Retroperitoneal and abdominopelvic sarcomas are rare heterogeneous malignancies. The only therapy proven to improve disease-free survival (DFS) is R0/R1 surgical resection. We sought to analyze whether additional factors such as radiation and systemic therapy were associated with DFS and abdominal recurrence-free survival (RFS).Retrospective review of adults (≥18) with resectable abdominopelvic and retroperitoneal sarcomas who underwent intent-to-cure surgery at a high-volume tertiary referral center between 1998 and 2015. The main outcome measures were DFS and abdominal RFS.Overall, 159 patients met the criteria for inclusion. Median follow-up was 4.8 years (range 0.1-18.9 years). The most common histology was liposarcoma (49%). Systemic therapy was administered to 48% of patients and was not associated with improved outcomes. The neoadjuvant radiotherapy group (11%) had improved adjusted DFS (5.46 years, 95% CI [3.68, 7.24] vs. 3.1 years, 95% CI [2.48, 3.73]) and abdominal RFS (6.14 years, 95% CI [4.38, 7.89] vs. 3.22 years, 95% CI [2.61, 3.84]). The adjuvant radiotherapy group (19%) had no improvement.In a cohort of patients undergoing resection for retroperitoneal or abdominopelvic sarcoma, neoadjuvant radiation improved DFS and abdominal RFS. A follow-up of over three years was needed to appreciate a difference in outcomes.

In calf-fed Holstein steers, the early growing phase is characterized by very high relative dry matter intake (DMI) accompanied with lower-than-expected efficiency of energy utilization. Although fat is commonly supplemented in growing–finishing diets, the comparative feeding value has not been investigated during the initial receiving–growing period. Eighty-four Holstein calves (127.7 ± 2.1 kg body weight) were used to evaluate the effects of including 3.5% of supplemental yellow grease (YG) in the diet on characteristics of growth performance and dietary energy utilization of calves during the early growth phase. Morbidity and mortality were not affected by dietary treatments. Yellow grease supplementation did not affect DMI, but tended to increase average daily gain (4.6%; P = 0.07), and increased (P ≤ 0.03) gain efficiency (5.8%) and dietary net energy for maintenance (NEm) and gain (NEg) by 4.1% and 5.3%, respectively. Based on performance data, the estimated NE value of supplemental YG was 4.67 and 3.68 Mcal/kg for NEm and NEg, respectively. These values are consistent with current tabular values assigned for vegetable oils (4.75 and 3.51 Mcal/kg), but markedly less (16.6%) than NEm value assigned for YG (5.60 Mcal NEm/kg; NASEM, 2016. During threat exposure, survival depends on defensive reactions. Prior works linked large glutamatergic populations in the midbrain periaqueductal gray (PAG) to defensive freezing and flight, and established that the overarching functional organization axis of the PAG is along anatomically-defined columns. Accordingly, broad activation of the dorsolateral column induces flight, while activation of the lateral or ventrolateral (l and vl) columns induces freezing. However, the PAG contains diverse cell types that vary in neurochemistry. How these cell types contribute to defense remains unknown, indicating that targeting sparse, genetically-defined populations may reveal how the PAG generates diverse behaviors. Though prior works showed that broad excitation of the lPAG or vlPAG causes freezing, we found in mice that activation of lateral and ventrolateral PAG (l/vlPAG) cholecystokinin-expressing (CCK) cells selectively caused flight to safer regions within an environment. Furthermore, inhibition of l/vlPAG-CCK cells reduced predator avoidance without altering other defensive behaviors like freezing. Lastly, l/vlPAG-CCK activity decreased when approaching threat and increased during movement to safer locations. These results suggest CCK cells drive threat avoidance states, which are epochs during which mice increase distance from threat and perform evasive escape. Conversely, l/vlPAG pan-neuronal activation promoted freezing, and these cells were activated near threat. Thus, CCK l/vlPAG cells have opposing function and neural activation motifs compared to the broader local ensemble defined solely by columnar boundaries. In addition to the anatomical columnar architecture of the PAG, the molecular identity of PAG cells may confer an additional axis of functional organization, revealing unexplored functional heterogeneity.

