Objectives To investigate whether the deiodinase inhibitor iopanoic acid (IOP) has chondroprotective properties, a mechanical stress induced model of human aged explants was used to test both repeated dosing and slow release of IOP. Methods Human osteochondral explants subjected to injurious mechanical stress (65%MS) were treated with IOP or IOP encapsulated in poly lactic-co-glycolic acid–polyethylene glycol nanoparticles (NP-IOP). Changes to cartilage integrity and signalling were determined by Mankin scoring of histology, sulphated glycosaminoglycan (sGAG) release and expression levels of catabolic, anabolic and hypertrophic markers. Subsequently, on a subgroup of samples, RNA sequencing was performed on 65%MS (n = 14) and 65%MS+IOP (n = 7) treated cartilage to identify IOP’s mode of action. Results Damage from injurious mechanical stress was confirmed by increased cartilage surface damage in the Mankin score, increased sGAG release, and consistent upregulation of catabolic markers and downregulation of anabolic markers. IOP and, though less effective, NP-IOP treatment, reduced MMP13 and increased COL2A1 expression. In line with this, IOP and NP-IOP reduced cartilage surface damage induced by 65%MS, while only IOP reduced sGAG release from explants subjected to 65%MS. Lastly, differential expression analysis identified 12 genes in IOP’s mode of action to be mainly involved in reducing metabolic processes (INSIG1, DHCR7, FADS1 and ACAT2) and proliferation and differentiation (CTGF, BMP5 and FOXM1). Conclusion Treatment with the deiodinase inhibitor IOP reduced detrimental changes of injurious mechanical stress. In addition, we identified that its mode of action was likely on metabolic processes, cell proliferation and differentiation.

It also explored differences in its validity according to age, sex and GOLD airflow obstruction levels. Methods: 62 participants (68 ± 8 years, 53 males, FEV1 51 ± 23%pred) completed the Portuguese IPAQ-sf, wore an accelerometer for 7 days and completed a second IPAQ-sf. Test-retest reliability/agreement was assessed with Intraclass Correlation Coefficient (ICC2,1), 95% Limits of Agreement (LoA), standard error of measurement (SEM) and minimal detectable change (MDC95) for continuous variables, and percentage of agreement (%agreement) for categories ("active"/"inactive"). Validity was assessed with 95% LoA and Spearman's correlations (ρ) between IPAQ-sf 2 (METs-min/week, time in vigorous [VPA], moderate PA [MPA] and walking) and accelerometry (time in MVPA, VPA, MPA and step counts) for continuous variables; %agreement, Cohen's kappa, and sensitivity specificity and±predictive values for categories. Correlations were also performed for age, sex and GOLD airflow obstruction grades. Results: Reliability was good (ICC2,1 = 0.707) with wide LoA (-6446-6409 METs-min/week). SEM and MDC95 were 1840 and 4971 METs-min/week, respectively. %agreement between the two IPAQ-sf was 84% (kappa = 0.660). Positive, moderate and significant correlations were found between IPAQ-sf and accelerometry (0.396 ≤ ρ ≤ 0.527, p < 0.001), except for VPA (p > 0.05). Here, we describe the synthesis of a novel type of rare-earth-doped nanoparticles (NPs) for multimodal imaging, by combining the rare-earth elements Ce, Gd and Nd in a crystalline host lattice consisting of CaF2 (CaF2: Ce, Gd, Nd). CaF2: Ce, Gd, Nd NPs are small (15–20 nm), of uniform shape and size distribution, and show good biocompatibility and low immunogenicity in vitro. In addition, CaF2: Ce, Gd, Nd NPs possess excellent optical properties. CaF2: Ce, Gd, Nd NPs produce downconversion emissions in the second near-infrared window (NIR-II, 1000–1700 nm) under 808 nm excitation, with a strong emission peak at 1056 nm. Excitation in the first near- infrared window (NIR-I, 700–900 nm) has the advantage of deeper tissue penetration power and reduced autofluorescence, compared to visible light. Thus, CaF2: Ce, Gd, Nd NPs are ideally suited for in vivo fluorescence imaging. In addition, the presence of Gd3+ makes the NPs intrinsically monitorable by magnetic resonance imaging (MRI). Moreover, next to fluorescence and MR imaging, our results show that CaF2: Ce, Gd, Nd NPs can be used as imaging probes for photoacoustic imaging (PAI) in vitro. Therefore, due to their biocompatibility and suitability as multimodal imaging probes, CaF2: Ce, Gd, Nd NPs exhibit great potential as a traceable imaging agent in biomedical applications.

