Background: Human blood contains a big variety of natural antibodies, circulating throughout life at constant concentration. Previously, we have found natural antibodies capable of binding to trisaccharide Galα1-4Galβ1-4Glc (P k ) practically in all humans. Intriguingly, the same trisaccharide is a key fragment of glycosphingolipid globotriaosylceramide (Gb3Cer) - normal component of erythrocyte and endothelial cell membrane, i.e. the antibodies and their cognate antigen coexist without any immunological reaction., Aim: To explain the inertness of human anti-P k antibodies towards own cells., Materials and Methods: We used a combination of immunochemical and molecular dynamics (MD) experiments. Antibodies were isolated using affinity media with P k trisaccharide, their epitope specificity was characterized using ELISA (enzyme-linked immunosorbent assay) with a set of synthetic glycans related to P k synthetic glycans and FACS (Fluorescence-Activated Cell Sorting) analysis of cells with inserted natural Gb3Cer and its synthetic analogue. Conformations and clustering of glycolipids immersed into a lipid bilayer were studied using MD simulations., Results: Isolated specific antibodies were completely unable to bind natural Gb3Cer both inserted into cells and in artificial membrane, whereas strong interaction took place with synthetic analogue differing by the presence of a spacer between trisaccharide and lipid part. MD simulations revealed: i) although membrane-bound glycans do not form stable long-living aggregates, their transient packing is more compact in natural Gb3 as compared with the synthetic analog, ii) similar conformation of P k glycan in composition of the glycolipids, iii) no effect on the mentioned above results when cholesterol was inserted into membrane, and iv) better accessibility of the synthetic version for interaction with proteins., Conclusions: Both immunochemical and molecular dynamics data argue that the reason of the "tolerance" of natural anti-P k antibodies towards cell-bound Gb3Cer is the spatial inaccessibility of P k glycotope for interaction. Directed transport of biological species across the surface of a substrate is essential for realizing lab-on-chip technologies. Approaches that utilize localized magnetic fields to manipulate magnetic particles carrying biological entities are attractive owing to their sensitivity, selectivity, and minimally disruptive impact on biomaterials. Magnetic domain walls in magnetic tracks produce strong localized fields and can be used to capture, transport, and detect individual superparamagnetic microbeads. The dynamics of magnetic microbead transport by domain walls has been well studied. However, demonstration of more complex functions such as selective motion and sorting using continuously driven domain walls in contiguous magnetic tracks is lacking. Here, a junction architecture is introduced that allows for branching networks in which superparamagnetic microbeads can be routed along dynamically-selected paths by a combination of rotating in-plane field for translation, and a pulsed out-of-plane field for path selection. Moreover, experiments and modeling show that the select-field amplitude is bead-size dependent, which allows for digital sorting of multiple bead populations using automated field sequences. This work provides a simple means to implement complex routing networks and selective transport functionalities in chip-based devices using magnetic domain wall conduits.

