The low bioavailability of hydrophobic organic compounds (HOCs) is one of the key sources of uncertainty in the implementation of in situbioremediation. Bioavailability of HOCs in the subsurface is affected by sorption/desorption processes in two important ways. First, sorption causes high organic concentrations in microporous regions and impermeable zones to which bacterial access is obstructed. Second, because desorption and immobile zone diffusion must occur before biodegradation can proceed, the overall rate of bioremediation can be limitedor even controlled by these mass transfer processes, not by the activity of the degrading microorganisms. Rate models that couple sorption/desorption--related mass transfer processes and biodegradation havebeen successfully applied to laboratory results and are beginning tooffer some insight into the problem. Specifically, the influence of sorption on biodegradation is quantified here by defining a bioavailability factor, Bf. However, many questions remain and predictive modeling is elusive, especially in the context of complicated heterogeneous natural systems. Challenges facing environmental engineers are to develop a better understanding of these processes at both laboratory and field scales and ultimately to use such understanding toward the development of more effective and economical remediation technologies. Experiments tracked the deposition of two model bacteria - Pseudomonas aeruginosa and Klebsiella pneumoniae - onto clean filter media (i.e., glass beads and filter sand) and the subsequent removal of the bacteria by backwashing. The ability of bacteria to accumulate and be retained in a frequently backwashed filter is critical for effective removal of biodegradable organic matter in a process termed biofiltration. Column filtration experiments showed that the bacteria, especially P. aeruginosa, readily attached to the filter media during filtration, despite unfavorable chemical conditions. Water backwash was not effective at removing bacteria from the filter media, with only 20-40percent removal during 10-min backwash cycles at hydraulic loading rates ranging from 23 to 53 m/h (9.3 to 22 gpm/sq ft) and bed expansions ranging from 15 to 50 percent. Incomplete removal of biomass during backwashing was beneficial for removal of biodegradable total organic carbon, typically allowing a backwashed biofilter to maintain treatment at levels similar to the period immediately preceding the backwash. Aerobic biodegradation of benzene, toluene and naphthalene was studied in pre-equilibrated soil-water slurry microcosms. The experiments were designed to simulate biodegradation at waste sites where sorption reaches equilibrium before biodegradation becomes important. Rates of biodegradation were reduced by the presence of soil. For example, nearly complete naphthalene biodegradation (1.28 mg/L) by indigenous soil bacteria occurred within 60 hours in aqueous solution (soil-free) while it took two weeks to degrade the same amount in the presence of 0.47 kg soil/L of water. The rate of biodegradation was observed to decrease with increasing organic compound hydrophobicity, soil/water ratio, soil particle size, and soil organic carbon content. These results clearly indicate that the rate of biodegradation is affected by both the extent and rate of sorption. Further analysis suggests that mass transfer could control the performance of in situ bioremediation for highly hydrophobic organic contaminants which exhibit a large extent of sorption and slow rate of desorption. The feasibility of aerobic in situ bioremediation is being investigated for use in a strategy to control subsurface coal tar contamination at the site of a former manufactured gas plant. As part of this investigation, anoxic aquifer sands collected between 11 and 25 m below ground surface were assayed in batch microcosms to measure the singular and combined effects of O2, NO3-,and PO43- on 14C-naphthalene mineralization. The influence of these additives varied considerably between sediments. A high initial concentration of O2 (21 mg/L) promoted the greatest extent of mineralization in the majority of active sediments. NO3- (85 mg/L) was observed to enhance, inhibit, or have no effect on the rate of naphthalene mineralization, although significant denitrification was observed in nearly all the active sediments. Data suggest that PO43- complexation and/or precipitation with sediment cations limited P bioavailability. The sediments that were incapable of mineralizing naphthalene were characterized by low pH (< 4.1), high SO42- (>500 mg/L), and moderate to high dissolved Fe(II) (30-265 mg/L) when equilibrated aerobically with water. Fe(II) likely exerted a significant O2 demand that reduced the O2 available as an electron acceptor for biodegradation. These experiments demonstrate that while aeration/oxygenation can be an effective strategy for enhancing subsurface bioremediation of aromatic hydrocarbons, the biodegradation response to aeration/oxygenation and nutrient addition may vary considerably within an aquifer. • Heavy metals in water accumulate within sediments that alter their behavior. • Manganese is a watershed contaminant associated with Parkinsonism in mammals. • Manganese sediment interaction does not alter total manganese in drinking water. • Manganese sediment interaction reduced motor impairment in mice exposed via drinking water. Heavy metals enter the aquatic environment and accumulate within water sediments, but these metal-sediment interactions remain to be explored within toxicity studies. We developed an exposure model in mice that encapsulates the aquatic microenvironment of metals before exposure. Male and female C57/BL6 mice were exposed via their drinking water to manganese contaminated sediment (Sed_Mn) or to manganese without sediment interaction (Mn) for six weeks. Sediment interaction did not alter weekly manganese ingestion from water in males or females. We analyzed motor impairment, a common feature in manganese-induced Parkinsonism, using the beam traversal, cylinder, and accelerating rotarod tests. Sed_Mn mice performed better overall compared to Mn mice and males were more sensitive to manganese than females in both Sed_Mn and Mn treatment groups. Our study indicates that metal-sediment interactions may alter metal toxicity in mammals and introduces a new exposure model to test the toxicity of metal contaminants of drinking water. One year after the Deepwater Horizon oil spill accident, semipermeable membrane devices (SPMDs) and polyethylene devices (PEDs) were deployed in wetland areas and coastal areas of the Gulf of Mexico (GOM) to monitor polycyclic aromatic hydrocarbons (PAHs). The measured PAH levels with the PEDs in coastal areas were 0.05-1.9 ng/L in water and 0.03-9.7 ng/L in sediment porewater. With the SPMDs, the measured PAH levels in wetlands (Barataria Bay) were 1.4-73 ng/L in water and 3.3-107 ng/L in porewater. The total PAH concentrations in the coastal areas were close to the reported baseline PAH concentrations in GOM; however, the total PAH concentrations in the wetland areas were one or two orders of magnitude higher than those reported in the coastal areas. In light of the significant spatial variability of PAHs in the Gulf's environments, baseline information on PAHs should be obtained in specific areas periodically.

