A questionnaire covering all aspects of grapevine propagation including cultural and sanitation practices in mother blocks and harvest and transport of cuttings from mother blocks to nurseries, nursery operations and field nursery management, was mailed to all Management Committee members of the European COST Action FA1303 “Sustainable Control of Grapevine Trunk Diseases” for distribution to the identifiable nurseries in each European country. The main objective was to develop understanding of the current propagation practices and to identify those likely to have the greatest impacts on the quality of planting material, especially with regard to the control measures used against fungal trunk pathogen infections. The questionnaire was sent to 666 vine nurseries, and 146 replies were received (21.9% response rate) The study identified several risks factors which could increase infection by fungal trunk pathogens during the propagation processes, as well as a clear need for further research into the effects of treatments on grapevine viability, including hot water treatment, and the potential of biological agents and other strategies such as ozonation to control grapevine trunk diseases in nurseries.

Wood decay of kiwifruit (Actinidia deliciosa) is a relatively new disease of complex etiology. This disease has gained much attention in recent years because it has become widespread in kiwifruit-growing areas. Foliar symptoms of the disease are associated with toxins produced by pathogens and develop presumably in response to certain environmental and physiological factors. Vineyard surveys were conducted over nine years in a main Italian kiwifruit growing area, to investigate a potential correlation between soil sodicity and the occurrence of leaf symptoms. Greenhouse trials were also carried out to evaluate physiological parameters potentially associated with the symptoms, as transpiration rate, stomatal conductance, net photosynthesis, and water potential of potted vines grown with different levels of soil sodicity. In the vineyard trials, an increase in soil sodicity significantly augmented the incidence of wood decay of kiwifruit. Higher levels of soil sodicity correlated to increased transpiration and decreased water potential in plant tissues, which in turn can increase toxin translocation and accumulation in the leaves.