Background: Profiling of donor's antibodies using glycan arrays demonstrated presence of antibodies capable of binding to >100 mammalian glycans or their fragments. For example, relatively high binding to Galα1-4Galβ1-4GlcNAc (P(1)), Galα1-4Galβ1-4Glc (P(k)), Galβ1-3GlcNAc (Le(c)), 4-O-SuGalβ1-4GlcNAc, and GalNAcα1-3GalNAc (Fs) was found in all tested individuals. Affinity isolation using hapten-specific chromatography in combination with epitope mapping revealed their glycotopes. Notably, a significant part of the antibodies was capable of recognizing a fragment of larger glycans, for example, -Galβ1-4Glc of glycolipids, or Fucα1-3GlcNAc motif of Le(X)/Le(Y) antigens. Their epitope specificity did not vary between different healthy individuals. Nominally, all the mentioned immunoglobulins could be classified as auto-antibodies., Methods: In this work we re-evaluated results published earlier and analyzed new data to address the question why autologous antibodies found in healthy individuals do not cause severe auto-immune reactions., Results: In all cases the presumably "auto" antibodies were found to bind short fragments "subtracted" from larger glycans whereas recognition of the same fragment in the context of the whole natural chain was completely abolished. Thus, in spite of numerous formally positive signals observed on the printed glycan array, we are yet unable to identify in blood serum of healthy individuals true auto-antibodies capable of binding carbohydrate chains in their naturally occurring form., General Significance: The identified natural anti-glycan antibodies were found to be specific, high-titer and population conservative immunoglobulins - all of this suggesting as yet unknown biological role(s) of the studied proteins. Polyacrylamide glycoconjugates, Glyc-PAA, having various tags or labels are convenient tools for analysis of cellular lectins. Adaptation of such glycoprobes for flow cytometry allows us to reveal lectins expressed on cell surface and analyze their carbohydrate specificity as well as functionality. Localization of lectins is visualized by labeling of cells with fluorescein-tagged glycoprobes, Glyc-PAA-fluo, in combination with fluorescent microscopy techniques. Additionally, biotinylated glycoprobes can be immobilized on magnetic particles making it possible to separate a cell population according to its carbohydrate-binding profile. Here, we exemplify application of glycoprobes in the study of cellular siglecs and galectins, as well as lectin patterning of tumor cells. The specificity of sialic acid binding membrane-anchored lectins, siglecs-1, -5, -7, -8 and -9 was determined using this methodology. To study the carbohydrate-binding profile of soluble galactoside-binding lectins, galectins-1 or -3, these were loaded on (initially galectin free) Raji cells and probed using Glyc-PAA-fluo. Lessons learned from this model system allowed us to study the galectin distribution pattern of tumors: cells obtained from mice carrying mammary adenocarcinoma or lymphoma were probed with Glyc-PAA-fluo using flow cytometry. Disaccharide 6OSuLacdiNAc was shown to be the most potent probe for adenocarcinoma cells, demonstrating that 6OSuLacdiNAc-binding molecules accumulate on cell surface in a patch-wise distribution., (Copyright 2009 Elsevier GmbH. Objective: To assess the impact of a structured intervention, the "primary intervention for memory structuring and meaning acquisition" (PIMSMA) performed randomly in the emergency department with survivors of suicide bombing attacks, on their medium-term mental health outcome., Design: Follow up and assessment 3-9 months postinjury, and 24 months thereafter., Setting: A tertiary referral general hospital in Tel Aviv, Israel., Participants: Injured survivors of 9 suicide bombing and suicide shooting, men and women aged 16-72 at the time of the incident., Main Outcome Measures: Diagnosis of posttraumatic stress disorder (PTSD) was made using the Hebrew validated version of the DSM-IV SCID-PTSD rating scale. Other psychiatric symptoms were assessed using the following rating scales: impact of event scale (IES), Hamilton rating scale for depression (HAM-D) and for anxiety (HAM-A), and the Pittsburgh sleep quality index (PSQI). Effects of PIMSMA and PTSD level of psychological distress were analyzed using ANOVA and for change over time for continuous variables repeated measured multivariate analyses was performed, and for categorical variables nonparametric-related sample McNemar. Logistic regression for variable associated with PTSD was performed., Results: Out of 213 eligible injured survivors evacuated to our ER, 129 were retrieved 3-9 months after the incident, and 53 were available for assessment 2 years later. Multivariate analyses for being PTSD vs non-PTSD at the first evaluation, being hospitalized OR = 5.6 (95 percent CI 1.1-27.6) and treated OR = 24.5 (95 percent CI 2.8-200) were the only predictors, with no effect (p = 0.9) for PIMSMA vs other supportive intervention. Predictor for PTSD at the second evaluation were IES severity score at first evaluation OR = 1.1 (95 percent CI 1.04-1.2)., Conclusion: The PIMSMA approach is as good as the nonspecific supportive treatment performed routinely in the ED with all survivors of traumatic events of any origin. Further studies are needed to establish valid, evidence-based treatment approaches for the acute aftermath of exposure to severe potentially traumatic events.