Electrical manipulation of spin textures inside antiferromagnets represents a new opportunity for developing spintronics with superior speed and high device density. Injecting spin currents into antiferromagnets and realizing efficient spin-orbit-torque-induced switching is however still challenging due to the complicated interactions from different sublattices. Meanwhile, because of the diminishing magnetic susceptibility, the nature and the magnitude of current-induced magnetic dynamics remain poorly characterized in antiferromagnets, whereas spurious effects further complicate experimental interpretations. In this work, by growing a thin film antiferromagnetic insulator, {\alpha}-Fe2O3, along its non-basal plane orientation, we realize a configuration where an injected spin current can robustly rotate the N\'eel vector within the tilted easy plane, with an efficiency comparable to that of classical ferromagnets. The spin-orbit torque effect stands out among other competing mechanisms and leads to clear switching dynamics. Thanks to this new mechanism, in contrast to the usually employed orthogonal switching geometry, we achieve bipolar antiferromagnetic switching by applying positive and negative currents along the same channel, a geometry that is more practical for device applications. In experiment 1, 150 calf-fed Holstein steers (119 ± 6 kg) were used to evaluate the effects of level of supplemental condensed tannin (0, 14, and 28 g/kg diet DM) in a conventional steam-flaked corn-based growing-finishing diet on feedlot growth performance. There were no treatment effects on growth performance during the initial 56-d period. However, during the subsequent 56-d period, supplemental condensed tannin tended to increase average daily gain (ADG; linear effect, P = 0.09). The increase in ADG was consistent with the tendency for increased dry matter intake (DMI; linear effect, P = 0.08). Hence, differences in gain efficiency during the second 56-d period were not appreciably affected (P = 0.80). Supplemental condensed tannin did not affect overall (112-d) ADG, DMI, gain efficiency, or dietary NE. In a second experiment, six Holstein steers (179.4 ± 7.9 kg) with cannulas in the rumen and proximal duodenum were used in a replicated 3 × 3 Latin square design to evaluate treatment effects on characteristics of ruminal and total tract digestion. Treatments were the same as in Trial 1. Tannin supplementation decreased (linear effect, P = 0.03) ruminal OM digestion and tended to decrease (linear effect, P = 0.06) ruminal feed N degradation. Tannin supplementation decreased (linear effect, P = 0.03) ruminal pH, but did not affect (P > 0.20) ruminal molar proportions of volatile fatty acids and estimated methane production. In current study, supplemental tannin had marginal effects on overall growth performance of calf-fed Holstein steers, but reduced ruminal, postruminal, and total tract nitrogen digestion.

Background In animal models, prenatal zinc deficiency induced epigenetic changes in the fetus, but data in humans are lacking. We aimed to examine associations between maternal zinc levels during pregnancy and DNA methylation in LINE-1 and Alu repetitive sequences in young adult offspring, as well as anthropometry and cardiometabolic parameters. Methods Participants were 74 pregnant women from the Chiang Mai Low Birth Weight cohort, and their offspring followed up at 20 years of age. Maternal plasma zinc concentrations were measured at approximately 36 weeks of gestation. DNA methylation levels in LINE-1 and Alu repetitive sequences were measured in the offspring, as well as anthropometry and cardiometabolic parameters (lipid profile, blood pressure, and glucose metabolism). Results Over half of mothers (39/74; 53%) were zinc deficient ( Conclusions Lower maternal zinc concentrations late in gestation were associated with changes in DNA methylation in later life. Thus, zinc deficiency during pregnancy may induce alterations in total LINE-1 methylation and LINE-1 hypermethylation loci. These results suggest a possible epigenetic link between zinc deficiency during pregnancy and long-term outcomes in the offspring.

