352. Examining the Use of Video Annotations in Debriefing Conversations during Video-Assisted Coaching Cycles

Abstract

This study examined how mathematics coaches leverage written annotations to support professional discourse with teachers about important classroom events during synchronous debriefing conversations. Coaches and teachers created the annotations while asynchronously watching video of an implemented lesson as part of online video-assisted coaching cycles. More specifically, this project examined the extent to which a coach and teacher discussed the annotations during a debrief conversation in a coaching cycle. We present a rationale for needing new knowledge about the relationships between video annotations and professional discourse as well as the potential implications of such knowledge. 353. Leveraging an Innovative Teaching Strategy to Identify Use of Biostatistics by Advanced Practice Nurses

Abstract

Advanced practice nurses (APNs) have a responsibility to translate research evidence into their clinical practice (i.e., evidence-based practice) for the promotion of patient and population health. A precursor to the adoption of evidence-based practice is APNs having sufficient knowledge and understanding of biostatistics. To promote APN students' motivation to learn biostatistics, nursing faculty teaching biostatistics need to know how biostatistics are used by APNs in their clinical practice. A teaching innovation was incorporated in a biostatistics course to describe the use of biostatistics by APNs. For this study, 118 students conducted interviews with APNs determining their use of biostatistics in clinical practice. Students posted interview summaries to Flipgrid, a video web-based blog. Blog posts were qualitatively analyzed; derived themes were Consuming the Literature, Translating the Evidence, and Conducting Biostatistical Analyses. The APN interview as implemented was an active learning strategy effective for generating the scientific premise to inform students of the need to learn biostatistics.

Abstract

BackgroundClarifying the relationship between depression symptoms and cardiometabolic and related health could clarify risk factors and treatment targets. The objective of this study was to assess whether depression symptoms in midlife are associated with the subsequent onset of cardiometabolic health problems.MethodsThe study sample comprised 787 male twin veterans with polygenic risk score data who participated in the Harvard Twin Study of Substance Abuse ('baseline') and the longitudinal Vietnam Era Twin Study of Aging ('follow-up'). Depression symptoms were assessed at baseline [mean age 41.42 years (s.d. = 2.34)] using the Diagnostic Interview Schedule, Version III, Revised. The onset of eight cardiometabolic conditions (atrial fibrillation, diabetes, erectile dysfunction, hypercholesterolemia, hypertension, myocardial infarction, sleep apnea, and stroke) was assessed via self-reported doctor diagnosis at follow-up [mean age 67.59 years (s.d. = 2.41)].ResultsTotal depression symptoms were longitudinally associated with incident diabetes (OR 1.29, 95% CI 1.07-1.57), erectile dysfunction (OR 1.32, 95% CI 1.10-1.59), hypercholesterolemia (OR 1.26, 95% CI 1.04-1.53), and sleep apnea (OR 1.40, 95% CI 1.13-1.74) over 27 years after controlling for age, alcohol consumption, smoking, body mass index, C-reactive protein, and polygenic risk for specific health conditions. In sensitivity analyses that excluded somatic depression symptoms, only the association with sleep apnea remained significant (OR 1.32, 95% CI 1.09-1.60).ConclusionsA history of depression symptoms by early midlife is associated with an elevated risk for subsequent development of several self-reported health conditions. When isolated, non-somatic depression symptoms are associated with incident self-reported sleep apnea. Depression symptom history may be a predictor or marker of cardiometabolic risk over decades.

Abstract

Medulloblastoma (MB) comprises a group of heterogeneous paediatric embryonal neoplasms of the hindbrain with strong links to early development of the hindbrain1-4. Mutations that activate Sonic hedgehog signalling lead to Sonic hedgehog MB in the upper rhombic lip (RL) granule cell lineage5-8. By contrast, mutations that activate WNT signalling lead to WNT MB in the lower RL9,10. However, little is known about the more commonly occurring group 4 (G4) MB, which is thought to arise in the unipolar brush cell lineage3,4. Here we demonstrate that somatic mutations that cause G4 MB converge on the core binding factor alpha (CBFA) complex and mutually exclusive alterations that affect CBFA2T2, CBFA2T3, PRDM6, UTX and OTX2. CBFA2T2 is expressed early in the progenitor cells of the cerebellar RL subventricular zone in Homo sapiens, and G4 MB transcriptionally resembles these progenitors but are stalled in developmental time. Knockdown of OTX2 in model systems relieves this differentiation blockade, which allows MB cells to spontaneously proceed along normal developmental differentiation trajectories. The specific nature of the split human RL, which is destined to generate most of the neurons in the human brain, and its high level of susceptible EOMES+KI67+ unipolar brush cell progenitor cells probably predisposes our species to the development of G4 MB.