The ON and OFF pathways that emerge at the first synapse in the retina are generally thought to be streamed in parallel to higher visual areas, but recent work shows cross talk at the level of retinal ganglion cells. The ON pathway drives inhibitory inputs onto some OFF ganglion cells, such that these neurons show "push-pull" convergence of OFF-excitation and ONdisinhibition. In this study we measure the spatial receptive field of excitatory and inhibitory inputs to OFF-sustained (OFF-S) retinal ganglion cells of mouse, establish how contrast adaptation modulates excitatory and inhibitory synaptic inputs, and show the pharmacology of the inhibitory inputs. We find that the spatial tuning properties of excitatory and inhibitory inputs are sufficient to determine the spatial profile of the spike output and that high spatial acuity may be particularly reliant on disinhibitory circuits. Contrast adaptation reduced excitation to OFF-S ganglion cells, as expected, and also unmasked an asymmetry in inhibitory inputs: disinhibition at light-off was immune to contrast adaptation, but inhibition at light-on was substantially reduced. In pharmacological experiments we confirm that inhibitory inputs are partly mediated by glycine, but our measurements also suggest a substantial role for GABA. Our observations therefore reveal functional diversity in the inhibitory inputs to OFF ganglion cells and suggest that in addition to enhancing operational range these inputs help shape the spatial receptive fields of ganglion cells. Biostimulants are compounds that influence physiological processes in plants, producing better growth and enhancing stress tolerance. The effect of some biostimulants on vines was investigated over a number of years to assess their effect both on the incidence of esca leaf symptoms in the vineyard and on the growth of Phaeomoniella chlamydospora artificially inoculated into potted vines. Field trials were carried out for 4 -7 years in five 15 - 20-year-old vineyards infected with esca proper. Potted plants were sprayed with biostimulants, after which the vine trunks were inoculated with P. chlamydospora, and then the vines were sprayed again with biostimulants in the following 2 or 3 growing seasons. On the whole, biostimulants in the field did not reduce foliar symptoms. The percentage of symptomatic vines that had shown symptoms in previous years was higher in the biostimulant-sprayed plots. In the greenhouse, a certain reduction of internal necrosis caused by P. chlamydospora was seen with three of the four biostimulants tested. The mammalian retina is considered an autonomous circuit, yet work dating back to Ramon y Cajal indicates that it receives inputs from the brain. How such inputs affect retinal processing has remained unknown. We confirmed brain-to-retina projections of histaminergic neurons from the mouse hypothalamus. Histamine application ex vivo altered the activity of various retinal ganglion cells (RGCs), including direction-selective RGCs that gained responses to high motion velocities. These results were reproduced in vivo with optic tract recordings where histaminergic retinopetal axons were activated chemogenetically. Such changes could improve vision of fast-moving objects (e.g., while running), which fits with the known increased activity of histaminergic neurons during arousal. An antihistamine drug reduced optomotor responses to high-speed moving stimuli in freely moving mice. In humans, the same antihistamine nonuniformly modulated visual sensitivity across the visual field, indicating an evolutionary conserved function of the histaminergic system. BACKGROUND: Among patients with stable coronary disease and moderate or severe ischemia, whether clinical outcomes are better in those who receive an invasive intervention plus medical therapy than in those who receive medical therapy alone is uncertain. METHODS: We randomly assigned 5179 patients with moderate or severe ischemia to an initial invasive strategy (angiography and revascularization when feasible) and medical therapy or to an initial conservative strategy of medical therapy alone and angiography if medical therapy failed. The primary outcome was a composite of death from cardiovascular causes, myocardial infarction, or hospitalization for unstable angina, heart failure, or resuscitated cardiac arrest. A key secondary outcome was death from cardiovascular causes or myocardial infarction. RESULTS: Over a median of 3.2 years, 318 primary outcome events occurred in the invasive-strategy group and 352 occurred in the conservative-strategy group. At 6 months, the cumulative event rate was 5.3% in the invasive-strategy group and 3.4% in the conservative-strategy group (difference, 1.9 percentage points; 95% confidence interval [CI], 0.8 to 3.0); at 5 years, the cumulative event rate was 16.4% and 18.2%, respectively (difference, -1.8 percentage points; 95% CI, -4.7 to 1.0). Results were similar with respect to the key secondary outcome. The incidence of the primary outcome was sensitive to the definition of myocardial infarction; a secondary analysis yielded more procedural myocardial infarctions of uncertain clinical importance. There were 145 deaths in the invasive-strategy group and 144 deaths in the conservative-strategy group (hazard ratio, 1.05; 95% CI, 0.83 to 1.32). CONCLUSIONS: Among patients with stable coronary disease and moderate or severe ischemia, we did not find evidence that an initial invasive strategy, as compared with an initial conservative strategy, reduced the risk of ischemic cardiovascular events or death from any cause over a median of 3.2 years. The trial findings were sensitive to the definition of myocardial infarction that was used. The worldwide increase in the number and use of agrochemicals impacts nearby soil and freshwater ecosystems. Beyond the excess in applications and dosages, the inadequate management of remnants and the rinsing water of containers and application equipment worsen this problem, creating point sources of pollution. Advanced oxidation processes (AOPs) such as photocatalytic and photo-oxidation processes have been successfully applied in degrading organic pollutants. We developed a simple prototype to be used at farms for quickly degrading pesticides in water solutions by exploiting a UV–H2O2-mediated AOP. As representative compounds, we selected the insecticide imidacloprid, the herbicide terbuthylazine, and the fungicide azoxystrobin, all in their commercial formulation. The device efficiency was investigated through the disappearance of the parent molecule and the degree of mineralization. The toxicity of the pesticide solutions, before and during the treatment, was assessed by Vibrio fischeri and Pseudokirchneriella subcapitata inhibition assays. The results obtained have demonstrated a cost-effective, viable alternative for detoxifying the pesticide solutions before their disposal into the environment, even though the compounds, or their photoproducts, showed different sensitivities to physicochemical degradation. The bioassays revealed changes in the inhibitory effects on the organisms in agreement with the analytical data.

