The aim of this work was to study the in situ and in vivo nasal absorption of borneol. A novel single pass in situ nasal perfusion technique was applied to examine the rate and extent of nasal absorption of borneol by rats. Experimental conditions, such as perfusion rate, pH and drug concentration, were investigated. The in situ experiments showed that the nasal absorption of borneol was not dependent on drug concentration, and fitted a first order process. The absorption rate constant, Ka, influenced with an increase in perfusion speed. The borneol was well absorbed in the conditions of the nasal cavity within the pH range and pH value of physiological conditions. In vivo studies of borneol absorption were carried out in rats and the pharmacokinetics parameters of intranasal (i.n.) was compared with intravenous (i.v.) administration. The bioavailabilities of borneol was 90.82% for i.n. while Tmax values were 10min. MRT (Mean Residence Time) were 262.55 +/- 67.35 min and 204.22 +/- 14.50 min for i.n. and i.v. methods, respectively. The results demonstrate that borneol could be absorbed promptly and thoroughly by i.n. administration in rats.

Objective: To establish a method for determination of geniposide in Beagle dogs plasma by high performance liquid chromatography (HPLC), and study the pharmacokinetics and bioavailability of geniposide in Beagle dogs after oral administration Xingnaojing., Method: To determine the geniposide in Beagle dogs plasma by HPLC after oral administration or intravenous injection Xingnaojing, and the pharmacokinetic parameters were calculated by the software of Kinetica., Result: The good linearity range of geniposide was 1.24 - 158.88 mg x L(-1). The main pharmacokinetic parameters after oral administration was as follows: Cmax (11.8 +/- 0.6) mg x L(-1), Tmax (52.0 +/- 4.5) min, AUC(1280.8 +/- 172.0) mg x min x L(-1), MRT(118.7 +/- 25.4) min, and these parameters after intravenous injection was follows: Cmax 107.4 +/- 6.3) mg x L(-1), AUC(7930.1 +/- 670.0) mg x min x L(-1), MRT(92.4 +/- 5.1) min. The bioavailability of geniposide in Beagle dogs after oral administration Xingnaojing was (6.46 +/- 0.87)%., Conclusion: The HPLC method had good applicability. The extract recovery, method recovery, intra-day precision and inter-day precision of the method were all met the requirements. The stability in conditions of room temperature and freeze-thaw cycle was good. The results indicated that the oral administration bioavailability of geniposide was in low degree.

Objective: To study the variation in plasma concentration after oral administration of Danshen multi-component osmotic pump tablets in Beagle dogs with conventional Danshen tablets as the control preparation, in order to assess the sustained release of Danshen multi-component osmotic pump tablets., Method: HPLC was adopted for determination, with Shimadzu's ODS-SP column (4.6 mm x 150 mm, 5 microm), flow rate of 1 mL x min(-1), and column temperature of 30 degrees C. Protein in plasma samples of tanshinone II(A) and salvianolic acid B was removed by ethyl acetate extract and acetone precipitation method respectively. Their pharmacokinetic parameters were calculated based on determination results., Result: The methodology study on the two constituents with different polarity showed conformity to the requirements. Compared with conventional Danshen tablets, Danshen multi-component osmotic pump tablets showed decrease in Cmax and extension in Tmax after oral administration in Beagle dogs., Conclusion: Danshen multi-component osmotic pump tablet has a good sustained-release effect compared with conventional Danshen tablets.

Objective: To study the transdermal absorbability of gentiopicroside, naringin and protosappanin B contained in Xuanbi gel plaster., Method: The Franz diffusing cells method was adopted for the in vitro model of rat belly skins. Three indexes, gentiopicroside, naringin and protosappanin B, residued in the accept liquid, skins and plaster were determined by HPLC., Result: The penetration rates of gentiopicroside, naringin and protosappanin B were respectively 3.47, 1.59, 2.13 microg x cm(-2) x h(-1). After 24 h, their penetration rates were 25.42%, 11.73%, 17.78%, respectively. The residual quantities of gentiopieroside, naringin and protosappanin B in skin were 0.231, 0.593, 0.568 microg x cm(-2), ith the retention rates of 0.027%, 0.227%, 0.475%, respectively. The amount of residue of gentiopicroside, naringin and protosappanin B in plaster were 2179, 674, 278 microg, with the retention rates of 81.36%, 81.92%, 73.83%, respectively., Conclusion: The in vitro transdermal behavior of Xuanbi gel plaster is close to a zero-order process. The residual quantity the retention rate in skins is much lower than the penetration rate and the residual rate in plaster.