Early life adversity (ELA) is a risk factor for early onset morbidities and mortality, a relationship that may be driven in part by immune system dysregulation. One mechanism of dysregulation that has yet to be fully examined in the context of ELA is alterations to immune cell dynamics in response to acute stress. Using a within-person between-group experimental design, we investigated stress-induced changes in immune cell populations, and how these changes may be altered in individuals with a history of ELA. Participants were young adults (N = 34, aged 18–25 years, 53% female, 47% with a history of ELA). Complete immune cell counts were measured at four time-points over a 5-hour window across two sessions (Trier Social Stress Test [TSST] vs. no-stress) separated by a week. Across all participants, total white blood cells increased over time (F(3,84)=38.97, p < .001) with a greater increase in response to the TSST compared to the no-stress condition at 240 minutes post-test (b = 0.43±.19; t(179)=2.22, p = .027). This pattern was mirrored by neutrophil counts. Lymphocyte counts were initially depressed by TSST exposure (b =−205±.67; t(184)=−3.07, p = .002) but recovered above baseline. ELA status was associated with higher stress-induced immune cell counts, a difference likely driven by increases in neutrophils (F(1,22)=4.45, p = .046). Overall, these results indicate differential immune cell dynamics in response to acute stress in individuals with a history of ELA. This points to altered immune system functioning in the context of stress, a finding that may be driving increased morbidity and mortality risk for ELA-exposed individuals.

Bans on the use of ionophores in several regions of the world has led to a need to identify alternative feed additivies to be added in cattle diets. Essential oil blends have been identified as a potential alternative to ionophores in feedlot diets. The objective of this study was to evaluate the effects of a supplemental a blend of essential oils and 25-hydroxyvitamin D3 on growth performance, energetic efficiency, and carcass characteristics in calf-fed Holstein steers. Ninety Holstein steer calves (123 ± 7 kg; 4 months old) were randomly assigned to 18 pens (5 steers/pen; 6 pens/treatment). Dietary treatments consisted of a steam-flaked corn-based diet supplemented with (DM basis): (1) no additives (CON); (2) 30 mg/kg DM of monensin (MON); (3) 200 mg/kg DM of a mixture of essential oils plus 25-hydroxyvitamin D3 (EO+HYD). There were no treatment effects (P > 0.05) on initial, intermediate and final cattle live weight; moreover, cattle had similar (P > 0.05) average daily gain (ADG) and dry matter intake (DMI) among dietary treatments. However, during the first 112 days of feed, calf-fed Holstein steers supplemented with EO+HYD had a greater (P ≤ 0.05) gain to feed ratio (G/F) than cattle fed the control diet but similar (P > 0.05) G/F to cattle supplemented with MON. However, there was no effect (P > 0.05) of dietary treatments on 112 to 286 d and the overall G/F ratio of calf-fed Holstein steers. Calf-fed Holstein steers supplemented with EO+HYD had greater (P ≤ 0.05) estimated net energy for maintenance (NEm) and net energy for gain (NEg) based on cattle growth performance than cattle fed the CON diet. Cattle supplemented with MON had an intermediate and similar (P > 0.05) NEm and NEg compared to the other two dietary treatments. However, when observed vs. expected NEm and NEg were calculated, cattle supplemented with MON and EO+HYD had greater efficiency of dietary energy utilization than cattle fed the CON diet. Calf-fed Holstein steers supplemented with MON had greater (P < 0.05) fat thickness than EO+HYD supplemented steers, and both were intermediate (P ≥ 0.05) to that of cattle fed the CON diet. There were no other effects (P > 0.05) on kidney, pelvic and heart fat, longissimus area, marbling score, and retail yield. The health status of cattle and liver abscesses or liver scars at slaughter were similar (P > 0.05). We conclude that supplementing calf-fed Holstein steers with MON or EO+HYD for over 285 days increased dietary net energy utilization for maintenance and gain of the diet by 3 and 4%, respectively, compared to non-supplemented steers.