Oxidative stress and inflammation determine retinal ganglion cell degeneration, leading to retinal impairment and vision loss. Muller glial cells regulate retinal repair under injury, through gliosis. Meanwhile, reactive gliosis can turn in pathological effects, contributing to neurodegeneration. In the present study, we tested whether Cord Blood Serum (CBS), rich of growth factors, might improve the viability of Muller cells under in vitro damage. BDNF, NGF, TGF-alpha, GDNF and EGF levels were measured in CBS samples by Human Magnetic Luminex Assay. CBS effects were evaluated on rat (rMC-1) and human (MIO-M1) Muller cells, under H2O2 and IL-1 beta damage. Cells grown with FBS or CBS both at 5% were exposed to stress and analyzed in terms of cell viability, GFAP, IL-6 and TNF-alpha expression. CBS was also administrated after treatment with K252a, inhibitor of the neurotrophin receptor Trk. Cell viability of rMC-1 and MIO-M1 resulted significantly improved when pretreated with CBS and exposed to H2O2 and IL-1 beta, in comparison to the standard culture with FBS. Accordingly, the gliosis marker GFAP resulted down-regulated following CBS priming. In parallel, we observed a lower expression of the inflammatory mediators in rMC-1 (TNF-alpha) and MIO-M1 (IL-6, TNF- alpha), especially in presence of inflammatory damage. Trk inhibition through K252a administration impaired the effects of CBS under stress conditions on MIO-M1 and rMC-1 viability, not significantly different from FBS condition. CBS is enriched with neurotrophins and its administration to rMC-1 and MIO-M1 attenuates the cytotoxic effects of H2O2 and IL-1 beta. Moreover, the decrease of the main markers of gliosis and inflammation suggests a promising use of CBS for neuroprotection aims. This study is a preliminary basis that prompts future investigations to deeply explore and confirm the CBS potential.

The progressive degeneration of retinal photoreceptors is one of the most significant causes of blindness in humans. Conjugated polymers represent an attractive solution to the field of retinal prostheses, and a multi-layer fully organic prosthesis implanted subretinally in dystrophic Royal College of Surgeons (RCS) rats was able to rescue visual functions. As a step toward human translation, we report here the fabrication and in vivo testing of a similar device engineered to adapt to the human-like size of the eye of the domestic pig, an excellent animal paradigm to test therapeutic strategies for photoreceptors degeneration. The active conjugated polymers were layered onto two distinct passive substrates, namely electro-spun silk fibroin (ESF) and polyethylene terephthalate (PET). Naive pigs were implanted subretinally with the active device in one eye, while the contralateral eye was sham implanted with substrate only. Retinal morphology and functionality were assessed before and after surgery by means of in vivo optical coherence tomography and full-field electroretinogram (ff-ERG) analysis. After the sacrifice, the retina morphology and inflammatory markers were analyzed by immunohistochemistry of the excised retinas. Surprisingly, ESF-based prostheses caused a proliferative vitreoretinopathy with disappearance of the ff-ERG b-wave in the implanted eyes. In contrast, PET-based active devices did not evoke significant inflammatory responses. As expected, the subretinal implantation of both PET only and the PET-based prosthesis locally decreased the thickness of the outer nuclear layer due to local photoreceptor loss. However, while the implantation of the PET only substrate decreased the ff-ERG b-wave amplitude with respect to the pre-implant ERG, the eyes implanted with the active device fully preserved the ERG responses, indicating an active compensation of the surgery-induced photoreceptor loss. Our findings highlight the possibility of developing a new generation of conjugated polymer/PET-based prosthetic devices that are highly biocompatible and potentially suitable for subretinal implantation in patients suffering from degenerative blindness.