Eighteen microalgae, including two local isolates, were evaluated for their ability to grow and remove nutrients from unsterilized primary or secondary wastewater effluents as well as wastewater supplemented with nutrient-rich anaerobic digester centrate (ADC). Most of the tested species except several phylogenetically clustered Chlorella sorokiniana including local isolates and Scenedesmus strains were unable to grow efficiently. This may reflect the presence of certain genetic traits important for robust growth in the unsterilized wastewater. The maximum algal-specific growth rates and biomass density obtained in these bacterial-contaminated cultures were in the range of 0.8-1 day(-1) and 250-350 mg L(-1), respectively. ADC supplementation was especially helpful to biologically treated secondary effluent with its lower initial macronutrient and micronutrient content. As a result of algal growth, total nitrogen and orthophosphate levels were reduced by as much as 90 and 70 %, respectively. Biological assimilation was estimated to be the main mechanism of nitrogen removal in primary and secondary effluents with ammonia volatilization and bacterial nitrification-denitrification contributing for cultures supplemented with ADC. Assimilation by algae served as the principal mechanism of orthophosphate remediation in secondary wastewater cultures, while chemical precipitation appeared also to be important for orthophosphate removal in primary wastewater. Overall, cultivation of microalgae in primary and primary + 5 % ADC may be more favorable from an economical and sustainability perspective due to elimination of the costly and energy-intensive biological treatment step. These findings demonstrate that unsterilized wastewater and ADC can serve as critical nutrient sources for biomass generation and that robust microalgae can be potent players in wastewater phytoremediation.

