- Abstract
Step recovery diode (SRD) is the kernel of ultra-wideband (UWB) fuze generating a narrow pulse signal, and the parameters of SRD can affect the amplitude and width of narrow pulse signal. A SRD model is established based on the semiconductor device theory and SRD operating states, and the effects of SRD parameters on forward bias admittance and reverse bias barrier capacitance are studied. According to narrow pulse generating equivalent circuit, a circuit transient analysis method is used to solve the expression of narrow pulse amplitude and width. The relationships among SRD minority carrier lifetime, reverse saturation current, bias junction capacitance, doping distribution coefficient, narrow pulse amplitude and pulse width are studied through simulation. The simulated results show that the amplitude and pulse width of narrow pulse can be adjusted by changing SRD parameters. The correctness of simulated results is verified by comparing with the generated narrow pulse. [ABSTRACT FROM AUTHOR]

- Abstract
Obejective: To investigate the effects of propofol combined with dexmedetomidine on the analgesia in patients undergoing radical resection of lung cancer and its effect on stress response. Methods: 78 patients with radical resection of lung cancer from August 2014 to July 2016 were divided into the observation group and the control group according to the order of admission. The control group was given midazolam and dexmedetomidine for anesthesia. The observation group was treated with propofol combined with dexmedetomidine. The analgesic effect, mean arterial pressure, heart rate, blood glucose, interleukin-6(IL-6), interleukin-10 (IL-10), tumor necrosis factor-α(TNF-α) and total white blood cell count (WBC) levels in the two groups were compared. Results: The visual analogue (VAS) scores of patients in the observation group were significantly lower than those of the control group on waking up, at 10min and 30min after waking up (P<0.05), the levels of serum IL-6, TNF-α, WBC, heart rate, mean arterial blood pressure and blood glucose were significantly lower than those of the control group at 24 hours after treatment(P<0.05), and serum interleukin(IL-10) level was significantly higher than that of the control group <0.05). There was no significant difference in the incidence of adverse reactions between the observation group and the control group (P>0.05). Conclusions: Propofol combined with dexmedetomidine had good analgesic effect in the radical resection of lung cancer, and could effectively relieve the stress reaction with high safety. - Abstract
Caffeine and theobromine as the major component of alkaloids in tea,while caffeine is the important taste compound. As the application of caffeine increased extensively in the fields of food and medicine,caffeine biosynthesis becomes a new hotspot. Nowadays,exploring the method of caffeine biosynthesis is of great significance,when caffeine synthesis mainly relies on chemical synthesis on the market. This research concatenated coffee xanthosine methyltransferase gene ( CaXM T) and four mutants of tea caffeine synthase gene ( TCS1) respectively in the same expression vector pM A L-c5X, then induced coexpression of the fusion protein,which was analyzed by SDS-PAGE gel electrophoresis,and chromatography were added into the crude enzyme solution by sonication methionine. At la s t,in vitro enzymatic reaction products were detected by high performance liquid. The results showed that only theobromine generated in the products of p MAL- CaXMT - T M 2 /3 /4 without caffeine. This research provides the information for establishing tandem gene coexpression vector which was used for the biosynthesis of caffeine and theobromine and new ideas to study on caffeine biosynthesis. - Abstract
Ultra-wide bandwidth(UWB) radio fuze receiver receives a target echo signal based on a correlation receiving principle. The signals from fuze receiver are obtained by the correlation calculation of echo signal and delayed sampling pulse signal. The sampling pulse width can be optimized to increase the detection sensitivity of UWB fuze. A correlation receiving mathematical model of UWB fuze is established based on the correlation receiving theory,and the relationship between the sampling pulse width and the amplitude correlation receiver function is analyzed. The received output signals are simulated when transmitted pulse width is 50 ps,100 ps and 200 ps. The simulated results show that the fuze-received output signal has the maximum amplitude when the sampling pulse width is the same as the transmitted pulse width. [ABSTRACT FROM AUTHOR]

319. 11 096 例体检者幽门螺杆菌感染现状分析.

