Abstract

Summary: There is an urgent need for an oral, efficient and safe regimen for high‐risk APL under the pandemic of COVID‐19. We retrospectively analysed 60 high‐risk APL patients. For induction therapy (IT), in addition to all‐trans retinoic acid (ATRA) and oral arsenic (RIF), 22 patients received oral etoposide (VP16) as cytotoxic chemotherapy (CC), and 38 patients received intravenous CC as historical control group. The median dose of oral VP16 was 1000 mg [interquartile rage (IQR), 650–1250]. One patient died during IT in the control group, 59 evaluable patients (100%) achieved complete haematological remission (CHR) after IT and complete molecular remission (CMR) after consolidation therapy. The median time to CHR and CMR was 36 days (33.8–44) versus 35 days (32–42; p = 0.75) and 3 months (0.8–3.5) versus 3.3 months (2.4–3.7; p = 0.58) in the oral VP16 group and in the control group. Two (9.1%) and 3 (7.9%) patients experienced molecular relapse in different group respectively. The 2‐year estimated overall survival and event‐free survival were 100% versus 94.7% (p = 0.37) and 90.9% versus 89.5% (p = 0.97) respectively. A completely oral, efficient and safe induction regimen including oral VP16 as cytoreductive chemotherapy combined with ATRA and RIF is more convenient to administer for patients with high‐risk APL. [Display omitted] • Host factors and pathways were screened out in asthma and COVID-19 comorbidity. • Host factors of COVID-19 and asthma comorbidity involved in multiple pathways. • Networks of ACE2-related host factors in COVID-19 and asthma were revealed. • Estradiol and so on were identified as potential drugs for comorbidity treatment. ACE2 as a functional receptor for SARS coronavirus plays an important role in COVID-19 infection of the host. Thus, we aimed to explore interaction networks between ACE2 and common host factors in COVID-19/Asthma comorbidity. We identified 1,191 protein targets closely related to ACE2, and integrated the GEO database and multiple public databases to identify 305 host factors related to ACE2 between COVID-19/Asthma comorbidity. Further enrichment analyses showed that metabolic processes, Th1 and Th2 cell differentiation and PPAR signaling pathways had vital significance in the comorbidity of asthma and COIVD-19. HRAS, IFNG, CAT, CDH1, FASN, ACLY, CCL5, VCAM1, SCD and HMGCR were the key host factors related to ACE2. Tissue-specific enrichment and correlation analysis with ACE2 showed that SCD, CAT and FASN were highly expressed in the lung; and these 10 molecules were closely related to ACE2 and specifically expressed in multiple tissues. We also identified a series of drugs, such as estradiol, tetrachlorodibenzodioxin, resveratrol, cyclosporine, perfluorooctanoic acid and so on. Finally, the expression levels and diagnostic performance of HRAS and SCD showed statistical significance in external databases. This study explored the interaction networks of ACE2-related host factors in COVID-19 and asthma and identified several potential drugs for COVID-19/Asthma comorbidity. 321. The power of informal institutions: The impact of clan culture on the depression of the elderly in rural China

Abstract

Exploration of ways to improve the subjective welfare of residents is an important area of current academic research. Using data from the China Family Panel Studies survey conducted in 2010, this paper investigated the impact of clan culture on the mental health of elderly people in rural China. The results demonstrated that clan culture can significantly decrease the depression score of the rural elderly. Further, there was no gender difference with respect to the impact of clan culture on the depression score of the elderly. At the same time, the positive effects of clan culture on the depression score of the elderly have gradually weakened with economic development. Exploration of the mechanisms involved indicated that in areas with stronger clan culture, older people receive more social support. This study enriches our understanding of the impact of informal institutions on the welfare of rural residents. At the same time, it can also provide a certain decision-making reference for the government to formulate relative poverty relief strategies in a new stage of poverty alleviation.

Abstract

100-kernel weight (KW) is one of the most important agronomic traits in maize (Zea mays L.), related to yield. To realize its genetic basis, in this study, a recombinant inbred line (RIL) population derived from the cross between Mo17 and Huangzao4 was used for quantitative trait locus (QTL) mapping for KW under high and low nitrogen (N) regimes. As a result, five QTLs were identified on chromosomes 3, 4, 7 and 9, of which three were detected under both N environments, while the other two QTLs were respectively detected under high and low N regimes. These QTLs could explain phenotypic variance rom 4.47 to 14.47%. Due to additive effects, the three QTLs from Mo17, including two on chromosome 3 and one on chromosome 4, could increase KW from 0.64 to 1.01 g, while the other two from Huangzao4 on chromosomes 7 and 9 could decrease KW from 0.62 to 1.07 g. These results are beneficial for understanding the genetic basis of KW and developing the markers linked with KW for marker-assisted selection breeding in maize.Key words: Maize (Zea mays L.), 100-kernel weight, quantitative trait locus (QTL), recombinant inbred line (RIL), nitrogen regime.