This study evaluated methane production and nutrient recovery from industrially produced, lipid extracted algal biomass (LEA) of Auxenochlorella protothecoides using semi-continuous anaerobic digestion (AD) at different organic loading rates (OLRs) and hydraulic retention times (HRTs). It was shown, that AD can improve biofuel production efficiency and sustainability, especially for scaled processes, through up to 30% increase in energy generation (up to 0.25 L of methane per g of LEA volatile solids) and partial nutrient recovery and recycling. The nutrient recycling with the AD effluent may reduce the cost of the supplied fertilizers by up to 45%. However, methane production was limited to nearly 50% of theoretical maxima potentially due to biomass recalcitrance and inhibition effects from the residual solvent in the LEA. Traveling bacterial waves are capable of improving contaminant remediation in the subsurface. It is fairly well understood how bacterial chemotaxis and growth separately affect the formation and propagation of such waves. However, their interaction is not well understood. We therefore perform a modeling study to investigate the coupled effects of chemotaxis and growth on bacterial migration, and examine their effects on contaminant remediation. We study the waves by using different initial electron acceptor concentrations for different bacteria and substrate systems. Three types of traveling waves can occur: a chemotactic wave due to the biased movement of chemotactic bacteria resulting from metabolism-generated substrate concentration gradients; a growth/decay/motility wave due to a dynamic equilibrium between bacterial growth, decay and random motility; and an integrated wave due to the interaction between bacterial chemotaxis and growth. Chemotaxis hardly enhances the bacterial propagation if it is too weak to form a chemotactic wave or its wave speed is less than half of the growth/decay/motility wave speed. However, chemotaxis significantly accelerates bacterial propagation once its wave speed exceeds the growth/decay/motility wave speed. When convection occurs, it speeds up the growth/decay/motility wave but slows down or even eliminates the chemotactic wave due to the dispersion. Bacterial survival proves particularly important for bacterial propagation. A critical factor in implementing microalgal biofuels for mass production is the nutrient requirements. The current study investigated the fate of macro- and micronutrients and their availability in a sequential phototrophic-heterotrophic production process for the lipid rich microalga Auxenochlorella protothecoides. More than 99 % (by weight) of overall process nutrients were supplied during the initial photoautotrophic stage reflecting its significantly larger volume. Under photoautotrophic growth conditions only 9-35 % of supplied Mn, S, Fe, N, Mg, and Cu and less than 5 % of P, Mo, Co, B, Zn, and Ca were consumed by the algae. The rest of these nutrients remain in the spent growth media during the culture concentration-down from an 800 L phototrophic pond to a 5 L heterotrophic fermenter. In contrast, Zn, Mo, Mn, Mg, Ca, and N were exhausted (90-99 % removal) during the first 25 h of the heterotrophic growth stage. The depletion of these key nutrients may have ultimately limited the final biomass density and/or lipid productivity achieved. Approximately 10-20 % of the total supplied S, Mn, Fe, N, and Cu and 5 % of Ca and Zn were assimilated into algal biomass. Several elements including N, P, Mn, B, Cu, Ca, Mg, S, and Fe were released back into the liquid phase by anaerobic digestion (AD) of the residual biomass after lipid extraction. The nutrients recovered from the AD effluent and remaining in the spent medium should be recycled or their initial concentration to the phototrophic stage decreased to enhance process economics and sustainability for future commercialization of algal-derived biofuels.