A specific apoptotic glycosylation pattern may play an assistant or even a causative role in phagocytosis of apoptotic bodies. To elucidate the role of macrophages in lectin-mediated phagocytosis, an experimental system was used, where monocyte-derived THP-1 cells engulf the apoptotic bodies from the melanoma cell line MELJUSO. A flow cytometry assay was performed to reveal lectin expression and quantify the phagocytosis of apoptotic bodies. Taking into account that siglecs, a mannose receptor and galectins expressed on macrophages could be involved in engulfment of apoptotic bodies we studied their potential expression on THP-1 cells by means of polyacrylamide glycoconjugates. A strong binding of the cells to siglec ligands (3'SiaLac, 6'SiaLac, [Neu5Acalpha2-8]2) and galectin ligands (LacNAc, GalNAcbeta1 - 4GlcNAc, Galbeta1 - 3GalNAcbeta and asialoGM1) was observed. To reveal the corresponding targets on apoptotic bodies, the carbohydrate pattern of MELJUSO cells was analyzed. The apoptotic membrane was characterized by a high level of glycans terminated by galactose or sialic acid. To study lectin-mediated phagocytosis of apoptotic bodies by THP-1 cells, an inhibitory phagocytosis assay was performed. Binding of Galbeta1 - 3GalNAc- or LacNAc-specific reagents (lectins and antibodies) to apoptotic bodies abolished their engulfment by the THP-1 cells whereas blocking of Neu5Acalpha2 - 6 or Neu5Acalpha2 - 3 sites by the corresponding lectins was not effective. Furthermore, Galbeta1 - 3GalNAcbeta-PAA or asialoGM1-PAA binding to the THP-1 cells decreased phagocytosis, whereas two other potent THP-1-binding probes, LacNAc-PAA and GalNAcbeta1 - 4GlcNAc-PAA did not inhibit phagocytosis. Thus, Galbeta1 - 3GalNAcbeta-terminated chains represented on the apoptotic bodies but not the other tested galectin ligands appear to be a target for THP-1 cells.

Our study deals with the interaction of CD33 related-siglecs-5,-7,-8,-9,-10 with gangliosides GT1b, GQ1b, GD3, GM2, GM3 and GD1a. Siglec-5 bound preferentially to GQ1b, but weakly to GT1b, whereas siglec-10 interacted only with GT1b ganglioside. Siglec-7 and siglec-9 displayed binding to gangliosides GD3, GQ1b and GT1b bearing a disialoside motif, though siglec-7 was more potent; besides, siglec-9 interacted also with GM3. Siglec-8 demonstrated low affinity to the gangliosides tested compared with other siglecs. Despite high structural similarity of CD33 related siglecs, they demonstrated different ganglioside selectivity, in particular to the Neu5Acalpha2-8Neu5Ac motif.

In the present study, the interaction between epileptogenesis and the immune system were studied in a kindling model. First, the effects of a single administration of TNF-alpha (5.0 microg/kg, i.p.) on seizure and EEG activity were investigated in amygdala-kindled rats. TNF-alpha treated rats showed more prolonged epileptiformic discharges than control rats. TNF-alpha also induced a decrease in the power of delta band and an increase in theta and alpha activity. In addition, a marked increase in the power of beta and gamma band was observed. The EEG changes were most numerous in the frontal cortex and amygdala. All effects were registered 24 h after TNF-alpha administration. Finally, electrical stimulation enhanced the level of TNF-alpha in blood serum from 1.9 +/- 1.5 to 12.7 +/- 3.8 pg/ml and in brain tissue 56.8 +/- 6.0 to 109.2 +/- 6.0 pg/mg, as was determined via the ELISA method. It can be concluded that there is a mutual facilitative interaction of both epileptogenic and cytokine-derived mechanisms on this type of seizure. The changes in the power spectrum of the EEG after TNF-alpha might contribute to intensify thalamic-derived facilitation of epileptic discharge in cortical structures.

