O projeto teve como objetivo completar, através da cultura material, lacunas da história da igreja Nossa Senhora da Conceição (1874) e da região onde está inserida. Foram realizados cortes experimentais na parte externa (fundos) e interna do templo (altar e nave central). O perfil estratigráfico do pátio apresentou cinco camadas, alcançando 1,90m de profundidade, já o interno revelou três camadas e a profundidade de 1,20m. Escavações foram realizadas ao redor de uma estrutura (poço) encontrada na nave lateral esquerda, junto ao altar, 0,60cm abaixo do piso atual. O poço mede 2,25m de diâmetro interno, 0,33m de espessura (comprimento dos tijolos) e 0,02m de reboco. Material resgatado: cerâmica (vasilhas das tradições Tupiguarani, Vieira e Neobrasileira e cachimbo da Neobrasileira); cerâmica colonial e colonial vidrada; louça (faiança, faiança fina, ironstone, salt-glazed, etc.); vidro (garrafas, taças, toucador, frascos de remédio, etc.); metal (lâmina de machado, colher, armadilha para animais, tesouras, chaves, cravos, pregos, panelas fragmentadas, moedas portuguesas e do Brasil Império etc.); osso (botões, cabos e restos de alimentação); lítico: material (afiador de faca, lousas, ponteiras, etc.); matéria-prima (ardósia, quartzo leitoso, granito, arenito etc.). O material arqueológico foi classificado, restaurado, analisado, fotografado, desenhado, foram confeccionadas tabelas e o material foi acondicionado conforme método Mentz Ribeiro (2004); em gabinete, realizaram-se estudos comparativos, a arte final e redação do presente artigo científico para publicação. Entre os resultados preliminares, concluímos que o poço pertencia a uma residência situada ao oeste, provavelmente junto à rua, ou seria um dos dez que abasteciam a cidade por volta de 1860. Com o início da construção da igreja (1872), o mesmo teria sido aterrado. Não foram encontrados vestígios do forte Jesus-Maria-José, de 1737, descartando uma tradição oral de que a igreja teria sido construída sobre aquela construção militar, marco da colonização portuguesa no Rio Grande do Sul.

Background: The duplication of genes can occur through various mechanisms and is thought to make a major contribution to the evolutionary diversification of organisms. There is increasing evidence for a large-scale duplication of genes in some chelicerate lineages including two rounds of whole genome duplication (WGD) in horseshoe crabs. To investigate this further, we sequenced and analyzed the genome of the common house spider Parasteatoda tepidariorum. Results: We found pervasive duplication of both coding and non-coding genes in this spider, including two clusters of Hox genes. Analysis of synteny conservation across the P. tepidariorum genome suggests that there has been an ancient WGD in spiders. Comparison with the genomes of other chelicerates, including that of the newly sequenced bark scorpion Centruroides sculpturatus, suggests that this event occurred in the common ancestor of spiders and scorpions, and is probably independent of the WGDs in horseshoe crabs. Furthermore, characterization of the sequence and expression of the Hox paralogs in P. tepidariorum suggests that many have been subject to neo-functionalization and/or sub-functionalization since their duplication. Conclusions: Our results reveal that spiders and scorpions are likely the descendants of a polyploid ancestor that lived more than 450 MYA. Given the extensive morphological diversity and ecological adaptations found among these animals, rivaling those of vertebrates, our study of the ancient WGD event in Arachnopulmonata provides a new comparative platform to explore common and divergent evolutionary outcomes of polyploidization events across eukaryotes.