- Abstract
Objective To investigate the distribution of Helicobacter pylori (Hp) infections among general population in Tianjin. Methods Individuals (n=11 096) who visited our hospital for medical examination and H. Pylori infection screen were included in the research objects. Rapid urease method was utilized to rapidly detect H. pylori infection among the popultion. Individuals were grouped based on their gender, age and occupations, and then were analyzed. Results H. pylori-positive rate was 30.52% (3 386/11 096) in 11 096 individuals, in which there were 34.77% (1 784/5 131) male and 26.86% (1 602/5 965) female respectively. Individuals were divided into 5 groups according to their ages: 19 to 29 years old group, 30 to 39 years old group, 40 to 49 years old group, 50 to 59 years old group, 60 to 69 years old group and 70 to 92 years old group. The positive rates of H. pylori increased with age: 19.50% ( 379/1 944 ) in 19-29 years old group, 25.82% (650/2 517 ) in 30-39 years old group, 31.59% ( 908/2 874 ) in 40-49 years old group, 37.48% (915/2 441 ) in 50-59 years old group, 41.09% ( 353/859 ) in 60-69 years old group and 39.18% (181/462) in 70-92 years old group. The differences in positive rates between different age groups were all of statistically significant (P < 0.05). Individuals were also divided into seven groups according to their occupations: medical worker group, worker group, teacher group, engineer group, clerk group, civil worker group and other career group. The positive rates of H. pylori infection were 25.42% (586/2 305), 29.35% (1 062/3 618) , 30.61% (360/1 176), 32.49% (116/357), 33.44% (205/613), 34.52% (455/1 318) and 35.23% (602/1 709) respectively. The positive rate was 25.42% (586/2 305) medical workers, which was lower than that of other occupation groups with significant difference (P < 0.002). Conclusion Good management of H. pylori diagnosis and treatment in middle age and elderly together with popularizing knowledge of H.pylori prevention can effectively reduce H.pylori incidence. Vegetative trapping of heavy rainfall is an important mechanism for mitigating frequent rain-induced flood disasters in the context of global warming. To investigate the role of natural vegetation in trapping heavy rainfall following the widespread implementation of the "the Grain for Green" project in the Loess Hilly and Gully area, an automatic soil moisture monitoring system was employed to continuously monitor the soil moisture content in the top 10 m on one shrub-covered slope and three herbaceous slopes during six heavy rainfall events occurring in the Fangta watershed of the Ansai District, Shaanxi Province, from 2019 to 2022. The results showed that the maximum infiltration depth of the herbaceous slopes was approximately 200 cm during the heavy rainfall events, while for the shrub-covered slope, it was only 100 cm. Furthermore, the shrub-covered slope exhibited superior rainfall interception compared to the herbaceous slopes. The trapping efficianncy of the herbaceous slopes improved with each successive year of "Grain for Green" project implementation. Additionally, natural shrub-covered slopes displayed a trapping efficiency of up to 60.8% during low-intensity, short-duration rainfall events, and 40.8% during high-intensity, long-duration rainfall events. Therefore, it is advisable to strategically conserve and allocate shrub-herbaceous ecosystems within the Loess Hilly and Gully area to enhance the storage and mitigation capacity of vegetation and soil in response to heavy rainfall. - Abstract
Objective To investigate the function of melatonin (MEL) on liver transplantation by detecting the effect of MEL on glucose regulated protein-78 (GRP-78) and CCAAT enhancer-binding protein homologous protein (CHOP) involved in the endoplasmic reticulum stress (ERS) pathways in livers of rats with liver transplantation. Methods The rat orthotopic liver transplantation model was established by using‘magnetic-loop method'. Male SD rats were randomized into 3 groups: the sham operation group (Sham group), the orthotopic liver transplantation group (OLT group) and the orthotopic liver transplantation + MEL treatment group (OLT + MEL group), 8 rats in each group. Serum sample and liver sample were collected at 24 h after operation. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were detected. Liver tissues were stained with hematoxylin-eosin (HE) and pathological changes of liver tissues of each group were observed. The messenger ribonucleic acid (mRNA) and protein expression levels of GRP-78 and CHOP were detected by polymerase chain reaction and Western blot. Results Compared with the Sham group, the serum ALT and AST of the OLT group increased significantly (both in P < 0. 01), liver tissues injury was serious, and mRNA and protein expression levels of GRP-78 and CHOP increased significantly (all in P < 0. 01). Compared with the OLT group, ALT and AST of the OLT + MEL group decreased significantly (both in P < 0. 05), liver tissues injury was alleviated,and mRNA and protein expression levels of GRP-78 and CHOP decreased significantly (all in P < 0. 05). Conclusions MEL may reduce mRNA and protein expression levels of GRP-78 and CHOP involved in the ERS pathways of transplant liver,which may be one of its mechanisms to alleviate liver injury after liver transplantation. Topologically nontrivial Fe-based superconductors attract extensive attentions due to their ability of hosting Majorana zero modes (MZMs) which could be used for topological quantum computation. Remarkably, the tunneling spectra show a sharp zero-bias peak (ZBP) with multiple integer-quantized states at the step edge under zero magnetic field. We propose that the increasing hole doping around step edges may drive the local superconductivity into a state with possible spontaneous time-reversal symmetry breaking. Consequently, the ZBP can be interpreted as an MZM in an effective vortex in the superconducting topological surface state by proximity to the center of a tri-junction with different superconducting order parameters. - Abstract
BACKGROUND: Pulsed electromagnetic fields, as an important physical therapy, are exactly effective in the treatment of osteoarthritis, but the mechanism has not been fully clarified. OBJECTIVE: To observe the effect of pulsed electromagnetic field on the degeneration of knee joint cartilage in aged rats. METHODS: Eight 6-month-old Sprague-Dawley rats were selected as the young group and were subjected to normal diet with no treatment. Sixteen 22-monthold Sprague-Dawley rats were randomly divided into old group (n=8) and pulsed electromagnetic field group (n=8). The rats in the pulsed electromagnetic field group were subjected to a pulsed electromagnetic field intervention, once a day, 5 days per week for continuous 8 weeks. The rats in the old group were given no treatment. All rats were anesthetized and executed after 8 weeks for the detection of relevant indexes. RESULTS AND CONCLUSION: Compared with the young group, serum type II collagen C-terminal peptide level was increased in the old group (P < 0.05); compared with the old group, serum type II collagen C-terminal peptide level was decreased in the pulsed electromagnetic field group (P < 0.05). Micro-CT showed that the bone volume fraction, bone mineral density, and number of bone trabeculae decreased (P < 0.05) and the trabecular separation increased (P < 0.05) in the tibia of rats in the aged group compared with the young group; and the bone volume fraction, bone density, and number of trabeculae increased (P < 0.05) and the trabecular separation decreased (P < 0.05) in the tibia of rats in the pulsed electromagnetic field group compared with the aged group. The tibial plateau Safranin O-fast green staining showed that the articular cartilage structure of rats in the aged group was disorganized, and the number of chondrocytes was obviously reduced, and the tidal line could not be distinguished. The above results were improved in the pulsed electromagnetic field group. RT-qPCR and western blot assay showed that the mRNA and protein expression levels of matrix metalloproteinase 1, matrix metalloproteinase 13, P53 and P21 in the articular cartilage and subchondral bone of rats were elevated in the aged group compared with the young group (P < 0.05) and decreased in the pulsed electromagnetic field group compared with the old group (P < 0.05). To conclude, pulsed electromagnetic fields may improve osteoarthritis in aged rats by