An SL2 lectin was isolated from human serum and characterized previously; cellular localization of the lectin was studied using polyclonal rabbit antibodies. According to cytofluorimetry, anti-SL2 antibodies bound only to lymphocytes and monocytes but not to other blood cells. Antibodies bound to Jurkat T cell lymphoma but did not interact with IM-9 cells of B cell origin. Moreover, the Jurkat cells bound oligosaccharides having the highest affinity to SL2 (GalNAcalpha_and Fucalpha1-2Gal), and this interaction was inhibited by anti-SL2 antibodies. Lysis of the Jurkat cells with subsequent electrophoresis and Western blotting indicates that anti-SL2 antibodies recognized a 14-kD protein.

A Mycoplasma agalactiae genomic fragment carrying four vsp-related genes (designated avg: agalactiae variable genes) was cloned, sequenced and compared to the vspA gene of Mycoplasma bovis. The following features were revealed: (i) the presence of a highly conserved vsp 5' upstream region; (ii) a highly homologous vsp N-terminal end encoding a putative lipoprotein signal sequence; (iii) sequence divergence of the rest of the mature proteins. By using avg specific probes in Southern blot analysis of genomic DNAs of M. agalactiae strains as well as of isolates from infected animals, marked DNA polymorphism of avg fragments was demonstrated. In addition, the avg genomic fingerprints were monitored for a period of 7 months, in isolates of M. agalactiae from an individual chronically infected animal. The results provided evidence that a chromosomal region of M. agalactiae, carrying vsp-related genes, undergoes rearrangements in vivo in the natural animal host during the course of infection.

This review describes new polymer materials for biomedical applications developed in the Polymers for Biology Laboratory of the Shemyakin-Ovchinnikov Institute of Bioorganic Chemistry, Russian Academy of Sciences. These include composite rigid sorbents for biochromatography, polymer dispersions for immunoassay, polymer hydrogels for immobilization of enzymes and cells, and polymer ultra thin films as biomembrane models and materials for biosensors. Some general and specific properties of these new materials and models as well as examples of their applications are discussed.

Carbohydrate (spacered saccharide residue, Glyc) probes with various tags were synthesized as analytical tools for study of cellular lectins, i.e., Glyc-polyacrylamide-3H, Glyc-PAA-biotin, Glyc-PAA-fluorescein (flu), and Glyc-PAA-digoxigenin, where PAA is a soluble polyacrylamide carrier of approximately 30 kDa. Binding of all types of probes, where Glyc is the sialyl Lewis X (SiaLeX) tetrasaccharide or a blank saccharide, was assessed using Chinese hamster ovary (CHO) cells either transfected with the E-selectin cDNA or mock-transfected. High binding of SiaLeX-PAA-3H to E-selectin-transfected cells and absence of binding to control cells (both native and permeabilized) allowed the conclusion that the polyacrylamide carrier and the spacer arm do not contribute significantly to the binding. The biotinylated probe showed a high level of nonspecific binding in cell enzyme-linked assays. A similarly built digoxigenin-labeled probe was significantly better. In flow cytometry assays, the fluorescein probe demonstrated a specific binding to E-selectin-transfected cells of a similar level to that given by an anti-E-selectin antibody. In addition, it could be inhibited by the anti-E-selectin antibody, further demonstrating specificity. Tumors were obtained from nude mice by injection of CHO E-selectin or mock-transfected cells. The fluorescent SiaLeX-PAA-flu probe could bind to tumor sections from E-selectin-positive CHO cells, but not from control CHO cells. These probes can thus be used to reveal specifically complex carbohydrate-binding sites on cells either in culture or on tissue sections., (Copyright 1998 Academic Press.)

Multiple restriction fragments, homologous to the previously described Mycoplasma bovis vspA gene, were identified in the chromosome of Mycoplasma agalactiae. The vspA, a representative variable surface lipoprotein gene of the vsp gene family, and four synthetic oligonucleotides, representing sequences complementary to selected regions of the vsp genes, were used as probes against digested chromosomal DNAs of several M. agalactiae clinical isolates. The resulting Southern blot analysis demonstrated a marked DNA polymorphism of multiple vspA-related fragments among the isolates. An oligonucleotide representing a conserved 5'-region common to all known vsp genes, was found to hybridize to multiple M. agalactiae genomic fragments while the other three oligonucleotides, representing distinct repetitive structures within the coding region of three known vsp genes (vspA, vspE, and vspF), failed to react. These results argue for the possible existence of a gene family in M. agalactiae analogous to the vsp system of M. bovis but comprised of diverse genes.

