Purpose: Our study intended to explore how low-dose anti-angiogenic drugs affected anti-tumor immunity of tumor-infiltrating exhausted CD8+T cells and achieved better clinical response when combined with immunotherapy. We set out to find potential targets or predictive biomarker on CD8+T cells for immunotherapy. Methods: We tested different doses of anti-VEGFR2 antibody combined with anti-PD1 antibody to treat LUAD in vivo and analyzed tumor-infiltrating CD8+T cells by flow cytometry. CD8+T cells overexpressing LAYN were co-cultured with LA795 cell lines to identify the function of LAYN in CD8+T cells. We also analyzed clinical samples from advanced LUAD patients treated with anti-angiogenesis therapy combined with immunotherapy. Results: Low-dose anti-VEGFR2 antibody combined with anti-PD1 antibody treatment delayed tumor growth and prolonged the survival time of tumor-bearing mice. The number of tumor-infiltrating CD8+T cells was reduced and the expression of LAYN was down-regulated in tumor-infiltrating CD8+T cells in the low-dose anti-VEGFR2 combination group. It was found that LAYN inhibited the killing function of CD8+T cells. In patients with advanced LUAD who received anti-angiogenesis therapy combined with immunotherapy, the LAYN+CD8+T cell subpopulation in good responders was significantly higher than that in poor responders. Furthermore, we demonstrated the expression of LAYN was regulated by upstream transcription factor NR4A1. Conclusion: Low-dose anti-VEGFR2 antibody combined with anti-PD1 antibody therapy promoted anti-tumor immunity and the downregulation of LAYN in tumor-infiltrating CD8+T cells played an important role in this process. These findings had implications for improving the efficacy of immune checkpoint blockade therapy and further optimized clinical treatment guidelines in advanced LUAD. [ABSTRACT FROM AUTHOR]

• Ring-core fiber with few modes has higher nonlinear power threshold than single-mode fiber, and lower dispersion effect between modes due to annular core structure than multimode fiber. In the sensing application, using the high-order core modes in RCF, can avoid environmental disturbances. In this work, we firstly proposed long-period grating (LPG) inscribed in RCF by CO 2 laser, which can convert fundamental mode to LP 11 mode with high conversion efficiency of 98 %. Moreover, the proposed RCF-LPG can realize vector curvature measurement with sensitivities of − 0.584, 0.879, and − 0.735 dB/m−1 in the bending directions of 0°, 90°, and 180°, respectively, in curvature range of 5–9.466 m−1, as well as vector torsion measurement with sensitivity of − 0.13 nm/(rad/m) and 0.554 dB/(rad/m) with the torsion rate changing from − 6.98 to 6.98 rad/m. The measured influence of temperature can be negligible. The proposed RCF-LPG could not only be used as high-efficiency mode converter in mode division multiplexing systems but also be potential in few-mode fiber based multiparameter sensing fields. With the rapid development of optical fiber sensor in physical measurement, it is of practical significant to realize multi-parameter measurement. In this work, we proposed and demonstrated a multiparameter sensor for curvature, torsion and temperature. The sensor is formed from CO 2 laser-inscribed long-period grating (LPG) in few-mode ring-core fiber (RCF), which can convert fundamental mode to LP 11 mode. One-side inscription enhances asymmetric index modulation in fiber, resulting in directionally sensitive to bend by intensity interrogation. The curvature sensitivities in various bending directions perform linearity in curvature range of 5–9.466 m−1. Curvature sensitivities are related to the bending directions. The intensity sensitivity achieves − 0.584 dB/m−1, 0.879 dB/m−1, and − 0.735 dB/m−1, respectively. And torsion sensitivity from - 6.98 rad/m to 6.98 rad/m displays linearity with −0.554 dB/(rad/m). The measured influence of temperature can be negligible. The joint design of the optical system and the downstream algorithm is a challenging and promising task. Due to the demand for balancing the global optima of imaging systems and the computational cost of physical simulation, existing methods cannot achieve efficient joint design of complex systems such as smartphones and drones. In this work, starting from the perspective of the optical design, we characterize the optics with separated aberrations. Additionally, to bridge the hardware and software without gradients, an image simulation system is presented to reproduce the genuine imaging procedure of lenses with large field-of-views. As for aberration correction, we propose a network to perceive and correct the spatially varying aberrations and validate its superiority over state-of-the-art methods. Comprehensive experiments reveal that the preference for correcting separated aberrations in joint design is as follows: longitudinal chromatic aberration, lateral chromatic aberration, spherical aberration, field curvature, and coma, with astigmatism coming last. Drawing from the preference, a 10% reduction in the total track length of the consumer-level mobile phone lens module is accomplished. Moreover, this procedure spares more space for manufacturing deviations, realizing extreme-quality enhancement of computational photography. Scrophularia ningpoensis Hemsl. (SN) is an herbal medicine used as a functional food