inhibiting chondrocyte senescence, alleviating articular cartilage degradation and inhibiting subchondral bone osteoporosis through suppressing the expression of P53/P21. In order to study the non-covalent interaction between peptides, the peptides were mixed in the molar rat io of 1 : 1, and the interact ion between peptides were analyzed by electrospray ionization mass spectrometry after equilibrium. The results showed that some of the pentapeptides did not interact with each other to form non-- covalent complexes, and another part of the pentapeptides could easily form noncovalent complexes through interaction. After the amino acid sequence of peptides were changed and the peptide chain were lengthened, the changes of peptides interaction were analyzed by electrospray ionization mass spectrometry, and the effects of sulfhydryl group, hydrogen bond and hydrophobic action on the non-covalent action of peptides were studied. The pentapeptide II -5 (Ile-Leu-Gly-Tyr-Ile) and IP-5 (lle-Phe-Thr-ThrPro) int eracted with each other and strong non-covalent complex peaks wer e obtained. The hydrophobic branching amino acid phenylalanine Phe in IP-5 was replaced by glycine Gly to become pentapeptide IP' -5 (Ile-Gly-Thr-Thr-Pro), and the hydroxyside chain threonine There was replaced by glycine Gly to become pentapeptide IP"-5 (lle-Phe-GlyThr-Pro), t he pen ta peptide II -5 interacted with IP' -5 and IP' -5, respectively, the spectral signals of non-covalent bond composites were weakened. The results showed that hydrogen bonding and hydrophobic interaction could enhance t he non-covalent binding between peptides. The pentapeptide AK-5(Ala-Val-Ile-Phe-Lys) and ED-S(Glu-Ile-CysAla-Asp) did not interact with each other. Thesemipatropine Cys containing sulfhydryl side chain in ED-5 was replaced by Ile without sulfhydryl side chain, and the amino acid sequence was changed to ED1 -5 (Glu-Ile-Ile-Ala-Asp). When AK-5 was mixed with ED'- S,the non-covalent effect of AK-5 and ED'-5 was stronger than that of AK-5 and ED-5. Increased the hydroxyl group and hydrophobic group in the polypeptide chain, and the influences of the peptide chain's length on the polypeptide interaction were studied. By lengthening the peptide chain, the pentapeptide ED-5 was changed into nine-peptide ED9 (Ala-Gl u-Ala-Ile-Cys-Ala-Asp-Pro-Lys), and AK-5 was changed into AK-9 (Lys-GluA la-Val-lle-Phe-Lys-Thr-Ile). Then AK-5 was mixed with ED-9, AK- 9 was mixed with ED-5, there was not non- covalent binding. The results showed that hydrogen bonding and hydrophobicity can enhance the non- covalent binding between peptides, and the thiol group is easy to form disulfide bonds between peptides, which hinders the non- covalent binding between different peptides. Objective: To investigate the distribution of pathogens in complicated abdominal infection and the prognostic evaluation value of acute physiology and chronic health II (APACHE II) score, sepsis related organ dysfunction (SOFA) score combined with procalcitonin (PCT) detection. Methods: 80 patients with complicated abdominal infection in Hefei Second People's Hospital from January 2016 to October 2020 were selected, the distribution of pathogenic bacteria in abdominal cavity were analyzed. According to the prognosis, the patients were divided into survival group (n=45) and death group (n=35). The APACHE II score, SOFA score, PCT, C-reactive protein (CRP), white blood cell count (WBC), neutrophil ratio and blood lactic acid levels were compared between the two groups. The influence factors of the prognosis of patients were analyzed, and apply the receiver-operating characteristic (ROC) curve analysis APACHE Ⅱ score, SOFA score, PCT and the value of combined detection of prognosis prediction. Results: A total of 112 strains of pathogens were isolated from 80 patients with complex abdominal infection, including 20 strains of gram-positive cocci (account for 17.86% ), 64 strains of gram-negative bacilli (account for 57.14% ) and 28 strains of fungi (account for 25.00% ). The infection rate of Candida albicans and Pseudomonas aeruginosa in the death group were significantly higher than those in the survival group, and the infection rate of Escherichia coli was significantly lower than that in the survival