expressed in myeloid immune cells and inhibiting the CD47-SIRPa signaling pathway leads to tumor cell phagocytosis. Data Sources: We organized and presented the preclinical research of anti-CD47 in the past 40 years, and we also reviewed the results of anti-CD47-based clinical trials from 2018 to 2022. Study Selection: All studies we cited focus on using anti-CD47 blocking agents for targeting CD47/SIRPa. Results: In preclinical studies, anti-CD47 showed effectiveness in inducing macrophage phagocytosis of both solid tumors and blood cancers. However, clinically, anti-CD47 can only produce effective therapeutic effects when combined with other drugs, especially in treating blood cancers. Anti-CD47 may depend on directly regulating macrophage phagocytosis, tumor antigen capture by dendritic cells, and open adaptive immunity through cross-priming. Conclusion: Ongoing studies are investigating anti-CD47 in combination with chemotherapy, radiotherapy, targeted therapy, and immunomodulatory agents.
