301. Effects of Morphine in Decomposing Bodies on the Development of Lucilia sericata(Diptera: Calliphoridae)

Abstract

This study concerns the effects of morphine in tissues on the rate of development of Lucilia sericata(Diptera: Calliphoridae) using those tissues as a food source. Lucilia sericatais a species of fly commonly found on human corpses in Europe during the early stages of decomposition and thus of forensic interest. Three rabbits were administered 12.5, 25.0 and 50.0 mg/h of morphine chlorhydrate via ear perfusion over a period of 3 h. These dosages and duration of perfusion were calculated to give tissue concentrations of morphine similar to those encountered in fatal human overdoses. A fourth rabbit was used as a control. Following administration of the drug, rabbits were sacrificed and 400 eggs of Lucilia sericata, all of the same age, were placed in the eyes, nostrils and mouth of each rabbit. Developing larvae were sampled daily to determine growth rate and weight. Puparia and emerging adult flies were also sampled. Data were analyzed using analysis of variance (ANOVA) and Student's T-test. Results of this study show that an underestimation of the postmortem interval of 24 h is possible if the presence of morphine in tissues is not considered. This study demonstrates again the necessity of considering the possible effects of drugs in tissues on insect growth rates when estimating the postmortem interval using entomological techniques.

302. Morphine Perfused Rabbits: A Tool for Experiments in Forensic Entomotoxicology

Abstract

In order to establish an animal model for entomotoxicological studies, the kinetics of morphine elimination from blood after a single intravenous injection of morphine and the concentrations of morphine in tissues following a continuous perfusion were studied. The aim of these experiments was to obtain controlled morphine tissue concentrations similar to those encountered in fatal human heroin overdoses. These tissues can be used as a food source for developing fly larvae in entomotoxicological studies. In the single injection experiment, seven rabbits were administered 1 or 2 mg/kg body weight of morphine chlorhydrate via the main ear artery. Blood samples of 200 μL were removed regularly via a catheter. Morphine concentration was determined using RIA techniques. Morphine was found to be first rapidly distributed and then slowly eliminated, following a two-exponential equation. Elimination of morphine from blood can be described as a two-compartment model. Constants of the equation were determined using the Kaleidagraph® program. Using those constants, the main pharmacokinetics parameters were calculated. Results of these parameters showed the following: clearance from 13.3 to 16.2 L.h-1, half-life of the distribution phase from 0.6 to 0.9 min, and half-life of the elimination phase from 21 to 26 min. These results were used to calculate the rate of perfusion of morphine for rabbits to obtain desired, controlled, and constant concentrations of morphine in tissues. In the second experiment, three rabbits received a perfusion of morphine intravascularly at a rate of 2 mg/kg/h for a period of 3 h. These rabbits were sacrificed and analyses performed on several abdominal and thoracic organs. Results showed that the concentrations of morphine differed according to the organ analyzed, but were reproducible for organs between animals. These concentrations were similar to those normally encountered in cases of human death due to heroin overdoses.

Abstract

The Micro Vertex Detector (MVD) of the future Compressed Baryonic Matter (CBM) experiment at FAIR will have to provide a spatial precision of ∼ 5 μ m in combination with a material budget of 0. 3 % – 0. 5 % X 0 for a full detector station. Simultaneously, it will have to handle the rate and radiation load of operating the fixed target experiment at an average collision rate of 100 kHz (4 – 10 AGeV Au+Au collisions) or 10 MHz (up to 28 GeV p-A collisions). The harsh requirements call for a dedicated detector technology, which is the next generation CMOS Monolithic Active Pixel Sensor MIMOSIS. We report about the requirements for the sensor, introduce the design approach being followed to cope with it and show first test results from a first sensor prototype called MIMOSIS-0. Abstract

The CBM experiment will investigate heavy-ion collisions at beam energies from 8 to 45 AGeV at the future accelerator facility FAIR. The goal of the experiment is to study the QCD phase diagram in the vincinity of the QCD critical point. To do so, CBM aims at measuring rare probes among them open charm. In order to identify those rare and short lived particles despite the rich combinatorial background generated in heavy ion collisions, a micro vertex detector (MVD) providing an unprecedented combination of high rate capability and radiation hardness, very light material budget and excellent granularity is required. In this work, we will discuss the concept of this detector and summarize the status of the R&D.

