307. miR-106b-5p Inhibits IRF1/IFN-β Signaling to Promote M2 Macrophage Polarization of Glioblastoma

Abstract

Purpose The microRNA (miRNA) profile changes in the tumor-associated macrophages. However, the role of miR-106b-5p in the glioblastoma-associated macrophages is poorly understood. Materials and Methods In our study, miR-106b-5p and M2 macrophage markers were detected by qRT-PCR and Western blotting in THP1 cells, with the conditioned medium from U251 cells or M2 macrophages in response to IL-4 stimulation and M1 macrophages stimulated by LPS and IFN-γ. IFN regulatory factor (IRF1) was identified as a target of miR-106b-5p in the glioma infiltrating macrophages by luciferase reporter assay. The molecular mechanisms involved in the miR-106b-5p-mediated regulation of M2 polarization were clarified by shRNA knockdown assay. Results Our results showed miR-106b-5p expression was upregulated in glioma-infiltrating macrophages. miR-106b-5p regulated M2 polarization of glioma infiltrating macrophages and enhanced the growth of glioma-infiltrating macrophages. IRF1 was identified as a target of miR-106b-5p. Furthermore, miR-106b-5p inhibited IRF1 expression by targeting IRF1/IFN-β pathway to promote M2 polarization of macrophages. Conclusion miR-106b-5p may inhibit IRF1/IFN-β signaling to promote M2 macrophage polarization of glioblastoma, and it may become a novel target for the treatment of glioblastoma.

Circular (circ) RNA is a member of the non-coding RNA family. However, clinical references to circRNAs in MCL are not clear.Methods: In this study, we detected the expression level of circCDYL in the plasma of MCL patients compared to healthy donors by the quantitative reverse transcription polymerase chain reaction. The diagnostic value of circCDYL was determined using a receiver operating characteristic (ROC) curve. We constructed a circCDYL short hairpin RNA plasmid and infected the MCL cell line, Z138, to detect its effect on cell proliferation.Results: CircCDYL was high expressed in the plasma of MCL patients. The ROC curve showed that circCDYL had diagnostic value (area under the curve (AUC) = 0.856). Functionally, circCDYL knockdown inhibited MCL cell proliferation. We conducted bioinformatics analyses and identified a circCDYL-micro (mi)RNA–mRNA/long non-coding (lnc)RNA network, highlighted by five miRNAs (hsa-miR-129-5p, hsa-miR-3163, hsa-miR-4662a-5p, hsa-miR-101-3p, and hsa-miR-186-5p), three lncRNAs (MALAT1, NEAT1, and XIST), and five mRNAs (NOTCH1, FMR1, ABCB1, TWIST1, and VEGFA).Conclusion: These findings indicate that circ-CDYL might serve as a potential diagnostic biomarker in clinical practice.Keywords: circular RNA, biomarker, AUC, diagnosis, non-coding RNA &nbsp

309. Temporal-attentive Covariance Pooling Networks for Video Recognition

Abstract

For video recognition task, a global representation summarizing the whole contents of the video snippets plays an important role for the final performance. However, existing video architectures usually generate it by using a simple, global average pooling (GAP) method, which has limited ability to capture complex dynamics of videos. For image recognition task, there exist evidences showing that covariance pooling has stronger representation ability than GAP. Unfortunately, such plain covariance pooling used in image recognition is an orderless representative, which cannot model spatio-temporal structure inherent in videos. Therefore, this paper proposes a Temporal-attentive Covariance Pooling(TCP), inserted at the end of deep architectures, to produce powerful video representations. Specifically, our TCP first develops a temporal attention module to adaptively calibrate spatio-temporal features for the succeeding covariance pooling, approximatively producing attentive covariance representations. Then, a temporal covariance pooling performs temporal pooling of the attentive covariance representations to characterize both intra-frame correlations and inter-frame cross-correlations of the calibrated features. As such, the proposed TCP can capture complex temporal dynamics. Finally, a fast matrix power normalization is introduced to exploit geometry of covariance representations. Note that our TCP is model-agnostic and can be flexibly integrated into any video architectures, resulting in TCPNet for effective video recognition. The extensive experiments on six benchmarks (e.g., Kinetics, Something-Something V1 and Charades) using various video architectures show our TCPNet is clearly superior to its counterparts, while having strong generalization ability. 310. A comparative study of Mg-Gd-Y-Zr alloy cast by metal mould and sand mould

Abstract

The differences of the microstructure and mechanical property between metal mould and sand mould cast Mg-10Gd-3Y-Zr alloy were investigated both under as-cast condition and after solution heat treatment. In the as-cast specimens, the microstructure is similar and composed of α-Mg solid solution and eutectic compound of α-Mg+ Mg24(Gd,Y)5; whereas the grain size using metal mould and sand mould is 27 μm and 71 μm, respectively.The eutectic compound of metal mould cast alloy was completely dissolved after solution treated at 500℃ for 8 h, however it needs higher temperature (525 ℃) and longer time (12 h) to achieve the absolute dissolving under sand mould condition. In contrast to metal mould, the peak time of sand mould alloy aged at 225 ℃ and 250 ℃ of was advanced by 4 h and 6 h, respectively. The precipitation reaction sequence in sand mould cast Mg-10Gd-3Y-Zr alloy during isothermal ageing at 250 ℃ follows S.S.S.S.→β″(D019)→β′(cboc)→β1(fcc)→β(fcc), which is similar to that in the alloy cast using metal mould.