Two galactose-binding lectins, SL1 and SL2, were isolated from human serum by two-step affinity chromatography on sorbents with immobilized disaccharides Gal beta 1-3GlcNAc beta (Lec) and Fuc alpha 1-2Gal beta (Hdi). The purification degree of the SL1 and SL2 preparations was 4000-6000 times and yields were 6.9 and 4.7 micrograms/ml of serum, respectively. Electrophoresis showed that both lectins are oligomers with molecular masses of about 440 kDa, which are composed of ca. 67 kDa subunits. The carbohydrate specificity of the lectins was assayed by hemagglutination inhibition using mono- and oligosaccharides. Both lectins showed specificity for Gal and GalNAc residues but differed in the interactions with oligosaccharides. In particular, SL1 showed maximum affinity for disaccharide Gal beta 1-3GalNAc beta (in the form of 4-nitrophenyl glycoside), 6'-sialyllactose, and disaccharide residues Fuc alpha 1-3Gal beta and Gal alpha 1-3Gal beta, whereas SL2 was specific for GalNAc alpha residue and its derivatives as well as for disaccharide Hdi.

These studies were dealing with muscle protein depletion in rats under the heart hypertrophy induced by aorta coarctation or under liver regeneration after partial hepatectomy. The last experimental model was also used in order to establish how far such protein mobilization from muscles is realized during their simultaneous adaptation to local overload or disuse induced by a surgical method. 3 and 7 days after induction of hypertrophy of organs investigated, the quantities of sarcoplasmic, myofibrillar and stroma proteins were measured in last-twitch (m. extensor digitorum longus, m. biceps brachii) and slow-twitch (m. soleus) muscles. The contributions of protein biosynthesis and/or protein degradation change into muscle protein loss were evaluated by specially elaborated radiotracer method. The data obtained demonstrate that the pattern of muscle protein mobilization depends on nature of organ and rate of its hypertrophy. The process occurs differently in muscle of different type and in various muscle structures. It appears to be potentiated in overloaded muscles and to a lesser extent in unused ones. The loss of muscle proteins is certainly associated with inhibition of their biosynthesis. Increased degradation (or release) of preformed (non-labelled) protein molecules may also contribute to this loss whereas degradation of newly-formed (labelled) molecules does not change appreciably, possibly because their distribution in muscle structures is non-uniform.

The paper deals with convenient and fast methods for purification of monoclonal antibodies (MAbs) to carbohydrate antigens Le(y) and Le(d) from the cell culture and ascite fluid by ion-exchange chromatography on S-Sepharose and salt-promoted chromatography on a "thiophilic" adsorbent. One-step procedure on S-Sepharose of MAbs to Le(y) (IgG and IgM) provides significant purification (over 90% of contaminants were removed), while a purification factor for IgM to Le(d) is much lower. Highly purified IgM to Le(d) could be obtained by a two-step purification procedure including "thiophilic-adsorption" chromatography and gel-filtration (90-98% of contaminants from the cell culture and ascite fluid were removed). The preparations of IgG and IgM retain their initial antibody activity.

Tetrasaccharide Fuc alpha 1-2Gal beta 1-4(Fuc alpha 1-3)GlcNAc is known as carbohydrate determinant of cancer- and AIDS-associated antigen Lewisy (Ley). Synthetic antigen to generate mouse monoclonal antibodies (MAbs) directed to Ley was prepared and constructed as a spacer-armed tetrasaccharide coupled with lipophilized polymer, Ley-PAA-PE, where PAA is a 30-kD polyacrylamide and PE is phosphatidylethanolamine. An efficient immune response was provided by using Ley-PAA-PE adsorbed on Salmonella minnesota. Positive hybridomas were screened by enzyme-linked immunosorbent assay (ELISA) using Ley-PAA as a coating agent. An inhibitory version of the same test system showed absolute specificity of two MAbs: only hapten Ley and Ley-PAA were strong inhibitors, in contrast to Leb, tri- and disaccharidic fragments of the mentioned tetrasaccharides, as well as their PAA-conjugates. MAbs obtained against synthetic antigen specifically stained the Ley (+) cell line A431.