Objective: To study the characteristics of intestinal absorption of icariin and epimedin C of Xianlinggubao capsules, and compare the absorption of Xianlinggubao capsules prepared by different processes., Method: Non everted gut sac method was applied to investigate the influence of absorption sites and concentration on icariin and epimedin C, which were determined by HPLC., Result: The absorption rate constants of epimedin C in duodenum were absolutely more than that in jejunum and ileum (P < 0.05). The absorption rate constants of icariin in jejunum were absolutely less than that in duodenum and ileum (P < 0.05). The absorption rate constants of epimedin C and icariin kept at the same level when the concentrations of drug solution were at high, middle and low level. The Ka of epimedin C at three levels were 0.040, 0.058, 0.061 h(-1) , respectively, and the Ka of icariin at three levels were 0.002, 0.007, 0.003 h(-1), respectively., Conclusion: Intestinal absorption of icariin and epimedin C is not effected by concentrations. The absorption rate constants of icariin and epimedin C in new Xianlinggubao capsules are higher.

Objective: To investigate the absorption characteristic of borneol., Method: Using single pass perfusion model, the active ingredient of borneol were detected by GC. The drug concentration, perfusion rate and pH value on the absorption of borneol were studied., Result: Perfusion rate on the absorption rate constants (Ka) had significant effects. Drug concentration and pH value on the absorption rate constants had no significant impact., Conclusion: the absorption of borneol is good by nasal. The absorption rate constants of borneol have no effected by drug concentration. The absorption of borneol is via a simple diffusion.

Objective: To study pharmacokinetic of puerarin in rats following different methods of administration of Tongqiao Sanyu prescription., Method: Tongqiao Sanyu prescription was administered to rats by caudal vein injection, nasal administration and oral administration. Plasma samples were extracted with methanol and the plasma concentration of puerarin was analyzed by RP-HPLC. The pharmacokinetic parameters and bioavailability were calculated with Kinetica software., Result: The main pharmacokinetic parameters were as follows: AUC(0-infinity) of caudal vein injection was (787.99 +/- 70.44) mg x min x L(-1); AUC(0-infinity) of nasal administration was (376.56 +/- 93.93) mg x min x L(-1); AUC(0-infinity) and oral administration (The dose was decuple higher than that of caudal vein injection and nasal administration) was (491.18 +/- 110.64) mg x min x L(-1). The absolute bioavailability of puerarin was 47.78% by nasal administration and 6.23% by oral administration., Conclusion: The bioavailability of nasal administration is higher than oral administration significantly, this result can provide some scientific foundantion for the method of administration and the reform of dosage form of Tongqiao Sanyu prescription.

Objective: To develop a GC-FID method for the determination of borneol concentration in rat plasma and to investigate the pharmacokinetics after injection of novel-Xingnaojing., Method: Novel-Xingnaojing was injected via by caudal vein injection. The blood samples were collected by posterior orbital venous plexus approach at 0.5, 1, 3, 5, 8, 12, 20, 30, 45 min. The drug in plasma was extracted with ethyl acetate and then detected by GC-FID, octadecane was used as the internal standard. The pharmacokinetic parameters were calculated by the software of Kinetica., Result: The calibration curve was good linear in the range of 1.67-16.67 mg x L(-1). The extraction recoveries of low, medium and high concentration were (92.81 +/- 1.11)%, (85.38 +/- 0.86)% and (84.58 +/- 0.58)%, respectivley. And the RSDs of within-day and between-day were below 3.00%. Plasma concentration of borneol was consistent with the two-compartment open model. The pharmacokinetic parameters were that the t1/2alpha was (1.18 +/- 0.20) min, the t1/2beta was (22.27 +/- 6.85) min, the C(max)(Calc) was (18.76 +/- 2.10) mg x L(-1), the MRT was (23.84 +/- 7.67) min(-1), and the AUC was (100.00 +/- 15.85) mg x min x L(-1)., Conclusion: The GC-FID method developed can be applied to determination and pharmacokinetics. The borneol in novel-Xingnaojing is distributed and metabolized fast after being administrated.