La producción de 87 de los principales cultivos alimentarios del mundo depende de los polinizadores (abejas, mariposas, abejorros, avispas, colibríes, murciélagos, entre otros). Sin embargo, éstos están en constante riesgo de disminuir sus poblaciones, lo cual provoca efectos negativos en los ecosistemas y la producción agrícola y de alimentos. Ante esta problemática, en 2021 el gobierno de México implementó la Estrategia Nacional para la Conservación y Uso Sustentable de los Polinizadores (ENCUSP), con el fin de asegurar la permanencia de las especies de polinizadores y plantas nativas en sitios urbanos. Esta iniciativa implicó la creación de jardines para polinizadores, de la cual “Oasis de las flores” forma parte y pretende fungir como un sitio de alimentación y mismo centro para incentivar la propagación y cultivo de especies de plantas nativas que ofrezcan, preferentemente, recursos florales para los diferentes tipos de polinizadores. “Oasis de las flores” como modelo de jardín (susceptible de replicarse en otros contextos y escalas urbanas), contribuye a mejorar la calidad de vida de las personas, ofreciendo espacios para el descanso, recreación, inspiración e incluso sanación. Este oasis vegetal ofrece alimento, agua y refugio a los polinizadores, absorbe partículas contaminantes del aire y produce oxígeno, filtra el agua de lluvia al subsuelo y reduce el ruido urbano gracias a la vegetación que lo compone, alberga una gran variedad de plantas nativas y aumenta la diversidad de especies animales. Por lo que el 14 de septiembre 2022, este jardín recibió por parte de la Comisión Nacional para el Conocimiento y Uso de la Biodiversidad (CONABIO), el Certificado como parte de la Red Poliniza que forma parte de la ENCUSP y promueve el diseño y desarrollo de jardines para polinizadores en ambientes urbanos y suburbanos, en bordes de caminos y cultivos, así como jardines regionales de polinizadores con vegetación nativa.

Objectives: Indicators of oral and periodontal diseases have previously been associated with handgrip strength and muscle mass and function in older adults in several populations. However, research in middle-aged adults on the relationship between periodontitis and handgrip strength and skeletal muscle mass – another important indicator of sarcopenia – has been limited. This study investigated the association between periodontitis and sarcopenia indicators in middle-aged adults from a U.S. population. Materials and Methods: Adults with complete periodontal and whole-body dual x-ray absorptiometry examinations from the 2013–2014 wave of the National Health and Nutrition Examination Survey (NHANES) were analysed using fully adjusted multiple linear regression models for associations between periodontitis diagnosis and severity and skeletal muscle mass index (SMMI) and combined handgrip strength (cHGS). Results: A subset of 1,912 individuals with a complete periodontal examination and whole-body dual x-ray absorptiometry scan from the NHANES 2013–2014 main cohort (n = 10,683) were included in the analysis. The median age of the participants was 43 years. Overall, 32% of the participants (n = 612) were determined to have periodontitis. Of these, 513 (26.8%) had non-severe (mild or moderate) periodontitis, and 99 (5.2%) had severe periodontitis. Both non-severe and severe periodontitis showed significant associations with cHGS (ß = -2.28, 95% CI -3.57–-0.98 and ß = -2.84, 95% CI -5.37–-0.31, respectively), but not with SMMI. Conclusion: The findings of the current study indicate that periodontitis and handgrip strength are interrelated. The present study highlights the need of further prospective research to investigate the nature and direction of the relationship between periodontitis and sarcopenia indicators. Clinical relevance: Future studies can support the screening, prevention and clinical management of sarcopenia and periodontitis, and emphasize the interdisciplinary and complementary approach between the disciplines of geriatric medicine and periodontology.

The removal of three acid pharmaceuticals—clofibric acid (CLA), diclofenac (DCL), and indomethacin (IND)—by a biological-photocatalytic sequential system was studied. These pharmaceutical active compounds (PhACs) are considered to persist in the environment and have been found in water and sewage, producing adverse effects on the aquatic environment. For the biological process, in batch experiments, a fixed bed bioreactor and activated sludge (hybrid bioreactor), under aerobic conditions, was used as pretreatment. The pretreated effluent was exposed to a photocatalytic process employing TiO2 nanotubular films (NTF-TiO2) with the following characteristics: an internal diameter of 112 nm, a wall thickness of 26 nm, nanotube length of 15 µm, a roughness factor of 1840 points, and an anatase-rutile crystalline structure. In the hybrid bioreactor, 39% IND and 50% ACL and DCL were removed. The biological-photocatalysis sequential system achieved the degradation of up to 90% of the initial concentrations of the three acid pharmaceuticals studied. This approach appears to be a viable alternative for the treatment of these non-biodegradable effluents.