The development of a digestive system is an essential feature of bilaterians. Studies of the molecular control of gut formation in arthropods have been studied in detail in the fruit fly Drosophila melanogaster. However, little is known in other arthropods, especially in noninsect arthropods. To better understand the evolution of arthropod alimentary system, we investigate the molecular control of gut development in the spider Parasteatoda tepidariorum (Pt), the primary chelicerate model species for developmental studies. Orthologs of the ectodermal genes Pt-wingless (Pt-wg) and Pt-hedgehog (Pt-hh), of the endodermal genes, Pt-serpent (Pt-srp) and Pt-hepatocyte-nuclear factor-4 (Pt-hnf4) and of the mesodermal gene Pt-twist (Pt-twi) are expressed in the same germ layers during spider gut development as in D. melanogaster. Thus, our expression data suggest that the downstream molecular components involved in gut development in arthropods are conserved. However, Pt-forkhead (Pt-fkh) expression and function in spiders is considerably different from its D. melanogaster ortholog. Pt-fkh is expressed before gastrulation in a cell population that gives rise to endodermal and mesodermal precursors, suggesting a possible role for this factor in specification of both germ layers. To test this hypothesis, we knocked down Pt-fkh via RNA interference. Pt-fkh RNAi embryos not only fail to develop a proper gut, but also lack the mesodermal Pt-twi expressing cells. Thus, in spiders Pt-fkh specifies endodermal and mesodermal germ layers. We discuss the implications of these findings for the evolution and development of gut formation in Ecdysozoans.

In nearly all metazoans, the earliest stages of development are controlled by maternally deposited mRNAs and proteins. The zygotic genome becomes transcriptionally active hours after fertilization. Transcriptional activation during this maternal-to-zygotic transition (MZT) is tightly coordinated with the degradation of maternally provided mRNAs. In Drosophila melanogaster, the transcription factor Zelda plays an essential role in widespread activation of the zygotic genome. While Zelda expression is required both maternally and zygotically, the mechanisms by which it functions to remodel the embryonic genome and prepare the embryo for development remain unclear. Using Cas9-mediated genome editing to generate targeted mutations in the endogenous zelda locus, we determined the functional relevance of protein domains conserved amongst Zelda orthologs. We showed that neither a conserved N-terminal zinc finger nor an acidic patch were required for activity. Similarly, a previously identified splice isoform of zelda is dispensable for viability. By contrast, we identified a highly conserved zinc-finger domain that is essential for the maternal, but not zygotic functions of Zelda. Animals homozygous for mutations in this domain survived to adulthood, but embryos inheriting these loss-of-function alleles from their mothers died late in embryogenesis. These mutations did not interfere with the capacity of Zelda to activate transcription in cell culture. Unexpectedly, these mutations generated a hyperactive form of the protein and enhanced Zelda-dependent gene expression. These data have defined a protein domain critical for controlling Zelda activity during the MZT, but dispensable for its roles later in development, for the first time separating the maternal and zygotic requirements for Zelda. This demonstrates that highly regulated levels of Zelda activity are required for establishing the developmental program during the MZT. We propose that tightly regulated gene expression is essential to navigate the MZT and that failure to precisely execute this developmental program leads to embryonic lethality.

Rhodnius prolixus not only has served as a model organism for the study of insect physiology, but also is a major vector of Chagas disease, an illness that affects approximately seven million people worldwide. We sequenced the genome of R. prolixus, generated assembled sequences covering 95% of the genome ( approximately 702 Mb), including 15,456 putative protein-coding genes, and completed comprehensive genomic analyses of this obligate blood-feeding insect. Although immune-deficiency (IMD)-mediated immune responses were observed, R. prolixus putatively lacks key components of the IMD pathway, suggesting a reorganization of the canonical immune signaling network. Although both Toll and IMD effectors controlled intestinal microbiota, neither affected Trypanosoma cruzi, the causal agent of Chagas disease, implying the existence of evasion or tolerance mechanisms. R. prolixus has experienced an extensive loss of selenoprotein genes, with its repertoire reduced to only two proteins, one of which is a selenocysteine-based glutathione peroxidase, the first found in insects. The genome contained actively transcribed, horizontally transferred genes from Wolbachia sp., which showed evidence of codon use evolution toward the insect use pattern. Comparative protein analyses revealed many lineage-specific expansions and putative gene absences in R. prolixus, including tandem expansions of genes related to chemoreception, feeding, and digestion that possibly contributed to the evolution of a blood-feeding lifestyle. The genome assembly and these associated analyses provide critical information on the physiology and evolution of this important vector species and should be instrumental for the development of innovative disease control methods.