group (P<0.05). The serum APACHE II score, SOFA score, PCT, CRP and blood lactic acid levels in the death group were significantly higher than those in the survival group, and the ratio of WBC and neutrophils were significantly lower than those in the survival group (P<0.05). Multivariate logistic regression analysis showed that Pseudomonas aeruginosa infection, Candida albicans infection, APACHE II score is greater than or equal to 20 scores, SOFA score is greater than or equal to 14 scores, PCT is greater than or equal to 7.00 ng/mL, CRP is greater than or equal to 100.00 mg/L, blood lactic acid is greater than or equal to 4 mmol/L, WBC is less than 6.00×109 the risk factors for patients death (P<0.05). - Abstract
A slope-gully system is one of the basic units in the soil/water loss and governance in watersheds. Among them, the runoff and sediment can greatly contribute to the restoration and reconstruction of the ecological environment in soil erosion areas. The runoff and sediment yield can also depend mainly on the pattern of sediment sources, sinks, and transport paths in the slope-gully system. But it is still lacking in this field. In this study, unmanned aerial vehicles (UAV) remote sensing and geographic information systems were used to determine the distribution and quantitative indicators of sediment source-pathwaysinks in different slope-gully systems. The monitoring data of rainfall and runoff was combined with the sediment yield of five slope-gully systems in the Fangta small watershed from 2016 to 2022, including two with no human disturbance (SG1 and SG2) and two with grazing disturbances (SG3 and SG4) of slope-gully systems with restored vegetation and natural vegetation, one slope-gully system with orchard and sloping cultivated land (SG5). A systematic investigation was made to clarify the properties of runoff depth and sediment yield in five slope and gully systems, together with their responses to sediment source pathway-sink patterns. The results show that the sediment yield modulus in SG1 and SG2 was significantly lower than that of the rest (P<0.05), and the maximum runoff depth and sediment yield were found in SG3 under rainfall events. The average annual runoff depth of SG1 and SG2 was significantly lower than that of SG5 (P<0.05), and the average annual yield was significantly lower than that of the rest (P<0.05). The runoff depth was significantly positively correlated with the maximum patch index both of the sediment source and sink area in the slope-gully system, valley land, and gully land (P<0.05), respectively. In addition, the degree of correlation between the sediment source and sink index and the depth of runoff in the valley were greater than those in the gully land. The runoff depth also showed a significant negative correlation (P<0.05) with the path number density and the bifurcation ratio in the total slope-gully system, and the path gradient both in the valley and the gully land. There was no significant correlation between the sediment yield modulus and each index of source and sink, but there was a significant positive correlation (P<0.05) between the path length density and the path bifurcation ratio in the valley land. In addition, the increasing path complexity also significantly reduced the runoff depth and sediment yield modulus. In short, the appropriately increasing complexity and transport distance of sediment transport paths can be expected to effectively reduce the soil and water loss in slope-gully systems. This finding can provide scientific support for the prevention and control of soil erosion in the slope and gully systems, the ecological protection, and the restoration of watersheds. Objective: To investigate the correlation between ultrasonic elastography parameters and deep vein thrombosis of lower limbs after orthopedic surgery. Methods: Between June 2021 and June 2022, 80 patients with postoperative deep vein thrombosis of lower limbs who were diagnosed and treated in the orthopaedic department of our hospital were selected as the observation group, and 60 healthy patients who underwent physical examination at the same time were selected as the control group. Compare the ultrasound elasticity scores and strain values between the two