The effect of various pretreatment strategies on methane yields following anaerobic digestion (AD) of five different microalgal strains was investigated. Pavlova&#95;cf sp., Tetraselmis sp. and Thalassiosira weissflogii exhibited substantial methane yields of 0.4-0.5L/g volatile solids (VS) without pretreatment, providing up to 75-80% of theoretical values. In contrast, methane yields from Chlorella sp. and Nannochloropsis sp. were around 0.35L/g VS, or 55-60% of the theoretical values, respectively. Alkali treatment was not effective and thermal pretreatment only enhanced Nannochloropsis methane yields. Thermochemical pretreatment had the strongest impact on biomass solubilization with methane yields increasing by 30% and 40% for Chlorella and Nannochloropsis, respectively. The lipid content had a strong beneficial impact on the theoretical and observed methane yields as compared to protein and carbohydrate content. Other features such as cell-wall composition are also likely to be important factors dictating algal biodegradability and methane yields addressed in part by thermochemical pretreatment., (Copyright © 2014 Elsevier Ltd. Pharmaceuticals and personal care products (PPCPs) have been detected in bodies of water worldwide, yet their effects on the environment are not fully understood. Recent toxicity studies suggest that mixtures of PPCPs at low concentrations may be detrimental to exposed organisms, highlighting the need to remove PPCPs from wastewater treatment plant effluent before it is discharged to the environment. In this study, the utility of biofilm-based PPCP removal as a means to prevent environmental PPCP contamination was investigated. The removal of 14 PPCPs, each at an initial concentration of 10 μg/L, was studied in laboratory sand columns inoculated with wastewater treatment plant effluent. The examined PPCPs included biosol, biphenylol, p-chloro-m-cresol, p-chloro-m-xylenol, chlorophene, sodium diclofenac, gabapentin, gemfibrozil, 5-fluorouracil, ibuprofen, ketoprofen, naproxen, triclosan, and valproic acid. Ten of the PPCPs were removed by greater than 95% during column passage, while the four other compounds proved more recalcitrant. The effect of the concentration (either 50 or 1000 μg/L) of an easily degradable primary substrate (acetate) supplied along with the mixture of PPCPs was examined. Most of the tested PPCPs were removed consistently by the biofilms regardless of the concentration of acetate, although the extent of removal for three compounds showed dependence on acetate concentration, and two behaved with no reproducible pattern over time. Biofilm protein measurements indicated that the mixture of PPCPs supplied to columns suppressed biofilm growth, suggesting toxicity of the PPCPs to the biofilm communities. This laboratory-scale experiment suggests that biofilm-based water treatment strategies, such as soil aquifer treatment and slow sand filtration, may be well-suited for the removal of many PPCPs from impacted water., (Copyright © 2012 Elsevier Ltd. A sensitive yet robust analytical method is presented for the simultaneous determination of 12 human pharmaceuticals (valproic acid, phenytoin, ibuprofen, gabapentin, acetaminophen, gemfibrozil, naproxen, ketoprofen, secobarbital, phenobarbital, 5-fluorouracil, and diclofenac) and 6 antiseptics (biosol, biphenylol, p-chloro-m-cresol, p-chloro-m-xylenol, chlorophene, and triclosan). The method employs solid-phase extraction (SPE) followed by a novel pentafluorobenzylation using a mixture of acetontrile/water (1/1, v/v). The method is simple to perform (derivatization can be completed in a single test tube) and eliminates the need for any solvent/SPE cartridge drying or blow-down. It affords excellent resolution, high sensitivity and reproducibility, and freedom from interference even for matrices as complex as untreated sewage. The method was applied to the analysis of sewage samples using 15 isotopically labeled surrogates, which resulted in the detection of 10 of the 12 pharmaceuticals and all of the antiseptics sought. Ten of 15 surrogates were synthesized from pure analytes by a simple H-D exchange reaction employing D(2)O and D(2)SO(4). Measured recoveries were sensitive to matrix effects and varied substantially among analytes, indicative of the limitations associated with using a single surrogate standard.

The presence of pharmaceuticals and other wastewater-derived micropollutants in surface and groundwaters is receiving intense public and scientific attention. Yet simple GC/MS methods that would enable measurement of a wide range of such compounds are scarce. This paper describes a GC/MS method for the simultaneous determination of 13 pharmaceuticals (acetaminophen, albuterol, allopurinol, amitriptyline, brompheniramine, carbamazepine, carisoprodol, ciclopirox, diazepam, fenofibrate, metoprolol, primidone, and terbinafine) and 5 wastewater-derived contaminants (caffeine, diethyltoluamide, n-butylbenzene sulfonamide, n-nonylphenol, and n-octylphenol) by solid phase extraction (SPE) and derivatization with BSTFA. The method was applied to the analysis of raw and treated sewage samples obtained from a wastewater treatment plant located in the mid-Atlantic United States. Pharmaceuticals and personal care products (PPCPs) have been the focus of much recent research as concerns rise about their occurrence in bodies of water worldwide. In an effort to characterize the risk and determine the prevalence of these micropollutants in lakes and rivers, many researchers are examining PPCP removal from impaired water during wastewater treatment and water recycling (soil passage) processes. Biodegradation studies and projects considering combinations of biodegradation and other removal processes have been conducted over a wide range of compound categories and therapeutic classes, as well as across different systems and scales of study. This review summarizes the extent of PPCP removal observed in these various systems.

Treated drinking water samples from 12 water utilities in the Midwestern United States were collected during Fall 2003 and Spring 2004 and were analyzed for selected neutral degradates of chloroacetamide herbicides, along with related compounds. Target analytes included 20 neutral chloroacetamide degradates, six ionic chloroacetamide degradates, four parent chloroacetamide herbicides, three triazine herbicides, and two neutral triazine degradates. In the fall samples, 17 of 20 neutral chloroacetamide degradates were detected in the finished drinking water, while 19 of 20 neutral chloroacetamide degradates were detected in the spring. Median concentrations for the neutral chloroacetamide degradates were approximately 2-60ng/L during both sampling periods. Concentrations measured in the fall samples of treated water were nearly the same as those measured in source waters, despite the variety of treatment trains employed. Significant removals (average of 40% for all compounds) were only found in the spring samples at those utilities that employed activated carbon.