Miniaturized mechanical testing based on small sample testing technology is a powerful technique to characterize the mechanical properties of different materials, and it is being used in different application fields. However, the small size of the specimens poses several challenges because the results are highly sensitive to measurement accuracy and the corresponding mechanical properties can change substantially due to the so-called specimen size effect. In this work, a novel testing device based on miniaturized specimens is presented. The equipment is designed to test materials in tensile and compressive loadings, but it is also capable of performing reverse-loading tests. Buckling of the specimen is an inherent phenomenon in compression loadings, especially for thin materials. Therefore, specimen geometry is properly studied and optimized to mitigate this effect. To evaluate the deformation of the specimen, the digital image correlation (DIC) technique is used to capture the full-field strain in the central gauge section of the sample. A sensitivity analysis of the DIC setting parameters was performed for this application. Background Pressure Injury (PI) management is challenging for dependent patients in Acute Care Wards (ACW) despite standard care (regular pressure relief measures, incontinence management, debridement, optimisation of hydration and nutrition). A Pressure Injury Care Bundle (PICB), introduced by the Department of Geriatrics, enhanced standard care by diligent and regular interdisciplinary team monitoring of patients with PIs following transfer to Long-Term Care (LTC) wards and thus may improve outcomes. Methods The PICB was delivered by multiple PI Nurse Champions with education of all nurse assistants and medical staff into PI aetiology/management. Progress was monitored with weekly PI measurements/photography and Nurse Champion-lead team review of all PIs. Data are presented as mean+/-1SD. After 96+/-103 days in the ACW, the PICB was applied to 30 consecutive patients aged 80+/-14 years, (60 % female; 19/30). All had stage 2-4 PIs (present in 83% on admission to ACW). On transfer to LTC wards, all had severe physical dependency with mean Charlson Comorbidity Index of 7+/-3, 27(90%) had palliative needs and 24(80%) were bedfast. Cognitive impairment was present in 22(68%) patients with 12(37%) dying due to advanced dementia. Patients were followed for 116+/-274 days. Results PI improvement by >2 stages occurred in 11(36%) patients after a mean of 103 days. Ulcers closed fully after 154+/-48 days in a further 15(50 %) patients. However, new ulcers emerged or preterminal ( Conclusion These results suggest an intensive multimodal intervention involving best practice enhanced by PI Nurse Champions and delivered by educated staff with regular PI team progress reviews results in significant improvement/healing of PIs in severely dependent patients with palliative needs. Expansion of the PICB to other wards with prospective evaluation is planned.

Plant biotechnology is rife with new advances in transformation and genome engineering techniques. A common requirement for delivery and coordinated expression in plant cells, however, places the design and assembly of transformation constructs at a crucial juncture as desired reagent suites grow more complex. Modular cloning principles have simplified some aspects of vector design, yet many important components remain unavailable or poorly adapted for rapid implementation in biotechnology research. Here, we describe a universal Golden Gate cloning toolkit for vector construction. The toolkit chassis is compatible with the widely accepted Phytobrick standard for genetic parts, and supports assembly of arbitrarily complex T-DNAs through improved capacity, positional flexibility, and extensibility in comparison to extant kits. We also provision a substantial library of newly adapted Phytobricks, including regulatory elements for monocot and dicot gene expression, and coding sequences for genes of interest such as reporters, developmental regulators, and site-specific recombinases. Finally, we use a series of dual luciferase assays to measure contributions to expression from promoters, terminators, and from cross-cassette interactions attributable to enhancer elements in certain promoters. Taken together, these publicly available cloning resources can greatly accelerate the testing and deployment of new tools for plant engineering.

Background: Muscle wasting, also known as myopenia, is frequent in rheumatoid arthritis (RA). To date, it is still unknown if the failure of pharmacologic therapies increases the risk of myopenia in RA. Objective: To identify if treatment failure with conventional synthetic DMARDs (csDMARDs) constitutes an independent risk factor of muscle wasting in women with RA. Methods: This was a cross-sectional study. We included 277 women with RA. Assessments in RA patients included: clinical, epidemiological, and therapeutic variables. The skeletal muscle index (SMI) was estimated by DXA, and myopenia was diagnosed if they had an SMI < 5.45 kg/m2. Multivariable logistic regression models identified risk factors of myopenia. Results: Muscle wasting was observed in 28.2% of patients with RA. The risk factors of myopenia in RA were menopausal (OR: 4.45, 95% CI: 1.86 to 10.64) and failure of combined therapy with csDMARDs (OR: 2.42, 95% CI: 1.15 to 5.07). The increased body mass index was protective (OR:0.81, 95% CI: 0.75 to 0.87). Conclusions: Around one of four patients with RA presented muscle wasting. Muscle wasting is related to treatment failure of combined csDMARDs; other factors influencing the presence of muscle wasting is being postmenopausal, whereas, the body mass index was a protective factor.