Objective: To study in vitro release of Xingnaojing microemulsion and to investigate the release mechanism., Method: The concentration of jasminoidin was determined by HPLC and the concentration of Aipian was determined by GC. In vitro release characteristics were conducted by dialysis technique. Model fitting was used to determine the kinetics and mechanism., Result: Jasminoidin released completely within 2 h, fitting the Weibell model best. The release of borneol fitted first order model., Conclusion: The release mechanisms of different types of medicines are quite different. The different types of medicines dissolve in the different phases in the microemulsion.

Objective: To develop a sensitive and specific LC-MS/MS method for determination of Yizhi osmotic pump tablets active ingredient in Beagle dog plasma., Method: Beagle dog plasma pre-treatment methods were established. Geniposide, notoginsenoside R1, ginsenoside Rg1 and Rb1 notoginsenoside molecular ions and fragment ions peaks were separated well and detected synchronously by LC-MS/MS with digoxin as internal standard., Result: Under the selected LC-MS/MS conditions, the characteristic fragment ions of the four components could be well separated and quantified, and the calibration curves showed good linearity within a certain concentration range of each component; extraction recoveries of those four compounds in plasma were higher than 75%, method recoveries were higher than 90%; day precision (RSD <10%) and inter-day precision (RSD <15%) were generally fine; freeze-thaw and room temperature experiments showed good stability (RSD <15%)., Conclusion: The method is proved to be suitable for pharmacokinetic studies of Yizhi active ingredients in Beagle dog plasma.

Objective: To study the characteristics of intestinal absorption of psoralen and isopsoralen of Xianlinggubao capsule, and compare the absorption of Xianlinggubao capsule prepared by different technologies., Method: Non everted gut sac method was applied to investigate the influence of absorption sites and drug concentration on psoralen and isopsoralen absorption, which were determined by HPLC., Result: Although the absorption rate constants of psoralen and isopsoralen in duodenum were more than that in jejunum and ileum, there was no significance difference between them. The absorption rate constants of psoralen kept at the same level when the concentrations of drug solution were at middle and low level, while the absorption rate constant at high level was absolutely lower than them (P < 0.05). The results of isopsoralen were the same as psoralen's., Conclusion: Intestinal absorption of psoralen and isopsoralen may be affected by the dissolution. The absorption rate constants of psoralen and isopsoralen in new Xianlinggubao capsules are higher. The absorptions of active components absorption has significant difference in different preparation processes of Xianlinggubao capsule.

Objective: To establish a sensitive HPLC method for determining the concentrations of paeonol in rat plasma and to evaluate its pharmacokinetic characteristics., Method: The paeonol from eortex Moutan was distilled by the way of water-vapor. A single i.v. dose of 4 mg x kg(-1) paeonol injection was given to 5 health rats. Paeonol was separated on a Diamonsil -C18 column with methanol-water (60: 40)as mobile phase. The plasma concentrations of paeonol were determined and its pharmacokinetic parameters were calculated and evaluated by using kinetica 4.0., Result: The linear range of the method for paeonol was 0.204-20.4 mg x L(-1) and the determination limit was 0.204 mg x L(-1). The main pharmacokinetic parameters, such as AUC, MRT, C(max), Kel, t(1/2kel), after a single dose of paeonol injection were (111.88 +/- 14.44) mg x L(-1) x min(-1), (23.25 +/- 5.86) min, (8.99 +/- 0.84) mg x L(-1), (0.082 +/- 0.015) min(-1) and (8.73 +/- 1.54) min, respectively., Conclusion: The HPLC method for determining paeonol concentration in plasma is simple, rapid, sensitive and suitable for pharmacokinetic studies.

