333. The role of autophagy in prostate cancer and prostatic diseases: a new therapeutic strategy

Abstract

Background: Autophagy is a well-conserved catabolic process that plays a key role in cell homeostasis. In the prostate, defective autophagy has been implicated in the genesis and progression of several pathological conditions. Aim: The present review explored the autophagy pathway in prostate-related dysfunctions, focusing on prostate cancer (PCa), benign prostatic hyperplasia (BPH) and prostatitis. Results: Impaired autophagy activity has been shown in animal models of BPH and prostatitis. Moreover, autophagy activation by specific and non-specific drugs improved both conditions in pre-clinical studies. Conversely, the efficacy of autophagy inducers in PCa remains controversial, depending on intrinsic PCa characteristics and stage of progression. Intriguingly, autophagy inhibitors have shown beneficial effects in PCa suppression or even to overcome chemotherapy resistance. However, there are still open questions regarding the upstream mechanisms by which autophagy is deregulated in the prostate and the exact role of autophagy in PCa. The lack of specificity and increased toxicity associated with the currently autophagy inhibitors limits its use clinically, reflecting in reduced number of clinical data. Conclusion: New therapeutic strategies to treat prostatic diseases involving new autophagy modulators, combination therapy and new drug formulations should be explored. Understanding the autophagy signaling in each prostatic disease is crucial to determine the best pharmacological approach.

Abstract

We present a fabrication scheme and testing results for epitaxial sub-micrometer Josephson junctions. The junctions are made using a high-temperature (1170 K) "via process" yielding junctions as small as 0.8 mu m in diameter by use of optical lithography. Sapphire (Al2O3) tunnel-barriers are grown on an epitaxial Re/Ti multilayer base-electrode. We have fabricated devices with both Re and Al top electrodes. While room-temperature (295 K) resistance versus area data are favorable for both types of top electrodes, the low-temperature (50 mK) data show that junctions with the Al top electrode have a much higher subgap resistance. The microwave loss properties of the junctions have been measured by use of superconducting Josephson junction qubits. The results show that high subgap resistance correlates to improved qubit performance.

Abstract

We consider a boundary value problem for the system of equations describing the stationary motion of a viscous nonhomogeneous asymmetric fluid in a bounded planar domain having a $C^2$ boundary. We use a stream-function formulation after the manner of N. N. Frolov [Math. Notes, \textbf{53}(5-6), 650--656, 1993] in which the fluid density depends on the stream-function by means of another function determined by the boundary conditions. This allows for dropping some of the equations, most notably the continuity equation. Abstract

We present a cosmological model with a solution that self-accelerates at late-times without signs of ghost instabilities on small scales. The model is a natural extension of the Brans-Dicke (BD) theory including a non-linear derivative interaction, which appears in a theory with the Galilean shift symmetry. The existence of the self-accelerating universe requires a negative BD parameter but, thanks to the non-linear term, small fluctuations around the solution are stable on small scales. General relativity is recovered at early times and on small scales by this non-linear interaction via the Vainshtein mechanism. At late time, gravity is strongly modified and the background cosmology shows a phantom-like behaviour and the growth rate of structure formation is enhanced. Thus this model leaves distinct signatures in cosmological observations and it can be distinguished from standard $\Lambda$CDM cosmology., Comment: references added, accepted for publication in PRD, 5 pages, 4 figures

Abstract

We study the spectrum of gravitational perturbations around a vacuum de Sitter brane in a 5D asymmetric braneworld model, with induced curvature on the brane. This generalises the stealth acceleration model proposed by Charmousis, Gregory and Padilla (CGP) which realises the Cardassian cosmology in which power law cosmic acceleration can be driven by ordinary matter. Whenever the bulk has infinite volume we find that there is always a perturbative ghost propagating on the de Sitter brane, in contrast to the Minkowski brane case analysed by CGP. We discuss the implication of this ghost for the stealth acceleration model, and identify a limiting case where the ghost decouples as the de Sitter curvature vanishes., Comment: 21 pages