groups; Pearson test was used to analyze the correlation between ultrasonic elasticity score, strain value and limb deep vein thrombosis; Multivariate logistic regression analysis was used to analyze the independent risk factors of limb deep vein thrombosis; The ROC was used to analyze the value of ultrasonic elasticity score and strain value in predicting deep limb vein thrombosis. Results: The ultrasound elasticity score and strain value in the observation group were higher than those in the control group (P<0.05). Pearson correlation test showed that ultrasonic elasticity score and strain value were positively correlated with lower limb deep vein thrombosis (r=0.785, 0.826, P<0.05). Multivariate logistic regression analysis showed that ultrasonic elasticity score and strain value were independent risk factors for lower limb deep vein thrombosis (OR=4.632, 4.768, P<0.05). ROC curve analysis results show that ultrasonic elasticity score and strain value are of great value in predicting lower limb deep vein thrombosis. Conclusion: With the formation of lower limb deep vein thrombosis, patients' ultrasonic elasticity scores and strain values increase. These two indicators have high value in predicting lower limb deep vein thrombosis. As a cheap and abundant renewable raw material, lignocellulosic biomass can be converted into cellulosic fuel ethanol through pretreatment, enzymatic hydrolysis and microbial fermentation, etc., which has widely attracted attention in recent decades. Poplar is a kind of fast-growing hardwood that is widely artificially cultivated and is mainly used in papermaking industry. Poplar is considered as an excellent lignocellulosic material for the production of cellulosic ethanol due to its high contents in cellulose and hemicellulose components. The application of poplar in cellulosic ethanol production was focused on in this paper, including briefly introducing the composition and structural characteristics of poplar, and importantly overviewing the research progress of poplar in pretreatment technology, enzymatic hydrolysis of pretreatment raw materials and microbial fermentation and so on. Finally, the technical obstacles and difficulties restricting the application of poplar in cellulosic ethanol industry were summarized, and the corresponding countermeasures were analyzed and the application were prospected. Objective: To investigate the relationship between serum osteopontin (OPN), thymidine kinase 1 (TK1), fibroblasts growth factor receptor 4 (FGFR4) and neoadjuvant chemotherapy in patients with locally advanced cervical cancer (LACC). Methods: 160 patients with LACC who were treated in the Affiliated Tumor Hospital of Guizhou Medical University from August 2011 to July 2019 were selected. They all received three cycles of paclitaxel and carboplatin neoadjuvant chemotherapy. According to the curative effect, the patients were divided into effective group and ineffective group. The levels of serum OPN, TK1 and FGFR4 were detected before chemotherapy, and the differences of the above indexes between the two groups were compared. The related data were collected, and the influencing factors of neoadjuvant chemotherapy in patients with LACC were analyzed by multivariate Logistic regression. The predictive value of OPN, TK1 and FGFR4 on the efficacy of neoadjuvant chemotherapy was analyzed by receiver operating characteristic (ROC) curve. Results: All 160 patients successfully completed neoadjuvant chemotherapy, with effective treatment in 129 cases (effective group), and ineffective treatment in 31 cases (ineffective group). The levels of serum OPN, TK1 and FGFR4 in ineffective group were higher than those in effective group (P<0.05). Multivariate Logistic regression analysis showed that FIGO stage Ⅲ, IVA and the high levels of serum OPN, TK1 and FGFR4 were the risk factors affecting the efficacy of neoadjuvant chemotherapy in patients with LACC (P<0.05). The areas under the ROC curve of serum OPN, TK1 and FGFR4 in predicting the efficacy of neoadjuvant chemotherapy in patients with LACC were 0.831, 0.695 and 0.767 respectively, and the areas under the curve predicted by the combination of three items were 0.893, which was higher than that detected by each index alone. Conclusion:The efficacy of neoadjuvant chemotherapy in patients with LACC is affected by FIGO stage and the levels of serum OPN, TK1 and FGFR4. Combined detection of the levels of serum OPN, TK1 and FGFR4 has a certain predictive value for the efficacy of neoadjuvant chemotherapy in patients with LACC. [ABSTRACT FROM AUTHOR]