356. Relationships between Depression and Executive Functioning in Adolescents: The Moderating Role of Unpredictable Home Environment.

Abstract

Although researchers have explored the link between depression and executive functioning (EF), the influence of early-life environmental and relational instability on this association has not been comprehensively assessed in adolescents. This cross-sectional study examined whether unpredictability of home environment in childhood moderated the relationship between depression and EF in adolescents. Participants were 138 adolescents aged 13 to 17 years (72% female; 47.8% White; 47.1% Hispanic). Diagnostic status (major depression versus healthy control) and depression severity were assessed using psycho-diagnostic interviews and self-reports from parents and adolescents. Participants also completed the Questionnaire of Unpredictability in Childhood (QUIC). EF was assessed using self-report (Behavior Rating Inventory of Executive Function, Second Edition; BRIEF2) and a battery of performance-based measures. Results showed that QUIC scores moderated the relationship between depression and BRIEF2 scores, such that high unpredictability was associated with poorer EF for adolescents exhibiting low depression severity. Participants with the highest levels of depression exhibited the poorest EF ratings, regardless of childhood unpredictability. Unpredictability was moderately associated with performance-based measures, but did not interact with depressive symptoms to predict complex performance-based EF. Recommendations include assessing for unpredictable childhood environment and acknowledging this risk factor for poor EF in youth with sub-threshold depression. Treatment implications are discussed with respect to family systems/parenting interventions, as mildly depressed adolescents growing up in unstable homes may be vulnerable to EF difficulties. Further, this study adds to the EF measurement literature by examining associations between self-report and performance-based EF instruments in a diverse sample of adolescents.

Abstract

Biodiversity monitoring is an almost inconceivable challenge at the scale of the entire Earth. The current (and soon to be flown) generation of spaceborne and airborne optical sensors (i.e., imaging spectrometers) can collect detailed information at unprecedented spatial, temporal, and spectral resolutions. These new data streams are preceded by a revolution in modeling and analytics that can utilize the richness of these datasets to measure a wide range of plant traits, community composition, and ecosystem functions. At the heart of this framework for monitoring plant biodiversity is the idea of remotely identifying species by making use of the 'spectral species' concept. In theory, the spectral species concept can be defined as a species characterized by a unique spectral signature and thus remotely detectable within pixel units of a spectral image. In reality, depending on spatial resolution, pixels may contain several species which renders species-specific assignment of spectral information more challenging. The aim of this paper is to review the spectral species concept and relate it to underlying ecological principles, while also discussing the complexities, challenges and opportunities to apply this concept given current and future scientific advances in remote sensing.

Abstract

PurposeTelomere maintenance is a hallmark of cancer. Most tumors maintain telomere length via reactivation of telomerase reverse transcriptase (TERT) expression. Identifying clinically translatable imaging biomarkers of TERT can enable noninvasive assessment of tumor proliferation and response to therapy.Experimental designWe used RNAi, doxycycline-inducible expression systems, and pharmacologic inhibitors to mechanistically delineate the association between TERT and metabolism in preclinical patient-derived tumor models. Deuterium magnetic resonance spectroscopy (2H-MRS), which is a novel, translational metabolic imaging modality, was used for imaging TERT in cells and tumor-bearing mice in vivo.ResultsOur results indicate that TERT expression is associated with elevated NADH in multiple cancers, including glioblastoma, oligodendroglioma, melanoma, neuroblastoma, and hepatocellular carcinoma. Mechanistically, TERT acts via the metabolic regulator FOXO1 to upregulate nicotinamide phosphoribosyl transferase, which is the key enzyme for NAD+ biosynthesis, and the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase, which converts NAD+ to NADH. Because NADH is essential for pyruvate flux to lactate, we show that 2H-MRS-based assessment of lactate production from [U-2H]-pyruvate reports on TERT expression in preclinical tumor models in vivo, including at clinical field strength (3T). Importantly, [U-2H]-pyruvate reports on early response to therapy in mice bearing orthotopic patient-derived gliomas at early timepoints before radiographic alterations can be visualized by MRI.ConclusionsElevated NADH is a metabolic consequence of TERT expression in cancer. Importantly, [U-2H]-pyruvate reports on early response to therapy, prior to anatomic alterations, thereby providing clinicians with a novel tool for assessment of tumor burden and treatment response in cancer.