Upconversion nanoparticles (UCNPs) represent a group of NPs that can convert near-infrared (NIR) light into ultraviolet and visible light, thus possess deep tissue penetration power with less background fluorescence noise interference, and do not induce damage to biological tissues. Due to their unique optical properties and possibility for surface modification, UCNPs can be exploited for concomitant antigen delivery into dendritic cells (DCs) and monitoring by molecular imaging. In this study, we focus on the development of a nano-delivery platform targeting DCs for immunotherapy and simultaneous imaging. OVA 254–267 (OVA24) peptide antigen, harboring a CD8 T cell epitope, and Pam3CysSerLys4 (Pam3CSK4) adjuvant were chemically linked to the surface of UCNPs by amide condensation to stimulate DC maturation and antigen presentation. The OVA24-Pam3CSK4-UCNPs were thoroughly characterized and showed a homogeneous morphology and surface electronegativity, which promoted a good dispersion of the NPs.In vitroexperiments demonstrated that OVA24-Pam3CSK4-UCNPs induced a strong immune response, including DC maturation, T cell activation, and proliferation, as well as interferon gamma (IFN-γ) production.In vivo, highly sensitive upconversion luminescence (UCL) imaging of OVA24-Pam3CSK4-UCNPs allowed tracking of UCNPs from the periphery to lymph nodes. In summary, OVA24-Pam3CSK4-UCNPs represent an effective tool for DC-based immunotherapy.Graphical abstract

The devices used to carry out Remote Handling (RH) manipulation tasks in radiation environments address requirements that are significantly different from common robotic and industrial systems due to the lack of repetitive operations and incompletely specified control actions. This imposes the need of control with human-in -the-loop operations. These RH systems are used on facilities such PRIDE, CERN, ESS, ITER or IFMIF-DONES, the reference used for this work. For the RH system is crucial to provide high availability, robustness against radiation, haptic devices for teleoperation and dexterous operation, and smooth coordination and integration with the centralized control room. To achieve this purpose is necessary to find the best approach towards a standard control framework capable of providing a standard set of functionalities, tools, interfaces, communications, and data formats to the different types of mechatronic devices that are usually considered for Remote Handling tasks. This previous phase of homogenization is not considered in most facilities, which leads towards a costly integration process during the commissioning phase of the facility.In this paper, an approach to the IFMIF-DONES RH Control framework with strong standard support based on protocols such as OPC UA has been described and validated through an experimental setup. This test bench includes a set of physical devices (PLC, conveyor belt and computers) and a set of OPC UA compatible software tools, configured and operable from any node of the University of Granada network. This proof-of-concept mockup provides flexibility to modify the dimension and complexity of the setup by using new virtual or physical devices connected to a unique backbone. Besides, it will be used to test different aspects such as control schemes, failure injection, network modeling, predictive maintenance studies, operator training on simulated/ real scenarios, usability or ergonomics of the user interfaces before the deployment. Unsupported, carbon-containing molybdenum sulfide (MoS2) and bimetallic iron-molybdenum sulfide (FeMoS) catalysts are obtained by in-situ decomposition of (a) ammonium thiomolybdate (ATM), (b) Fe(III)-containing ATM, (c) hexyltrimethylammonium thiomolybdate (HTMATM), and (d) Fe(III)-containing HTMATM, labeled Mo-1, Mo-2, Mo-3 and Mo-4 catalysts. Their activity and selectivity are tested in the hydrodesulfurization (HDS) of dibenzothiophene (DBT). HDS activity of the novel bimetallic Mo-4 catalyst is nearly two times greater than that of the reference Mo-1 catalyst, with a less favorable selectivity towards hydrogenation products compared to the reference. Surface iron and carbon contents of the catalysts are found to affect their activity as follows; Mo-4 > Mo-3 ≈ Mo-1 > Mo-2.

Skin photosynthesis is the main source of vitamin D. Limited sun exposure and insufficient dietary vitamin D supply justify vitamin D supplementation in certain age groups. In older adults, recommended doses for vitamin D supplementation vary between 200 and 2000 IU/day, to achieve a goal of circulating 25-hydroxyvitamin D (calcifediol) of at least 50 nmol/L. The target level depends on the population being supplemented, the assessed system, and the outcome. Several recent large randomized trials with oral vitamin D regimens varying between 2000 and 100,000 IU/month and mostly conducted in vitamin D-replete and healthy individuals have failed to detect any efficacy of these approaches for the prevention of fracture and falls. Considering the well-recognized major musculoskeletal disorders associated with severe vitamin D deficiency and taking into account a possible biphasic effects of vitamin D on fracture and fall risks, an European Society for Clinical and Economic Aspects of Osteoporosis, Osteoarthritis and Musculoskeletal Diseases (ESCEO) working group convened, carefully reviewed, and analyzed the meta-analyses of randomized controlled trials on the effects of vitamin D on fracture risk, falls or osteoarthritis, and came to the conclusion that 1000 IU daily should be recommended in patients at increased risk of vitamin D deficiency.