Sprouted barley grains, the waste product of malt extract production, were incriminated as the cause of a lethal (96% mortality) neurotoxic syndrome in sheep fed the grains. Clinical manifestations, comprising tremors, lameness, abnormal gait, paralysis and death indicated a tremorgenic mycotoxicosis. Whilst 50% of the flock died within 17 days, mortality continued over more than 5 months. Pathological findings were limited to neuronal degeneration and necrosis in the midbrain. Germinating grains were shown to be contaminated with growth of Aspergillus clavatus.

Exogenous application of indoleacetic acid results in a significant suppression of the linear growth that is promoted by exogenous gibberellic acid in Avena stem segments in a fashion similar to that previously noted in Avena leaf base segments (van Overbeek and Dowding, 1961, Fourth International Conference Plant Growth Regulation). Treatment with the auxin transport inhibitors, methyl-2-chloro-9-hydroxyfluorene-(9)-carboxylate (CFM) or 2,3,5-triiodobenzoic acid (TIBA), alone promotes elongation growth of the stem segments over that of control growth. This effect is interpreted as being due to the interference in the transport of native indoleacetic acid by CFM and TIBA, thus removing the inhibitory effect of native indoleacetic acid on gibberellin-promoted growth in the internodal intercalary meristem. This results in a greater promotion of internodal growth by native gibberellins. In the presence of (2-chloroethyl) trimethylammonium chloride (CCC), the growth-promoting effects of CFM and TIBA are decreased, and the antiauxin, PCIB (4-chloro-phenoxyisobutyric acid), has no growth-promoting effects whatsoever. These results indicate that the CFM and TIBA-promoted growth require the continuous presence of gibberellins. They further support the view that native indoleacetic acid acts as a growth suppressor hormone in its regulation of gibberellin-promoted internodal extension in Avena shoots.

Histone metabolism in liver studied within 72-hour period of liver regeneration after partial hepatectomy in 24 hours after the injection of 14C-amino acids in rats. The increase in radioactivity of f2a, f3 and f2b histones and the simultaneous decrease in f1 histone radioactivity was observed in regenerating rat liver as compared with the level of radioactivity estimated for the respective histones in ectomized liver lobes. These changes, which are characteristic for regenerating liver, were not observed after the shame operation and they did not eliminate after the injection of respective unlabelled amino acid. Possible correlation between the increase in specific radioactivity of most nuclear histones under regeneration process and a migration of pre-synthesized labelled histone molecules into nucleus, and also a transformation into histones of other nuclear proteins is discussed.

Protein synthesis in the heart, lungs, liver, pancreas, thymus, kidneys, skeletal muscles and testes of the 22-day Cosmos-605 flight and synchronous rats was studied with the aif of 14C-amino acid incorporation. On the 2nd postflight day both flight and synchronous rats showed an increased synthesis of heart sarcoplasmatic proteins. On the 26th postflight day the rate of incorporation was normal. On the 2nd and 26th days the synthesis of myofibrillar proteins in the m. quadriceps femoris was inhibited in flight animals postflight day the rate of incorporation was normal. On the 2nd and 26th days the syn-only. On the 2nd and 26th days the liver of the flight and synchronous rats showed qualitatively similar changes in protein metabolism of subcellular structures. No changes in the protein synthesis of other organs tested were detected.