Monitoring and troubleshooting distributed systems is notoriously difficult; potential problems are complex, varied, and unpredictable. The monitoring and diagnosis tools commonly used today—logs, counters, and metrics—have two important limitations: what gets recorded is defined a priori, and the information is recorded in a component- or machine-centric way, making it extremely hard to correlate events that cross these boundaries. This article presents Pivot Tracing, a monitoring framework for distributed systems that addresses both limitations by combining dynamic instrumentation with a novel relational operator: the happened-before join. Pivot Tracing gives users, at runtime, the ability to define arbitrary metrics at one point of the system, while being able to select, filter, and group by events meaningful at other parts of the system, even when crossing component or machine boundaries. We have implemented a prototype of Pivot Tracing for Java-based systems and evaluate it on a heterogeneous Hadoop cluster comprising HDFS, HBase, MapReduce, and YARN. We show that Pivot Tracing can effectively identify a diverse range of root causes such as software bugs, misconfiguration, and limping hardware. We show that Pivot Tracing is dynamic, extensible, and enables cross-tier analysis between inter-operating applications, with low execution overhead. [ABSTRACT FROM AUTHOR]

Rhipicephalus microplus - Abstract
Cyclin-dependent kinases (CDKs) are a family of serine/threonine kinases essential for cell cycle progression. Herein, we describe the participation of CDKs in the physiology of Rhipicephalus microplus, the southern cattle tick and an important disease vector. Firstly, amino acid sequences homologous with CDKs of other organisms were identified from a R. microplus transcriptome database in silico. The analysis of the deduced amino acid sequences of CDK1 and CDK10 from R. microplus showed that both have caspase-3/7 cleavage motifs despite their differences in motif position and length of encoded proteins. CDK1 has two motifs (DKRGD and SAKDA) located opposite to the ATP binding site while CDK10 has only one motif (SLLDN) for caspase 3–7 near the ATP binding site. Roscovitine (Rosco), a purine derivative that inhibits CDK/cyclin complexes by binding to the catalytic domain of the CDK molecule at the ATP binding site, which prevents the transfer of ATP’s cphosphoryl group to the substrate. To determine the effect of Rosco on tick CDKs, BME26 cells derived from R. microplus embryo cells were utilized in vitro inhibition assays. Cell viability decreased in the Rosco-treated groups after 24 hours of incubation in a concentration-dependent manner and this was observed up to 48 hours following incubation. To our knowledge, this is the first report on characterization of a cell cycle protein in arachnids, and the sensitivity of BME26 tick cell line to Rosco treatment suggests that CDKs are potential targets for novel drug design to control tick infestation.

This paper analyzes the power relations in the Captaincy of São José do Piauí, as well as strategies for establishing a local political power. To do so, exposes the contrast between the configuration of stockyard’s society, since the end of the century XXVII, and the context of the government's installation of the captaincy, in the second half of the eighteenth century. It is considered one of the oldest documents in the history of Piaui: a Description of the Piauí’s hinterland, written by priest Miguel de Carvalho in 1697. Then, from documents produced in the sphere of colonial administration, seeking to investigate the strategies adopted in the formation of a local elite that served the purposes of the metropolis. Among these strategies, we highlight the economic benefits and social prestige as important elements that have come to mediate power relations.

