301. The optimization of model ensemble composition and size can enhance the robustness of crop yield projections

Abstract

Linked climate and crop simulation models are widely used to assess the impact of climate change on agriculture. However, it is unclear how ensemble configurations (model composition and size) influence crop yield projections and uncertainty. Here, we investigate the influences of ensemble configurations on crop yield projections and modeling uncertainty from Global Gridded Crop Models and Global Climate Models under future climate change. We performed a cluster analysis to identify distinct groups of ensemble members based on their projected outcomes, revealing unique patterns in crop yield projections and corresponding uncertainty levels, particularly for wheat and soybean. Furthermore, our findings suggest that approximately six Global Gridded Crop Models and 10 Global Climate Models are sufficient to capture modeling uncertainty, while a cluster-based selection of 3-4 Global Gridded Crop Models effectively represents the full ensemble. The contribution of individual Global Gridded Crop Models to overall uncertainty varies depending on region and crop type, emphasizing the importance of considering the impact of specific models when selecting models for local-scale applications. Our results emphasize the importance of model composition and ensemble size in identifying the primary sources of uncertainty in crop yield projections, offering valuable guidance for optimizing ensemble configurations in climate-crop modeling studies tailored to specific applications.

302. An experimental study on grammatical sensitivity and production competence in Chinese and Spanish EFL learners and its implications on EFL teaching methods

Abstract

Implicit knowledge acquired by L2 learners determines their language competence; however, it remains an issue to what extent advanced EFL learners can acquire implicit language knowledge. This study aims at finding out whether advanced EFL learners from two different L1s could acquire a level of implicit knowledge of English questions by using the modified Elicited Oral Imitation Task. A quantitative, experimental study with the Elicited Oral Imitation Task experimental tool was designed. A total number of 91 participants were recruited via the online experimental platform from October to November, 2021, distributed into a native speaker group, a Chinese EFL learner group, and a Spanish EFL group. The study evaluated participants’ implicit language knowledge by assessing two indicators: the grammatical sensitivity index and the production index. Independent-sample t-test and one-way analysis of variance (ANOVA) were applied to examine the differences in the two indices among different groups. Results revealed that both EFL groups displayed a significant difference with the native speaker group in their degree of implicit knowledge of English questions in general. A further comparison of the two indicators showed that while both EFL groups displayed a relatively high grammatical sensitivity to morpho-syntactic errors in English questions, their corrective production rate of ungrammatical sentences was notably lower. These results indicate that advanced EFL learners had difficulty in acquiring implicit knowledge of English questions at native speaker’ level. 311. Multi-plane PIV using depth of field for in-cylinder flow measurements

Abstract

Extending the planar Particle Image Velocimetry (PIV) technique to enable measurements on multiple planes simultaneously allows for some of the 3 dimensional nature of unsteady flow fields to be investigated. This requires less hardware and retains the typically higher spatial resolution of planar PIV compared to fully 3-dimensional PIV techniques. Performing multi-plane PIV measurements requires the light scattered from the different measurement planes to be distinguishable. This may be achieved by using different laser wavelengths which adds significantly to the expense and complexity of the system, by using different light sheet polarisations which is challenging for engine measurements through windows due to stress-induced birefringence, or by making alternating measurements of each plane which sacrifices the simultaneity of the flow measurement across multiple planes. In this work, simultaneous PIV measurements on two parallel offset planes are performed by modifying a standard two-dimensional two-component (2D2C) PIV system consisting of a single high-speed camera and a dual-cavity PIV laser. Image splitting optics with a variable optical path length were used to image two measurement planes onto the single camera sensor. The degree of defocus of the droplet images arising from the narrow depth of field was used to distinguish between the measurement planes. The robustness of the degree of defocus method for plane discrimination was evaluated using both representative test targets and experimentally obtained PIV images of the in-cylinder flow field of a spark ignition internal combustion engine (ICE). This depth of field based method enables simultaneous multi-plane PIV measurements of in-cylinder flows to be performed with only minor alterations to a standard 2D2C PIV system using off-the-shelf optics.