With the aid of C14-leucine incorporation into proteins the protein synthesis in the red (m. soleus) and white (m. extensor dig. longus) skeletal muscles of flight and synchronous rats from the Cosmos-605 experiment was investigated. On the 2nd postflight day the total synthesis of proteins (as calculated per whole muscle) in the red muscle of flight rats remained unaltered. Therefore, distinct atrophy of this muscle that was seen only in flight rats can be attributed to increased degradation of proteins. On the 2nd postflight day the white muscle of both flight and synchronous rats showed moderate atrophy, not only increased protein degradation but also inhibited protein synthesis contributes to this.

Vaccination trials against Mycoplasma mycoides subsp. mycoides in suckling and weaned kids are described. A formalin-killed vaccine with adjuvant was used. Challenge to the vaccinated suckling kids was by ingestion of dam's infected milk and by subcutaneous inoculation of 10(6) organisms, and to the vaccinated weaned kids, by subcutaneous inoculation alone. The weaned 6-week-old kids were vaccinated, given a booster 3 weeks later and challenged 1 month after that. Serological monitoring was by enzyme-linked-immunosorbent assay (ELISA). The six control kids remained serologically negative and after challenge were pyrexic and mycoplasmaemic. All died or were killed in extremis, with a characteristic cellulitis at the site of inoculation, arthritis and swollen spleen. In contrast, the vaccinated kids remained healthy. There was a modest antibody response to the initial vaccination and a further steep rise after the booster. After challenge the organism was recovered only from the lymph nodes proximal to the site of challenge in some animals, and from the liver and kidney in two animals. All the vaccinated suckling kids were unaffected by ingestion of their dam's infected milk. The kids that received only one vaccine inoculation on the day of birth, and one control kid, succumbed to subcutaneous challenge, whereas the kids that were given a booster, except for one, remained healthy.

Diamond films ( approximately 0.7 micrometer thick) have been epitaxially grown on Si(111) substrates at room temperature with mass-selected 120-electronvolt C(+) ions. The diamond reflections observed in x-ray diffraction are well localized at their predicted positions, indicating that (i) the diamond(111) and (220) planes are parallel to the Si(111) and (220), respectively; (ii) the diamond rotational spread around its (111) normal is approximately 1.7 degrees ; and (iii) the mosaic block size is approximately 150 A. The film growth is discussed in terms of subplantation-a shallow subsurface implantation model. This discovery is an important step toward diamond semiconductor devices.

Using a radioindicator method the authors studied the effects of hydrocortisone on the synthesis and breakdown of sarcoplasmic and myofibrillar proteins in white (m. extensor digitorum longus) and red (m. soleus) functioning and idle skeletal muscles of rats. It was found that in addition to the catabolic effects, i.e. inhibition of the protein synthesis and/or stimulation of their breakdown the hormone also exhibited an "anticatabolic" effect that manifested itself in an inhibition of the protein breakdown. This effect was observed in all the cases (except the white functioning muscle) simultaneously with the protein synthesis inhibition, thus counteracting to this or that degree the development of atrophic reactions in the muscles.

Radiotracer studies in vivo on protein biosynthesis have been made in red (m. soleus) and white (m. extensor digitorum longus) rat shin muscles during their disuse atrophy caused by foot exarticulation. The rate of protein biosynthesis (as calculated per mass unit of total RNA) was increased in the white muscle on the 4th and 14th days, being unaltered on the 7th day after the surgery, but this rate was decreased in the red muscle at all these stages. The data obtained suggest that all these stages. The data obtained suggest that the atrophy, more pronounced in the red muscle than in the white one, can be mainly attributed in the former to decreased protein biosynthesis but in the latter to the increased protein degradation.

Using a radioindicator method, the metabolism of sarcoplasmic and myofibrillar proteins was studied in vivo at different stages of hypokinesia in rats. It was shown that the true muscle atrophy and the lowered content of both protein fractions during the first two weeks are due to sharp inhibition of sarcoplasmic protein biosynthesis as well as to deceleration of biosynthesis and acceleration of degradation of actomyosin. In hypokinesia the muscle mass does not increase within 3-8 weeks largely due to the acceleration of degradation of the de novo synthesized components of contractile proteins. In early hypokinesia a stressory mechanism plays an essential role in the pathogenesis of protein metabolism disturbances.