Bioavailability often controls the fate of organic contaminants in surface and subsurface aquatic environments. Bioavailability can be limited by sorption, mass transfer, and intrinsic biodegradation potential and can be further altered by the presence of other compounds. This paper reviews current perspectives on the processes influencing subsurface contaminant bioavailability, how these processes are modeled, and how the relative role of the various processes can be assessed through bioavailability indices. Although these processes are increasingly well understood, the use of sophisticated models and indices often are precluded by an inability to estimate the many parameters that are associated with complex models. Nonetheless, the proper representation of sorption, mass transfer, biodegradation, and co-solute effects can be critical in predicting bio-attenuation. The influence of these processes on contaminant fate is illustrated with numerical simulations for the simultaneous degradation of toluene (growth substrate) and trichloroethylene (nongrowth cometabolite) in hypothetical, aerobic, solid-water systems. The results show how the relative impacts on contaminant fate of the model's various component processes depends upon system conditions, including co-solute concentrations. Slow biodegradation rates increase the inhibition effects of a cometabolite and suppress the rate enhancement effects of a growth substrate. Irrespective of co-solute effects, contaminant fate is less sensitive to biodegradation processes in systems with strong sorption and slow desorption rates. Bioavailability indices can be used to relate these findings and to help identify appropriate modeling simplifications. In general, however, there remains a need to redefine such indices in order that bioavailability concepts can be better incorporated into site characterization, remediation design, and regulatory oversight.

Four chloroacetamide herbicides and 20 neutral chloroacetamide derivatives (known to occur as their environmental degradates) were subjected to simulated drinking water treatment (coagulation, oxidation and adsorption). Coagulation with alum and ferric chloride, at doses for optimum turbidity removal, provided little to no (<10%) removal of parent herbicides or neutral degradates. Chlorination with 6 mg/L applied free chlorine for 6 h was able to achieve 100% removal of those degradates lacking an acetanilide substituent; compounds possessing this functional group exhibited low (0-16%) removal efficiencies. Products were generally not identified, except in the case of dimethenamid and its deschloro degradate, both of which formed a single ring-chlorination product on their ready reaction (84% and 96% removal, respectively) with aqueous chlorine species. Treatment with ozone at an applied dose of 3 mg/L for 30 min proved effective (60-100%) at transforming all of the compounds under investigation to unidentified products. The parent herbicides and neutral degradates underwent adsorption by powdered activated carbon (PAC). Adsorption capacities (Freundlich K constants) correlated with K(ow) values.

This study was conducted to assess the applicability of alkaline hydrolysis as an alternative ex situ technology for remediating 2,4,6-trinitrotoluene (TNT)-contaminated water. TNT reactivity had a strong dependence on the reaction pH (11-12) and initial TNT (5-25 mg L(-1)) in batch systems, resulting in pseudo first-order transformation rate, k ranging between 1.9 x 10(-3) and 9.3 x 10(-5) min(-1). In continuous flow stirred-tank reactor (CFSTR) systems with initial TNT of 1 mg L(-1), the highest 74% of TNT reduction was achieved at the reaction pH of 11.9 and 2-day hydraulic retention time under steady-state condition. Oxalate was produced as the major hydrolysate in the CFSTRs, indicating a ring cleavage during alkaline hydrolysis. It was also believed that TNT alkaline hydrolysis occurred through the production of color-forming intermediates via dimerization. It is concluded that alkaline hydrolysis can be an alternative treatment technology for remediation of TNT-contaminated water.

