Abstract

Unlike most other fungi, molds of the genus Trichoderma (Hypocreales, Ascomycota) are aggressive parasites of other fungi and efficient decomposers of plant biomass. Although nutritional shifts are common among hypocrealean fungi, there are no examples of such broad substrate versatility as that observed in Trichoderma. A phylogenomic analysis of 23 hypocrealean fungi (including nine Trichoderma spp. and the related Escovopsis weberi) revealed that the genus Trichoderma has evolved from an ancestor with limited cellulolytic capability that fed on either fungi or arthropods. The evolutionary analysis of Trichoderma genes encoding plant cell wall-degrading carbohydrate-active enzymes and auxiliary proteins (pcwdCAZome, 122 gene families) based on a gene tree / species tree reconciliation demonstrated that the formation of the genus was accompanied by an unprecedented extent of lateral gene transfer (LGT). Nearly one-half of the genes in Trichoderma pcwdCAZome (41%) were obtained via LGT from plant-associated filamentous fungi belonging to different classes of Ascomycota, while no LGT was observed from other potential donors. In addition to the ability to feed on unrelated fungi (such as Basidiomycota), we also showed that Trichoderma is capable of endoparasitism on a broad range of Ascomycota, including extant LGT donors. This phenomenon was not observed in E. weberi and rarely in other mycoparasitic hypocrealean fungi. Thus, our study suggests that LGT is linked to the ability of Trichoderma to parasitize taxonomically related fungi (up to adelphoparasitism in strict sense). This may have allowed primarily mycotrophic Trichoderma fungi to evolve into decomposers of plant biomass.

318. Effect of N2content in shielding gas on formation quality and microstructure of high nitrogen austenitic stainless steel fabricated by wire and arc additive manufacturing

Abstract

The composition of shielding gas exerts a crucial influence on high nitrogen austenitic stainless steel (HNASS) deposited by gas metal arc additive manufacturing. The proportion of N2in shielding gas greatly affected the formation quality and microstructure of deposited parts. As N2proportion improved, the formation quality became worse gradually and the nitrogen concentration of deposition metal increased. The maximum nitrogen concentration of deposition metal was more than 1%. However, N2porosity in the deposition metal became serious when the N2content was over 10 %. The microstructure of the deposition metal changed from two phases of austenite and ferrite in shielding gas of 0∼10 %N2to complete austenite with 15 %∼25 %N2content in shielding gas. With optimal shielding gas of 5%N2+2%O2+10%He+83%Ar, the deposited parts of HNASS demonstrated good formation and excellent properties. The average ultimate tensile strength of 920 MPa was achieved and the elongation was 27.7%.

319. Multi-scale simulation of grain growth during laser beam welding of nickel-based superalloy

Abstract

A multi-scale model integrating a macro-scale finite element model and micro-scale phase field models is proposed to simulate the grain growth in the molten pool of laser beam welding of the Inconel 718 alloy. The calculated welding profile using the macro-scale model agrees well with experimental measurements. According to the calculated macro-scale temperature field, the temperature gradient G, the solidification rate R in the molten pool are calculated. The micro-scale phase field models of macro-scale G and R are coupled are used to simulate the grain growth in the molten pool. Simulated columnar grains are dendritic, which gives agreements with experimental observations. The simulation results and experimental measurements show that the columnar grains spacing increases slightly from the fusion edge to the weld. A power-law function of λs∝(G × R) −0.433is found from the simulation results, which is similar to the power-law function of λs∝(G × R) −0.431from the experimental measurements. Simulated microstructural patterns of the competitive growth match experimental findings well. The spacing of favorable oriented grains is a bit larger than that of unfavorable oriented grains in the competitive growth.

320. A quality diagnosis method of GMAW based on improved empirical mode decomposition and extreme learning machine

Abstract

Due to the non-stationary and nonlinear characteristics of arc signal in gas metal arc welding (GMAW), results in the difference of frequency distribution. In this study, a method for evaluate weld quality based on complete ensemble empirical mode decomposition with adaptive noise (CEEMDAN) and extreme learning machine (ELM) is proposed. First, the current signal is decomposed into intrinsic mode functions (IMFs) of different frequency bands by CEEMDAN, and then the energy entropy of IMFs is extracted. Because of the energy of each IMF under different weld quality is varies, the energy entropy and normalized energy of IMFs are used as a feature vector to classify the weld quality combined with extreme learning machine (ELM). The result shows that CEEMDAN and ELM can be used to identify the weld quality types of GMAW accurately.