Abstract

Flexoelectricity is an electromechanical phenomenon produced in a dielectric material, with or without centrosymmetric microstructure, undergoing a non-uniform strain. It is characterized by the fourth-order flexoelectric tensor, which links the electric polarization vector with the gradient of the second-order strain tensor. Our previous work [H. Le Quang and Q.-C. He, Proc. R. Soc. A 467, 2369 (2011)] solved the fundamental theoretical problem of determining the number and types of all rotational symmetries that the flexoelectric tensor can exhibit. In the present study, compact explicit matrix representations of the flexoelectric tensor are provided so as to facilitate the use of it with any possible rotational symmetry. The number and types of all reflection symmetries that the flexoelectric tensor can have are also determined. To identify the rotational symmetry and reflection symmetry of a given flexoelectric tensor, a simple and efficient graphic method based on the concept of pole figures is presented and illustrated. The objective of this study was to identify genes associated with osteoporosis and reveal potential therapeutic targets.Methods: Expression profiles from GSE13850 and GSE56815 datasets were combined for differential expression analysis. Extraction of intersecting genes from the combined datasets and the differentially expressed genes in GSE56814 were performed to construct a multi-scale embedded gene co-expression network analysis (MEGENA) to obtain module genes. Module genes with an area under the receiver operating characteristic curve (AUC) > 0.60 were chosen to construct the least absolute shrinkage and selection operator (LASSO) model to obtain feature genes. A regulated network was constructed using differentially expressed micro-RNAs (miRNAs) in GSE74209 and feature genes. Finally, key genetic pathways and pathways of the Kyoto Encyclopedia of Genes and Genomes were identified and explored.Results: The commonly identified differentially expressed genes involve oxidative phosphorylation and caffeine metabolism. We identified 66 modules with 2354 module genes based on MEGENA. CARD8, FOXO4, IL1R2, MPHOSPH6, MPRIP, MYOM1, PRR5L and YIPF4 were identified as feature genes by the LASSO model. Furthermore, predicted miRNA target genes included 8 genes associated with PMOP. The largest AUC was observed for FOXO4, which was found at the nexus of feature genes and miRNA-regulated genes and which correlated with the upregulation of dendritic cells. Moreover, FOXO4 was found to be involved in ABC transporters, as well as cocaine and nicotine addiction.Conclusion: FOXO4 may serve as potential biomarker and therapeutic target for PMOP.Keywords: postmenopausal osteoporosis, microRNAs, bioinformatics analysis, FOXO4, biomarker

However, current treatments for t-NEPC are limited and far from satisfactory. According to our limited knowledge, report regarding the management of t-NEPC related hemorrhage is rare. Here, we report a case of t-NEPC formation after chronic hormonal therapy accompanying with severe bleeding in primary tumor and share our experiences to deal with the severe hematuria resulting from the progression of t-NEPC tumor.Case Presentation: An 80-year-old man with a significantly high prostate-specific antigen was diagnosed via pathology as advanced AdPC due to multiple bone metastases. He then received ADT including bicalutamide and goserelin. After 20 months of stable disease, the cancer rapidly progressed and presented with severe gross hematuria caused by bleeding of the primary tumor. The histopathologic analysis of a secondary biopsy of the primary tumor confirmed neuroendocrine prostate cancer, and subsequent genetic testing revealed germ-line mutations in the RB1 and FOXA1. To control the bleeding and relieve symptoms, the patient was treated with superselective prostate artery embolization (PAE). After the left internal pudendal artery and the right prostatic artery were embolized, hematuria was quickly alleviated and disappeared. However, the patient was not a suitable candidate to platinum-based chemotherapy due to weak constitution. Goserelin was continuously applied to maintain castration level of serum testosterone. Meanwhile, palliative radiotherapy to the prostate tumor, high-risk lymph node drainage areas (including iliac and para-aortic lymph nodes, internal iliac lymph nodes, presacral lymph nodes and obturator nerve lymph nodes) and bone metastases (right sacroiliac joint and thoracic vertebra) was performed and relieved the pain. Abstract

Conventional superconductors naturally disfavor ferromagnetism because the supercurrent-carrying electrons are paired into anti-parallel spin singlets. In superconductors with strong Rashba spin-orbit coupling, impurity magnetic moments induce supercurrents through the spin-galvanic effect. As a result, long-range ferromagnetic interaction among the impurity moments may be mediated through such anomalous supercurrents in a similar fashion as in itinerant ferromagnets. Fe(Se,Te) is such a superconductor with topological surface bands, previously shown to exhibit quantum anomalous vortices around impurity spins. Here, we take advantage of the flux sensitivity of scanning superconducting quantum interference devices to investigate superconducting Fe(Se,Te) in the regime where supercurrents around impurities overlap. We find homogeneous remanent flux patterns after applying a supercurrent through the sample. The patterns are consistent with anomalous edge and bulk supercurrents generated by in-plane magnetization, which occur above a current threshold and follow hysteresis loops reminiscent of those of a ferromagnet. Similar long-range magnetic orders can be generated by Meissner current under a small out-of-plane magnetic field. The magnetization weakens with increasing temperature and disappears after thermal cycling to above superconducting critical temperature; further suggesting superconductivity is central to establishing and maintaining the magnetic order. These observations demonstrate surface anomalous supercurrents as a mediator for ferromagnetism in a spin-orbit coupled superconductor, which may potentially be utilized for low-power cryogenic memory.

