Abstract

The pH dependence of the O photointermediate of the bacteriorhodopsin (bR) photocycle was examined using wild-type (WT) pigment and mutants of Glu-194, Glu-204, and residues of the proton release group (PRG) in bR. Results showed that in WT, the rate constant of O to bR transition declined 30-fold upon a decrease of the pH from 6 to 3 with a pKa of around 4.3. D2O was also demonstrated to slow the rise and decay of the O intermediate in the WT at PH 3.5 by a factor of 5.5. Further analysis indicated that the rate of the O to bR transition at low pH is guided by the deprotonation of the PRG.

Abstract

Probiotic supplement might be beneficial for gestational diabetes mellitus. However, the results remained controversial. We conducted a systematic review and meta-analysis to explore the efficacy of probiotic supplement in gestational diabetes mellitus.PubMed, Embase, Web of science, EBSCO, and Cochrane Library databases were systematically searched. Randomized controlled trials (RCTs) assessing the effect of probiotic supplement in gestational diabetes mellitus were included. Two investigators independently searched articles, extracted data, and assessed the quality of included studies. The primary outcome were fasting serum insulin and fasting plasma glucose. Meta-analysis was performed using the fixed-effect or random-effect model.Six RCTs involving 830 patients were included in the meta-analysis. Overall, compared with control intervention in gestational diabetes mellitus, probiotic supplementation intervention was found to significantly reduce fasting serum insulin (Std. mean difference = -0.95; 95% CI = -1.73 to -0.17; p .00001) and HOMA-IR (Std. mean difference = -1.12; 95% CI = -2.05 to -0.18; p = .02), but showed no substantial impact on fasting plasma glucose (Std. mean difference = -0.49; 95% CI = -1.05-0.08; p = .09), gestational age (Std. mean difference = 0.07; 95% CI = -0.20-0.34; p = .63), and gestational weight (Std. mean difference = -0.11; 95% CI = -0.38-0.16; p = .43).Compared with control intervention in gestational diabetes mellitus, probiotic supplementation was found to significantly reduce insulin resistance (HOMA-IR) and fasting serum insulin, but had no substantial influence on fasting plasma glucose, gestational age and gestational weight.

Abstract

4‐(2‐Chlorobenzyl)‐1‐(furan‐2‐yl)‐[1,2,4]triazolo[4,3‐a]quinazolin‐5(4H)‐one is a triazoloquinazolinone compound with broad‐spectrum biological activity. In this study, the four‐step method was used to synthesize the title compound, and its structure was confirmed by Fourier‐transform infrared spectroscopy (FT‐IR), proton nuclear magnetic resonance (1H NMR), 13‐carbon nuclear magnetic resonance (13C NMR) spectroscopy, and mass spectroscopy (MS). Further, its single crystal was characterized by X‐ray diffraction. The density functional theory was used to calculate the optimal structure of the molecule. The results demonstrated that the analyzed structure was consistent with the crystal structure determined by single‐crystal diffraction. In addition, discrete Fourier transform was used to study the molecular electrostatic potential and Frontier molecular orbital of the title compound, revealing a few of the compound's physicochemical properties. Abstract

A series of novel N‐(3‐((6‐bromopyrido[2,3‐d]pyrimidin‐4‐yl)oxy)phenyl)pyrrolidine‐1‐carboxamide and 1‐(3‐((6‐bromopyrido[2,3‐d]pyrimidin‐4‐yl)oxy)phenyl)‐3‐propylurea derivatives were synthesized. Their antitumor activities against human breast carcinoma cells (MCF‐7) and human colon cancer cells (HCT‐116) in vitro were evaluated, using sorafenib as a positive control drug. Anticancer bioassays indicated that several compounds exhibited appreciable anticancer activity against MCF‐7 and HCT‐116 cells. Particularly, compounds 9g and 8b demonstrated the most significant inhibitory effect against HCT‐116 and MCF‐7 cells, with inhibition ratios of 25.56% and 26.46%, respectively. Additionally, the synthesized pyridine[2,3‐d]pyrimidine derivatives containing a urea group moieties exhibited antitumor activities against MCF‐7 and HCT‐116 cells in vitro. [ABSTRACT FROM AUTHOR]

Abstract

Sickle cell disease (SCD) is a genetic disorder predominantly affecting people of African descent and is associated with significant morbidity and mortality. To improve SCD outcomes, the National Heart Lung and Blood Institute funded eight centers to participate in the SCD Implementation Consortium. Sites were required to each recruit 300 individuals with SCD, over 20 months. We aim to describe recruitment strategies and challenges encountered. Participants aged 15–45 years with confirmed diagnosis of SCD were eligible. Descriptive statistics were used to analyze the effectiveness of each recruitment strategy. A total of 2432 participants were recruited. Majority (95.3%) were African American. Successful strategies were recruitment from clinics (68.1%) and affiliated sites (15.6%). Recruitment at community events, emergency departments and pain centers had the lowest yield. Challenges included saturation of strategies and time constraints. Effective recruitment of participants in multi-site studies requires multiple strategies to achieve adequate sample sizes. [ABSTRACT FROM AUTHOR]

