Abstract

We propose a baryogenenesis mechanism that uses a rotating condensate of a Peccei-Quinn (PQ) symmetry breaking field and the dimension-five operator that gives Majorana neutrino masses. The rotation induces charge asymmetries for the Higgs boson and for lepton chirality through sphaleron processes and Yukawa interactions. The dimension-five interaction transfers these asymmetries to the lepton asymmetry, which in turn is transferred into the baryon asymmetry through the electroweak sphaleron process. QCD axion dark matter can be simultaneously produced by dynamics of the same PQ field via kinetic misalignment or parametric resonance, favoring an axion decay constant fa ≲ 1010 GeV, or by conventional misalignment and contributions from strings and domain walls with fa ∼ 1011 GeV. The size of the baryon asymmetry is tied to the mass of the PQ field. In simple supersymmetric theories, it is independent of UV parameters and predicts the supersymmtry breaking mass scale to be $$ \mathcal{O} $$ O (10 − 104) TeV, depending on the masses of the neutrinos and whether the condensate is thermalized during a radiation or matter dominated era. The high supersymmetry breaking mass scale may be free from cosmological and flavor/CP problems. We also construct a theory where TeV scale supersymmetry is possible. Parametric resonance may give warm axions, and the radial component of the PQ field may give signals in rare kaon decays from mixing with the Higgs and in dark radiation.

309. The Student-Generated Reasoning Tool (SGRT): Linking medical knowledge and clinical reasoning in preclinical education

Abstract

The simultaneous integration of knowledge acquisition and development of clinical reasoning in preclinical medical education remains a challenge. To help address this challenge, the authors developed and implemented the Student-Generated Reasoning Tool (SGRT)—a tool asking students to propose and justify pathophysiological hypotheses, generate findings, and critically appraise information. In 2019, students in a first-year preclinical course (n = 171; SGRT group) were assigned to one of 20 teams. Students used the SGRT individually, then in teams, and faculty provided feedback. The control group (n = 168) consisted of students from 2018 who did not use SGRT. Outcomes included academic performance, effectiveness of collaborative environments using the SGRT, and student feedback. Students were five times more likely to get questions correct if they were in the SGRT group versus control group. Accuracy of pathophysiological hypotheses was significantly lower for individuals than teams. Qualitative analysis indicated students benefited from generating their own data, justifying their reasoning, and working individually as well as in teams. This study introduces the SGRT as a potentially engaging, case-based, and collaborative learning method that may help preclinical medical students become aware of their knowledge gaps and integrate their knowledge in basic and clinical sciences in the context of clinical reasoning.

310. Freeze-in, glaciation, and UV sensitivity from light mediators

Abstract

Dark matter (DM) freeze-in through a light mediator is an appealing model with excellent detection prospects at current and future experiments. Light mediator freeze-in is UV-insensitive insofar as most DM is produced at late times, and thus the DM abundance does not depend on the unknown early evolution of our universe. However the final DM yield retains a dependence on the initial DM population, which is usually assumed to be exactly zero. We point out that in models with light mediators, the final DM yield will also depend on the initial conditions assumed for the light mediator population. We describe a class of scenarios we call "glaciation" where DM freezing in from the SM encounters a pre-existing thermal bath of mediators, and study the dependence of the final DM yield on the initial temperature of this dark radiation bath. To compute DM scattering rates in this cosmology, we derive for the first time an exact integral expression for the Boltzmann collision term describing interactions between two species at different temperatures. We quantify the dependence of the DM yield on the initial dark temperature and find that it can be sizeable in regions near the traditional (zero initial abundance) freeze-in curve. We generalize the freeze-in curve to a glaciation band, which can extend as much as an order of magnitude below the traditional freeze-in direct detection target, and point out that the DM phase space distribution as well as the yield can be strongly dependent on initial conditions., Comment: 31 pages, 5 figures

Abstract

Rotations of an axion field in field space provide a natural origin for an era of kination domination, where the energy density is dominated by the kinetic term of the axion field, preceded by an early era of matter domination. Remarkably, no entropy is produced at the end of matter domination and hence these eras of matter and kination domination may occur even after Big Bang Nucleosynthesis. We derive constraints on these eras from both the cosmic microwave background and Big Bang Nucleosynthesis. We investigate how this cosmological scenario affects the spectrum of possible primordial gravitational waves and find that the spectrum features a triangular peak. We discuss how future observations of gravitational waves can probe the viable parameter space, including regions that produce axion dark matter by the kinetic misalignment mechanism or the baryon asymmetry by axiogenesis. For QCD axion dark matter produced by the kinetic misalignment mechanism, a modification to the inflationary gravitational wave spectrum occurs above 0.01 Hz and, for high values of the energy scale of inflation, the prospects for discovery are good. Abstract