in China. SN has traditionally been used to treat diabetes mellitus (DM), but its mechanism of action is not well understood. Our previous study indicated that the aqueous extract of SN can reduce hepatic insulin resistance and correct the irregular shape of pancreatic islets in diabetic mice. This study aims to investigate whether the aqueous extract of SN can regulate pyroptosis in pancreatic β-cells and explore its underlying mechanisms. Db/db mice were orally given an aqueous extract of SN for eight weeks. Subsequently, their pancreatic tissues were collected for histopathological analysis and immunoblotting. Rat insulinoma (INS-1) cells were cultured either alone or with the aqueous extract of SN and evaluated for cell viability, pyroptotic body formation, AMP-activated protein kinase (AMPK) activity, the NACHT, LRR, and PYD domain-containing protein 3 (NLRP3) inflammasome and gasdermin D (GSDMD) activation, and reactive oxygen species (ROS) production under high glucose (HG) exposure conditions. The aqueous extract of SN reduced insulitis and pancreatic β-cell death in db/db mice. Increased AMPK phosphorylation and decreased NLRP3 inflammasome and GSDMD activation were observed in pancreatic tissues after administration of the aqueous extract of SN. In vitro , the aqueous extract of SN increased cell viability and AMPK phosphorylation in a concentration-dependent manner. Additionally, the aqueous extract of SN corrected excessive ROS generation, NLRP3 inflammasome and GSDMD activation, and pyroptotic body formation. Inhibition of AMPK negated these beneficial effects of the aqueous extract of SN on INS-1 cells. This study aims to examine the trends and disparities in the combined prevalence of overweight and obesity among children in China by regions and specific groups over the last 30 years. PubMed, Wanfang, CNKI, VIP database, official websites of national governments and professional institutions/associations were systematically searched, for related reports and studies published up until December 2021. Reports based on nationally representative survey data were also examined and included. In mainland China, overweight and obesity decreased in pre‐school children (≤6 years old) and increased in school‐aged children (7–18 years old) between 2010 and 2013 to 2015 and 2017, but long‐term trends predict future increases in both groups. The prevalence of overweight and obesity in primary school students in Hong Kong dropped from 21.4% in 2007–2008 to 17.6% in 2016–2017. In Macao, the prevalence of overweight and obesity in children aged 6–18 years was reported by age stratification; the lowest and highest prevalence was 13.4%, 29.3% and 9.2%, 21.7% for boys and girls in latest survey in 2015, respectively. The prevalence of overweight and obesity in primary, middle and high school students in Taiwan dropped from 28.2%, 27.1% and 31.1% in 2010 to 26.7%, 25.2% and 26.7% in 2013, respectively. Furthermore, in mainland China, the prevalence in rural left‐behind boys and girls was 10.7% and 7.0%, respectively, in 2011, and the prevalence among migrant pre‐school children was about 5.0% around 2010. In summary, the prevalence of overweight and obesity in school‐aged children has increased in mainland China and Macao, but decreased in Hong Kong and Taiwan prior to 2017. The projections suggest that the highest prevalence of overweight and obesity in pre‐school children will be in rural children in the future. Several prior studies have highlighted the promise of mesenchymal stem cells (MSCs) as tools for treating myocardial infarction (MI) patients. While MSCs were initially thought to mediate post-MI repair through differentiation and replacement of injured cells, they are now thought to function by releasing exosomes carrying important cargos which can prevent apoptosis and facilitate revascularization in the context of MI. Herein, we comprehensively survey prior preclinical studies examining MSC-derived exosomes (MSC-Exos) utility for the repair of MI-related tissue injury. In total, 24 relevant studies were identified in the PubMed, Web of Science, Embase, and Cochrane Library databases as per the PRISMA guidelines. In most studies, exosome-treated rodents exhibited improved cardiac function and angiogenesis together with decreased apoptotic cell death. MSC-Exos thus offer beneficial therapeutic efficacy when treating MI injury. However, further work will be necessary to standardize experimental preclinical models and to validate these results. Objective: To verify the association between CD44 and CD133 expression levels and the prognosis of patients with lower grade gliomas (LGGs) and constructing radiomic models to predict those two genes' expression levels before surgery. Materials & methods: Genomic data of patients with LGG and the corresponding T2-weighted fluid-attenuated inversion recovery images were downloaded from The Cancer Genome Atlas and The Cancer Imaging Archive, which were utilized for prognosis analysis, radiomic feature extraction and model construction, respectively. Results & conclusion:CD44 and CD133 expression levels in LGG can significantly affect the prognosis of patients with LGG. Based on the T2-weighted fluid-attenuated inversion recovery images, the radiomic features can effectively predict the expression levels of CD44 and CD133 before surgery. [ABSTRACT FROM AUTHOR]