Microbial monitoring was conducted over a period of more than 1 year at three full-scale riverbank filtration (RBF) facilities, located in the United States along the Ohio, Missouri, and Wabash Rivers. Results of this study demonstrated the potential for RBF to provide substantial reductions in microorganism concentrations relative to the raw water sources. Cryptosporidium and Giardia were detected occasionally in the river waters but never in any of the well waters. Average concentrations and log reductions of Cryptosporidium and Giardia could not be accurately determined due to the low and variable concentrations in the river waters and the lack of detectable concentrations in the well waters. Average concentrations of aerobic and anaerobic spore-forming bacteria, which have both been proposed as potential surrogates for the protozoans, were reduced at the three facilities by 0.8 to > 3.1 logs and 0.4 to > 4.9 logs, respectively. Average concentrations of male-specific and somatic bacteriophage were reduced by > 2.1 logs and 3.2 logs, respectively. Total coliforms were rarely detected in the well waters, with 5.5 and 6.1 log reductions in average concentrations at the two wells at one of the sites relative to the river water. Average turbidity reductions upon RBF at the three sites were between 2.2 and 3.3 logs. Turbidity and microbial concentrations in the river waters generally tracked the river discharge; a similar relationship between the well water concentrations and river discharge was not observed, due to the low, relatively constant well water turbidities and lack of a significant number of detections of microorganisms in the well waters. Further research is needed to better understand the relationships among transport of pathogens (e.g., Cryptosporidium, Giardia, viruses) and potential surrogate parameters (including bacterial spores and bacteriophage) during RBF and the effects of water and sediment characteristics on removal efficiency.

Effects of H(2)O(2) dosage (0, 10, 50, 100 and 300 mg/l), reaction pH (11.9, 6.5 and 2.5) and initial color intensity (85, 80 and 60 color unit) on decolorization of alkaline 2,4,6-trinitrotoluene (TNT) hydrolysis effluents were investigated at a fixed UV strength (40 W/m(2)). Results indicated that UV/H(2)O(2) oxidation could efficiently achieve decolorization and further mineralization. Pseudo first-order decolorization rate constants, k, ranged between 2.9 and 5.4 h(-1) with higher values for lower H(2)O(2) dosage (i.e., 10 mg/l H(2)O(2)) when the decolorization occurred at the reaction pH of 11.9, whereas a faster decolorization was achieved with increase in H(2)O(2) dosage at both pH 6.5 and 2.5, resulting in the values of k as fast as 15.4 and 26.6 h(-1) with 300 mg/l H(2)O(2) at pH 6.5 and 2.5, respectively. Difference in decolorization rates was attributed to the reaction pH rather than to the initial color intensity, resulting from the scavenging of hydroxyl radical by carbonate ion. About 40% of spontaneous mineralization was achieved with addition of 10 mg/l H(2)O(2) at pH 6.5. Efficient decolorization and extension of H(2)O(2) longevity were observed at pH 6.5 conditions. It is recommended that the colored effluents from alkaline TNT hydrolysis be neutralized prior to a decolorization step.

Inactivation of Klebsiella pneumoniae cultures by chlorine and chloramine was evaluated under different growth conditions by varying nutrient media dilution, concentrations of essential inorganic nutrients (FeCl3, MgSO4, phosphate, and ammonium salts), and temperature. All inactivation assays were performed at room temperature (22-23 degrees C) and near neutral pH (7.2-7.5). C*T(99.9) values for chlorine increased >20-fold and for chloramine increased 2.6-fold when cells were grown in 100-fold diluted nutrient broth (2NB) solutions (final TOC of 35-40 mg/L). Background levels of Mg: 6.75 x 10(-2) mM and Fe: 3.58 x 10(-5) mM or high levels of FeCl3 (0.01 mM) and MgSO4 (1 mM) during growth resulted in the highest resistances to chlorine with C*T(99.9) values of 13.06 (+/-0.91) and 13.78 (+/-1.97) mg-min/L, respectively. Addition of low levels of FeCl3 (0.001 mM) and MgSO4 (0.1 mM) to K. pneumoniae cultures during growth resulted in the lowest bacterial resistances to inactivation; C*T(99.9) values ranged from 0.28 (+/-0.06) to 1.88 (+/-0.53)mg-min/L in these cultures. Increase in growth temperature from 22.5 degrees C to 35 degrees C for unamended 2NB cultures resulted in a 42-fold decrease in C*T(99.9) values for chlorine. A similar change in temperature resulted in no significant change in C*T(99.9) values for chloramine. These results indicate that inactivation of K. pneumoniae cultures by chlorine was highly sensitive to changes in growth conditions unlike inactivation by chloramine.