This thesis looks at food policy and its implementation under the notion of modernism as it guides the performance of agricultural services, that are based on science and technology. The research starts from the study of food sovereignty policy established by the State and implemented by its extension agents nationwide, and moves through the regional public institution in charge of agricultural development in Santo Domingo to conclude with the analysis of the policy performance by the State, and what are the effects and affects generated by this public policy implementation. Qualitative material including concepts and quotations involving peasant farmers' practices is presented to establish that traditional practices are imperative to food sovereignty contrary to the policy's implementation. Essentially, this thesis argues that once food sovereignty policy is translated through the state apparatus, it generates intended and/or unintended exclusion among the aimed beneficiaries of such policy.I aim to understand the peasant farmers' agricultural practices during situations of state intervention using ethnographic methods for data collection. This required examining various elements that form the assemblage of milk production in Santo Domingo and understand how these elements interact with each other and with external elements that derive from a food production process.The theoretical approach is explained in Chapter 2, where the main concepts used for this thesis's analysis are described. These are rural development interfaces, intersubjectivity, assemblages, and corporeality. The following chapters depict the empirical work as they focus on the agricultural practices of peasant farmers for milk and dairy products and are analyzed through these concepts.The concept of food sovereignty originates from the rural social movements, and it is gradually inserted into the policy apparatus of the Ecuadorian State, being constitutionalized and legalized for its implementation

Abstract

Despite no apparent defects in T cell priming and recruitment to tumors, a large subset of T cell rich tumors fail to respond to immune checkpoint blockade (ICB). We leveraged a neoadjuvant anti-PD-1 trial in patients with hepatocellular carcinoma (HCC), as well as additional samples collected from patients treated off-label, to explore correlates of response to ICB within T cell-rich tumors. We show that ICB response correlated with the clonal expansion of intratumoral CXCL13+CH25H+IL-21+PD-1+CD4+T helper cells (“CXCL13+TH”) and Granzyme K+PD-1+effector-like CD8+T cells, whereas terminally exhausted CD39hiTOXhiPD-1hiCD8+T cells dominated in nonresponders. CD4+and CD8+T cell clones that expanded post-treatment were found in pretreatment biopsies. Notably, PD-1+TCF-1+(Progenitor-exhausted) CD8+T cells shared clones mainly with effector-like cells in responders or terminally exhausted cells in nonresponders, suggesting that local CD8+T cell differentiation occurs upon ICB. We found that these Progenitor CD8+T cells interact with CXCL13+THwithin cellular triads around dendritic cells enriched in maturation and regulatory molecules, or “mregDC”. These results suggest that discrete intratumoral niches that include mregDC and CXCL13+THcontrol the differentiation of tumor-specific Progenitor exhasuted CD8+T cells following ICB.

Abstract

Introduction: Right Ventricular (RV) dysfunction in pulmonary arterial hypertension (PAH) is the major cause of mortality. Myocardial architecture determines the basis of heart function and can be assessed by generalized Q-space imaging by MRI (GQI), yet remains understudied in the RV. Therefore, we assessed myocardial fiber architecture patterns and correlated them with disease severity in two models of PAH. Methods: PAH was induced in Sprague-Dawley (SD) and Fischer (CDF) male rats by SU5416 (20 mg/kg; s.c.) followed by 3 wks of hypoxia (10% FiO2). Controls received vehicle and were kept at normoxia. After 4 wks, echocardiography and hemodynamics were performed. Whole hearts were resected in some animals (SD n=9, CDF n=8) and imaged via high-angular resolution GQI using a 7T magnet (512 directions, b=750 s/mm2). Patterns of inter-voxel coherence (tractography) were reconstructed, and transmural helix angle distribution (HAD) were assessed at 3 points across the LV and RV (Panel A) and normalized to a common vector. Finite-element models were developed via registering myofiber direction to anatomical mesh. Results: Both animal strains developed PAH, however CDF rats had lower pulmonary artery acceleration time (14±0.6 vs. 19±1.3 ms, p Conclusion: Myoarchitectural analysis of the RV in the setting of PAH reveals that myocardial HAD relates to functional parameters and may be a marker of maladaptation.

Abstract

Introduction: Pulmonary arterial hypertension (PAH) is characterized by severe pulmonary vascular remodeling. Transition of endothelial cells (EC) to mesenchymal cells (EndMT) contributes to vascular remodeling; however, the role of EndMT and underlying mechanisms in PAH remain unclear. While nicotinic acetylcholine (Ach) receptor (nAChR)-mediated pathway regulates epithelial to mesenchymal transition and promotes mesenchymal cell proliferation, its role in EndMT is unknown. In this study, we investigate EndMT in PAH and delineate the mechanisms. Methods: PAH was induced in Sprague Dawley rats by SU5416 (20 mg/kg; s.c.), followed by 3 wks of hypoxia (3 wk PAH) or 3wks of hypoxia with additional 4 wks of normoxia (7 wk PAH). Rats without SU5416 and kept at normoxia served as controls. At the end of experiments, hemodynamic measurements were performed. Lung EC were then isolated and purified using CD31 antibody conjugated beads and passage 3-4 were used. EndMT was assessed by dual staining of EC markers (von Willebrand factor and Griffonia simplicifolia) and α-smooth muscle actin (α-SMA) and by the mRNA expression of EC and mesenchymal genes. Lung ACh was measured by ELISA. Results: PAH rats had elevated PA pressures at both 3 wk (PAH vs. CON in mm Hg: 77.3 vs. 24.8, pIn vitro , treatment with ACh markedly induced EndMT in lung EC from normal rats, which was assessed by increased dual staining of EC markers and α-SMA and increased expression of mesenchymal genes (α-SMA, PAI1, and pro-collagen 1, p Conclusions: EndMT in PAH is associated with increased lung ACh levels and α7nAChR expression in lung ECs. ACh/nAChR-mediated EndMT may contribute to the vascular remodeling in PAH.

