With the aim to explore the ability of diarylheptanoids to reduce oxidative changes in human plasma proteins, a phytochemical investigation of the MeOH extract of Corylus avellanaleaves was perfomed. Analysis by LC-ESI/LTQOrbitrap/MS/MSnguided the isolation of two new diarylheptanoid derivatives, giffonins W (1) and X (2). The structures 1and 2were assigned by analysis of NMR data combined with a QM (quantum mechanical)/NMR approach. The absolute configurations of 1and 2were established by analysis of electronic circular dichroism (ECD) spectra compared with the TDDFT-simulated curves. The antioxidant activity of the new and known giffonins was evaluated by inhibition of human plasma lipid peroxidation. Giffonins with the highest inhibitory activity were tested for their ability to reduce oxidation of thiol groups and carbonylation in plasma proteins, and some of them exhibited higher antioxidant activity than curcumin.

This study extends the literature on how transparency influences news credibility perceptions by examining trust signals at the news outlet level, rather than at the story level, as earlier research has done. Experiments in the United States (n= 1,037) and Germany (n= 1,000) found that exposure to trust signals in a Google search about a known news brand, rather than an unknown brand, and the German cultural context increased news credibility perceptions. Participants were more likely to click on trust signals that gave background about the news brand or offered ways to engage with a news outlet.

Localization of single fluorescent molecules is key for physicochemical and biophysical measurements, such as single-molecule tracking and super-resolution imaging by single-molecule localization microscopy. Over the last two decades, several methods have been developed in which the position of a single emitter is interrogated with a sequence of spatially modulated patterns of light. Among them, the recent MINFLUX technique outstands for achieving an ∼10-fold improvement compared with wide-field camera-based single-molecule localization, reaching ∼1–2 nm localization precision at moderate photon counts. Here, we present a common framework for this type of measurement. Using the Cramér-Rao bound as a limit for the achievable localization precision, we benchmark reported methods, including recent developments, such as MINFLUX and MINSTED, and long-established methods, such as orbital tracking. In addition, we characterize two new proposed schemes, orbital tracking and raster scanning, with a minimum of intensity. Overall, we found that approaches using an intensity minimum have a similar performance in the central region of the excitation pattern, independent of the geometry of the excitation pattern, and that they outperform methods featuring an intensity maximum.

Breast carcinoma is a rare disease in men, and bilateral cases are extremely uncommon. The rarity of male breast carcinoma and the small number of large studies on this topic have made it necessary to extrapolate treatment standards and outcomes from those established for women. Between 1997 and 2007, 75 men with breast cancer were referred to our institute, and the bilateral case we present here was the only one we have observed since 1994. The goal of our work was to contribute to the available literature with this extremely unusual presentation of the disease.

The association of the STH gene polymorphism with Alzheimer disease (AD) is debated. In the analysis of two genetically and diagnostically distinct groups of Alzheimer patients from the USA and Italy, the authors did not find an association with the STHpolymorphism. However, the APOE-4-associated risk of AD greatly increased if the STH-Gallele was also present. The STH-Gallele appears to be a risk modifier for AD.

Physostigmine was administered orally 1 mg q.i.d. for one month to 8 patients with a clinical diagnosis of Alzheimer's presenile dementia. The possible beneficial effects of the drug were evaluated by means of a neuropsychological battery administered to all patients before and after treatment. The performances obtained by demented patients on vetest did not show any difference in comparison with performances obtained on the first neuropsychologic assessment. Some implications of these negative results are briefly discussed.

We tested DNA from 15 centrally infected cases of iatrogenic Creutzfeldt-Jakob disease (CJD) (dura mater or corneal homografts and stereotactic EEG electrodes), 11 peripherally infected cases (native human growth hormone or gonadotrophin), and 110 control individuals for the presence of mutations in the chromosome 20 amyloid gene. No patient or control had any of the known pathogenic point or insert mutations found in familial disease, but allelic homozygosity at polymorphic codon 129 was present in all but two (92) of the 26 patients, compared with 54 (50) of the 110 controls (p< 0.001). Pooled data from all identified and tested cases of iatrogenic disease yielded a worldwide total of 56 patients, of whom all but four were homozygous at codon 129 (p< 0.001). These findings support the thesis that homozygosity at codon 129 enhances susceptibility to iatrogenic infections of both central and peripheral origin, with evident implications for the population of dura mater homograft and pituitary hormone recipients whose lives have been complicated by the possibility of exposure to the infectious agent of CJD.