Purpose Human cord blood serum (CBS) is enriched with neurotrophins endowed of neuroprotective functions. Muller cells orchestrate retinal functioning, but their hyper-activation can exacerbate retinal damage under oxidative stress, which typically occurs during neurodegenerative diseases. In this study we tested whether CBS could regulate human Muller cell activation under H2O2- and IL-1?- induced damage. Methods Selected neurotrohpins (BDNF, TGF-?, NGF, GDNF, EGF) were analysed in CBS (Human Magnetic Luminex Screening Assay). The human Muller cell line Moorfields/Institute of Ophtalmology-Muller 1 (MIO-M1) was obtained from the UCL Institute of Ophtalmology, London, UK1. MIO-M1 were seeded in growth media enriched with standard FBS or CBS, both at 5%. After 24 h, MIO-M1 were exposed to H2O2 and IL-1? for 24 h. CBS effects were compared to FBS control cells in terms of cell survival (MTT assay), activation (GFAP mRNA) and inflammation (TNF-? and IL-6 mRNA). Results Median levels of neurotrophins in CBS were: BDNF 1.5 ng/ml, NGF 3.0 pg/ml, GDNF 1.5 pg/ml, TGF-? 36.3 pg/ml, EGF 820 pg/ml. MIO-M1 pre-cultured with CBS and exposed to H2O2, underwent a 50% decrease of GFAP mRNA in comparison to FBS control cells. CBS also mitigated the inflammatory activation, as shown by a significant 50% reduction of IL-6 and TNF-? mRNA levels. The CBS ability to modulate the inflammatory pathway was confirmed by the significant decrease of IL-6 and TNF-alpha genes (6- and 4- fold lower than FBS, respectively) in MIO-M1 exposed to IL-1beta (50 ng/ml). Conclusion CBS protects retinal Muller cells from damage under in vitro stress conditions. The main targets of this neuroprotecting stretegy are GFAP, marker of Muller cell activation, and the inflammatory cytokines, closely related to the neurodegenerative diseases. Among the fungi associated with the kiwi wood diseases, the vascular pathogen Phaeoacremonium minimum can infect plants already at nursery stage, without any external symptoms. At the moment, there are not effective control strategies. The effect of soil treatments applicable in organic agriculture was evaluated in two-years experiments on potted kiwi plants artificially inoculated with P. minimum. The soil treatments were based on commercial formulations of iron chelate, silicon, neem paste, Trichoderma afroharzianum strain T22, and cover cropping with selected perennial Poaceae. Cover cropping and iron chelate treatments enhanced the iron availability for the plants and significantly reduced wood necrosis caused by the pathogen. Both treatments also produced an increase of hairy root proliferation, so the plants were able to better cope with stress conditions. Laboratory assays showed the role of iron on the pathogen growth and its pathogenesis enzyme activities.

The mechanism underlying visual restoration in blind animal models of retinitis pigmentosa using a liquid retina prosthesis based on semiconductive polymeric nanoparticles is still being debated. Through the application of mathematical models and specific experiments, we developed a coherent understanding of abiotic/biotic coupling, capturing the essential mechanism of photostimulation responsible for nanoparticle-induced retina activation. Our modeling is based on the solution of drift-diffusion and Poisson-Nernst-Planck models in the multi-physics neuron-cleft-nanoparticle-extracellular space domain, accounting for the electro-chemical motion of all the relevant species following photoexcitation. Modeling was coupled with electron microscopy to estimate the size of the neuron-nanoparticle cleft and electrophysiology on retina explants acutely or chronically injected with nanoparticles. Overall, we present a consistent picture of electrostatic depolarization of the bipolar cell driven by the pseudo-capacitive charging of the nanoparticle. We demonstrate that the highly resistive cleft composition, due to filling by adhesion/extracellular matrix proteins, is a crucial ingredient for establishing functional electrostatic coupling. Additionally, we show that the photo-chemical generation of reactive oxygen species (ROS) becomes relevant only at very high light intensities, far exceeding the physiological ones, in agreement with the lack of phototoxicity shown in vivo .