Abstract

A hydrolyte of magnesium 7-methoxyl-4′-hydroxylisoflavone-3′-sulfonate ([Mg(H2O)6](C16H11O4SO3)2·10H2O) was synthesized and its crystal structure was determined by X-ray diffraction analyses. The crystal belongs to triclinic crystal system, space group P-1 with cell constants a = 0.7359(1) nm, b = 0.9935(1) nm, c = 1.6335(2) nm, α = 91.16(1)°, β = 92.07(1)°, γ = 106.53(1)°, V = 1.1436(2) nm5 and Z = 1. The results show that the title compound consists of [Mg(H2O)6]2+, C16H11O4SO3- and H2O. Mg(II) is located at the symmetry center of the title compound. [Mg(H2O)6]2+, C16H11SO3− and H2O form fourteen kinds of hydrogen bonds. The distance of 0.351 nm between parallel isoflavone rings shows a #-# stacking interaction. The hydrogen bonds and #-# stacking interaction lead to supramolecular formation with three-dimensional network structure. The title compound was also characterized by IR and 1H NMR spectra.

Aerosol indirect effects on warm clouds are estimated in the Grid-point Atmospheric Model of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics (IAP LASG) (GAMIL) with a new two-moment cloud microphysics scheme using two different physically-based aerosol activation parameterizations: Abdul-Razzak and Ghan, and Nenes and Seinfeld. The annual global mean changes in shortwave cloud forcing from preindustrial times to present day (a measure of the aerosol indirect effects) estimated from these two parameterizations are remarkably similar: 0.76 W m-2 with the Abdul-Razzak and Ghan parameterization, and 0.78 W m-2 with the Nenes and Seinfeld parameterization. Physically-based parameterizations can provide robust representations of aerosol effects on droplet nucleation, meaning that aerosol activation is no longer the most un-certain factor in modeling aerosol indirect effects.

Abstract

Summary: The Skp1‐Cul1‐F‐box‐protein (SCF) ubiquitin ligases are important parts of the ubiquitin system controlling many cellular biological processes in eukaryotes. However, the roles of SCF ubiquitin ligases remain unclear in phytopathogenic Magnaporthe oryzae. Here, we cloned 24 F‐box proteins and confirmed that 17 proteins could interact with MoSkp1, showing their potential to participate in SCF complexes. To determine their functions, null mutants of 21 F‐box‐containing genes were created. Among them, the F‐box proteins MoFwd1, MoCdc4 and MoFbx15 were found to be required for growth, development and full virulence. Fluorescent‐microscopy observations demonstrated that both MoFbx15 and MoCdc4 were localized to the nucleus, compared with MoFwd1, which was distributed in the cytosol. MoCdc4 and MoFwd1 bound to MoSkp1 via the F‐box domain, the deletion of which abrogated their function. Race tube and qRT‐PCR assays confirmed that MoFwd1 was involved in circadian rhythm by regulating transcription and protein stability of the core circadian clock regulator MoFRQ. Moreover, MoFWD1 also orchestrates conidial germination by influencing conidial amino acids pools and oxidative stress release. Overall, our results indicate that SCF ubiquitin ligases play indispensable roles in development and pathogenicity in M. oryzae. [ABSTRACT FROM AUTHOR]

Abstract

The performance of semi-dry anaerobic co-digestion of corn stover and vegetable waste was investigated using batch reactors and leaching bed reactors (LBRs). Six mixing ratios of corn stover and vegetable waste were employed to investigate optimal methane production using batch reactors, and process stability was assessed using the two LBRs. Kinetic assessment, methane production, volatile solid (VS) reduction, and digestate characteristics were used to evaluate the performance of anaerobic co-digestion. After 50 days of anaerobic co-digestion in the batch reactor, methane yields reached 314.5–323.4 mL/g VS, and increase in methane production rate indicated that synergistic effects occurred during anaerobic co-digestion. The corn stover and vegetable waste with a total solids (TS) ratio of 14:1 yielded the highest methane production efficiency and rate, as well as VS removal efficiency. However, serious acidification at the initial stage of digestion in the LBR experiments suggests that the scaling-up process from batch reactor to LBR was unstable, which may be attributed to high TS concentration and substrate characteristics.