Abstract

The European physical journal / C 81(5), 397 (2021). doi:10.1140/epjc/s10052-021-09135-3, The physics goal of the strong interaction program of the NA61/SHINE experiment at the CERN Super Proton Synchrotron (SPS) is to study the phase diagram of hadronic matter by a scan of particle production in collisions of nuclei with various sizes at a set of energies covering the SPS energy range. This paper presents differential inclusive spectra of transverse momentum, transverse mass and rapidity of $\pi ^{-}$ mesons produced in central${}^{40}$Ar+${}^{45}$Sc collisions at beam momenta of 13A, 19A, 30A, 40A, 75A and 150$A\,\text{ Ge }\text{ V }\!/\!\textit{c}$. Energy and system size dependence of parameters of these distributions – mean transverse mass, the inverse slope parameter of transverse mass spectra, width of the rapidity distribution and mean multiplicity – are presented and discussed. Furthermore, the dependence of the ratio of the mean number of produced pions to the mean number of wounded nucleons on the collision energy was derived. Abstract

Measurements of multiplicity and transverse momentum fluctuations of charged particles were performed in inelastic p+p interactions at 20, 31, 40, 80, and 158 GeV /c beam momentum. Results for the scaled variance of the multiplicity distribution and for three strongly intensive measures of multiplicity and transverse momentum fluctuations Δ[P T ,N], Σ[P T ,N] and Φ p T are presented. For the first time the results on fluctuations are fully corrected for experimental biases. The results on multiplicity and transverse momentum fluctuations significantly deviate from expectations for the independent particle production. They also depend on charges of selected hadrons. The string-resonance Monte Carlo models Epos and Urqmd do not describe the data. The scaled variance of multiplicity fluctuations is significantly higher in inelastic p+p interactions than in central Pb+Pb collisions measured by NA49 at the same energy per nucleon. This is in qualitative disagreement with the predictions of the Wounded Nucleon Model. Within the statistical framework the enhanced multiplicity fluctuations in inelastic p+p interactions can be interpreted as due to event-by-event fluctuations of the fireball energy and/or volume.

Abstract

The production cross section of 30.92 GeV/c protons on carbon is measured by the NA61/SHINE spectrometer at the CERN SPS by means of beam attenuation in a copy (replica) of the 90-cm-long target of the T2K neutrino oscillation experiment. The employed method for direct production cross-section estimation minimizes model corrections for elastic and quasi-elastic interactions. The obtained production cross section is σ$_{prod}$ = 227.6 ± 0.8(stat)$\frac{+1.9}{−3.2}$(sys) − 0.8(mod) mb. It is in agreement with previous NA61/SHINE results obtained with a thin carbon target, while providing improved precision with a total fractional uncertainty of less than 2%. This direct measurement is performed to reduce the uncertainty on the T2K neutrino flux prediction associated with the re-weighting of the interaction rate of neutrino-yielding hadrons.

Abstract

The European physical journal / C 81(2), 961 (1-21) (2021). doi:10.1140/epjc/s10052-020-08514-6, The NA61/SHINE collaboration studies at the CERN Super Proton Synchrotron (SPS) the onset of deconfinement in hadronic matter by the measurement of particle production in collisions of nuclei with various sizes at a set of energies covering the SPS energy range. This paper presents results on inclusive double-differential spectra and mean multiplicities of $\pi ^{-}$ mesons produced in the 5% most central$^7$Be + $^9$Be collisions at beam momenta of 19A, 30A, 40A, 75A and 150$A\,\text{ GeV }\!/\!c$ obtained by the so-called $h^-$ method which does not require any particle identification. The shape of the transverse mass spectra differs from the shapes measured in central Pb + Pb collisions and inelastic p+p interactions. The normalized width of the rapidity distribution decreases with increasing collision energy and is in between the results for inelastic nucleon–nucleon and central Pb + Pb collisions. The mean multiplicity of pions per wounded nucleon in central$^7$Be + $^9$Be collisions is close to that in central Pb + Pb collisions up to 75$A\,\text{ GeV }\!/\!c$. However, at the top SPS energy the result lies between those for nucleon–nucleon and Pb + Pb interactions. Abstract

Physical review / D 103(1), 012006 (1-13) (2021). doi:10.1103/PhysRevD.103.012006, The production cross section of 30.92 GeV/c protons on carbon is measured by the NA61/SHINE spectrometer at the CERN Super Proton Synchrotron by means of beam attenuation in a copy (replica) of the 90-cm-long target of the T2K neutrino oscillation experiment. The employed method for direct production cross-section estimation minimizes model corrections for elastic and quasielastic interactions. The obtained production cross section is σprod=227.6±0.8(stat)-3.2+1.9(sys)−0.8(mod) mb. It is in agreement with previous NA61/SHINE results obtained with a thin carbon target, while providing improved precision with a total fractional uncertainty of less than 2%. Abstract