Abstract

Bats play an essential role in maintaining ecosystems. Their unique characteristics increase the likelihood of interactions with various species, making them a potential source for the emergence and spread of infectious diseases. Hantaviruses are continuously expanding their range of hosts. This study presents the identification of a partial genome associated with Hantavirus in samples collected from neotropical bats. We conducted a metagenomic study using samples from Carollia perspicillata in Maranhão, Brazil. Tissue fragments were used for RNA extraction and subsequent sequencing. The resulting data was subjected to bioinformatic analysis. A sequence showing an identity of 72.86% with the L gene in the reference genome was obtained. The phylogenetic analysis revealed the study sequence, denoted as Buritiense, clustering within the Mobatvirus clade. The intragroup analysis showed a broader dispersion and were markedly asymmetric. This observation suggests the possibility that Buritiense could potentially represent a new species within the bat-borne hantaviruses, but further analyses are needed to provide additional insights if bats plays a role as reservoirs and the potential for transmission to human populations. Abstract

Priapism, defined as a prolonged and often painful penile erection occurring without sexual stimulation or desire, is a common complication in sickle cell disease (SCD), affecting up to 48% of male patients. This condition presents significant clinical challenges and can lead to erectile dysfunction if not properly managed. Current pharmacological treatments for SCD-related priapism are primarily reactive rather than preventative, highlighting a gap in effective medical intervention strategies. A critical factor in developing priapism is the reduced basal bioavailability of nitric oxide (NO) and cyclic guanosine monophosphate (cGMP) in erectile tissues. New prevention strategies should ideally target the underlying pathophysiology of the disease. Compounds that stimulate and activate soluble guanylate cyclase (sGC) emerge as potential therapeutic candidates since these compounds have the property of inducing cGMP production by sGC. This review explores the potential of sGC stimulators and activators in treating priapism associated with SCD. We discuss the advantages of these agents in the face of the challenging pathophysiology of SCD. Additionally, the review underscores the impact of intravascular hemolysis and oxidative stress on priapism pathophysiology in SCD, areas in which sGC stimulators and activators may also have beneficial therapeutic effects. Abstract

Recently, approach-avoidance tendencies and visual perception biases have been increasingly studied using bistable point-light walkers (PLWs). Prior studies have found a facing-the-viewer bias when one is primed with general threat stimuli (e.g. angry faces), explained by the "error management theory", as failing to detect a threat as approaching is riskier than the opposite. Importantly, no study has explored how disease threat – linked to the behavioural immune system – might affect this bias. This study aimed to explore whether disease-signalling cues can alter how we perceive the motion direction of ambiguous PLWs. Throughout 3 experiments, participants indicated the motion direction of a bistable PLW previously primed with a control or disease-signalling stimuli – that is, face with a surgical mask (Experiment 1), sickness sound (Experiment 2), or face with a disease cue (Experiment 3). Results showed that sickness cues do not significantly modulate the perception of approach-avoidance behaviours. However, a pattern emerged in Experiments 2 and 3, suggesting that sickness stimuli led to more facing away percepts. Unlike other types of threat, this implies that disease-related threat stimuli might trigger a distinct perceptual bias, indicating a preference to avoid a possible infection source. Abstract

Background: The involvement of the KMT2 methyltransferase family in the pathogenesis of head and neck squamous cell carcinoma (HNSCC) remains elusive. Method: This study adhered to the PRISMA guidelines, employing a search strategy in the LIVIVO, PubMed, Scopus, Embase, Web of Science, and Google Scholar databases. The methodological quality of the studies was assessed by the Joanna Briggs Institute. Results: A total of 33 studies involving 4294 individuals with HNSCC were included in this review. The most important alteration was the high mutational frequency in the KMT2C and KMT2D genes, with reported co‐occurrence. The expression of the KMT2D gene exhibited considerable heterogeneity across the studies, while limited data was available for the remaining genes. Conclusions: KMT2C and KMT2D genes seem to have tumor suppressor activities, with involvement of cell cycle inhibitors, regulating different pathways that can lead to tumor progression, disease aggressiveness, and DNA damage accumulation. Abstract