Microstructure transformation characteristics by continuous cooling of SA508Gr. 4N steel for next generation nuclear pressure vessels were subjected to physical simulation experiment, microstructure observation and hardness measurement. Results show that critical cooling rate of SA508Gr. 4N steel for bainitic transformation is 0.5 °C /s. When cooling rate is 1 ~30 qC/s, transformation products are lath martensite with hardness up to 470 HV. When cooling rate is 0. 5 ~0. 02 °C /s, bainite and martensite are transformation products, and as cooling rate decreases,bainite content increases gradually, granular and short rod shaped fine carbides can be seen in the bainite structure, and hardness value is as low as 400 HV. Koistinen Marburger equation is used to describe martensitic transformation kinetics of SA508Gr. 4N steel, fitting results showed that initial martensitic transformation temperature is 345.4 °C and kinetic parameter is 0. 031 4,which can accurately predict the relationship between martensitic transformation and temperature with transformed amount exceeding 20%. OBJECTIVE: To probe into the effect of high mobility group protein 1 ( HMGB1)-mediated inflammatory pathway in reducing hepatotoxicity of licorice-processed Tripterygium wilfordii, so as to provide evidence for elucidating the mechanism of licorice-processed Tripterygium wilfordii in reducing hepatotoxicity. METHODS: SD rats were randomly divided into the blank control group, Tripterygium wilfordii group ( 16 g/ kg unprocessed Tripterygium wilfordii ), and licorice-processed Tripterygium wilfordii group ( 16 g/ kg unprocessed Tripterygium wilfordii), with 8 cases in each group. Three groups were given continuous intragastric administration for 3 weeks, and the blank control group was given intragastric administration with 0. 9% sodium chloride solution. Serum biochemical indicators of rats [aspartate aminotransferase (AST), alanine aminotransferase (ALT)] were detected weekly. At the end of the third week of administration, the levels of inflammatory factors HMGB1, interleukin (IL)1β, IL-2 and tumor necrosis factor α (TNF-α) in serum of rats were detected by enzyme linked immunosorbent assay. The protein expression levels of HMGB1, nuclear factor κB (NF-κB) and Toll-like receptor 4 (TLR4) in rat liver were detected by western blotting. Histopathological changes of rat liver were observed after Hematoxylin-Eosin staining. RESULTS: Compared with the Tripterygium wilfordii group, the levels of ALT and AST in licorice-processed Tripterygium wilfordii group were significantly lower (P<0. 01), the damage of liver pathological structure decreased, the levels of TNF-α, IL-1β and HMGB1 decreased significantly (P<0. 01), and the levels of IL-2 decreased (P<0. 05),the expressions of HMGB1 pathway related proteins TLR4 and NF-κB decreased (P<0. 05), and the expression of HMGB1 decreased (P<0. 01), with statistically significant differences. Compared with the blank control group, there was no statistical significance in the above indicators in the licorice-processed Tripterygium wilfordii group (P>0. 05). Objective To investigate the effect of serum uric acid (SUA) on prognosis of patients with maintenance hemodialysis (MHD) complicated with mineral metabolism disorder. Methods A total of 192 patients with MHD complicated with mineral metabolism disorder were selected. According to the level of SUA, patients were divided into the group Q1 (SUA≤363 μmol/L, n=47), the group Q2 (SUA 364-459 μmol/L, n=48), the group Q3 (SUA 460-559 μmol/L, n= 49) and the group Q4 (SUA≥560 μmol/L, n=48). The clinical data were compared between the 4 groups. Patients were followed up. Spearman method was used to analyze the correlation between SUA and other index. Kaplan-Meier method was used to compare the survival rates of the 4 groups, and Cox regression model was used to analyze the effect of SUA on the prognosis of the patients. Results Compared