A presente tese investiga a incidência de interdições discursivas, explícitas e não explícitas, na história política do Brasil, com destaque para a interdição diluidora do abolicionismo em meio à discursividade republicana e anticolinialista da Conjuração Baiana de 1798. Tal interdição teria se efetivado a partir da práxis discursiva dos revolucionários baianos, que lhes demandou a constituição de palanques enunciativos virtualmente capazes de tirá-los da invisibilidade, da mudez e do non sens de sua existência política e de resistir aos efeitos de captura dos discursos dominantes. As outras duas análises, que servem de contraponto à primeira, investigam a interdição (explícita) ao Partido Comunista Brasileiro na cassação de seu registro partidário em 1947 e a interdição (não-explícita) ao movimento comunitário na promoção ao Orçamento Participativo de Porto Alegre. A tese propõe o desenvolvimento de procedimentos capazes de lidar proveitosamente com os estudos históricos no interior dos dispositivos de análise do discurso propostos por Michel Pêcheux em 1969 e depois revisitados por ele e Catherine Fuchs em 1975. Considera também uma série de intervenções do fundador da Análise do Discurso nos seus últimos anos de vida, nas quais uma compreensão mais fina da língua e da história neutralizam as tentações homogeneizantes que incidiam sobre os estudos discursivos. Algumas proposições de Jacques Rancière sobre a prática política, os modos de subjetivação e o desentendimento servem de base às propostas teórico-metodológicas desenvolvidas na tese, assim como uma reconsideração crítica do legado de Louis Althusser no bojo da teoria do discurso de Michel Pêcheux, com destaque à sua teoria da ideologia e à simples alusão à história, sem deixar de reconhecer e seguir trabalhando o sentido materialista de sua intervenção no marxismo. Conclui-se que também as classes dominantes não podem escapar à necessidade e à contingência de fazer política, contra os fatores de transformação social que atravessam os muitos furos e instabilidades da ordem, e que não há interdição que se sustente sem a promoção de alianças (reais, possíveis e imaginárias) no tecido social. As lesões no ligamento cruzado anterior (LCA) são frequentes no âmbito esportivo. O estudo dos mecanismos de lesão demonstrou que a maioria das lesões é oriunda tanto de aterrissagens unilaterais quanto bilaterais. Apesar de estudos terem demonstrado diferenças no comportamento cinemático de membros inferiores entre homens e mulheres, não há evidências de como homens se comportam em diferentes condutas. O objetivo neste estudo foi comparar a cinemática angular e temporal dos membros inferiores, manifesta em aterrissagens de saltos com propulsão bilateral, realizadas sobre um e dois membros inferiores. Aterrissagens unilaterais e bilaterais no plano frontal e sagital de quinze atletas masculinos de voleibol foram registradas e processadas por meio de técnicas videogramétricas. Os resultados indicam que, na aterrissagem unilateral, os atletas apresentaram menor flexão do quadril e joelho, assim como maior valgismo do joelho, quando comparada com a aterrissagem bilateral. A diferença no tempo de aterrissagem entre as duas condutas não foi estatisticamente significante. Em conclusão, os resultados deste estudo apóiam a premissa de que a cinemática dos membros inferiores se altera em função da configuração da conduta motora realizada em homens. Estudos futuros são necessários para explorar o impacto das diferenças cinemáticas encontradas na tensão ligamentar e relacioná-las com os mecanismos de lesões no LCA em homens. Anterior cruciate ligament (ACL) injuries are common in sports. Studies investigating injury mechanisms have demonstrated that most injuries arise from landing tasks. Despite the demonstration of differences between male and female kinematics, there are no studies showing how males behave during different landing tasks. The objective of this study was to compare the angular and temporal kinematics of the lower limbs between two different landing tasks. Double leg and single leg landings were recorded in the frontal and sagittal plane in 15 male volleyball athletes by videogrammetry. Reduced hip and knee flexion and increased knee valgus were observed in the single leg landing task compared to the double leg landing task. No significant difference in landing time was observed between the two tasks. In conclusion, the results support the premise that lower limb kinematics change according to the task performed. Further studies are necessary to explore the impact of these kinematic differences on knee loading and to relate them to ACL injury mechanisms in men.

The main goal of the present study was to identify the ideal number of repetitions for two different intensities for lower limbs power training. Moreover, it was investigated if the rest interval affected power production between sets. Sixteen volunteers were evaluated in three sets of either four or twelve repetitions of half-squat exercise, with loads corresponding to 40% or 60%1RM. Two different rest intervals were attributed between first and second set and second and third set. Power produced when subjects were evaluated at 60% 1RM was significantly greater than at 40% 1RM (p O principal objetivo deste estudo foi identificar o número ideal de repetições para duas diferentes intensidades de treinamento de potência muscular dos membros inferiores. Além disso, investigou-se a influência da duração do intervalo de recuperação entre as séries de estímulos de treinamento sobre a potência produzida. Para isso testou-se um grupo de 16 sujeitos em três momentos, realizando três séries de quatro ou doze repetições no exercício de meio-agachamento, com as cargas de 40% ou 60% de 1RM. Para testar o efeito dos intervalos de recuperação, dois intervalos distintos foram estabelecidos: entre a primeira e a segunda séries e entre a segunda e a terceira séries.