Objective: To study the absorption mechanism of tanshinones in the roots of Salvia miltiorrhiza as well as the interaction between the components in the effective fraction., Method: The model of intestinal absorption in vivo was used and the concentrations of the tanshinones were determined by HPLC, the apparent absorption constant (K(a)) and half life of absorption (t1/2) were obtained, the other components in the extract on the absorption of the there components were also studied., Result: With the increase of dosage of tanshinone II A or cryptotanshinone, K(a) decreased and absorption half life t1/2 increased, the drug absorption rate constant (K(a)) had little significant effect on duodenum, Jejunum and ileum. Tanshinone I has a good absorbing in the rat small intestine. The absorption of tanshinone II A and cryptotanshinone was promoted by the other components in tanshinones extract while tanshinone I' was decreased., Conclusion: When the concentration increases to a certain level, the uptake of tanshinone II A and cryptotanshinone do not increase. The transport mechanism of tanshinone II A and cryptotanshinone in vivo is similar to active transport or facilitated diffusion. They could be well absorbed in general intestinal tract without specific absorption site, and the complex constituents in extract could significant effect the absorption of the pharmaceutical effective constituents.

Objective: To develop a RP-HPLC method for the simultaneous determination of five components including ligustrazine, ferulic acid, butylphthalide, ligustilide and butene-NBP in Rhizoma Chuanxiong, a Chinese herbal medicine., Method: The chromatographic analysis was carried out by using a grace smart RP C18 column with the mobile phase consisted of acetonitrile and 0.1% phosphoric acid (volume fraction), which was in gradient elution. A DAD detector was used to detect the components. The peak area was chosen under their maximum absorption wavelength for different components, and quantitated by using the external standard method., Result: The calibration curves were linear within the range of 0.008 35-0.668, 0.020 6-1.648, 0.012 2-0.976 , 0.050 75-4.06, 0.015 7-1.256 microg for ligustrazine, ferulic acid, butylphthalide, ligustilide and butene-NBP, respectively. The average recoveries of the five components were 101.98%, 99.91%, 96.94%, 100.85% and 99.04%, respectively., Conclusion: This method is simple, quick, reproducible, with high recovery, and has been successfully applied to the simultaneous determination of the five components in Rhizoma Chuanxiong. This method can be used to control the quality of Rhizoma Chuanxiong and its preparations.

Objective: To establish a method to determine the dissolution of Tanshinone IIA in xianling gubao capsules, and compare the dissolution of Xianling Gubao capsules prepared by different processes., Method: The small cup method was used at a rotary speed of 100 r x min(-1), the dissolubility was inspected in the following 6 mediums: (1) water; (2) 0.1 mol x L(-1) hydrochloric acid solution; (3) pH 6.8 phosphate buffer; (4) pH 6.8 phosphate buffer solution + 0.5% cholic acid; (5) pH 6.8 phosphate buffer solution + 1.0% cholic acid; (6) pH 7.6 phosphate buffer solution + 4. 0% cholic acid. A HPLC method was used to determine the dissolution samples of tanshione IIA in different dissolution mediums. The accumulative dissolution percentages were calculated, and the resemblance of release curves were compared by similarity factors (f2)., Result: The accumulative dissolution percentage was the highest while pH 7.6 phosphate buffer solution + 4.0% cholic acid was used as medium. The similarity factor values (f2) of tanshinone IIA dissolution curves of different preparation processes of Xianling Gubao capsules were all less than 50., Conclusion: There is significant difference in tanshinone IIA dissolution between Xianling Gubao capsules of different preparation processes. The accumulative dissolution percentages of the tanshinone IIA in new Xianling Gubao capsules is higher than that in the capsules prepared by other preparation processes.

Objective: To develop an HPLC method for the determination of plasma concentration of jasminoidin and study the pharmacokinetics of jasminoidin in rabbits administered Xingnaojing naristillae by nasal medication., Method: After sampling blood from the left arteria carotis of rabbits which were administered Xingnaojing naristillae medication by nasal by 12 mg x kg(-1) (counted by gardenia extract) at 1, 3, 5, 10, 20, 30, 45, 60, 90, 120, 240 min, the plasma samples were dealt with acetonitrile precipitation and HPLC was used to determine the plasma concentration of jasminoidin. The pharmacokinetic parameters were computed by Kinetica software., Result: The calibration curve was linear (r = 0.999 6) within the range of 0.136 5-2.73 mg x L(-1) for jasminoidin in plasma. The average recovery was (97.14 +/- 3.78)%, (95.06 +/- 2.95)%, (91.50 +/- 1.82)%. The within-day and between-day precision met the requirements, because the RSD were both less than 4%. Jasminoidin was fitted to a two-compartment open pharmacokinetic model in rabbits. The mainly pharmacokinetic parameters were: C(max) = (2.013 +/- 0.563) mg x L(-1), T(max) = (6.405 +/- 1.764) min, K(e) = (0.032 5 +/- 0.013 3) min(-1), CL = (0.059 3 +/- 0.0246) L x min(-1) x kg(-1), AUC = (116.89 +/- 50.19) mg x min(-1) x L(-1), MRT = (84.447 +/- 19.420) min., Conclusion: The method can be used to determine the concentration and to investigate the pharmacokinetics of jasminoidin in rabbits. Jasminoidin was absorbed rapidly by nasal medication and has a good perspective.