The mitochondrion is intimately linked to energy and overall metabolism and therefore the morphology of mitochondrion can be very informative for inferring the metabolic state of cells. In this study we report an approach for automatic classification of mitochondrial morphologies using supervised machine learning to efficiently classify them from a large number of cells at a time. Fluorescence microscopy images of the chronic encysted form of parasite Toxoplasma gondii were used for this development. Manually classifying these morphologies from the hundreds of parasites within typical tissue cysts is tedious and error prone. In addition, because of inherent biological heterogeneity in morphologies, there can be variability and lack of reproducibility in manual classification. We used image segmentation to detect mitochondrial shapes and used features extracted from them in a multivariate logistic regression model to classify the detected shapes into five morphological classes: Blobs, Tadpoles, Lasso/Donuts, Arcs, and Other. The detected shapes from a subset of images were first used to obtain consensus classification among expert users to obtain a labeled set. The model was trained using the labeled set from five cysts and its performance was tested on the mitochondrial morphologies from ten other cysts that were not used in training. Results showed that the model had an average overall accuracy of 87%. There was high degree of confidence in the classification of Blobs and Arcs (average F scores 0.91 and 0.73) which constituted the majority of morphologies (85%). Although the current development used microscopy images from tissue cysts of Toxoplasma gondii, the approach is adaptable with minor adjustments and can be used to automatically classify morphologies of organelles from a variety of cells.

Objectives were to evaluate the relationship between pen mud accumulation and growth performance of calf-fed Holstein steers during the extreme heat characteristic of the months of August and September in the Southern California desert region. Holstein steers (n = 105; initial BW = 443 ±17 kg) were utilized in 28-d experiment (August 17 – September 14, 2021). Steers were blocked by initial shrunk weight (220 d before the initiation of the experiment) and randomly assigned to 21 pens (5 steers per pen). Pens were 62 m2 with 25 m2 overhead shade, automatic waterers and 2.4 m fence-line feed bunks. All steers were fed a steam-flaked corn-based growing-finishing diet. A bolus measuring temperature activity (smaXtec Basic, Smaxtec Animal Care GmbH, Austria) was placed in the rumen of each steer 55 d before the initiation of the experiment (June 22, 2021). Mud scores were recorded weekly by visual inspection. Mud score was determined as the percentage of pens surface area covered with mud. Scores were determined as follows: 1 – 0 to 20% of pen covered by mud; 2 – 21 to 40% of pen covered by mud; 3 – 41 to 60% of pen covered by mud; 4 – 61 to 80% of pen covered by mud; 5 – 81 to 100% of pen covered by mud. Ambient temperature averaged 33.2 ±2.3° C for the experimental period. Temperature of dry pen surfaces was greater than that of muddy surfaces (50.3 vs 20.4° C, respectively, P < 0.01). The relationship between ruminal temperature and pen mud score was not appreciable (P = 0.56). However, ADG was greater with a pen mud score of 2. Average daily gain decreased as pen mud score increased (r2 = 0.55). Similarly, feed efficiency was greatest in pens with a pen mud score of 2. Greater pen mud scores resulted in decreased feed efficiency (r2 = 0.56).

Since 2004, national guidelines have supported the omission of sentinel lymph node biopsy (SLNB) and radiotherapy for women ≥ 70 years of age with early-stage, hormone receptor-positive (HR+) breast cancer, but many women continue to receive at least one of these services. Provider- and patient-level factors may contribute to persistent utilization, but the role of facility-level factors is unknown. We aimed to determine facility-level variation of SLNB and adjuvant radiotherapy utilization in older women with early-stage, HR+ breast cancer undergoing breast-conserving surgery (BCS). Additionally, we aimed to explore factors associated with SLNB and radiotherapy utilization and the intra-facility correlation in their utilization.We conducted a retrospective cohort study using a statewide registry of claims data. We included women ≥70 years of age diagnosed with breast cancer who underwent BCS from 2012 to 2019 at 80 hospitals in the Michigan Value Collaborative. The main outcome was inter-facility rates and variation of SLNB and radiotherapy, as well as intra-facility correlation in their utilization.The cohort included 7253 women (median age 77 years). Only 20% (n = 1440) underwent BCS alone, whereas 71% (n = 5122) underwent SLNB and 52% (n = 3793) received radiotherapy. Inter-facility rates of SLNB ranged from 35 to 82% (median 70%), and radiotherapy ranged from 19 to 72% (median 49%). SLNB and radiotherapy were positively correlated (r = 0.27, p = 0.016).SLNB and radiotherapy rates remain high with significant variation in utilization at the facility level. High utilizers of SLNB are likely to be high utilizers of radiotherapy, suggesting the opportunity for strategic targeting of these facilities and their clinicians.

