303. Pharmacokinetic interaction between itraconazole and metformin in rats: competitive inhibition of metabolism of each drug by each other via hepatic and intestinal CYP3A1/2

Abstract

Fungal infection is prevalent in patients with diabetes mellitus. Thus, we investigated whether a pharmacokinetic interaction occurs between the anti-fungal agent itraconazole and the anti-glycaemic drug metformin, as both drugs are commonly administered together to diabetic patients and are metabolized via hepatic CYP3A subfamily in rats.Itraconazole (20 mg·kg(-1)) and metformin (100 mg·kg(-1)) were simultaneously administered i.v. and p.o. to rats. Concentrations (I) of each drug in the liver and intestine, maximum velocity (V(max)), Michaelis-Menten constant (K(m)) and intrinsic clearance (CL(int) ) for the disappearance of each drug, apparent inhibition constant (K(i) ) and [I]/K(i) ratios of each drug in the liver and intestine were determined. Also the metabolism of each drug in rat and human CYPs was measured in vitro.After simultaneous administration of both drugs, either i.v. or p.o., the total area under the plasma concentration-time curve from time zero to infinity (AUC)s of itraconazole and metformin were significantly greater than that of either drug administered alone. The metabolism of itraconazole and metformin was significantly inhibited by each other via CYP3A1 and 3A2 in rat and 3A4 in human microsomes.The significantly greater AUCs of itraconazole and metformin after i.v. administration of both drugs are probably due to competitive inhibition of the metabolism of each drug by each other via hepatic CYP3A1/2. Whereas after oral administration of both drugs, the significantly greater AUCs of each drug administered together than that of either drug alone is mainly due to competitive inhibition of intestinal metabolism of each drug by each other via intestinal CYP3A1/2.

318. Effects of Slurry Composting and Biofiltration Liquid Fertilizer on Growth and Fruiting of Chestnut

Abstract

The study was conducted to investigate the availability of Slurry Composting and Biofiltration (SCB) liquid fertilizer instead of chemical fertilizer for growth and fruiting of chestnut trees. SCB liquid fertilizer is made from livestock manure through Composting and filtration through sawdust and is used as fertilizer. The experiment was conducted from April 2008 to september 2009 using seven-year-old grafting trees 'Tsukuba'. We used SCB liquid fertilizer, chemical fertilizer and organic fertilizer to compare the effect of various fertilizers. In 2009, height and basal diameter showed 8.04% and 25.90% increment compared to those of the first year by the treatment of SCB liquid fertilizer. The number of burr per bearing mother branch showed no significant increase, but fruiting rate of SCB liquid fertilizer treatment was higher than any other treatments. The results demonstrate not only excellence of SCB liquid fertilizer on growth and fruiting of chestnut trees but also possibility on substitution of chemical fertilizer for SCB liquid fertilizer., Proceedings of the 18th European Biomass Conference and Exhibition, 3-7 May 2010, Lyon, France, pp. 280-282

321. Application of Unit Time Block Entropy to Fetal Distress Heart Rate

Abstract

Recently, multiple time scale characteristics of heart dynamics have received much attention for distinguishing healthy and pathologic cardiac systems. Despite structural peculiarities of the fetal cardiovascular system, the fetal heart rate(FHR) displays multiple time scale characteristics similar to the adult heart rate due to the autorhythmicity of its different oscillatory tissues and its interaction with other neural controllers. In this paper, we investigate the event and time scale characteristics of the normal and two pathologic fetal heart rate groups with the help of the new measure, called the Unit Time Block Entropy(UTBE), which approximates the entropy at each event and time scale based on symbolic dynamics. This method enables us to match the measurement time and the number of words between fetal heart rate data sets simultaneously. We find that in the small event scale and the large time scale, the normal fetus and the two pathologic fetus are completely distinguished. Abstract

The Microvariability and Oscillations of stars (MOST) satellite has detected low-amplitude light variations ($\Delta m\sim$1 mmag) in the Be star $\beta$ CMi (B8Ve). The observations lasted 41 days and the variations have typical periods $\sim 0.3$ days. We demonstrate that the dominant frequencies are consistent with prograde high-order g-modes of $m=-1$ excited by the Fe-bump of opacity in an intermediate-mass ($\approx 3.5 M_\odot$) star with a nearly critical rotation period of 0.38 days. This is the first detection of nonradial g-mode pulsations in a Be star later than B6 leading to the possibility that pulsations are excited in all classical Be stars., Comment: 17 pages, 6 figures; Astrophysical Journal part 1 in press

Built by a 16-year-old high school student named Jack DeWitt, the first radio station in Nashville went on the air in 1922. Three years later, DeWitt helped start WSM, arguably one of the nation�s greatest radio stations, and in 1950, he and WSM put Nashville�s first television station on the air. Over the years, Nashville has had its share of local radio personalities, such as Noel Ball, Coyote McCloud, and Gerry House, as well as television personalities like Jud Collins, Bill Jay, and Larry Munson. Nationally recognized stars such as Dinah Shore, Oprah Winfrey, Pat Sajak, and Pat Boone started their careers in Nashville as well. Here are the stories and images of the people heard on transistor radios and the programs�including Five O�Clock Hop, Ruffin� Reddy, and The Mickey Mouse Club�watched by children while they did their homework.