Abstract

Summary: CO2 reduction to produce value‐added chemicals/fuels is generally considered as an effective way to solve the energy crisis and global warming. However, the greatest challenge to the technology is the lack of high‐performance catalysts as this process is quite sluggish under mild conditions. Herein, we have carefully prepared a series of bimetallic PbxCdy nanoparticles (NPs) as well as their monometallic counterparts via a facile chemical reduction. Electrocatalytic performances of obtained catalysts were evaluated with respect to chemical composition, electrolytic potential, and so on. All of them exhibit high performance and achieve high faradaic efficiency for HCOOH production (FEHCOOH) over a voltage window of −1.16 to −1.26 V vs the reversible hydrogen electrode (VRHE). Among them, Pb3Cd1 achieved a maximum FEHCOOH of 96% at −1.16 VRHE and a desirable stability over 12 hours of continuous electrolysis. The excellent performance of Pb3Cd1 can be attributed to the huge specific area of NPs and the synergistic effect of the components. This study shows that efficient bimetallic PbxCdy electrocatalysts have a large application potential for highly selective HCOOH production from CO2. 338. Preparation of Pinocembrin-Loaded F127/MPEG-PDLLA Polymer Micelles and Anti-Osteoporotic Activity.

Abstract

Pinocembrin (PCB) is 5,7-dihydroxyl flavanone and has multiple pharmacological activities, namely, anti-inflammation, anti-osteoporotic, and so on. However, low water solubility and bioavailability have hindered its application. Herein, we aimed to increase its bioavailability through preparation of F127/MPEG-PDLLA polymer micelles (PCB-M). We characterized the micelles through appropriate attributes such as analysis of particle size (PS), polydispersity (PDI), transmission electron microscopic (TEM) image, stability test, and evaluation of in vitro release of drug. After physical characterization, the respective PS, PDI, and entrapment efficiency (EE) of PCB-M were estimated to be 27.63 ± 0.17 nm, 0.055 ± 0.02, and 90.53 ± 0.01%. Fluorescence probe method was employed to measure critical micelle concentration (CMC) of PCB-M, we observed CMC was low, thereby suggesting that PCB-M had good stability. In vitro release analysis indicated that the rate of cumulative PCB release from PCB-M was greater than 90% in each medium compared with free PCB, which was less than 40%, thus pointing to a significantly improved solubility of PCB. In vivo pharmacokinetic results showed that oral biological availability of PCB-M increased 5.3 folds comparable to free PCB. The effects of PCB on osteoblasts and ALP activities were investigated; subsequently, zebrafish osteoporotic model was established with prednisolone to study the anti-osteoporotic effects of PCB and PCB-M. The results showed that PCB improved osteoporosis with PCB-M being more effective than free PCB. Finally, PCB-M can be used as a promising method to improve the solubility of PCB, while the bioavailability and anti-osteoporotic effect of PCB could be improved, thus laying a foundation for clinical use in the future. Abstract

The electron collection efficiency (CE) of the photomultiplier tube based on microchannel plates (MCP-PMT) is limited by the MCP open area fraction. Coating MCP with a high secondary yield material is supposed to be an effective approach to improve CE. Both typical and coated MCP-PMTs are developed. A relative measurement method is proposed to characterize the collection efficiency performance. Abstract

As a neuronal transmembrane protein, leucine‐rich repeat and fibronectin type‐III domain‐containing protein 2 (LRFN2) can recruit and combine with N‐methyl‐d‐aspartate receptors (NMDARs) to promote nerve growth. Genetic studies suggest that mutations in LRFN2 are associated with various cancers. However, the role and mechanism of LRFN2 in the progression of ESCC have not been elucidated. In this study, we demonstrated that LRFN2 was significantly downregulated in ESCC tissues by qRT‐PCR and immunohistochemistry. Low LRFN2 expression was an adverse prognostic factor in patients with ESCC. Overexpression of LRFN2 effectively suppressed the proliferation, migration, invasion, and epithelial‐to‐mesenchymal transition in vitro and tumor growth in vivo. Bioinformatics analysis indicated that Wnt/β‐catenin signaling regulation was one of the most potential mechanisms and studies confirmed that overexpression of LFRN2 obviously downregulated the expression of β‐catenin, c‐Myc, and cyclin D1 in ESCC cells and tumor tissues. Further studies revealed that LRFN2 plays an anti‐ESCC role by binding with NMDAR‐GRIN2B and this effect can be weakened by NR2B‐selective NMDA antagonist‐NMDA‐IN‐1. Moreover, the bioinformatics analysis showed that the interaction of GRIN2B and GSK3β affects the NF‐κB pathway, which was demonstrated by western blot experiments. Collectively, our results indicate that LRFN2 binding to NMDARs inhibits the progression of ESCC by regulating the Wnt/β‐catenin and NF‐κB pathway, which provides a new therapeutic target for improving the prognosis of patients with ESCC. Abstract

Transformer models are not only successful in natural language processing (NLP) but also demonstrate high potential in computer vision (CV). Despite great advance, most of works only focus on improvement of architectures but pay little attention to the classification head. For years transformer models base exclusively on classification token to construct the final classifier, without explicitly harnessing high-level word tokens. In this paper, we propose a novel transformer model called second-order transformer (SoT), exploiting simultaneously the classification token and word tokens for the classifier. Specifically, we empirically disclose that high-level word tokens contain rich information, which per se are very competent with the classifier and moreover, are complementary to the classification token. To effectively harness such rich information, we propose multi-headed global cross-covariance pooling with singular value power normalization, which shares similar philosophy and thus is compatible with the transformer block, better than commonly used pooling methods. Then, we study comprehensively how to explicitly combine word tokens with classification token for building the final classification head. For CV tasks, our SoT significantly improves state-of-the-art vision transformers on challenging benchmarks including ImageNet and ImageNet-A. For NLP tasks, through fine-tuning based on pretrained language transformers including GPT and BERT, our SoT greatly boosts the performance on widely used tasks such as CoLA and RTE.