Santa Catarina is an important tomato‐producing state in Brazil, but there is little data regarding the prevalence and characterization of viruses and insect vectors in tomato plants in this state. This study surveyed viruses and their vectors in commercial tomato‐producing areas, as well as alternative hosts and weeds in the surrounding areas. Sixty‐five samples of young leaves were collected between 2019 and 2023, and viruses were detected with generic or specific PCR primers targeting several virus genera. In addition, 397 virus vector insect samples were collected and identified. The orthotospoviruses tomato chlorotic spot virus (TCSV) and groundnut ringspot virus (GRSV) were the most prevalent and distributed in several regions, whilst the crinivirus tomato chlorosis virus (ToCV) and begomovirus tomato severe rugose virus (ToSRV) were restricted to the metropolitan region of Florianópolis. This is the first report of ToSRV in Solanum americanum in Brazil and of TCSV in zucchini and cucumbers worldwide. A survey of insect vectors found whiteflies, Mediterranean cryptic species of Bemisia tabaci (MED) and Trialeurodes vaporariorum were predominant, while Frankliniella occidentalis and F. schultzei thrips were often found cohabiting in the sampled crops. These results highlight the need to adapt management strategies in Santa Catarina, especially in the Santo Amaro da Imperatriz region, where a greater frequency and diversity of viral species and vectors were found. In addition, both the thrips and whitefly species (especially MED) found are a cause for concern, as they readily transmit viruses that infest crops in the open fields of Santa Catarina. Abstract

Human bacterial infections significantly contribute to the increase in healthcare-related burdens. This scenario drives the study of novel techniques for the early and precise diagnosis of infectious processes. Some alternatives include Nuclear Medicine- and Molecular Imaging-based strategies. However, radiopharmaceuticals that are available for routine assessments are not specific to differentiating infectious from aseptic inflammatory processes. In this context, [68Ga]Ga-DOTA-Ubiquicidin29-41 was synthesized using an automated module and radiochemical; in vivo and in vitro studies were performed. The radiopharmaceutical remained stable in saline (up to 180 min) and in rodent serum (up to 120 min) with radiochemical purities > 99 and 95%, respectively. Partition coefficient and serum protein binding at 60 min were determined (−3.63 ± 0.17 and 44.06 ± 1.88%, respectively). Ex vivo biodistribution, as well as in vivo microPET/CT images in mice, showed rapid blood clearance with renal excretion and reduced uptake in other organs in Staphylococcus aureus-infected animals. Higher uptake was observed in the target as compared to the non-target tissue (p < 0.0001) at 60 min post administration. The presented in-human clinical case demonstrates uptake of the radiopharmaceutical by Staphyloccocus aureus bacteria. These results indicate the potential of [68Ga]Ga-DOTA-Ubiquicidin29-41 as a radiopharmaceutical that can be obtained in a hospital radiopharmacy for the diagnosis of infectious processes using PET/CT. Abstract

The cultivation of açai palm, which yields tasteful and nutritional fruits, has been stimulated by the high commercial values of açai pulps; however, the occurrence of anthracnose (a disease caused by the fungus Colletotrichum gloeosporioides) on açaí nursery and orchards has been a challenge for the shift from açaí extractivism to its cultivation. The interaction among endophytic fungi and host can change the plant disease severity, facilitating infection by phytopathogens and increasing its damage to plants. Therefore, this study aimed to investigate the antagonistic activity of five endophytic fungi against C. gloeosporioides, as well as to evaluate the anthracnose severity in açaí seedlings in response to the inoculation of the most promising of the tested endophytic fungus. First, in vitro evaluation showed that most of the fungi were able to grow over C. gloeosporioides mycelia. The endophytic fungi Graphium sp. 2.4765 and Hypoxylon anthochroum 2.4996 recorded the highest rates of growth inhibition, 79.3% and 77.0%, respectively. Because H. anthochroum 2.4996 presented better in vitro growth and spore production, it was chosen for subsequent evaluations. Second, in planta evaluation showed that the inoculation of H. anthochroum 2.4996 in diseased plants enhanced both the necrotic area in leaflets and disease symptoms. Third, comparative analyses based on the cultural, micromorphological and molecular characteristics have shown that such strain is related to H. anthochroum.