Uptake of 123I-Tyr-3-octreotide (TOCT) by hormone-secreting abdominal tumors was studied to compare scintigraphic observations with the reduction in hormone levels brought about by a brief therapeutic test.A prospective study was conducted on 17 patients, totalizing 46 proven lesions, with endocrine tumors of the pancreas (10 patients, 20 lesions) and/or carcinoid metastases (8 patients, 26 lesions). Tumor hormonal hypersecretion was inhibited by octreotide.There was good agreement between the results of these examinations.The detection of abdominal tumors using this radiotracer is strongly related to its functional characteristics. Variations in the scintigraphic and test results according to different tumor types were in agreement with published data on the density of somatostatin receptors measured by in vitro studies or scintigraphy and by the therapeutic effects of octreotide.

Abstract

The presence of somatostatin receptors on carcinoid tumors mediates imaging of tumor extent and inhibition of tumor growth and marker secretion. This prospective study aimed to evaluate radiolabeled somatostatin analog scans in the therapeutic workup of carcinoids.Twenty-one patients with carcinoids underwent 26 scans with iodine octreotide or indium pentetreotide. The results for tumor and metastases imaging were analyzed and compared with those of a short inhibition test of marker secretion and with those of metaiodobenzylguanidine scan.The sensitivity for obtaining images of the overall 43 tumor sites was 72%. We had no false-positive results. Unknown tumor sites were discovered in three patients. The results were slightly better with indium pentetreotide and metastases imaging. A positive scan did not always preclude responsiveness to the functional effect of octreotide. Results of somatostatin analog scans were better than those with metaidobenzylguanidine. The two techniques were complementary in one patient.The choice of treatment for patients with carcinoid tumors should benefit from functional inhibition test with octreotide and from indium pentetreotide and metaidobenzylguanidine scans.

Abstract

The European physical journal / C 81(5), 384 (1-19) (2021). doi:10.1140/epjc/s10052-021-09107-7, Measurements of multiplicity fluctuations of identified hadrons produced in inelastic p+p interactions at 31, 40, 80, and 158 $\text {Ge}\text {V}/c$ beam momentum are presented. Three different measures of multiplicity fluctuations are used: the scaled variance $\omega $ and strongly intensive measures $\Sigma $ and $\Delta $. These fluctuation measures involve second and first moments of joint multiplicity distributions. Data analysis is preformed using the Identity method which corrects for incomplete particle identification. Strongly intensive quantities are calculated in order to allow for a direct comparison to corresponding results on nucleus–nucleus collisions. The results for different hadron types are shown as a function of collision energy. Abstract

This study concerns the determinations of morphine concentrations in fly larvae reared on rabbits administered different concentrations of morphine and a correlation between concentrations of the drug in larvae and tissues. Three rabbits (R1, R2 and R3) were given dosages of 12.5, 25.0 and 50.0 mg/h of morphine over a 3 h period via continuous ear artery perfusion. These dosages and time of perfusion were calculated to create tissue concentrations of morphine similar to those encountered in human deaths due to overdose. Morphine blood level plateau was attained after 1 h of perfusion. A fourth rabbit was used as a control. To evaluate drug concentrations, tissues were sampled using a coelioscopic technique. Approximately 400 eggs of Lucilia sericata, all of the same age category, were placed in eyes, nostrils and mouth of each rabbit carcass. Larvae and puparia were regularly collected from each rabbit for toxicological analysis. The concentrations of the drug in the tissues sampled were determined to be similar to those normally encountered in human overdoses and were correlated with the dosage of morphine that had been administered. Morphine was detected in all larvae and pupae fed on tissues from carcasses administered morphine, except for puparia from the colony fed on the R1 animal which received 12.5 mg/h dosage of morphine. All samples from the control rabbit were negative for morphine. Concentrations of morphine in larvae reared on rabbit carcasses containing morphine were 30 to 100 times lower than the concentrations found in the tissues. A correlation between the tissue concentrations and larval concentrations was found in only 3rd instar larvae (80 to 140 h following hatching). No correlations were found between administered dosages, tissue concentrations and younger larvae, prepuparial larvae or puparia.