Objective: To prepare puerarin deoxycholate/phospholipid mixed micelle to increase the solubility of puerarin., Method: Sodium deoxycholate and soybean phospholipids were used to prepare puerarin mixed micelle through orthogonal design experiments. With the solubility, shape and particle size as the response indexes, the preparing process of puerarin mixed micelle was optimized., Result: The optimized process for the puerarin deoxycholate/phospholipid mixed micelle was that the puerarin, soya phosphatidylcholine and sodium deoxycholate with the mole ratio of 3:2:4 should be dissolved in methanol-chloroform (1:1), and the solvents should be evaporated rotatively at 30 degrees C. The particle diameter of the mixed micelle was (64.8 +/- 13) nm (volume-weighted particle size distribution), and the solubility was 0.811 1 g x L(-1) in water at the room temperature, which was 22.3 times as that of the raw puerarin (0.036 4 g x L-1)., Conclusion: The puerarin deoxycholate/phospholipid mixed micelle can improve the solubility of puerarin significantly.

Objective: To study the dissolution characteristics of seven active components of Xianlinggubao capsules in vitro, and compare the dissolution of Xianlinggubao capsules prepared by different processes., Method: The dissolution of Xianlinggubao capsules was determined by small cup method with the rotating speed of 100 r x min(-1) and phosphate buffer (pH 6.8) as dissolution medium. The different dissolution rates in vitro of seven kinds of active components (icariin, epimedin C, asperosaponin VI, psoralen, isopsoralen, salvianolic acid B and tanshinone IIA) of Xianlinggubao capsules were investigated. The contents of active components were determined by HPLC. The accumulative dissolution percentages were calculated, and the resemblance of release curves were compared by similarity factors (f2)., Result: The results of determination showed that the similarity factor values (f2) of the dissolution curves of seven active components after different preparation processes were all less than 50., Conclusion: There is significant difference in dissolution of active components between Xianlinggubao capsules of different preparation processes. The accumulative dissolution percentages of the active components in new Xianlinggubao capsules are higher than that of Xianlinggubao capsules.

Systemic mastocytosis with associated clonal haematological non-mast cell lineage disease (SM-AHNMD) is a heterogeneous group of mast cell disorders with different clinical, pathologic and underlying molecular characteristics. While myelomonocytic/myeloid neoplasia overwhelmingly predominates the AHNMD component, lymphoproliferative disorders rarely occur as an AHNMD component of SM-AHNMD. Here we report two cases of SM-AHNMD, in which the AHNMD component is chronic lymphocytic leukemia in one case, and concurrent chronic lymphocytic leukemia as well as plasma cell myeloma in another case. To the best of our knowledge, this is the first case report of SM-AHNMD with chronic lymphocytic leukemia and plasma cell dyscrasia simultaneously.

