To better understand the impact of warming on heavy metals (HM) associated with plant litter decomposition in streams, we investigated the impact of high and low HM (Cu and Zn) levels and different water temperatures (10,15 and 20oC) on microbial decomposition of TyphaangustifoliaL.litter and the associated extracellular enzyme activities. During a 100-day incubation, changes in litter mass losses, chemical composition (lignin and total carbohydrate), and extracellular enzyme activity were determined. The decomposition rates were accelerated by the low HM levels at 20oC (0.0051 day–1 at CK vs 0.0061 day–1 at low HM levels). The negative effects of Cu and Zn on Typha litter decomposition were more pronounced at lower temperatures (10 and 15°C). The enhanced enzyme activities of cellulase and β-glucosidase and the higher lignin/litter weight loss and lignin/carbohydrate ratios were found at 20oC and low HM treatment. The enzyme activities of β-glucosidase and cellulase were positively correlated with litter mass losses at 20oC and low HM levels. These results suggest that a 5oC increase in water temperature may attenuate the inhibition of low HM level on litter decomposition.

A field experiment was conducted in 2006-2008 to evaluate the control effects of three mowing frequencies in combining with planting three tree species with three densities on the Chromolaena odorata in southwestern Karst region of Guangxi. In all treatments, the relative coverage, height, density, and aboveground biomass of C. odorata were decreased by 89.7% -99.3%, 41.6% -81.2%, 61.4% -83.2% and 91.7% -97.8%, respectively, and the capitulum number was significantly lesser than that in the control (P<0.05). The control effects on the growth of C. odorata were in the order of mowing frequency > tree species > planting density, and the optimal control mode was mowing twice one year and planting four plants of Delavaya yunnanensis per plot (4 m×4 m). Lonicerae japonicae Flos is a traditional Chinese medicine that has the effect is used for clearing away heat and detoxification. When stored improperly, this medicine is susceptible to mold growth, causing fungal contamination, reducing its safety and clinical efficacy. In order to understand the fungal contamination of Lonicerae japonicae Flos, we utilized the Illumina NovaSeq6000 platform to characterize fungal communities associated with five moldy Lonicerae japonicae Flos samples from warehouses in China via the sequencing of fungal internal transcribed spacer 2 regions. These analyses led us to identify 3 phyla, 5 classes, 7 orders, 9 families, 13 genera, and 20 species of fungi in these samples. Of these, Ascomycota was the dominant phylum, while Cysticomycetes, Ascomycetes, and Staphylococci were relatively dominant at the class level, and the most prevalent genera were Aspergillus, Penicillium, Xanthomonas, Microcystis, Talaromyces, and Erysiphe with relative abundance values of 79%, 21%, 94%, 100%, and 98%. In summary, in the present study we conducted the ITS-based comprehensive characterization of fungal communities associated with Lonicerae japonicae Flos for the first time. This approach aims to facilitate the early detection of fungal contamination in Lonicerae japonicae Flos, providing a theoretical basis for the study of anti-mold technologies.

Chemical ligation of peptides is increasingly used to generate proteins not readily accessible by recombinant approaches. However, a robust method to ligate "difficult" peptides remains to be developed. Here, we report an enhanced native chemical ligation strategy mediated by peptide conjugation in trifluoroacetic acid (TFA). The conjugation between a carboxyl-terminal peptide thiosalicylaldehyde thioester and a 1,3-dithiol-containing peptide in TFA proceeds rapidly to form a thioacetal-linked intermediate, which is readily converted into the desired native amide bond product through simple postligation treatment. The effectiveness and practicality of the method was demonstrated by the successful synthesis of several challenging proteins, including the SARS-CoV-2 transmembrane Envelope (E) protein and nanobodies. Because of the ability of TFA to dissolve virtually all peptides and prevent the formation of unreactive peptide structures, the method is expected to open new opportunities for synthesizing all families of proteins, particularly those with aggregable or colloidal peptide segments. Virus assembly, which takes place during the late stage of viral replication, is essential for virus propagation. However, the underlying mechanisms remain poorly understood, especially for viruses with complicated structures. Here, we use correlative light and electron microscopy to examine the formation of cytoplasmic virion assembly compartments (cVACs) during infection by a γ-herpesvirus. These cVACs are membraneless organelles with liquid-like properties. Formation of cVACs during virus infection is mediated by ORF52, an abundant tegument protein. ORF52 undergoes liquid–liquid phase separation (LLPS), which is promoted by both DNA and RNA. Disrupting ORF52 phase separation blocks cVACs formation and virion production. These results demonstrate that phase separation of ORF52 is critical for cVACs formation. Our work defines herpesvirus cVACs as membraneless compartments that are generated through a process of LLPS mediated by a tegument protein and adds to the cellular processes that are facilitated by phase separation. Effect of salicylic acid (SA) on chilling injury (Cl) of sponge gourd during storage (9 days, 9°C) plus shelf life (2 days, 20°C) was evaluated in this study. SA treatment at the concentration of 1.5 mmol L−1 significantly reduced postharvest Cl of sponge gourds. Besides, the application of SA could effectively decrease the electrolyte leakage, reduce the accumulation of malondialdehyde (MDA) and total phenolics, enhance the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), and inhibit the activities of phenylalanine ammonia-lyase (PAL) and polyphenol oxidase (PPO). The beneficial effects of SA could be attributed to preserved membrane integrity, inhibited membrane peroxidation, enhanced antioxidant system and suppressed activities of browning related enzymes. In a sense, SA as a postharvest tool may be commercially used in alleviating Cl of sponge gourd.

