Abstract

Quercetin-3-O-acyl esters (I-VI) were synthesized and their usefulness as quercetin topical prodrugs was evaluated. Quercetin esters were assayed to determine their water stability and solubility, their susceptibility to undergoing enzymatic hydrolysis and their permeation through excised human skin. Quercetin ethyl (I) and hexyl (IV) esters proved poorly stable in aqueous media and they were not assayed further. Among the derivatives tested, quercetin propyl (II) and butyl (III) esters were more water-soluble than the parent drug. Esters II, III and V were readily hydrolyzed by human plasma and esters II and III penetrated excised human skin better than quercetin from aqueous saturated solutions. On the basis of the results obtained, esters II and III could be regarded as promising quercetin topical prodrugs.

Abstract

What is known and objective Paritaprevir/ritonavir/ombitasvir+dasabuvir (Pr OD) is a direct-acting antiviral ( DAA) approved for the treatment of chronic hepatitis C virus. We report on a probable interaction between Pr OD with ribavirin and warfarin. Case description Two weeks after the start of Pr OD with ribavirin, the patient's international normalized ratio ( INR) became subtherapeutic. Eleven weeks into therapy and following a 125% total increase in the weekly warfarin dose, therapeutic INR was achieved. Thirteen days after DAA therapy was completed and discontinued, the patient's INR became critically supratherapeutic. What is new and conclusion Patients on Pr OD plus ribavirin with warfarin should have INR followed closely upon initiation and discontinuation of therapy due to a probable drug interaction. Abstract

Little information is available regarding the relationship between the fibrotic evolution of pulmonary tuberculosis (PTB) and cytokine levels in human bronchoalveolar lavage fluid (BALF) and serum.To evaluate correlations between profibrotic cytokine levels and post-treatment lung fibrotic evolution.BALF and serum amounts of pro- or anti-inflammatory cytokines were obtained by ELISA before and 6 months after the start of anti-tuberculosis chemotherapy in 13 subjects with PTB. BALF levels were recalculated as ELF (epithelial lining fluid) levels by the urea method. High resolution computed tomography (HRCT) of both lungs was performed at the same time.When comparing pre- and post-treatment radiological data, the scores for 2-10 mm nodules, consolidation and fibrosis presented significant differences (P0.05). Concomitantly, pre-treatment vs. 6 month concentrations of ELF IFN-gamma and TNF-alpha were decreased (P0.05), while those of IL-4 and IL-10 were increased (P0.012). At serum level, IFN-gamma decreased, as did TNF-alpha, TGF-beta1 and PDGF-BB (P0.05). When the patients were subdivided into two groups, 1) stable or 2) increasing HRCT fibrosis score, significant increases in the second group were observed for ELF/ serum values of TGF-beta1 and ELF PDGF-BB (P0.05) at 6 months post-treatment. Only serum TGF-beta1 values were significantly associated with the same group before treatment.

334. Introducing Solar Thermal “Net Metering” in an Actual small-scale District Heating System: A Case-study Analysis.

Abstract

An energy and economic analysis on an hypothesis of introduction of prosumers in an actual small-scale district heating system (DHS) in Northern Italy has been carried out. The study was performed by means of dynamic simulations of the DHS. The investigated configurations include the transformation of one or more customers in prosumers, producing and self-consuming solar thermal heat, and supplying the DHS with the excess heat produced. For the considered case-study the solar heat fed into the DHS is basically antagonist of the CHP production. This leads to a decrease in the non-renewable energy share for the whole DHS, to a decrease in the profit of the DHS utility and to a profitable investment for the prosumers. [ABSTRACT FROM AUTHOR]

Abstract

The potential therapeutic advantages of the encapsulation of idebenone within pegylated liposomes were investigated in vitro on primary cortical astrocytes of rats. In particular, both the concentration-dependent effects and the therapeutic effectiveness toward excitotoxic injury, elicited by chronic treatment with ethanol (100 microM) for 12 days, were evaluated. The following parameters were taken into consideration to assay free or liposomally entrapped idebenone: lactic dehydrogenase release, respiratory capacity measured by tetrazolium salt conversion, glutamine synthetase, and the levels of constitutive and inducible 70-kDa heat shock proteins. To evaluate the effects on astrocytes, three different drug concentrations were used (0.5 microM, 5 microM, and 50 microM). At the highest concentration used (50 microM), a toxic effect of the free and liposomally entrapped drug was observed. Toxic effects seem to be due to a cellular membrane perturbation, as demonstrated by (45)Ca(2+) permeation. The therapeutic effect of free or liposomally entrapped idebenone on ethanol-induced injury of primary cortical astrocytes was evaluated as a function of the drug concentration. The drug liposome formulation was much more effective than the free drug in counteracting the ethanol-induced damage in astrocytes, i.e., 10-times-lower doses of liposomally entrapped idebenone are able to provide a greater protective action than the free drug. The improved action of idebenone-loaded liposomes is probably due to the greater drug bioavailability at the cellular level.