Abstract

The LHC BSM Reinterpretation Forum: et al., We report on the status of efforts to improve the reinterpretation of searches and measurements at the LHC in terms of models for new physics, in the context of the LHC Reinterpretation Forum. We detail current experimental offerings in direct searches for new particles, measurements, technical implementations and Open Data, and provide a set of recommendations for further improving the presentation of LHC results in order to better enable reinterpretation in the future. Abstract

Genetic variants are the primary driver of congenital heart disease (CHD) pathogenesis. However, our ability to identify causative variants is limited. To identify causal CHD genes that are associated with specific molecular functions, the study used prior knowledge to filter de novo variants from 2,881 probands with sporadic severe CHD. This approach enabled the authors to identify an association between left ventricular outflow tract obstruction lesions and genes associated with the WAVE2 complex and regulation of small GTPase-mediated signal transduction. Using CRISPR zebrafish knockdowns, the study confirmed that WAVE2 complex proteins brk1, nckap1, and wasf2 and the regulators of small GTPase signaling cul3a and racgap1 are critical to cardiac development.

Abstract

We propose a baryogenenesis mechanism that uses a rotating condensate of a Peccei-Quinn (PQ) symmetry breaking field and the dimension-five operator that gives Majorana neutrino masses. The rotation induces charge asymmetries for the Higgs boson and for lepton chirality through sphaleron processes and Yukawa interactions. The dimension-five interaction transfers these asymmetries to the lepton asymmetry, which in turn is transferred into the baryon asymmetry through the electroweak sphaleron process. QCD axion dark matter can be simultaneously produced by dynamics of the same PQ field via kinetic misalignment or parametric resonance, favoring an axion decay constant $f_a \lesssim 10^{10}$ GeV, or by conventional misalignment and contributions from strings and domain walls with $f_a \sim 10^{11}$ GeV. The size of the baryon asymmetry is tied to the mass of the PQ field. In simple supersymmetric theories, it is independent of UV parameters and predicts the supersymmtry breaking mass scale to be $\mathcal{O}(10-10^4)$ TeV, depending on the masses of the neutrinos and whether the condensate is thermalized during a radiation or matter dominated era. We also construct a theory where TeV scale supersymmetry is possible. Parametric resonance may give warm axions, and the radial component of the PQ field may give signals in rare kaon decays from mixing with the Higgs and in dark radiation., 71 pages, 6 Figures

Abstract

IntroductionAbdominoscrotal hydroceles (ASH) are uncommon occurrences in boys and usually treated similarly to a hernia with the assumption that there is an associated patent processus vaginalis. Treatment in this manner may be challenging due to sac size, extension and adherence to the spermatic cord. Due to the rarity of ASH, the literature is mostly limited to small, single-institution case studies.ObjectiveOur goal was to evaluate two techniques in large number. We hypothesized a simplified scrotal technique with eversion, Jaboulay procedure, would demonstrate less complications and equivalent efficacy to standard excision.MethodsWe retrospectively reviewed medical records at three tertiary children's hospitals to identify boys who underwent surgical repair of ASH between 1998 and 2018. Group 1 had excision and/or ligation of the hydrocele sac. Group 2 had a scrotal incision with limited excision and then eversion of the hydrocele sac (Jaboulay procedure). Variables that were analyzed included preoperative imaging, surgical technique, surgical findings, length of follow up, complications and recurrence of swelling.ResultsWe identified 61 boys, who had 77 abdominoscrotal hydroceles. Group 1 included 38 patients with 48 hydroceles. Group 2 included 23 patients with 29 hydroceles. Complications were more common in Group 1 patients (18% vs 0%) but complication rate and operative time were not statistically associated with surgery type or age. No patient in either group had recurrence of hydrocele.DiscussionAlthough this is a large study for this rare condition, the analysis is limited by number and its retrospective nature.ConclusionFor the rare and difficult to treat abdominoscrotal hydrocele, we were unable to prove with statistical significance that a simplified technique of eversion via the scrotum is safer. However, this large series did demonstrate that the simplified procedure provides equal efficacy as excision.

Abstract

Axions have for some time been considered a plausible candidate for dark matter. They can be produced through misalignment, but it has been argued that when inflation occurs before a Peccei-Quinn transition, appreciable production can result from cosmic strings. This has been the subject of extensive simulations. But there are reasons to be skeptical about the possible role of axion strings. We review and elaborate on these questions, and argue that parametrically strings are already accounted for by the assumption of random misalignment angles.