The antifungal activity of chitosan on a common fungal phytopathogen, Sclerotinia sclerotiorum, and the control effect on sclerotinia rot of carrot were investigated. Mycelial growth and fungal biomass were strongly inhibited by chitosan. Using propidium iodide stain combined with fluorescent microscopy, the plasma membrane of chitosan-treated S. sclerotiorum mycelia was observed to be markedly damaged. Concomitantly, protein leakage and lipid peroxidation was also found to be significantly higher in chitosan-treated mycelia compared to the control. Chitosan provided an effective control of sclerotinia rot of carrot, with induction of activity of defense-related enzymes including polyphenoloxidase and peroxidase. These data suggest that the effects of chitosan on sclerotinia rot of carrot may be associated with the direct damage to the plasma membrane and lipid peroxidation of S. sclerotiorum, and the elicitation of defense response in carrot.

Puccinia triticina Eriks. (Pt), the causal agent of wheat (Triticum aestivum L.) leaf rust, is the most widespread disease of common wheat worldwide. In the present study, 83 wheat cultivars from three provinces of China and 36 tester lines with known leaf rust resistance (Lr) genes were inoculated in the greenhouse with 18 Pt pathotypes to identify seedling effective Lr genes. Field tests were also performed to characterize slow leaf rusting responses at the adult plant growth stage in Baoding and Zhoukou in the 2014–15 and 2015–16 cropping seasons. Twelve Lr genes, viz. Lr1, Lr26, Lr3ka, Lr11, Lr10, Lr2b, Lr13, Lr21, Lr34, Lr37, Lr44, and Lr46 either singly or in combination were identified in 41 cultivars. Known Lr genes were not detected in the remaining 42 cultivars. The most commonly identified resistance genes were Lr26 (20 cultivars), Lr46 (18 cultivars), and Lr1 (eight cultivars). Less frequently detected genes included Lr13, Lr34, and Lr37 (each present in four cultivars), Lr10 (three cultivars), and Lr3ka and Lr44 (each in two cultivars). Evidence for the presence of genes Lr11, Lr2b, and Lr21 (each in one cultivar) was also obtained. Seventeen cultivars were found to have slow rusting resistance in both field growing seasons. The properties of k-rotation symmetric Boolean functions (k-RSBF) were analyzed.It was presented that the Walsh spectrum and auto-correlation value were invariant when the parameters of a k-rotation symmetric Boolean function were under k-circular translation of indices were presented.The analysis of the properties showed that many properties of k-RSBF could be described by their orbits,and the counting formulas of long cycles and short cycles on k-RSBF’ orbits were given.

Artemisinin is an effective antimalarial drug isolated from the medicinal plant Artemisia annua L. Due to its increasing market demand and the low yield in A. annua, there is a great interest in increasing its production. In this paper, in an attempt to increase artemisinin content of A. ANNUA by suppressing the expression of β-caryophyllene synthase, a sesquiterpene synthase competing as a precursor of artemisinin, the antisense fragment (750 bp) of β-caryophyllene synthase cDNA was inserted into the plant expression vector pBI121 and introduced into A. annua by Agrobacterium-mediated transformation. PCR and Southern hybridization confirmed the stable integration of multiple copies of the transgene in 5 different transgenic lines of A. annua. Reverse transcription PCR showed that the expression of endogenous CPS in the transgenic lines was significantly lower than that in the wild-type control A. annua plants, and β-caryophyllene content decreased sharply in the transgenic lines in comparison to the control. The artemisinin content of one of the transgenic lines showed an increase of 54.9 % compared with the wild-type control. The present study demonstrated that the inhibition pathway in the precursor competition for artemisinin biosynthesis by anti-sense technology is an effective means of increasing the artemisinin content of A. annua plants., (© Georg Thieme Verlag KG Stuttgart · New York.)