Abstract

The process of osteoblast switching to alternative mesenchymal phenotypes is incompletely understood. In this study, we tested the ability of the osteoblast/preosteocyte osteogenic cell line, MLO-A5, to also differentiate into either adipocytes or chondrocytes. MLO-A5 cells expressed a subset of skeletal stem cell markers, including Sca-1, CD44, CD73, CD146, and CD166. Confluent cultures of cells underwent differentiation within 3 days upon the addition of osteogenic medium. The same cultures were capable of undergoing adipogenic and chondrogenic differentiation under lineage-appropriate culture conditions, evidenced by lineage-specific gene expression analysis by real-time reverse-transcription-PCR, and by Oil Red O and alcian blue (pH 2.5) staining, respectively. Subcutaneous implantation of MLO-A5 cells in a gel foam into NOD SCID mice resulted in a woven bone-like structure containing embedded osteocytes and regions of cartilage-like tissue, which stained positive with both alcian blue (pH 2.5) and safranin O. Together, our findings show that MLO-A5 cells, despite being a strongly osteogenic cell line, exhibit characteristics of skeletal stem cells and display mesenchymal lineage plasticity in vitro and in vivo. These unique characteristics suggest that this cell line is a useful model with which to study aging and disease-related changes to the mesenchymal lineage composition of bone. Abstract

We investigated if time between injury and surgery affects cancellous bone properties in patients suffering tibial plateau fractures (TPF), in terms of structural integrity and gene expression controlling bone loss. A cohort of 29 TPF, operated 1–17 days post‐injury, had biopsies from the fracture and an equivalent contralateral limb site, at surgery. Samples were assessed using micro‐computed tomography and real‐time RT‐PCR analysis for the expression of genes known to be involved in bone remodeling and fracture healing. Significant decreases in the injured vs control side were observed for bone volume fraction (BV/TV, −13.5 ± 6.0%, p = 0.011), trabecular number (Tb.N, −10.5 ± 5.9%, p = 0.041) and trabecular thickness (Tb.Th, −4.6 ± 2.5%, p = 0.033). Changes in these parameters were more evident in patients operated 5–17 days post‐injury, compared to those operated in the first 4 days post‐injury. A significant negative association was found between Tb.Th (r = −0.54, p < 0.01) and BV/TV (r = −0.39, p < 0.05) in relation to time post‐injury in the injured limb. Both BV/TV and Tb.Th were negatively associated with expression of key molecular markers of bone resorption, CTSK, ACP5, and the ratio of RANKL:OPG mRNA. These structure/gene expression relationships did not exist in the contralateral tibial plateau of these patients. This study demonstrated that there is a significant early time‐dependent bone loss in the proximal tibia after TPF. This bone loss was significantly associated with altered expression of genes typically involved in the process of osteoclastic bone resorption but possibly also bone resorption by osteocytes. 342. Wind tunnel study on the wake bubble of model trucks

Abstract

Heavy traffic volume makes tailgating a common picture on the road today. Wake interference, particularly in the scenario when a relatively small sedan drives into the wake of a large truck, may raise some serious highway safety concerns. In this study, the characteristics of separation bubble in the wake of model trucks with various degrees of model details are studied. The objective is to uncover the impact of truck model details on the characteristics of the wake bubble. Furthermore, how the spacing between the leading sedan and truck affects the bubble is also investigated. The hotwire measurements as well as flow visualization using a tuft grid have been conducted in a closed-loop wind tunnel at Re=2–7×105. The wind tunnel results revealed that both the degree of model details and the spacing between sedan and truck have significant impact on the three dimensional structure of the wake bubble.

Abstract

The surface temperature of the thin-walled parts deposited by gas metal arc welding (GMAW)- and double-electrode gas meal arc welding (DE-GMAW)-based additive manufacturing (AM) techniques was captured through the infrared thermography technique. The dimensions of the molten pool in the deposition process including length, depth, and width were reduced in different degrees by the bypass arc during the DE-GMAW-based AM. The volume of the molten pool can decrease by about 30% in DE-GMAW under the same deposition rate. The volume of the high-temperature metal for the deposited parts during DE-GMAW-based AM was smaller than that during GMAW-based AM when depositing the high layer. The mean temperature of the deposited parts after the cooling process in DE-GMAW-based AM was lower under the same condition. The bypass arc in DE-GMAW-based AM is conducive to the reduction of the heat accumulation in the deposited thin-walled parts without a corresponding decrease in the deposition rate.