with the group Q1, serum creatinine (Scr), urea nitrogen (BUN) and blood phosphorus (P) were increased in the groups Q2, Q3 and Q4, glomerular filtration rate (eGFR) and high density lipoprotein (HDL-C) were decreased, and albumin (ALB) was increased in the groups Q2 and Q4 (P<0.05). Compared with the groups Q2 and Q3, the levels of Scr, BUN and P were decreased in the group Q4 (P<0.05). SUA was positively correlated with ALB, BUN, Scr, P, and magnesium (Mg), and negatively correlated with calcium (Ca) (P<0.05). The mean follow-up time was 16.0 (11.0, 23.8) months. A total of 45 patients (23.4%) had outcome events, including 22 patients (11.5%) with cardiovascular death. Kaplan-Meier survival curve showed that the all-cause mortality and cardiovascular disease mortality were higher in the group Q1 (Log-rank χ2 =9.707 and 7.912, P<0.05). Univariate Cox regression analysis showed that low ALB, low blood Mg, and CVD were risk factors for cardiovascular disease death in MHD patients with mineral metabolism disorder, while higher uric acid (the Q4 groups) was its protective factor (P<0.05). Multivariate Cox regression analysis showed that advanced age, low blood Mg and cardiovascular disease were independent risk factors for all-cause death in MHD patients with mineral metabolism disorder, while higher uric acid (the groups Q3,Q4) was its protective factor (P< 0.05). Conclusion Higher SUA can reduce all-cause mortality in MHD patients with mineral metabolism disorders. [ABSTRACT FROM AUTHOR]

- Abstract
Objective To investigate the effects of erythropoietin (EPO) intervening in human umbilical cord mesenchymal stem cells (HUMSCs) on the fibrosis of human renal tubular epithelial cell line HK-2. Methods HUMSCs and HK-2 cells were cultured, respectively. The optimal concentration of EPO in HUMSCs migration and proliferation was 100 IU/mL by Transwell assay and MTT assay, and the optimal concentration of transforming growth factor-β1 (TGF-β1)in HK-2 cell fibrosis was 10 μg/L by RT-PCR and Western blotting. The HK-2 cells were divided into 4 groups: control group (simple medium was in the upper chamber, and HK-2 cells and medium were in the lower chamber), TGF-β1 group (simple medium was in the upper chamber, and HK-2 cells added with 10 μg/L TGF-β1 were in the lower chamber), HUMSCs intervention group (HUMSCs and medium were in the upper chamber, and HK-2 cells added with 10 μg/L TGF-β1 medium were in the lower chamber), and EPO+HUMSCs intervention group (HUMSCs added with 100 IU/mL EPO were in the upper chamber, and HK-2 cells added with 10 μg/L TGF-β1 were in the lower chamber). After 72 h of culture, the morphology and growth of cells were observed under the microscope, and the number of transmembrane cells was counted. RT-PCR was used to detect the relative mRNA expression levels of fibrosis factors TGF-β1, fibronectin (FN), Col Ⅳ, α-smooth muscle actin (α-SMA), and homing factors stromal cell-derived factor1 (SDF-1) and its receptor CXCR4stem cell factor (SCF) and its receptor c-Kit in the HK-2 cells; the relative protein expression levels of fibrosis factors and homing factors were determined by Western blotting. Results Compared with the control group, after TGF-β1 intervention, HK-2 cells lost their original epithelial cell morphology and formed epithelioid cells and myofibroblasts with migration ability in the TGF-β1 group. Compared with the control group, the number of cell migration increased in the TGF-β1 group, HUMSCs intervention group, and EPO+HUMSCs intervention group; the number of cell migration in the EPO+ HUMSCs intervention group was lower than those in the HUMSCs intervention group and TGF-β1 group (all P<0. 01). Compared with the control group, the mRNA and protein levels of TGF-β1, FN, ColⅣ, α-SMA, CXCR4, SDF-1, c-Kit and SCF in the TGF-β1 group were the highest, followed by those in the HUMSCs intervention group, and those in the EPO+HUMSCs intervention group were the lowest (P<0. 05 or P<0. 01). Conclusion EPO can promote the proliferation and migration of HUMSCs, direct the migration of HUMSCs to the damaged HK-2, inhibit the differentiation of HK-2 into non-functional epithelial cells with migration ability, and delay the progression of fibrosis of renal tubular epithelial cells.