Abstract

The study of open charm meson production provides an efficient tool for the investigation of the properties of hot and dense matter formed in nucleus–nucleus collisions. The interpretation of the existing di-muon data from the CERN SPS suffers from a lack of knowledge on the mechanism and properties of the open charm particle production. Due to this, the heavy-ion programme of the NA61/SHINE experiment at the CERN SPS has been extended by precise measurements of charm hadrons with short lifetimes. A new Vertex Detector for measurements of the rare processes of open charm production in nucleus–nucleus collisions was designed to meet the challenges of track registration and high resolution in primary and secondary vertex reconstruction. A small-acceptance version of the vertex detector was installed in 2016 and tested with Pb + Pb collisions at 150 A GeV / c . It was also operating during the physics data taking on Xe + La and Pb + Pb collisions at 150 A GeV / c conducted in 2017 and 2018. Abstract

Neriifolin is a cardenolide structurally closed to digoxin and is contained in the fruit kernel of the red-eye-sea mango tree (Cerbera manghas), on which the coconut crab feeds. We previously reported lethal neriifolin poisonings following the consumption of New Caledonian coconut crabs Birgus latro(Maillaud etal., Toxicon 2010). This led us to consider digoxin-specific Fab antibody fragments (Fab) as a specific treatment. This presentation focuses on two cases of successful Fab use.

Abstract

The ultra-thin and highly granular CMOS Monolithic Active Pixel Sensors (MAPS) are typically optimized for high rate high precision tracking, which implies the use of a very thin active medium and digital readout. Both features hamper using the devices for identifying low momentum particles by means of dE/dx. Still, MAPS feature charge sharing and typically clusters of more than one fired pixel per impinging particles are formed. It was previously shown that the number of fired pixels per cluster scales with the dE/dx, which allowed identifying highly ionizing nuclear fragments [1]. Assuming a sufficiently strong response to different dE/dx, this approach could also be considered for distinguishing minimum ionizing particles (MIP) from light fragments like alpha particles in tracking detectors. In this work, we study this response with particle beams with a dE/dx of up to four times the ones of MIPs, for non-irradiated and irradiated chips, with different sensing nodes as implemented in the MIMOSIS-1 prototype used for the vertex detector of the CBM experiment. Abstract

Background Although EGFR mutant tumors exhibit low response rates to immune checkpoint blockade overall, some EGFR mutant tumors do respond to these therapies; however, there is a lack of understanding of the characteristics of EGFR mutant lung tumors responsive to immune checkpoint blockade. Patients and methods We retrospectively analyzed de-identified clinical and molecular data on 171 cases of EGFR mutant lung tumors treated with immune checkpoint inhibitors from the Yale Cancer Center, Memorial Sloan Kettering Cancer Center, University of California Los Angeles, and Dana Farber Cancer Institute. A separate cohort of 383 EGFR mutant lung cancer cases with sequencing data available from the Yale Cancer Center, Memorial Sloan Kettering Cancer Center, and The Cancer Genome Atlas was compiled to assess the relationship between tumor mutation burden and specific EGFR alterations. Results Compared with 212 EGFR wild-type lung cancers, outcomes with programmed cell death 1 or programmed death-ligand 1 (PD-(L)1) blockade were worse in patients with lung tumors harboring alterations in exon 19 of EGFR (EGFRΔ19) but similar for EGFRL858R lung tumors. EGFRT790M status and PD-L1 expression did not impact response or survival outcomes to immune checkpoint blockade. PD-L1 expression was similar across EGFR alleles. Lung tumors with EGFRΔ19 alterations harbored a lower tumor mutation burden compared with EGFRL858R lung tumors despite similar smoking history. Conclusions EGFR mutant tumors have generally low response to immune checkpoint inhibitors, but outcomes vary by allele. Understanding the heterogeneity of EGFR mutant tumors may be informative for establishing the benefits and uses of PD-(L)1 therapies for patients with this disease. [ABSTRACT FROM AUTHOR]

Abstract

Abstract: A new type of active pixel sensors (APSs) to track charged particles for particle physics experiments or to count number of electrons that cross any pixel at the focal plane of electron microscopes is described. The electric field of desirable shape is created inside the active volume of the pixel introducing the drift component in the movement of the signal electrons towards charge collecting electrodes. The electric field results from the flow of ∼100mA/cm2 hole currents within individual pixels of the sensor. The proposed sensor is produced using a standard industrially available complementary metal oxide silicon (CMOS) process. There are two main advantages of the proposed detectors when compared to the present (February 2005) state-of-the-art, i.e. field-free APS sensors. The first advantage of a field-assisted transport mechanism is the reduction of the charge collection time and of the sharing of the signal electrons between adjacent pixels by diffusion. The second advantage is the freedom to use both kinds of MOS transistors within each pixel of the sensor. Thus, the full functional power of CMOS circuits can be embedded in situ. As an example, 16-bit scalers will be implemented in each pixel of the sensor for electron microscopy. The reduced collection time combined with the state-of-the-art electronics within each pixel provides the most complete information about the position and the timing of incident charged particles for particle physics experiments. Position resolution of new sensors was computationally simulated to be a few microns, that is, the same as the resolution of standard APSs. Moreover, the active depth of the sensor and the associate electronics is less than about 20μm and a thinned down sensor together with its beryllium backing can have a total thickness of less than 0.1% of one radiation length. The reduction of the thickness of the detector reduces the amount of multiple scattering within the detector. The determination of the momentum and of the origin of a particle can be improved in particle physics experiments and images from electron microscopes can be sharper. [Copyright &y& Elsevier]