341. Be star outbursts: transport of angular momentum by waves

Abstract

The Be phenomenon, that is the ejection of matter from Be stars into a circumstellar disk, has been a long lasting mystery. In the last few years, the CoRoT (Convection, Rotation and planetary Transits) satellite brought clear evidence that Be outbursts are directly correlated with pulsations. We found that it may be the transport of angular momentum by waves or pulsation modes that brings the already rapid stellar rotation to its critical value at the surface, and allows the star to eject material. The recent discovery of stochastically excited gravito-inertial modes by CoRoT in a hot Be star strengthens this scenario. We present the CoRoT observations and modeling of several Be stars and describe the new picture of the Be phenomenon which arose from these results., Comment: 6 pages, proceedings of a conference held in Hakone, in press

342. Optical Refraction Measurements Across Chesapeake Bay

Abstract

The refraction of light was measured across a 16.2 km over-water path across Chesapeake Bay in order to recover the low-level temperature profiles. The apparent elevation of buoys and lights on a tower are observed from a number of different heights (ranging from 3 m to 30 m). This report describes the refractive measurements, the statistics of over 500 observations and their relationship to the observed meteorological conditions taken over an 18-month period. An analysis of refraction under stable and unstable conditions is discussed and a non-dimensional approach for displaying the results is shown. In addition, a technique for monitoring the height variations of the water surface is discussed.

Abstract

The polymorphism, KLF6-IVS1-27A, in the Krüppel-like factor 6 (KLF6) transcription factor gene enhances its splicing into antagonistic isoforms and is associated with delayed histological progression of nonalcoholic fatty liver disease (NAFLD). To explore a potential role for KLF6 in the development of insulin resistance, central to NAFLD pathogenesis, we genotyped KLF6-IVS1-27 in healthy subjects and assayed fasting plasma glucose (FPG) and insulin sensitivities. Furthermore, we quantified messenger RNA (mRNA) expression of KLF6 and glucokinase (GCK), as an important mediator of insulin sensitivity, in human livers and in liver tissues derived from a murine Klf6 knockdown model (DeltaKlf6). Klf6 overexpression studies in a mouse hepatocyte line were utilized to mechanistically link KLF6 with Gck promoter activity. KLF6-IVS1-27Gwt (i.e., less KLF6 splicing) was associated with stepwise increases in FPG and insulin and reduced hepatic insulin sensitivity. KLF6 binds to the liver-specific Gck promoter and activates a GCK promoter-reporter, identifying GCK as a KLF6 direct transcriptional target. Accordingly, in DeltaKlf6 hepatocytes Gck expression was reduced and stable transfection of Klf6 led to up-regulation of Gck. GCK and KLF6 mRNAs correlate directly in human NAFLD tissues and immunohistochemistry studies confirm falling levels of both KLF6 and GCK in fat-laden hepatocytes. In contrast to full-length KLF6, splice variant KLF6-SV1 increases in NAFLD hepatocytes and inversely correlates with glucokinase regulatory protein, which negatively regulates GCK activity. Conclusion: KLF6 regulation of GCK contributes to the development of hepatic insulin resistance. The KLF6-IVS1-27A polymorphism, which generates more KLF6-SV1, combats this, lowering hepatic insulin resistance and blood glucose. © 2011 American Association for the Study of Liver Diseases.

The three mass-squared differences that are needed to accommodate the atmospheric neutrino data (through $\nu_\mu - \nu_s$ oscillation), the solar neutrino data via MSW mechanism (through $\nu_e - \nu_\tau$ oscillation), and the positive result of $\nu_\mu - \nu_e$ oscillation from LSND can be generated by this mechanism, whereas the vacuum oscillation solution to the solar neutrino problem is disfavored. We find that the electron and tau neutrino masses are of order $10^{-3}$ eV, and the muon neutrino and a sterile neutrino are almost maximally mixed to give a mass of order 1 eV. Two heavy sterile neutrinos have a mass of order 1 keV which can be obtained by the double seesaw mechanism with an intermediate mass scale $\sim 10^5$ GeV.