Abstract

Minimal residual disease that persists after chemotherapy is the most valuable prognostic marker for haematological malignancies and solid cancers. Unfortunately, our understanding of the resistance elicited in minimal residual disease is limited due to the rarity and heterogeneity of the residual cells. Here we generated 161,986 single-cell transcriptomes to analyse the dynamic changes of B-cell acute lymphoblastic leukaemia (B-ALL) at diagnosis, residual and relapse by combining single-cell RNA sequencing and B-cell-receptor sequencing. In contrast to those at diagnosis, the leukaemic cells at relapse tended to shift to poorly differentiated states, whereas the changes in the residual cells were more complicated. Differential analyses highlighted the activation of the hypoxia pathway in residual cells, resistant clones and B-ALL with MLL rearrangement. Both in vitro and in vivo models demonstrated that inhibition of the hypoxia pathway sensitized leukaemic cells to chemotherapy. This single-cell analysis of minimal residual disease opens up an avenue for the identification of potent treatment opportunities for B-ALL.

Abstract

SiO2 aerogel @ carbon nanotube anchored on graphene sheet (SiO2/CNT/GE) has been produced by a mechanical mixing method. The SiO2 nanoparticle and carbon nanotube are dispersed on graphene sheet. When the current density of the SiO2/CNT/GE material is 120 mA g−1 for lithium ion semi-battery, the first discharge and charge capacities are 839.3 mA h g−1 and 303.5 mA h g−1, respectively. After 200 cycles, its discharge capacity retains 624.3 mA h g−1. When the SiO2/CNT/GE material is used as a negative electrode and LiMn2O4 as a positive electrode for a full cell, the first discharge capacity is 732 mA h g−1, and the first coulomb efficiency is 93.18%. After 200 cycles, the discharge capacity can still be maintained at 217.9 mA h g−1, indicating its potential as a negative electrode material for lithium ion battery. [ABSTRACT FROM AUTHOR]

Abstract

The L1 protein of Human papillomavirus (HPV), the main capsid protein, induces the formation of neutralizing antibodies. In this study, HPV52 L1 protein was induced to be expressed. Monoclonal antibody (mAb) 6A7 against L1 protein were screened by cell fusion techniques. Western Blot and immunofluorescence assay (IFA) demonstrated the specificity of the mAb. The L1 protein was truncated for prokaryotic expression (N1∼N7) and Dot-ELISA showed that 6A7 recognized N3 (aa 200–350). The immunodominant regions were truncated again for expression, with 6A7 recognizing N6 (aa 251–305). The N6 proteins were further truncated and then were constructed an four-segment eukaryotic expression vector. IFA showed that 6A7 could recognize amino acid 262–279. Amino acid 262–279 was selected to be truncated into short peptides P1 and P2. Finally, Peptide-ELISA and Dot-ELISA showed that the epitope regions of mAb 6A7 were amino acid 262–273. The mAbs with defined epitopes can lay the foundation for the analysis of antigenic epitope characteristics and promote the development of epitope peptide vaccines. • Monoclonal antibodies against HPV52 L1 protein were prepared to screen the antigen epitope. • Amino acid 262–273 were screened and identified by truncating L1 protein four times. • This study enriched the epitope information library of HPV52 L1 protein. 394. How Academic Achievement Moderates the Relationship Between Self-Stigma and Depression Among College Students

Abstract

Due to the prevalence and seriously negative consequences associated with mental illness stigma, a number of interventions have been developed to ameliorate the situation. Theatric performance and contact-based approaches represent promising modes of stigma-reduction intervention. One potential intervention that combines the contact and the artistic aspect is administering videos of people narrating about their experience of stigma, but no empirical evaluations have been published. This study examined the efficacy of a video of an individual diagnosed with bipolar disorder describing some general stereotypes and daily labeling experiences. Participants (N=38) were students from the USA, Taiwan, and India recruited by snowball sampling. They completed a pretest with a stigma and depression measure, along with questions about demographics and their GPA, then were randomly assigned to one of two video conditions: (1) the intervention video described above, or (2) an informational video with some facts about mood disorder. All then completed a depression and stigma measure post-test. Self-stigma scores increased in both conditions. Academic ability did not moderate the relationship between pretest and posttest scores in both conditions, although academic ability itself and the pretest stigma scores correlated positively and significantly with posttest scores. Brief video exposure requires additional careful evaluation of its efficacy before implementing widely.