Objective: To determine the O/W partition coefficient of panax pseudo-ginseng saponin (PNS) and investigate the absortion kinetics of it in whole small intestine and different intestinal segments of rats., Method: The shake-flask method was employed to determine the O/W partition co-efficient of geniposide, and an in situ intestinal perfusion model was employed to investigate the absorptive kinetics of geniposide., Result: The partition coefficient (P) of R1, Rg1 and Rb, of PNS were 1.0814, 6.3104 and 0.2743, respectively, and their logP were 0.0340, 0.8001, -0.5618, the absorptive rate constants (Ka) of R1, Rg1 and Rb1 of PNS at the concentration of 0.2, 0.5, 1.0 g x L(-1) were (0.135 +/- 0.006), (0.110 +/- 0.002), (0.095 +/- 0.016), (0.144 +/- 0.015), (0.110 +/- 0.006), (0.099 +/- 0.011), (0.238 +/- 0.013), (0.140 +/- 0.008), (0.137 +/- 0.012)h(-1), respectively. The Kb of R1, Rg1 and Rb1 of PNS were (0.030 +/- 0.006), (0.033 +/- 0.004), (0.033 +/- 0.007), (0.032 +/- 0.006), (0.044 +/- 0.012), (0.044 +/- 0.011), (0.042 +/- 0.007), (0.065 +/- 0.007), (0.044 +/- 0.014)h(-1) at duodenum, jejunum, ileum, respectively. The absorption rate of Rb1 was higher than R1 and Rg1., Conclusion: According to the P and the logP, it can be conjectured that the absorption of R1 and Rg1 are better than Rb1. The absorption rate is decreased with the increase of the PNS concentration. Their absorption is the passive diffusion mechanism and other transport may also take part in the transport process. PNS is absorbed at all small-intestinal segments of rats, there are no significant differences between the three sections.

Objective: To observe the influence of ingredients in Xingnaojing,such as moschus, borneol and radix curcumae on intestine absorption kinetics of gardenia extract., Method: An in situ intestinal perfusion model of rats was employed to investigate the absorption of geniposide in gardenia extract., Result: While gardenia extract was administered solely, the absorptive rate constant (K) of geniposide was (0.055 +/- 0.006) h(-1); But while the extract was co-administered with radix curcumae,moschus and borneol, the K were (0.060 +/- 0.001), (0.066 +/- 0.008), (0.072 +/- 0.010) h(-1), respectively. The K was (0.076 +/- 0.011) h(-1) while the extract in total formulation for Xingnaojing., Conclusion: The K, while the extract is co-administered with borneol or total formulation is significantly higher than solely used. Borneol and complex prescription can significantly increase the intestinal absorption of geniposide in gardenia extract.

Objective: To study the in vitro mucosal permeation properties of paeonol and the factors that affect such permeability., Method: Bullfrog skin was selected as the permeation model, and the modified Franz diffusion cell was adopted to investigate the percutaneous adsorption in vitro. Content of paeonol was quantified by HPLC. Kinds of receiving solution, parts of bullfrog skin and impacts of drug concentration on percutaneous adsorption in vitro were systematically studied., Result: Percutaneous permeation of paeonol was to some extent affected by osmotic pressure and pH value of the receiving solution. Abdominal skin, lateral abdominal skin and dorsal skin, with the permeability coefficient form high to low, had great influence on the permeation of paeonol. There existed a good percutaneous permeation of paeonol. And the relationship between the cumulative permeability rate and time was coincided with the first order kenetical process., Conclusion: Take the in vitro abdominal skin of bullfrog as the permeation model, some similarities are revealed between in vitro percutaneous permeation of paeonol and mucosal adsorption.

Objective: To optimize the extraction technology for Psoralea corylifolia and Salvia miltiorrhiza., Method: An orthogonal test was adopted in this study. The contents of Tanshinone IIA, Salvianolic acid B, Psoralen and Isopsoralen were used as indices of appraisal. Data processing was carried out with the multiple guidelines grading method for optimizing the extraction condition., Result: The best extraction technology was to extract for 2 times with 8 fold ethanol solution (70%) and 1.5 h each time., Conclusion: The optimized extraction technology was rational and the extraction rate of the active principles is high.

Objective: To determine the O/W partition coefficient of geniposide in fructus gardeniae extract and investigate the absortion kinetics of geniposide in whole small intestine and different intestinal segments of rats., Method: The shake-flask method was employed to determine the O/W partition coefficient of geniposide; an in situ intestinal perfusion model was employed to investigate the absorptive kinetics of geniposide., Result: The partition coefficient (P) of geniposide was 0.1077, logP was -0.9678; the absorptive rate constants (K) of geniposide at the concentration of 0.078, 0.311, 0.780 g x L(-1) were (0.130 +/- 0.007), (0.056 +/- 0.003), (0.031 +/- 0.006) h, respectively. The K of geniposide were (0.019 +/- 0.003), (0.015 +/- 0.002), (0.012 +/- 0.002) h at duodenum, jejunum, ileum, respectively., Conclusion: According to the P and the logP, it could be indicated that the absorption of geniposide at small intestine was poor absorption; The absorption rate was increased with the decrease of the extract concentration; Their absorption was first-order process besides the passive diffusion mechanism, and facilitated diffusion and active transport may also take part in the transport process. Geniposide was absorbed at all smallintestinal segments of rats, but the duodenal absorption ratewas higher more than other section.