Hazelnut represents one of the most consumed food, containing a large range of bioactive and health-promoting compounds. Investigation of the fatty component of C. avellananut has been widely carried out, but little is known about its phenolic content. In order to achieve deeper insight into the polar fraction of the n-butanol extracts of both fresh and roasted kernels of C. avellanacultivar, “Tonda di Giffoni” a PGI (Protected Geographical Indication) product of Campania region (Italy), a phytochemical investigation was carried out.High performance liquid chromatography coupled to electrospray ionization, multiple-stage linear ion-trap and orbitrap high-resolution mass spectrometry (LC-ESI/LTQOrbitrap/MS/MSn) profile of the fresh and roasted hazelnut led to the identification of eleven phenolic compounds corresponding to diarylheptanoid derivatives, flavonoid O-glycosides and ellagic acid. Moreover, the antioxidant activity of isolated compounds was evaluated by TEAC assay which showed that ellagic acid and flavonoid derivatives possessed the highest free-radical-scavenging activity.

Santolina chamaecyparissusis an aromatic evergreen herb growing in North Africa and near the Mediterranean coasts of Southern Europe. Literature data focused mainly on the investigation of the essential oil content of the aerial parts of S. chamaecyparissus, but few is known on the chemical composition of its polar extract. Total phenolic content was determined by the Folin-Ciocalteau method and the antioxidant activity of the n-butanol and chloroform fractions of S. chamaecyparissuswas tested by Trolox equivalent antioxidant capacity (TEAC) and diphenyl-2-picrylhydrazyl radical (DPPH•) assays. Difference of the phenolic content and of the antioxidant activity between the two extracts prompted us to investigate their chemical composition. Five compounds, eupatilin (1), circimaritin (2), 4’-hydroxy-5,6,7-trimethoxyflavone (3), 1,3-propanediol,2-amino 1-(4-hydroxy-3-methoxyphenyl) (4) and luteolin (5) have been isolated from the chloroform extract, while the investigation of butanolic extract led to the isolation of seven compounds, prunasin (6), luteolin 7-O-β-D-glucopyranoside (7), syringoside (8), junipeionoloside (9), citroside A (10), lonicerin (11) and apigenin 7-O-β-D-neohesperidoside (12). Their structures were elucidated by extensive spectroscopic methods including 1D- (1H, 13C and TOCSY) and 2D-NMR (DQF-COSY, HSQC, HMBC) experiments as well as ESIMS analysis.

Roehybridine β-N-oxide (RNO) is a proaporphine-tryptamine dimer alkaloid from Roemeria hybrida. RNO possesses five stereogenic centers and so far its absolute configuration has not been determined. In this study, the absolute configuration of RNO was established as 6aS,7aR,6R,9S,10Rby comparing experimental and TDDFT calculated electronic circular dichroism (ECD) spectra. In addition, we have reported an amendment indicating exact 1H and 13C chemical shifts of RNO. RNO exhibited cytotoxic activity when tested in vitroagainst prostate cancer cell lines including PC3 and DU145. The IC50values of RNO against PC3 and DU145 were 13.3 and 15.1 μg/mL, respectively.