Abstract

We compared short stories by autistic (n = 19) and non-autistic (n = 23) university students. We used automated software and content analysis to code students' stories. We found that writings were more similar than different. However, autistic students' stories were rated at a higher reading level (p = .013) than non-autistic students'. Autistic students' stories contained fewer grammatical errors (p = .02) but were less likely to include a climax (p = .026). Autistic students reported more positive writing affect than non-autistic students (p = .026). Higher writing affect was associated with writing highly fictional texts (p = .03) that contained more sentences (p = .005). 385. Examining How Teachers Enact the Suggestions of a Coach: Critique of a Methodology

Abstract

In this methodology paper, we present a methodology for characterizing how teachers use coaches' suggestions. We identified suggestions from planning conversations and explored the extent to which the teachers implemented the suggestions in enacted lessons. The planning conversations took place within online content-focused coaching cycles. A primary challenge confronting content-focused coaches when working one-on-one with teachers is finding a productive balance between giving suggestions and inquiring into teachers' practices through reflective questioning. This paper articulates a process for identifying suggestions made by a coach during a planning conversation and an analytic process for examining how a teacher takes up the suggestion during lesson implementation. We discuss the methodological challenges we encountered and tradeoffs in our decisions related to low- and high-inference claims. [For the complete proceedings, see ED629884.]

386. Coaches' and Teachers' Noticing through Annotations: Exploring Analytic Stance across Coaching Cycles

Abstract

We share results of a study on the analytic stances of coaches' and teachers' as they annotated key moments from classroom video of the teacher's lessons. In the analysis, emphasis was on the analytic stances of the coaches and how their annotations related to trends in teachers' annotations. Findings indicate differences in how coaches and teachers noticed across the coaching cycles, suggesting the annotations were influenced by the interactions between the coaches and teachers and the teachers' perceptions of coaching process. As a result of our analysis, we characterized one coach as having a high ratio of questions to suggestions, another as having annotations coded as interpretation, another as having more evaluations and suggestions, and the fourth as having asked more questions. Some teachers mirrored the analytic stance of their coach over time and other teachers shifted their analytic stance in ways that suggest they were responsive to their coach's analytic stance. Abstract

We consider products of $\psi$ classes and products of $\omega$ classes on $\overline{M}_{0,n+3}$. For each product, we construct a flat family of subschemes of $\overline{M}_{0,n+3}$ whose general fiber is a complete intersection representing the product, and whose special fiber is a generically reduced union of boundary strata. Our construction is built up inductively as a sequence of one-parameter degenerations, using an explicit parametrized collection of hyperplane sections. Combinatorially, our construction expresses each product as a positive, multiplicity-free sum of classes of boundary strata. These are given by a combinatorial algorithm on trees we call 'slide labeling'. As a corollary, we obtain a combinatorial formula for the $\kappa$ classes in terms of boundary strata. For degree-$n$ products of $\omega$ classes, the special fiber is a finite reduced union of (boundary) points, and its cardinality is one of the multidegrees of the corresponding embedding $\Omega_n: \overline{M}_{0,n+3}\to \mathbb{P}^1\times \cdots \times \mathbb{P}^n$. In the case of the product $\omega_1\cdots \omega_n$, these points exhibit a connection to permutation pattern avoidance. Finally, we show that in certain cases, a prior interpretation of the multidegrees via tournaments can also be obtained by degenerations.

Let $R$ be any ring with identity. We show that the homotopy category of all acyclic chain complexes of pure-projective $R$-modules is a compactly generated triangulated category. We do this by constructing abelian model structures that put this homotopy category into a recollement with two other compactly generated triangulated categories: The usual derived category of $R$ and the pure derived category of $R$. Abstract

We give a combinatorial proof of a recent geometric result of Farkas and Lian on linear series on curves with prescribed incidence conditions. The result states that the expected number of degree-$d$ morphisms from a general genus $g$, $n$-marked curve $C$ to $\mathbb{P}^r$, sending the marked points on $C$ to specified general points in $\mathbb{P}^r$, is equal to $(r+1)^g$ for sufficiently large $d$. This computation may be rephrased as an intersection problem on Grassmannians, which has a natural combinatorial interpretation in terms of Young tableaux by the classical Littlewood-Richardson rule. We give a bijection, generalizing the well-known RSK correspondence, between the tableaux in question and the $(r+1)$-ary sequences of length $g$, and we explore our bijection's combinatorial properties. We also apply similar methods to give a combinatorial interpretation and proof of the fact that, in the modified setting in which $r=1$ and several marked points map to the same point in $\mathbb{P}^1$, the number of morphisms is still $2^g$ for sufficiently large $d$.