Conventional therapies for OCCC include surgery and chemotherapy. However, these OCCC treatment approaches are characterized by a high risk of relapse and drug resistance resulting in poor prognosis. Therefore, alternative therapeutic approaches are required to achieve better outcomes. In this study, a PIK3CA p.R88Q mutation and PD-L1 expression with a tumor proportion score of 10% was explored in a patient who presented with rapid recurrence after surgery and unsuccessful postoperative chemotherapy. Based on the clinical condition and the patient preference, she was administered a novel combinatorial therapy comprising mTOR inhibitor everolimus, which is a well-known and potent inhibitor of the PI3K/AKT signaling pathway, and the anti-PD-1 antibody toripalimab. Treatment with this combinatorial therapy showed good prognosis, with more than eight months of disease control, and no severe adverse events were observed. The findings of this study provide a novel and effective strategy for OCCC patients. To the best of our knowledge, this is the first study to report a new combination regimen of immunotherapy (everolimus plus toripalimab) for solid tumors. Everolimus is not only an antitumor targeted drug but also an immunosuppressant; it’s combination with immunotherapy is controversial. The BBOX1-AS1–miR-3940-3p and miR-3940-3p-KPNA2 interplay was verified via luciferase reporter and RNA pull-down assays.Results: High BBOX1-AS1 levels were detected in the nasopharyngeal carcinoma tissues. BBOX1-AS1 silencing considerably suppressed the proliferative, migratory, and invasive abilities of NPC cells in vitro. Interestingly, BBOX1-AS1 could specifically bind to miR-3940-3 and abrogate the inhibition of KPNA2 induced by miR-3940-3. Additionally, analysis of tissue samples showed that miR-3940-3 was inversely correlated with BBOX1-AS1 and KPNA2.Conclusion: Our findings revealed that the BBOX1-AS1/miR-3940-3/KPNA2 axis is pro-oncogenic in NPC progression, uncovering novel insights into targeted therapy for this disorder.Keywords: migration, proliferation, invasion, nasopharyngeal carcinoma

This study aimed to evaluate the impact of metabolic syndrome on the clinical outcome of patients with nasopharyngeal carcinoma (NPC) and its mechanism.Methods: A cohort of 2003 NPC patients with a median follow-up time of 96.3 months (range: 4.1– 120.0 months) were enrolled in this analysis. Kaplan–Meier curves and the Log rank test were used to determine the differences in progression-free survival (PFS), cancer specific survival (CSS) and overall survival (OS). Univariate and multivariable analyses were used to identify independent prognostic predictors. Untargeted metabolomics (LC-HRMS) was used to detect the serum metabolic profiles of 10 well-matched patients with or without metabolic syndrome. Differential metabolite-based enrichment analysis and pathway analysis were performed to identify the potential mechanism of metabolic syndrome in NPC.Results: A total of 171/2003 (8.5%) patients were diagnosed with metabolic syndrome, and these patients tended to be male (P < 0.001) and older (P = 0.003). Patients with metabolic syndrome had poorer PFS (P = 0.011), CSS (P = 0.003) and OS (P = 0.001) than those without metabolic syndrome. Univariate and multivariable analyses showed that metabolic syndrome was a statistically significant and independent predictor for PFS (HR: 1.34, 95% CI: 1.03– 1.75, P = 0.032), CSS (HR: 1.53, 95% CI: 1.12– 2.08, P = 0.008), and OS (HR: 1.50, 95% CI: 1.13– 2.00, P = 0.006). The current mainstream therapies for MPE, ie, indwelling pleural catheter and pleurodesis, have some drawbacks. In this retrospective study, we explored the efficacy and safety of medical thoracoscopic thermal ablation (argon plasma coagulation, APC) therapy for metastatic pleural tumors with MPE.Patients and Methods: A total of 176 patients were enrolled and divided into catheter pleural drainage (CPD) group (n = 77), non-ablation group (n = 46), and thermal ablation group (n = 53). Propensity score matching (PSM) was used for between-group comparisons to minimize bias. The primary endpoints were pleural effusion objective response rate (ORR) and time to progression (TTP); secondary endpoints included overall survival (OS), chest-tube duration, and safety.Results: Thermal ablation group and non-ablation group showed significantly higher ORR and shorter chest-tube duration versus the CPD group (ORR: thermal ablation, 88.2% vs 66.7%, P = 0.004; non-ablation, 88.4% vs 64.4%, P = 0.042; chest-tube duration: thermal ablation, 4.90 vs 7.24 days, P < 0.001; non-ablation, 5.73 vs 7.33 days, P = 0.010). Thermal ablation group exhibited longer TTP than the CPD group (median, 13.7 vs 7.3 months, P = 0.001) and the non-ablation group (median, 13.6 vs 10.3 months, P = 0.037). OS in the thermal ablation group was numerically longer than that in the CPD group with marginally significant difference (P = 0.055).