Medemia argunis a mysterious and little known monotypic fan palm from the Nubian Desert Oases of southern Egypt and northern Sudan. Its fruits have been found in the tombs from the 5th Dynasty (ca. 2500 BC) to Roman times (6–7th century AD), including the celebrated tomb of Tutankhamun. In ancient Egypt, the fruits of this palm were widely distributed and were highly valued, as confirmed by their frequent occurrence in offerings in the tombs. İn order to elucidate the chemical composition of the phenolic fraction, phytochemical investigation of the BuOH extract of fruits was carried out to afford eight compounds (1–8), among which was the new 2,4-dihydroxy-6-methylacetophenone 2-O-β-D-glucopyranoside (1). With the aim to investigate if the high shelf life of M. argunfruits could be related to the occurrence of antioxidant principles that were able to prevent oxidative reactions, the evaluation was carried out of the in vitroantioxidant activity by Trolox equivalent antioxidant capacity (TEAC) assay of the extract and isolated compounds.

A new C-6 flavone glycoside (6), together with seven known compounds, cryptochlorogenic acid (1), chlorogenic acid (2), 2-O-trans-caffeoylhydrocitric acid (3), isovitexin 7-β-D-glucopyranoside (4), 7,4′-dihydroxy-5-methoxyflavone-6-C-β-D-glucopyranoside (5), 3,5-O-dicaffeoylquinic acid (7) and 4,5-O-dicaffeoylquinic acid (8), were isolated from the aerial parts of Knautia arvensis.Their structures were elucidated by extensive spectroscopic methods including 1D- (1H, 13C and TOCSY) and 2D-NMR (DQF-COSY, HSQC, HMBC) experiments, as well as ESIMS analysis. Compounds 1, 3-5and 8are reported for the first time in Knautia arvensis.

Two new derivatives of phenylpropenoic acids, N-trans-feruloyl-L-DOPA and O-trans-caffeoylmalic acid dimethyl ester, along with four known N-trans-caffeoyl-L-DOPA (clovamide), N-trans-caffeoyl-L-DOPA-methyl ester, O-trans-caffeoylmalic acid, O-trans-feruloyl-malic acid and quercetin 3-O-β-D-glucopyranoside were isolated from the aerial parts of Trifolium pallidum.Their structures were elucidated by extensive spectroscopic methods including 1D- (1H, 13C) and 2D-NMR (DQF-COSY, HSQC, HMBC) experiments as well as mass spectrometry analysis.

The UN1 monoclonal antibody recognized the UN1 antigen as a heavily sialylated and O-glycosylated protein with the apparent molecular weight of 100–120 kDa; this antigen was peculiarly expressed in fetal tissues and several cancer tissues, including leukemic T cells, breast, and colon carcinomas. However, the lack of primary structure information has limited further investigation on the role of the UN1 antigen in neoplastic transformation. In this study, we have identified the UN1 antigen as CD43, a transmembrane sialoglycoprotein involved in cell adhesion, differentiation, and apoptosis. Indeed, mass spectrometry detected two tryptic peptides of the membrane-purified UN1 antigen that matched the amino acidic sequence of the CD43 intracellular domain. Immunological cross-reactivity, migration pattern in mono- and bi-dimensional electrophoresis, and CD43 gene-dependent expression proved the CD43 identity of the UN1 antigen. Moreover, the monosaccharide GalNAc-O-linked to the CD43 peptide core was identified as an essential component of the UN1 epitope by glycosidase digestion of specific glycan branches. UN1-type CD43 glycoforms were detected in colon, sigmoid colon, and breast carcinomas, whereas undetected in normal tissues from the same patients, confirming the cancer-association of the UN1 epitope. Our results highlight UN1 monoclonal antibody as a suitable tool for cancer immunophenotyping and analysis of CD43 glycosylation in tumorigenesis.

The highly N‐methylated linear polypeptides koshikamide C‐E and the cyclic analogues F‐H are isolated from Theonella swinhoei and Theonella cupola.

Two new rhamnocitrin glycosides (1and 2) were isolated from the aerial parts of Astragalus vogelii, along with one known rhamnocitrin glycoside (3). Two known flavonol glycosides (4and 5) and four known flavone derivatives (6-9) were isolated from the aerial parts of Astragalus eremophilus. Their structures were elucidated by extensive spectroscopic methods including 1D- (1H, 13C and TOCSY) and 2D-NMR (DQF-COSY, HSQC, HMBC) experiments, as well as ESIMS analysis.
