Abstract

We conducted a meta-analysis of Parkinson's disease genome-wide association studies using a common set of 7,893,274 variants across 13,708 cases and 95,282 controls. Twenty-six loci were identified as having genome-wide significant association; these and 6 additional previously reported loci were then tested in an independent set of 5,353 cases and 5,551 controls. Of the 32 tested SNPs, 24 replicated, including 6 newly identified loci. Conditional analyses within loci showed that four loci, including GBA, GAK-DGKQ, SNCA and the HLA region, contain a secondary independent risk variant. In total, we identified and replicated 28 independent risk variants for Parkinson's disease across 24 loci. Although the effect of each individual locus was small, risk profile analysis showed substantial cumulative risk in a comparison of the highest and lowest quintiles of genetic risk (odds ratio (OR) = 3.31, 95% confidence interval (CI) = 2.55-4.30; P = 2 × 10(-16)). We also show six risk loci associated with proximal gene expression or DNA methylation.

Abstract

The clinical delineation of dementia with Lewy bodies (DLB) from Alzheimer disease (AD) remains unclear.To compare neuropsychological profiles in DLB and AD among Caribbean Hispanic family members and participants in a population-based epidemiologic sample using extended neuropsychological test batteries and to explore whether these differences were related to heritable factors.Cross-sectional study.Clinics in northern Manhattan (New York City), the Dominican Republic, and Puerto Rico.We compared measures of memory, orientation, language, and executive and visuospatial functioning between patients with DLB vs AD in 2 Caribbean Hispanic cohorts, including a family sample (89 patients with DLB and 118 patients with AD) and an epidemiologic sample (70 patients with DLB and 157 patients with AD). Patients with DLB in the family sample were further categorized as patients having at least 2 family members with DLB or as patients having 1 family member with DLB.To determine whether observed differences in cognitive profiles were driven by heritable factors, we repeated analyses in the epidemiologic sample after excluding familial cases. We applied general linear models adjusted for age, sex, educational level, disease duration, and apolipoprotein E epsilon4 (OMIM 104310) genotype.Patients with DLB in both samples were more severely impaired in orientation, visuoconstruction, and nonverbal reasoning after controlling for potential confounders. Patients having at least 2 family members with DLB had the most severe impairment in memory, followed by patients having 1 family member with DLB, and then by patients with AD. After excluding familial AD and DLB cases in the epidemiologic sample, the differences between the groups persisted but were attenuated.Compared with patients having AD, patients having DLB are more severely impaired in various cognitive domains, particularly orientation and visuospatial functioning. The difference seems stronger in familial DLB than in sporadic DLB. Whether this divergence in cognitive functions is caused by gene-gene or gene-environmental interactions remains unclear.

322. Effects of Urban Expansion and Anthropogenic Heat Enhancement on Tropical Cyclone Precipitation in the Greater Bay Area of China

Abstract

The impact of urban expansion and anthropogenic heat (AH) enhancement on tropical cyclone precipitation (TCP) in the Guangdong–Hong Kong–Macau Greater Bay Area (GBA) of China is investigated by using the Weather Research and Forecasting (WRF) model. Sensitivity experiments are conducted during the landfall periods of two tropical cyclones, Hato (2017) and Mangkhut (2018), by artificially varying the surface AH flux from 0 to 600 W/m2and the urban land surface from 1985 to 2017, respectively. Results show that the TCP in the GBA's urban region increases with both urban expansion and AH enhancement. However, in the urban downstream region, only AH enhancement causes an increase in TCP, whereas urban expansion has no significant influence on TCP. AH enhancement and urban expansion affect surface temperature, surface sensible heat flux, and surface latent heat flux, leading to changes in atmospheric instability and surface water evaporation, resulting in changes in low‐level moisture convergence. The change patterns of the vertically integrated moisture flux divergence between 850 and 910 hPa in the urban and urban downstream regions of the GBA are consistent with those of the TCP changes in this area, indicating that low‐level moisture convergence is the key factor determining the TCP changes in the region. This study examines the impact of urban expansion and anthropogenic heat (AH) enhancement on tropical cyclone precipitation (TCP) in the Guangdong–Hong Kong–Macau Greater Bay Area. Through sensitivity experiments by the Weather Research and Forecasting model, we find that both urban expansion and AH enhancement increase TCP in the urban region. However, AH enhancement favors TCP in the urban downstream region, whereas urban expansion has no significant influence on TCP in the region. Abstract

To investigate whether mortality risk is influenced by apolipoprotein E (APOE) genotype and whether the risk differs by ethnicity, we compared the mortality risk in 2,112 individuals ≥ 65 years of age residing in northern Manhattan in New York. Mortality risks associated with the APOE genotype, adjusted for sex, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and triglycerides, differed significantly by ethnic group. Among Caucasian and Hispanics, the E2/E3 genotype was associated with the lowest mortality risk in the multivariate Cox proportional hazards modeling, adjusted for lipid levels, whereas mortality risk did not differ substantially between the E4/E3 and E3/E3 genotypes. Among African-Americans, the E2/E3 genotype was not associated with the lowest mortality risk, but the E4/E3 genotype was. Adjustment for heart disease, diabetes, and stroke reduced mortality risk associated with each genotype by about 50% for all ethnic groups, but the patterns remained the same. Although we cannot rule out the possibility of a healthy survival bias, our analyses designed to examine healthy survival by comparing risk of mortality in groups who were younger or older at entry do not support this possibility. Our findings suggest that the APOE genotype is associated with mortality and that the genotypic risks differ by ethnic group. Nearly 50% of the mortality risk associated with the APOE genotype appears to act through major chronic diseases, but those diseases only partially explain the mechanism by which the genotypic risk acts. To better understand the observed ethnic differences in mortality risk by genotype, a detailed prospective study is needed to examine the relationships among APOE, other candidate genes, health conditions, and eventual death.