Animals must learn through experience which foods are nutritious and should be consumed, and which are toxic and should be avoided. Enteroendocrine cells (EECs) are the principal chemosensors in the GI tract, but investigation of their role in behavior has been limited by the difficulty of selectively targeting these cells in vivo. Here we describe an intersectional genetic approach for manipulating EEC subtypes in behaving mice. We show that multiple EEC subtypes inhibit food intake but have different effects on learning. Conditioned flavor preference is driven by release of cholecystokinin whereas conditioned taste aversion is mediated by serotonin and substance P. These positive and negative valence signals are transmitted by vagal and spinal afferents, respectively. These findings establish a cellular basis for how chemosensing in the gut drives learning about food.

For cancer survivors, insomnia is prevalent, distressing, and persists for years if unmanaged. Cognitive behavioral therapy for insomnia (CBT-I) is an effective treatment yet can be difficult to access and may require modification to address survivorship-specific barriers to sleep. In this 2-phase study, the authors adapted and assessed the feasibility, acceptability, and preliminary effects of synchronous, virtual CBT-I adapted for cancer survivors (the Survivorship Sleep Program [SSP]).From April to August 2020, cancer survivors with insomnia (N = 10) were interviewed to refine SSP content and delivery. From October 2020 to March 2021, 40 survivors were recruited for a randomized controlled trial comparing 4 weekly SSP sessions with enhanced usual care (EUC) (CBT-I referral plus a sleep hygiene handout). Feasibility and acceptability were assessed by enrollment, retention, attendance, fidelity, survey ratings, and exit interviews. Insomnia severity (secondary outcome), sleep quality, sleep diaries, and fatigue were assessed at baseline, postintervention, and at 1-month follow-up using linear mixed models.The SSP included targeted content and clinician-led, virtual delivery to enhance patient centeredness and access. Benchmarks were met for enrollment (56% enrolled/eligible), retention (SSP, 90%; EUC, 95%), attendance (100%), and fidelity (95%). Compared with EUC, the SSP resulted in large, clinically significant improvements in insomnia severity (Cohen d = 1.19) that were sustained at 1-month follow-up (Cohen d = 1.27). Improvements were observed for all other sleep metrics except sleep diary total sleep time and fatigue.Synchronous, virtually delivered CBT-I targeted to cancer survivors is feasible, acceptable, and seems to be efficacious for reducing insomnia severity. Further testing in larger and more diverse samples is warranted.

Telomeres are the protective DNA-protein sequences appearing at the ends of chromosomes; they shorten with each cell division and are considered a biomarker of aging. Shorter telomere length and greater erosion have been associated with compromised physical and mental health and are hypothesized to be affected by early life stress. In the latter case, most work has relied on retrospective measures of early life stressors. The Dutch research (n = 193) presented herein tested 3 hypotheses prospectively regarding effects of sensitive-insensitive parenting during the first 2.5 years on telomere length at age 6, when first measured, and change over the following 4 years. It was predicted that (1) less sensitive parenting would predict shorter telomeres and greater erosion and that such effects would be most pronounced in children (2) exposed to prenatal stress and/or (3) who were highly negatively emotional as infants. Results revealed, only, that prenatal stress amplified parenting effects on telomere change-in a differential-susceptibility-related manner: Prenatally stressed children displayed more erosion when they experienced insensitive parenting and less erosion when they experienced sensitive parenting.