Contemporary theoretical discussions of exploitation are dominated by thinkers in the liberal and Marxian traditions. Exploitation: From Practice to Theory, pushes past these traditional and binary explanations, to focus on unjust practises that both depend on and perpetuate inequalities central to exploitation. Using real-world examples, the chapters in this collection address key questions, including, in what ways are exploitation practices globalised, racialized and gendered? How do cases of organ selling, price gouging and commercial gestational surrogacy change our understandings of exploitation? What possible social and economic remedies do these new conceptions prescribe? Abstract

Purpose: We present a phase I/II first-in-human trial evaluating the safety and efficacy of 50 mg and 200 mg doses of linvoseltamab, a B-cell maturation antigen × CD3 bispecific antibody in relapsed/refractory multiple myeloma (RRMM)., Methods: Phase II eligible patients had RRMM that either progressed on/after ≥three lines of therapy including a proteasome inhibitor (PI), an immunomodulatory drug (IMiD), and an anti-CD38 antibody or was triple-class (PI/IMiD/anti-CD38) refractory. Phase II treatment was once a week through week 14 and then once every 2 weeks. Phase II 200 mg patients who achieved a ≥very good partial response by week 24 received linvoseltamab once every 4 weeks. The primary end point in phase II was overall response rate (ORR)., Results: Among the 117 patients treated with 200 mg, the median age was 70 years, 39% had high-risk cytogenetics, and 28% had penta-refractory disease. At a median follow-up of 14.3 months, the ORR was 71%, with 50% achieving ≥complete response (CR). In 104 patients treated with 50 mg at a median follow-up of 7.4 months, the ORR was 48%, with 21% achieving ≥CR. The median duration of response (DOR) for 200 mg patients (n = 83) was 29.4 months (95% CI, 19.2 to not evaluable). Among 200 mg patients, the most common adverse events included cytokine release syndrome (35.0% Gr1, 10.3% Gr2, 0.9% Gr3), neutropenia (0.9% Gr2, 18.8% Gr3, 23.1% Gr4), and anemia (3.4% Gr1, 4.3% Gr2, 30.8% Gr3). Immune effector cell-associated neurotoxicity syndrome occurred in 7.7% of patients (2.6% each Gr1, Gr2, Gr3). Infections were reported in 74.4% of patients (33.3% Gr3, 2.6% Gr4); infection frequency and severity declined over time., Conclusion: Linvoseltamab 200 mg induced deep and durable responses, with a median DOR of 29.4 months, in patients with RRMM with an acceptable safety profile.

Abstract

Immunoglobulin E (IgE) is a key driver of type 1 hypersensitivity reactions and allergic disorders, which are globally increasing in number and severity. Although eliminating pathogenic IgE may be a powerful way to treat allergy, no therapeutic strategy reported to date can fully ablate IgE production. Interleukin-4 receptor α (IL-4Rα) signaling is required for IgE class switching, and IL-4Rα blockade gradually reduces, but does not eliminate, IgE. The persistence of IgE after IL-4Rα blockade may be due to long-lived IgE + plasma cells that maintain serological memory to allergens and thus may be susceptible to plasma cell-targeted therapeutics. We demonstrate that transient administration of a B cell maturation antigen x CD3 (BCMAxCD3) bispecific antibody markedly depletes IgE, as well as other immunoglobulins, by ablating long-lived plasma cells, although IgE and other immunoglobulins rapidly rebound after treatment. Concomitant IL-4Rα blockade specifically and durably prevents the reemergence of IgE by blocking IgE class switching while allowing the restoration of other immunoglobulins. Moreover, this combination treatment prevented anaphylaxis in mice. Together with additional cynomolgus monkey and human data, our studies demonstrate that allergic memory is primarily maintained by both non-IgE + memory B cells that require class switching and long-lived IgE + plasma cells. Our combination approach to durably eliminate pathogenic IgE has potential to benefit allergy in humans while preserving antibody-mediated immunity.