338. Fostering Indigenous Leadership through Redefining Christian Relationship at Southlands Christian Schools

Abstract

Doctor of Ministry (DMin), The goal of this project was to pioneer a class which would nurture indigenous leadership among high school students in a Christian school by redefining leadership and Christian relationships, focusing on power relationships. This project was meant to encourage students to re-imagine what healthy power relationships should look like and how healthy ministry could be done by adolescents. The laboratory for this experiment took the form of a ministry focus class that was first offered during the 2009-2010 academic year at Southlands Christian Schools in Walnut, California. The class focused on the theological themes of theologia crucis, missio Dei, priesthood of all believers and spiritual gifts. These topics were discussed to lay a foundation of what healthy relationships and power dynamics among Christians might look like. Surveys were taken of this class and also of the school at large. The students then embarked on ministry projects within the school in order to get a taste of leadership and also to see how healthy power relationships might operate. At the conclusion of these projects, the students were asked to reflect upon their thoughts of leadership and power relationships. This project illustrated that a majority of the students in the class saw that their ideas of power and relationships were not Christian but more corporate or religious. It demonstrated that a better way to practice relationships and leadership existed. While the first year of this class did not completely change the culture at Southlands Christian Schools, it did start the discussion of how to continue this shift and to sustain such a shift in the culture of the school. The definitive version was published in Marine Pollution Bulletin 57 (2008): 335-348, doi:10.1016/j.marpolbul.2008.01.020., In 2001, the Hong Kong government implemented the Harbor Area Treatment Scheme (HATS) under which 70% of the sewage that had been formerly discharged into Victoria Harbor is now collected and sent to Stonecutters Island Sewage Works where it receives chemically enhanced primary treatment (CEPT), and is then discharged into waters west of the Harbor. The relocation of the sewage discharge will possibly change the nutrient dynamics and phytoplankton biomass in this area. Therefore, there is a need to examine the factors that regulate phytoplankton growth in Hong Kong waters in order to understand future impacts. Based on a historic nutrient data set (1986-2001), a comparison of ambient nutrient ratios with the Redfield ratio (N:P:Si=16:1:16) showed clear spatial variations in the factors that regulate phytoplankton biomass along a west (estuary) to east (coastal/oceanic) transect through Hong Kong waters. Algal biomass was constrained by a combination of low light conditions, a rapid change in salinity, and strong turbulent mixing in western waters throughout the year. Potential stoichiometric Si limitation (up to 94% of the cases in winter) occurred in Victoria Harbor due to the contribution of sewage effluent with high N and P enrichment all year, except for summer when the frequency of stoichiometric Si limitation (48%) was the same as P, owing to the influence of the high Si in the Pearl River discharge. In the eastern waters, potential N limitation and N and P co-limitation occurred in autumn and winter respectively, because of the dominance of coastal/oceanic water with low nutrients and low N:P ratios. In contrast, potential Si limitation occurred in spring and a switch to potential N, P and Si limitation occurred in eastern waters in summer. 349. Particle Tracking Model for Sedimentation from Buoyant Jets.

Abstract

Sediment-laden turbulent buoyant jets are commonly found in natural and engineered environments. Examples include volcanic eruptions, deep sea hydrothermal vents, discharge of partially-treated wastewater, and dredging operations. A horizontal sediment buoyant jet is characterized by a horizontal momentum jet, rising plume, and a horizontal surface gravity current. The settling of particles from a sediment buoyant jet depends on the complex interaction of particles with turbulent fluctuations and the mean flow--in particular particle reentrainment due to the external irrotational flow induced by the jet. A three-dimensional (3D) stochastic particle tracking model is proposed to predict the sedimentation from an arbitrarily inclined sediment-laden buoyant jet in a stagnant ambient. The simple model predicts the entire 3D jet flow field via a coupling of semianalytical models for the mean flow as well as turbulent fluctuations. The mean flow for the turbulent buoyant jet and the surface gravity current are determined using well-validated integral models, while the external jet-induced irrotational flow field is computed by a distribution of point sinks along the jet trajectory. Turbulent velocity fluctuations are modeled by a Lagrangian particle velocity autocorrelation function that mimics the trapping and loitering of sediment particles in turbulent eddies. The turbulent fluctuations are stochastically generated from a self-similar profile of the turbulent kinetic energy derived from computational fluid dynamics (CFD) solution of jets and plumes. Predictions of the particle tracking model are in excellent agreement with experimental data over the entire jet-plume regime for a wide range of particle sizes (58-621 µm) and properties. The model provides physical insight in delineating the modes of sediment fall out. The fraction of sediment mass fall-out from the jet lower boundary is found to be a function of the ratio between the jet momentum--buoyancy length scale ls = Mo3/4 / Bo1/2 and the momentum-settling length scale lm = Mo1/2 /ws, where Mo and Bo are the source momentum and buoyancy fluxes, and ws is the particle settling velocity in quiescent fluid. The particle distribution in the vertical centerline plane of the buoyant jet is also well-predicted.