- Abstract
High density multielectrode arrays (MEAs) based on CMOS technology (CMOS-MEAs) can simultaneously record extracellular spiking activity in neuronal cultures from 4096 closely spaced microelectrodes. This allows for a finer investigation of neuronal network activity compared to conventional MEAs with a few tens of electrodes. However, the sensing properties of these devices differ. To highlight this aspect, here we investigate and discuss the differences observed when quantifying spontaneous synchronized bursting events (SBEs) in datasets acquired with conventional MEAs and high-density MEAs from comparable hippocampal cultures. We found that datasets acquired with high-density MEAs exhibit collective dynamics similar to conventional arrays, but are characterized by a higher percentage of random spikes, i.e. spikes that are not part of a burst, most probably resulting from the larger recording capability. Additionally, the percentage of electrodes that record a burst is remarkably small on high-density MEAs compared to what can be observed on conventional MEAs and SBEs appear to be propagating in time across the electrode array, by involving shorter sequences of spikes per electrode. Overall, these results highlight a lower level of network synchronization involved in SBEs compared to what has been debated for several decades based on conventional MEA recordings from cell cultures.

High density multielectrode array recordings with CMOS-MEAs allow to monitor cell culture activity with unprecedent details respect to previous recording techniques. This is clarifying how network activity develops and is motivating the development of novel data analysis tools. Here, in order to advance in the exploitation of the richness of these large-scale array recordings, we introduce a principal component analysis approach that aims at improving on existing methodologies to describe neural activity events within large networks.

- Abstract
Early sensory experience shapes the functional and anatomical connectivity of neuronal networks. Light deprivation alters synaptic transmission and modifies light response properties in the visual system, from retinal circuits to higher visual centers. These effects are more pronounced during a critical period in juvenile life and are mostly reversed by restoring normal light conditions. Here we show that complete light deprivation, from birth to periods beyond the critical period, permanently modifies the receptive field properties of retinal ganglion cells. Visual deprivation reduced both the strength of light responses in ganglion cells and their receptive field size. Light deprivation produced an imbalance in the ratio of inhibitory to excitatory inputs, with a shift toward larger inhibitory conductances. Ganglion cell receptive fields in visually deprived animals showed a spatial mismatch of inhibitory and excitatory inputs and inhibitory inputs were highly scattered over the receptive field. These results indicate that visual experience early in life is critical for the refinement of retinal circuits and for appropriate signaling of the spatiotemporal properties of visual stimuli, thus influencing the response properties of neurons in higher visual centers and their processing of visual information.

- Abstract
Background: Physical training is believed the primary treatment of claudication symptoms. Although exercise therapy is self-effective, some drugs improving functional capacity have additive effects. TASC (Trans Atlantic Society Consensus) considers Propionil-L-Carnitine (PLC) and prostaglandin-E1 (PGE1) as poorly studied drugs with potential benefits in improving claudication. This retrospective, observational study was performed to compare the efficacy of PGE1 and PLC, and to evaluate both the immediate results of an intensive, short-course rehabilitation program and the outcome at one year follow-up., Methods: Twenty-five patients with severe-moderate claudication were selected. All patients were subjected to an intensive, supervised exercise program for 4 weeks in combination with either PGE1 (10 patients) or PLC (15 patients). Drugs were infused i.v. before every exercise session: 60 microg PGE1 within 2 hours and 600 mg PLC within 10 minutes. Patients were trained with the same supervised tread-mill walking-exercise program. At the end of the rehabilitation period, patients were instructed to keep walking (advised home exercise). Initial claudication distance (ICD) and absolute claudication distance (ACD) were evaluated during a constant treadmill test (3 km/hour speed, 10% grade) at entry, after 4 weeks and at one year follow-up. A patient was considered as no-responder if his/her improvement in ACD was <33%., Results: A significant increase of both ICD and ACD was detected after both 4 weeks (+269% and +135%, respectively, in PGE1 group; +245% and +125%, respectively, in PLC group) and one year (+364% and +202%, respectively, in PGE1 group; +279% and +176%, respectively, in PLC group). No-responder patients after the intensive training period (2 in PGE1 group and 4 in PLC group) remained no-responders also at one year follow-up. Both PGE1 and PLC treatments were well tolerated. No serious drug-related side effect requiring interruption of the treatment was observed., Conclusions: A short-course of intensive exercise treatment plus PLC or PGE1 may enhance walking ability. The result was confirmed at one year follow-up.