Objective: To observe the influence of natural borneol and synthetic borneol on mucosal permeability of Gardenia extract., Method: Taken frog skin as a vitro model to study the vitro mucosal permeation the impacts of the natural borneols and synthetic borneols on the P(app) of the Jasminoidin were studied, and the effect of different borneols on the stability of Jasminoidin were investigated. Compared the 10 h accumulated infiltration rate of each group the effects of influence factors,such as C(Ge), C(B) and rotation speed on P(app) were investigated by using response surface method., Result: The P(app) of Jasminoidin of natural borneol and synthetic borneol group were 1.44 fold and 1.77 fold of control group (P < 0.01). For two borneol groups, the results also showed a significant difference too (P < 0.05). Jasminoidin began to degrade about 8 h after the effect of frog skin for control group and synthetic borneol group, but was stable within 12 h for natural borneol group. The accumulated permeation rate of 10 h was same for different borneol groups. It was about 1.3 fold of control group. The C(Ge) had a salinence influence on the P(app) (P < 0.01) and C(B) had a salience influence on time-lag (P < 0.01)., Conclusion: Both the natural borneol and synthetic borneol can accelerate the permeation of Jasminoidin and the synthetic borneol has stronger effect on the P(app). Both two different borneol can reduce the degradation effect of frog skin to Jasminoidin, but the natural borneol has a better protect effect on it. By using more natural borneol, the mucosal permeability of Gardenia extract can be increased, the time-lag can be reduced, and Jasminoidin has better stability.

Objective: To study the factors that affect mucosal absorption of gardenia extract., Method: Take vitro frog skin as a model to study the vitro mucosal permeation. The impacts of the osmotic pressure and the pH value of permeation medium on the Papp of the Jasminoidin were studied, and the effect of frog skin on the stability of Jasminoidin was investigated also., Result: The Papp of Jasminoidin were (0.53 +/- 0.01), (0.21 +/- 0.05), (0.44 +/- 0.12), (0.42 +/- 0.13), (0.26 +/- 0.03) cm x min(-1) by using the normal saline (pH 6.88), pure water, 1.8 % NaCl solution, normal saline (pH 4.05) and normal saline (pH 10.05) as permeation medium for each. The accumulated permeation rate was (55.69 +/- 9.81)% by 12 h, using normal saline as permeation medium respectively, and there was no obvious time lag. Jasminoidin began to degrade around 8 h by affectedof frog skin, the constant of degradation rate (K) was 1.999, and the t1/2 was 0.347 h., Conclusion: The mucosal permeability of gardenia extract by using the vitro model of frog skin is good, and consistent with zero level absorption process. The osmotic pressure and pH value significantly affected the permeation and the isotonic and partial neutral permeation medium are more conducive to the permeation and absorption of Jasminoidin. The degradation effect of frog skin to the Jasminoidin will not affect mucosal permeation research. In vitro model of frog skin is a suitable way to simulate mucosal permeation process of the gardenia extract.

Musk is one of the rare traditional Chinese medicinal materials and is usually sophisticated with different adulterants. Some new adulterants have been detected by GC-MS, SPME and HS2000.

We describe a noninvasive, quantitative, and tomographic method to visualize lymphocytes within the whole animal. We used positron-emission tomography (PET) to follow the localization of adoptively transferred immune T lymphocytes. Splenic T cells from animals that had rejected a Moloney murine sarcoma virus/Moloney murine leukemia virus (M-MSV/M-MuLV)-induced tumor were marked with a PET reporter gene, injected into tumor-bearing mice, and imaged in a microPET by using a substrate specific for the reporter. Specific localization of immune T cells to the antigen-positive tumor was detected over time, by sequential imaging of the same animals. Naive T cells did not localize to the tumor site, indicating that preimmunization was required. Autoradiography and immunohistochemistry analysis corroborated the microPET data. The method we have developed can be used to assess the effects of immunomodulatory agents intended to potentiate the immune response to cancer, and can also be useful for the study of other cell-mediated immune responses, including autoimmunity.
